1.Expression of nuclear factor-kappaB in mouse uterus during peri-implantation.

To investigate the expression of the subunit p65 of NF-kappaB and inhibitor kappa B alpha (IkappaBalpha) in mouse uterus during peri-implantation, thereby investigating whether transient activation of nuclear factor-kappaB (NF-kappaB) takes place during embryo implantation in mice. Immunohistochemical technique was used to examine the expression and localization of p65 in endometrium or deciduas, and Western blot analysis was employed to detect the levels of IkappaBalpha protein in mouse endometrium or deciduas. P65 protein was detected in stromal cells, epithelial cells of endometrium as well as in myometrium. Staining was predominately seen in the cytoplasm of the cells. Staining intensity for p65 was stronger in the epithelial compartment than the stromal compartment and myometrium. Staining intensity increased slightly during pregnancy, and it reached a high level on pregnancy day 5 and day 8. In contrast to p65, the level of IkappaBalpha protein was lowest on pregnancy day 5 in all groups. Our results suggested that NF-kappaB may regulate embryo implantation by its transient activation in mice.

The activity of matrix metalloproteinases (MMPs) in the uterine flushing and endometrial tissue of normal adult women wearing FCu-IUD (fixed Cu-IUD) or FICu-IUD (indomethacin-releasing FCu-IUD) was observed by using zymography on SDS-PAGE containing gelatin. The results showed that the activity and kinds of MMPs in FCu-IUD group were increased significantly as compared with themselves before being inserted FCu-IUD. However, compared with the FCu-IUD group, the activity of some kinds of MMPs in the FICu-IUD group was decreased significantly. These data suggest that IUD can enhance the activity of MMPs in human endometrium, intermediated by prostaglandins, and MMPs may have relation to IUD-induced menorrhagia and indomethacin reduces IUD-induced menorrhagia by partly inhibiting MMPs synthesis.

5.A study of Modic changes'association with the volume and morphology of the endplate defects in the lumbar spine and their correlations with low back pain

Objectives:To analyze the association between the volume of Modic changes(MC)in the lumbar spinewith the volume and morphology of endplate defects(ED),and investigate the correlation between the associated endplate lesions and low back pain(LBP)symptoms.Methods:The clinical and imaging data of 401 patients with ED examined with lumbar MRI at the Second Affiliated Hospital of Soochow University between October 2020 and October 2023 were retrospectively analyzed.The patients were divided into MC group(204 patients)and non-MC group(197 patients)according to whether or not combined with MC,and the volume of ED was measured in the two groups of patients.The morphology of ED was categorized into typi-cal group(depressed,triangular,and round)and atypical group(rectangular and irregular).The length,width and height of MC were measured in patients of the MC group,and the volume fraction was calculated.The vol-ume fraction of MC was analyzed for correlation with the volume fraction of ED and morphological grouping.Then the intervertebral disc degeneration(IDD)grade of all the patients were evaluated and grouped,and the correlations between the volume fraction of MC and volume fraction and morphology grouping of ED for each IDD group of patients were analyzed separately.The low back pain(LBP)condition was followed-up,and the prevalence of LBP in patients with different ED morphology and with or without MC was compared.Results:In the typical morphology group,there were 369 vertebrae,and 152 vertebrae were combined with MC;In the atypical morphology group,there were 191 vertebrae,and 149 vertebrae were with combined MC.The mean volume fraction of ED in the group with MC was 0.017±0.014,and the mean volume fraction of ED in the group without MC was 0.008±0.007.The mean volume fraction of MC in patients with ED in the atypical group was 0.20±0.13,which was 0.11±0.10 in patients with ED in the typical group,with statistically signifi-cant difference(P<0.001).Overall MC volume fraction correlated linearly and positively with changes in ED volume fraction(P<0.001).The volume of MC correlated with the volume and morphology of ED in patients with mild and moderate IDD(P<0.05);In patients with severe IDD,there was a correlation between the vol-umes of MC and ED(P<0.001),but there was no correlation between the range of MC and the morphology of ED(P>0.05).The prevalence of LBP was higher in patients with ED combined with MC than in patients with ED alone(P<0.05).65 patients were followed up to record the LBP condition,and 30 patients had LBP(24 cases in the MC group and 6 cases in the non-MC group),There was no significant difference in the preva-lence of LBP between patients with different ED morphology(P>0.05).Conclusions:The volume fraction of lumbar MC shows a significant linear and positive correlation with ED volume fraction;In patients with mild and moderate disc degeneration,the patients with ED of atypical group of morphology tend to have a greater volume fraction than the patients in typical group of morphology;Patients combined with MC have a higher prevalence of LBP.
6.Investigation and analysis of the current situation of case reporting ethical review and patient informed consent reports in comprehensive journals of clinical medicine in China

Objective:To investigate and analyze case reporting ethical review and patient informed consent reports published in the comprehensive journals of clinical medicine in China in 2022.Methods:According to the data from the 2022 Edition of the Chinese Science and Technology Journal of the Citation Reports(Extended Version),the case reports published in comprehensive journals of clinical medicine in 2022 were selected as the research objects.The information on ethics and patient informed consent was extracted from the case reports that met the selection criteria,and Microsoft Excel 2021 and SPSS 21.0 were used to sort out and analyze the data.Results:A total of 587 case reporting articles were published in the 42 included journals in 2022,of which 36(6.13%)reported on science and technology ethics and/or informed consent.Case reports reporting on science and technology ethics and/or informed consent mostly came from the key magazine of China technology(88.89%Vs.65.88%),and the proportion of manuscripts involving science and technology ethics on the official website of the journal was relatively high(86.11%Vs.63.88%),and the difference was statistically significant(P<0.01).Conclusion:The proportion of case reports of science and technology ethics and/or informed consent in journals of comprehensive discipline classification of clinical medicine was relatively low.Currently,most international journals are required to obtain the informed consent of patients or legal guardians before publishing case reports.Compared with this,there are still certain gaps in China,which need to be paid great attention to.
