Objective: To investigate the inhibitory effects of Curcumin on A? generation in vitro and explore its potential mechanisms involing BACE1 and PPAR?.Methods: Plasmids APPswe and BACE1-mychis were transiently co-transfected into SHSY5Y cells by LipofectaminTM2000,then treated with Curcumin at 0(control Group),1.25,5.0,20.0 ?mol/L for 24 hr,or with Curcumin at 5 ?mol/L for 0,12,24 and 48 hrs for the time course assay.RT-PCR was performed to measure the endogenous levels of BACE1 mRNA and PPAR?.Western blot were used to detect the protein expression of BACE1 and PPAR?.The major cleavage product——A?40 and A?42 was detected by ?-amyloid 1-40 or 42 Colorimetric ELISA assay.Results: The mRNA level and protein level of BACE1 were decreased obviously in a dose-and time-dependent manner after treated with Curcumin in the cell line(P

Alzheimer's disease (AD) is a common progressive degenerative disease of the central nervous system. It's also a main cause for dementia in presenium and senectitude. In the past few years, the discoveries in vitro expansion of neural stem cells (NSCs) indicate a new way for recovery and replacement of damaged neurons as well as reconstruction of the neural circuit, and offer a useful future therapy for AD. There are two main promising approaches in NSCs replacement therapy. They are endogenetic approach, inducing proliferation and differentiation of endogenetic NSCs, improving self-repair of central nervous system, and exogenous approach, ransplantation of exogenous tissue and promoting proliferation of endogenous NSCs. However, it may not become truth before the mechanism of NSCs' proliferation, migration, differentiation and it's integration with the host tissues to be elucidated.

6. Phase II study of adoptive immunotherapy for advanced melanoma

Objective: To observe the efficacy and safety of chemotherapy combined with cytokine-induced killer (CIK) cells in the treatment of metastatic melanoma (MM). Methods: Fifty three chemotherapy-naive MM patients were administered fotemustine at 100 mg/m2 on d 1-5; dacarbazine were given at 400 mg/d on d 2-6; CIK were infused on d 7, d 14, d 16. Twenty-eight days were regarded as one cycle. The clinical efficay was evaluated every 2 cycles. Results: Thirty-four out of 53 patients were eligible to be evaluated. The overall response rate (ORR) was 23.5% including 1 case with complete response (CR) (2.9%) and 7 cases with partial response (PR) (20.6%). Fourteen cases had stable disease (44%). The clinical benefit response was 67.5%. Median progression free survival (PFS) was 8 months. Median overall survival (OS) was 11 months. The patients with normal lactate dehydrogenase (LDH) had longer OS; those with stable disease had longer PFS and OS. Adverse reaction included grade III/IV thrombopenia (41%), decrease in the number of WBC (23.5%), and hyperreactivity (2 cases). No treatment-related death occurred. Conclusion: Chemotherapy followed by the infusion of CIK was tolerable. The response rate was higher than common chemotherapy. It could prolong the survival time of tumor patients with elevated LDH and non progression disease. Phase III study is needed to confirm the results.

OBJECTIVE: To discuss the challenges and opportunities for traditional pharmacies in digital network time. METHODS: Aiming at the problems occurring in the work of pharmacy and pharmaceutical care, we analyzed the meaning and deep impact of the using of network information technology in pharmacy .RESULTS: It is clear that the drug supply ori- ented pharmacies should be transformed to pharmaceutical care oriented pharmacies. It is very important to use computer high text to handle pharmaceutical information efficiently.CONCLUSION: The digital networks can extend the work of pharma- ceutical care in space and time and provide an opportunity to the development of pharmacies.
8.Contribution of Lu Zhi-jun to the undertakings of acupuncture and moxibustion.

Comrade LU Zhi-jun, a famous surgeon and acupuncture expert, was a leading cadre in health field with outstanding achievements, and a leader of western medicine doctors learning traditional Chinese medicine in the Chinese Communist Party, and also he was a main establisher of The China Academy of Chinese Medical Sciences and a founder of TCM scientific researches and integrated Chinese and western medicine in new China. The present paper reviews the course of LU Zhi-jun in learning and applying acupuncture and moxibustion therapy and a part of works carried out in faithfully implementing our party's policies for uniting Chinese and Western medicine, and in development of TCM including acupuncture and moxibustion in past tens years.

9.Analysis of the therapeutic effects of cataract extraction and intraocular lens implantation in silicon oil stored eyes with proliferative diabetic retinopathy

Objective To compare the postoperative effect of pars plana vitrectomy,phacoemulsification and intraoeular lens (IOL)implantation with silicon oil removed,phacoemulsification and IOL implantation.Design A retrospective case-control study.Par- ticipants 91 cases(104 eyes)of proliferative diabetic retinopathy with tractive retinal detachment or vitreous hemorrhage were per- formed cataract surgery with IOL implantation.Methods All patients were followed up average 6 months,in whom 52 eyes with pars plana vitrectomy,phacoemulsification and IOL implantation(combined group),and the other 52 eyes with silicon oil removed,pha- coemulsification and IOL implantation(sequential group).The differences of the postoperative visual acuity and inflammatory reactions of the anterior chamber between the two groups were analyzed.Main outcome measures Visual acuity and inflammatory reactions of the anterior chamber.Results There were 14/52 eyes with best corrected visual acuity more than 0.1 in combined group and 8/52 eyes in the sequential group after the surgeries(X~2=-6.87,P=0.07) .Corneal edema happened in 16/52 eyes in combined group and 6/52 eyes in sequential group 1 week after the surgeries(X~2=11.53,P=0.001).Keratic precipitate happened in 20/52 eyes in combined group and 9/52 eyes in sequential group after the surgeries(X~2=5.79,P=0.019) .Conclusions The incidence of postoperative anterior chamber in- flammation is higher in Phacoemulsification combined with pars plana vitrectomy group.

P53 abnormality or mutation is commonly seen in patients with hepatocellular carcinoma (HCC), and therefore, restoration of P53 function has become a research hotspot in the treatment of HCC.This article reviews the association of P53 with Bcl-2 protein family, microRNA, TGFβ, HBV, HCV, and AKT and the role of P53 in regulating cell apoptosis, in order to provide clues for improving the therapeutic outcome of HCC.
