1.The clinical analysis of anti-infection treatment after 492 cases of orthopedics operation

Objective To investigate the effect after operation infection in the department of orthopedics in our hospital,and to explore the use of antibiotics to prevent infection treatment best administration time,duration of drugs and the effect of several drug combination therapy.Methods 492 patients were randomly divided into A,B,C groups,A group included 163 cases in β-lactam and Aminoglycosides,B group was 162 cases in β-lactam and Macrolide,C group 167 cases were in two β-lactam.Comparison of three groups of anti-infection treatment.Results Preoperative administration of 0.5 ~ 2h efficiency was 97.4%,preoperative 3 ~ 24h efficiency was 94.5%,both difference was statistically significant(x2 =4.97,P <0.05) ; medication persistence time < 24h effective rate reached 92.3%,medication duration 24h to 3d efficiency 96.8%,use duration of 4 ~7d effective rate was 83.5%,the use of antibiotics of short duration group had significantly higher efficiency long service group,significant difference (x2 =6.19,P <0.05).A,B,C group combined medication against infection of total effectiveness respectively was 95.3%,94.5%,95.2%,which was not statistically significant (F =0.16,P > 0.05).Conclusion Operation before the use of antibiotic prophylaxis against infection effect is good,several antibiotics combined with medication regimens has no significant difference in efficiency.
3.Effects of the Traditional Chinese Drug Chaiyuwendantang(CYWDT) and Its Component on the Plasm Levels of ACTH,COR and cAMP in Hippocampus of Depressed Mode

[Objective] To explore the effects of the traditional Chinese drugs Chaiyuwendantang(CYWDT) and its component on the plasm levels of ACTH,COR and cAMP in hippocampus of depressed mode.[Methods] Eighty——four male rats were divided into seven groups: control group,depressed model group,group of CYWDT,group of HuaTan,group of TiaoQiXue,group of YangXinPi,group of maprotiline equally and made up models in chronic unpredictable mild stress(CUMS).The plasm levels of ACTH,COR and the level of cAMP in hippocampus were detected by radioimmunoassay.[Results] We found that the plasm levels of ACTH,COR were significantly increase and cAMP in hippocampus was significantly decreasd in depression model of rat.CYWDT and TiaoQiXue could significantly decreased the level of ACTH(P
4.Research progress on HOTAIR in tumor biological behaviors

Long non-coding RNA (lncRNA) is a cluster with over 200 bp in length and has no protein-encoding product RNA, which is in-volved in cellular physiological or pathological process, especially in human oncogenesis. HOX transcript antisense RNA (HOTAIR), which is 2158 bp in length, is one of the most well-studied lncRNAs. Overexpression of HOTAIR is correlated with oncogenesis or me-tastasis in numerous epithelium original human cancers, including breast and colorectal cancers. Inhibiting HOTAIR expression could suppress cell growth and invasive ability of tumors. This review provides a brief summary of the latest progress in lncRNA-based can-cer research.
5.Effects of hypoxia inducible factor-2α on promoting angiogenesis of residual hepatocellular carcinoma after high-intensity focused ultrasound ablation.

OBJECTIVETo investigate the dynamic features of angiogenesis in residual tumors after high intensity focused ultrasound (HIFU),and to determine the temporal effect and mechanism of hypoxia inducible factor-2 alpha (HIF-2a) in the angiogenic process of residual tumors.
METHODSXenograft tumors of HepG2 cells were generated by subcutaneously inoculating athymic BALB/c nu/nu mice with the hepatoma cells.About 30 days after inoculation,all mice (except in the control group) were treated by HIFU and assigned randomly to the following 7 groups according to various time intervals post-treatment:1st,3rd,5th day and 1st,2nd,3rd,4th week when the residual tumor tissues were obtained from the experimental groups.Protein levels of HIF-2a and vascular growth factor A (VEGF-A) were quantified by immunohistochemistry and western blotting,and mRNA levels were measured by (real-time quantitative) qPCR. Microvascular density (MVD) was calculated by counting the CD31-positive vascular endothelial cells identified by means of an immunohistochemical staining method.
RESULTSCompared with results from the control group,the protein and mRNA levels of HIF-2a expression reached the highest level in the experimental mice at the 2nd week (P=0.000 and P < 0.01 respectively),and were decreased thereafter(3rd week and 4th week, P=0.000 and P < 0.05).VEGF-A expression in the residual tumor tissues group that received HIFU was significantly decreased,compared with the control group,at all time points uPto 1 week (all P=0.000 and P < 0.01),but the levels increased compared to controls in the 2nd through 4th week (all P=0.000, P < 0.05). Similar results were obtained for MVD.
CONCLUSIONHIFU treatment can inhibit angiogenesis in residual hepatoma tissues in the short-term (1 to 2 weeks post-treatment) in mice with hepatocellular carcinoma,but can promote angiogenesis overtime (2 to 4 weeks post-treatment); the angiogenic process may involve the HIF-2α/VEGFA pathway.

6.Performance comparison among glucose detection reagents with different chromogens

Objective To preliminarily evaluate the performance of glucose detention reagents with three kinds of different chro-mogens and to investigate their anti-interference performance according to NCCLS document.Methods According to the protocol EP10-A2 provided by the National Committee for Clinical Laboratory Standards(NCCLS),the samples with low,middle and high level of glucose were detected by the glucose reagent kits with 3 kinds of different chromogens.The bias,total imprecision,inter-cepts,slope rates,nonlinearities,carryover rate and drifts were calculated.The interference evaluation test was performed according to the document EP7-A2.Results The bias and total imprecision of three kinds of reagent kits were all within allowed ranges.No statistically significant differences were showed in intercepts,slope rates,nonlinearities,carryover rate and drifts.1450 turbidity chyle,5 g/L hematoglobin and 0.03 g/L vitamin C did not interfere with the assay of three kinds of glucose reagent kits with differ-ent chromogens.342 μmol/L free bilirubin,342 μmol/L conjugated bilirubin did not interfere with the detection of reagent with MAOS.Conclusion The glucose detention reagents with three different chromogens have good accuracy and precision,and various performance indexes all conform to the clinical application requirements,reagent with chromogen MAOS is better than other chro-mogenic reagents in the anti-interference performance.
7. Studies on sesquiterpenes from Vladimiria souliei

Objective: To study the sesquiterpenes from the roots of Vladimiria souliei. Methods: The compounds were isolated by silica gel, Sephadex LH-20, medium pressure chromatographic column, preparative-TLC, and preparative-HPLC. And their structures were identified by means of chemical methods and spectroscopic analyses. Results: Nine compounds were isolated from V. souliei, and named as 15-acetoxy-11αH-germacra-1(10)E, 4E-diene-12, 6α-olide (1), 15-acetoxy-11βH-germacra-1(10)E, 4E-diene-12, 6α-olide (2), 12-hydroxy-5α(H), 7α(H)-eudesm-4(14), 11(13)-diene-9-one (3), oplodiol (4), dihydroestafiatone (5), 10-oxo-dihydroxy-11β, 13-dihydrode hydrocostuslactone (6), (11S)-13-hydroxyl-guaia-4(15), 10(14)-diene-12, 6α-olide (7), 9-oxonerolidol (8), and vernopolyanthone (9). Conclusion: Compounds 1 and 3 are new compounds, named as vlasoudide and vlasoudione; and compounds 4-9 are isolated from the roots of V. souliei for the first time.
8.Metallo-?-lactamase Produced by Carbapenem-resistant Pseudomonas aeruginosa

OBJECTIVE To study the metallo-?-lactamases of 5 carbapenem resistant Pseudomonas aeruginosa isolates,which were recovered at 2006 in the Third People's Hospital of Yueqing. METHODS K-B method was used to determine the antimicrobial agents susceptibility in 5 isolates. The minimal inhibitive concentrations (MICs) of antimicrobial agents to these strains were determined by agar dilution method. Double disk synergy test was used to detect the metallo-?-lactamase. Molecular screening for blaIMP,blaVIM,and blaSPM was carried out using PCR method. The PCR product was sequenced. RESULTS One out of the 5 carbapenem resistant P. aeruginosa was positive for MBL double disk synergy test,and confirmed to contain blaVIM-2 gene. CONCLUSIONS A blaVIM-2-producing isolate of P. aeruginosa is identified. This carbapenem-resistant isolate is all multi-drug resistant.
9.The genetic immunization in mice with three constructs of Toxoplasma gondii SAG1 gene

Three forms of the major surface antigen (SAG1)of Toxoplasma gondii, that is the membrane form, the secrete form and the intracellular form, were constructed and used to immunize BALB/c mice. The humoral response in the mice immunized with the membrane form and the secrete form of SAG1 appeared earlier and stronger than those mice immunized with the intracellular form. Result from the challenging infection demonstrated that the protection in the mice immunized with the membrane and the secrete forms was also stronger than in the mice immunized with the intracellular form. We suggest that the immune efficiency of the three forms of SAG1 in the mouse model is different.
10.Diagnosis and prognosis of 280 patients with pancreatic carcinoma

Objective To evaluate the factors influencing early diagnosis and prognosis in patients with pancreatic carcinoma.Methods The clinical data of 280 patients who had complete follow-up data with pancreatic carcinoma treated from January 2002 to January 2007 were reviewed retrospectively.The medical history and follow-up data were collected from all patients.Survival rate was calculated by the life table method and the Kaplan-Meier estimation.Log-rank test was used for univariate prognostic analysis and Cox regression was used for multivariate prognostic analysis.Results 91.8%of the patients were more than 40 years old and the peak age was 50~73 years old;the major presentations were abdominal pain and jaundice.Major imaging tests included B-ultrasound and CT,the sensitivity was 70.6%,95.3%,respectively;89.3%of patients had combined B-ultrasound and CT examination.The sensitivity of CA19-9 was 81.1%.The median survival time was(7.0±0.5)months.Overall survival rates at 1~5 year survival rates were 28%,9%,6%,2%,and 1%.Univariate analysis suggested that age>65 years old,CA19-9>mean value,TNM Ⅲ or Ⅳ stage,lymph nodes invasion,vascular invasion,and metastasis of two or more organs,non-surgical treatment,KPS score<60 points,weight loss≥5 kg were poor prognostic factors;Cox multivariate analysis showed that treatment modalities,age,TNM stage,KPS score and ascites were independent risk factors for dismal prognosis.Conclusions The age,ascites,tumor stage and treatment modalities affected the prognosis of patients with pancreatic cancer.Early diagnosis and treatment was important to improve the survival time of patients with pancreatic cancer.
