1.Antagonism Function of Anti-Sense Nucleic Acids on IL-6 Signal Transduction in Myeloma Cells

Objective: To investigate which step of IL-6 signal transduction pathways in myeloma cells can be taken as the acting target of the antagonists for IL-6. Methods: EMSA and immunoprecipitation were used to detect the activation of transcription factors(TFs)-STAT3, NF-IL-6 and protein kinase ERK in a myeloma cell line-Sko-007 by IL-6. then anti-sense expression plasmids and anti-sense ODN for these signal moleculars were constructed and designed,the effects of these anti-sense nucleic acids on IL-6 signal transduction in Sko-007 cells were analyzed by the same methods. Results: IL-6 signal transduction could be antagonized by these anti-sense nucleic acids at different extent.Conclusion: TFs or protein kinases in IL-6 signal trareduction pathways can be taken as the target for antagonists design.
2.Antagonistic effect between two IL-6 signal transduction pathways in a human myeloma cell line-U266

Objective To investigate the IL-6 signal transduction pathways and their regulation mechanism in a human myeloma cell line-U266. Methods Electrophoretic mobility shift assay (EMSA) was used to detect the activation of the transcription factors (TFs)-STAT3 and NF-IL-6 by IL-6. Cells were treated with chemical agents or transfected with the expression plasmids for the two TFs or the anti-sense oligonucleotide for protein kinase involved in one IL-6 signal transduction pathway. The change of the activation state of another IL-6 signal transduction pathway was also exhibited by EMSA. Results Two IL-6 signal transduction pathways (JAK/STAT and Ras/NF-IL-6) can be activated by IL-6 of different dose in U266 cells. When one of the two signal transduction pathways was up-regulated, the other one was down-regulated. Conclusion There is an antagonistic effect between the activation of two IL-6 signal transduction pathways in U266 cells.
3.Constituents of Volatile Oil from New Hybrid of Amomum villosum Lour. Spring No.1-F4

Objective To compare the contents of main compounds and the composition of volatile oil from the new hybrid of Amomum villosum Lour. Spring No.1-F4 with those in its female parent Longfruit No.2. Methods Steam distillation, gas chromatography (GC) and gas chromatography-mass spectrometry (GC-MS) methods were used for analyzing the content differences of main compounds and compositions of volatile oil between Spring No.1-F4 and Longfruit No.2. Results The contents of volatile oil and bornyl acetate from Spring No.1-F4 and Longfruit No.2 met the standard required by Chinese Pharmacopoeia. The content of volatile oil from Spring No.1-F4 was higher than that of Longfruit No.2, and there were some differences between the two in the composition of volatile oil and their relative contents. Conclusion The effective constituents in Spring No.1-F4, a new hybrid species of Amomum villosum, has reached the standard required by Chinese Pharmacopoeia, which is expected for solving the problems of difficulty in pollination and for improving the yield and quality of Amomum villosum.

Preimplantation genetic diagnosis is the integration of both assisted reproductive technologiesand molecular genetic technologies.Since the birth of the first healthy females after PGD in 1990,re-markable advances have been achieved in this field.Most research in PGD is focused on new methods toimprove the sensitivity and accuracy of single cell analysis.The principal problems in single cell PCR in-clude amplification failure,ADO and contamination.Fluorescent PCR with multiplex amplifications ofhighly polymorphic markers is a highly effective strategy to avoid contamination and detect ADO.The ad-vantages and disadvantages of fluorescence in situ hybridization to detect age-related aneuploidy are stillunder debate.We summarize the most recent developments in this review,and also introduce our own ex-periences in PGD.
6.Clipping of anterior communicating artery aneurysms via supraorbital keyhole approach from the side of non-dominant A1

ObjectiveTo investigate the surgical method, experience, and skills in clipping anterior communicating artery aneurysm (ACoAA) via supraorbital keyhole approach.MethodsThe ACoAA case data of selectively clipping via non-dominant supraorbital keyhole approach according to the A2 open plane formed by the anterior communicating artery and the bilateral A2 segments were analyzed retrospectively.The surgical method and experience were analyzed.The indications and advantages and disadvantages of this method were summarized.ResultsACoAA in 12 patients were completely clipped via supraorbital keyhole approach from the side of non-dominant A1, and the dominant A1 segment arteries were well exposed.The patients were followed up for 4-29 months after procedure.No recurrence or rupture of the aneurysms was found.The Glasgow Outcome Scale score was 5 in 11 patients and 4 in 1 patient.Conclusions ACoAA can be completely clipped via supraorbital keyhole approach from the side of non-dominant A1.It is a minimally invasive surgical approach with good efficacy.
7.Value of levator shortening for correction of high supratarsal fold after cosmetic upper blepharoplasty

Objective To correct the high supratarsal fold after cosmetic upper blepharoplasty is a challenge because of the shortage of both upper eyelid skin and fat.Levator shortening has been a procedure of choice selected by the surgeons to correct this condition.Methods Under local anesthesia, the desired crease was marked according to the characteristics of eyelid anatomy and aesthetic consideration for Asian women.The incision was made through the marked new skin crease and the old incision with surrounding scar tissue was incised.The adhesion between the skin and the levator aponeurosis was completely released.The levator aponeurosis was then divided above the tarsal plate.It could be shortened and then reattached to the tarsal plate depending on the amount of the upper eyelid skin and the degree of the ptosis.The skin incision was then closed to form new but lowered fold.Results Between 2003 to 2015, a total of 34 Chinese women underwent bilateral levator shortening for correction of high supratarsal fold after previous unsatisfactory upper blepharoplasty by other surgeons.There were no surgical complications postoperatively in this series and 32 patients (94.1%) were satisfactory for the outcome of the proper height of the supratarsal fold with at least 1 month follow-up.Only one patient (2.9%) required surgical revision for asymmetry of supratarsal folds and one patient (2.9%) for correction of deepened supratarsal fold.Conclusions The levator shortening can be an effective procedure to correct high supratarsal fold after unsatisfactory upper blepharoplasty in Asian women.It is especially useful to correct such a condition where there is shortage of the eyelid skin combined with the eyelid ptosis.
8.Induction of CD45 expression in a myeloma cell line by IL-6

Objective:To investigate the relationship between “activation of IL 6 signal transduction pathways” and “induction of CD45 expression” in a myeloma cell line—Sko 007 by IL 6.Methods:Electrophoretic mobility shift assay(EMSA) was used to detect the activation of the transcription facotrs STAT3 and NF IL 6 by IL 6,which can represent the activation state of the two IL 6 signal transduction pathways respectively;Then the induction of CD45 mRNA and protein expression by IL 6 were detected by RT PCR and FACS.The effect of antisense expression plasmid of STAT3 pAT STAT3 on STAT3 activation and CD45 expression was shown by EMSA and RT PCR,respectively.Results:①Both two IL 6 signal transduction pathways JAK/STAT and Ras/NF IL 6 can be activated by IL 6 in Sko 007 cells.②The expression level of CD45 mRNA and protein is Sko 007 cells can be up regulated by IL 6.③The activation of JAK/STAT signal transduction pathway and induction of CD45 mRNA expression by IL 6 can be inhibited in pAT STAT3 transfected Sko 007 cells.Conclusion:IL 6 induced CD45 expression in Sko 007 cells in mediated by the activation of JAK/STAT signal transduction pathway.
9.Inhibition of dexamethasone-induced apoptosis in human myeloma cells by IL-6

Objective:To investigate the mechanism of dexamethasone(DEX) induced apoptosis of human myeloma cell line Sko 007 and the inhibitory effect of IL 6 on apoptosis.Methods:Effect of DEX on the growth of Sko 007 cells was measured by MTT assay;Apoptosis of Sko 007 cells induced by DEX was determined by flow cytometry with propidium iodide(PI) staining of nuclei;The expression of three anti apoptotic Bcl 2 family proteins Bcl 2,Bcl X L and Mcl 1 in Sko 007 cells with or without DEX were determined by immunblot assay.Results:DEX inhibited the proliferation of Sko 007 cells and induced a significant cell apoptosis.In addition,degradation of Bcl 2 can be observed in Sko 007 cells in the presence of Dex.IL 6 stimulation prevented down regulation of Bcl 2 and the apoptosis of Sko 007 cells induced by DEX.Conclusion:IL 6 inhibits DEX induced apoptosis of Sko 007 cells through the specific regulation of Bcl 2.
10.Clinical study of renal blood flow and endothelin in cirrhotic patients.

OBJECTIVESTo find the relationship between hepatic dysfunction and renal blood flow (RBF).
METHODS49 patients with hepatic cirrhosis were studied via Color-Coded Duplex Ultrasonography detecting the pulsatile index (PI), resistive index (RI), peak systolic velocity (PS), peak diastolic velocity (PD) and peak systolic velocity/peak diastolic velocity (PS/PD) in the interlobar and arcuate arteries. The plasma endothelin (ET) levels were also evaluated.
RESULTSAccording to a modified Child's classification which assesses the severity of liver cirrhosis, we compared different groups and found that the worse the function of liver appeared coincidently with the higher PI and RI, especially RI (0.60+/-0.09, 0.66+/-0.06, 0.72+/-0.07, respectively, P < 0.01); The results also indicated that PI, RI increased obviously followed by the rise of ascites (PI: 1.14+/-0.20, 1.31+/-0.29, 1.42+/-0.36, respectively; RI: 0.61+/-0.09, 0.68+/-0.07, 0.77+/-0.05, respectively). The evaluation of plasma ET level demonstrated that its increment is intimately correlated with PI and RI.
CONCLUSIONIn patients with cirrhosis, RBF is directly correlated with the hepatic function. The correlation between ascites and RBF is also discovered. In patients who have liver cirrhosis, the plasma ET level obviously increased, indicating that ET probably is one of the active factors of renal vascular contraction.
