1.Metabolic Effects of Nonsteroidal Anti-Inflammatory Drugs on Chondrocytes in Human Osteoarthritis

Objective To observe the effects of nonsteroidal anti-inflammatory drugs (NSAIDs) on the proliferation of chondrocytes and metabolism of human osteoarthritis (OA) in short-term (48 hours). Methods The human knee OA chondrocytes were cultured in the presence or absence of NSAIDs (celecoxib, diclofenac and ibuprofen). BrdU assays were used to evaluate the effects of NSAIDs on the proliferation of OA chondrocytes. ELISA was used to detect the contents of pro-teoglycan and type-Ⅱcollagen in chondrocytes and culture supernatant. The differences of those indexes were compared be-tween groups. Results The positive rate of BrdU labelling index of chondrocytes increased significantly in ibuprofen group than that of other groups (P<0.05). The content of type-Ⅱcollagen in the culture supernatant increased significantly in di-clofenac group than that of other groups (P<0.05 or P<0.01). The content of proteoglycan in the culture supernatant in-creased significantly in ibuprofen group than that of other groups (P<0.05 or P<0.01). There were no significant differences in the content of type-Ⅱcollagen and proteoglycan between chondrocytes of NSAIDs groups (P>0.05). Conclusion Ibu-profen may stimulate the proliferation and the secretion of proteoglycan of human OA chondrocytes. Diclofenac may stimu-late the secretion of type-Ⅱcollagen of human OA chondrocytes. There were no effects of celecoxib on the proliferation and metabolism of human OA chondrocytes.
2.Chemical Dynamic Change of Volatile Oil and Its Sensibilization Activity Constituent CSY of Root of Changium Smyrnioides Wolff During Processing

Objective: To clarify chemical dynamic change of volatile oil of Root of Changium Smyrnioides Wolff during Processing, to analyse and study the change of constituent proportion to see if new constituents produce or original constituents disappear, to discuss the processing mechanism of root of Changium smyrnioides . Method: The constituents and contain of volatile oils of root of Changium smyrnioides before, during and after processing are comparatively analyzed by using techniques of combined capillary gas chromatography mass spectrometry computer. Result: 34 chemical compounds are identified, among them there were 29 compounds isolated from volatile oil of fresh root of Changium smyrnioides, with a proportion of 97.873% in total oil, its main constituent is CSY and its content reaches 70.946%; 34 compounds were isolated from volatile oil of fresh decorticated root of Changium smyrnioides, with a propoction of 93.452%, main constituent is CSY, its content reaches 75.909%; and 19 compounds from volatile oil of fresh root cortices of Changium smyrnioides, 96.878% , main constituent is CSY, content 70.977%; 11 compounds from volatile oil of processed root of Changium smyrnioides, 88.839%, main constituent is CSY, content 67.234%. Conclusion: Main reasons of non sensibilization of processed root of Changium smyrnioides are considered to be the content decrease and qualitative change of the volatile oils, and the content of snsibilization activity constituent——CSY decreased from 0.0497% to 0.0134%.
3.Analysis of clinical distribution and resistance of methicillin-resistant Staphylococcus aureus in primary hospital

Objective To analyze the distribution and drug resistance rate of methicillin-resisitant Staphylococcus aureus(MRSA) and methicillin-susceptible Staphylococcus aureus(MSSA) isolated from different departments in a certain primary hospital during 2009 to 2012,and to provide scientific evidences for clinical application of antibiotics.Methods Pathogens and bacterial resistance to antibiotics were identified using the VITEK 2 compact equipment.The data were got from WHONTES.5 and analyzed by SPSS 13.0.Results There were 517 Staphylococcus aureus strains were detected(MRSA 135 strains,MSSA 382 strains).The rate of MRSA was 24.5%,27.7%,24.8%,27.0% during the four years.MRSA was mainly found in the department of oncology,orthopaedicsand ICU.MRSA was mainly isolated from pus,secretion,sputum and blood.The 517 Staphylococcus aureus strains showed high sensitive to vancomycin,linezolid and teicoplanin,the sensitive rate was 100%.Conclusion Establishing a more comprehensive MDRO monitoring and hospital infection control system in the primary hospital,and rational using antibacterial drugs at the based of the antibiotics susceptibility test in the treatment can be effective in preventing MRSA resistance rates increasing and hospital-borne.
4.Effects of resveratrol on astrocyte activation after cerebral ischemia/reperfusion injury in rats

Aim To investigate the effect of resveratrol on astrocyte activation after cerebral ischemia/reperfusion injury in rats.Methods 45 male Sprague-Dawley rats were divided into sham,control and resveratrol groups, 15 rats in each group.Resveratrol or solvent was intra-peritoneally perfused into the rats for 7 consecutive days after 3h for MCA I /R.Cerebral infarction volume with TTC,neurological function score with Longa score, and pathological change with HE staining,the protein expressions of Caspase-3 with immunohistochemistry at 24h after MCA I /R were examined.The protein ex-pressions of GFAP and NeuN in penumbra were detec-ted with immunohistochemistry and immunofluores-cence at 14d after MCA I /R.Results Resveratrol treatment significantly improved the neurological func-tion score,decreased the infarct volume,reduced neu-ron loss,and inhibited the activation and proliferation of astrocytes,compared with the control group.Con-clusion Resveratrol can inhibit the excessive activa-tion of astrocytes,protect neurons in penumbra and im-prove neurological function.
