Microdialysis probes were implanted into the rabbit cerebral cortex and caudate nucleus followed by perfusion with Ringer solution at a flow rate of 3.0?l/min. Cerebral ischemia was induced by electrical stimulation of the unilateral superior cervical ganglion for 1h. Dialysate samples were analyzed by high performance liquid chromatography (HPLC). Multiple myeloma is a malignancy of plasma cells,which is still incurable now.Despite dramatic improvement of the response rate and overall survival have been made due to the application of target therapy and stem cell transplantation,the treatment effect is difficult to maintain,nearly all patients will eventually relapse and become refractory.At relapse,the challenge is to select the appropriate treatment for each patient.The decision will be both disease-dependent and patient-dependent.
4.The clinical research of the effect of live combined bacillus subtilis and enterococcus faecium granules with multivitamines and nonnutritive suck on the premature infant

Objective To study the effect of live combined bacillus subtilis and enterococcus faecium granules with multivitamines (Medilac-Vita) and nonnutritive suck on the premature infant.Methods Eighty premature infants without asphyxia and digestive tract malformation were randomly divided into control group(without Medilac-Vita and nonnutritive suck group) and observation group( Medilac-Vita and nonnutritive suck group) including 40 cases each.The gastrointestinal dysfunction and growth-development index were observed in two groups.Results In observation group,the occurrence rate of vomit,abdominal distention,gastric retention,hemorrhage of digestive tract,meconium peritonitis and necrotizing enterocolitis were all lower than that in control group.The cleaning lime of meeoninm in observation group was lower than that in control group [(3.20±1.01 )d and (6.23±2.56)d],the removing time of canalis ventriculi [(4.03±2.11 )d and (7.10±2.91 )d],the time of the jaundice fading away [( 11.10±5.36)d and ( 14.30±6.82)d ],the time of staying in hospital [(15.02±3.12 )d and (20.00 + 5.11)d],the growth of the head circumference and weight in seven and fourteen days were significantly different between two groups.The incidence rates of intracranial hemorrhage,apnea and death in observation group were 12.5% (5/40),10.0% (4/40) and 0 (0/40),which in control group were 45.0%(18/40),37.5% (15/40) and 5.0% (2/40).Conclusion The treatment of Medilac-Vita and nonnutritive suck on the premature infants can promote the maturity of stomach and intestine function,reduce the occurrence of complications,promote growth and development.
5.The relationship between serum osteopontin and disease activity in rheumatoid arthritis and the role in interstitial pulmonary fibrosis

ObjectiveTo investigate the association between serum level of osteopotin(OPN) and disease activity of rheumatoid arthritis (RA) patients, and explore the importance of OPN in the pathogenesis of interstitial lung disease (ILD) in RA. MethodsSixty-five RA patients and 20 healthy controls were pros-pectively enrolled. RA patients were divided into active group(n=43) and inactive group(n=22), and ILD groups (n=24) and non-ILD group (n=41). Enzyme-linked immunosorbent assay (ELISA) was used to detect the serum levels of OPN in patients with RA and healthy controls, and the relationship between OPN and other clinical and laboratory findings were analyzed.Results① Serum OPN tended to be significantly higher in RA patients (median, 18.0 ng/ml) than in the healthy controls (median, 14.3 ng/ml), P<0.01; ②The serum level of OPN in RA patients showed a significant positive correlation with the course of disease, numbers of tender joints , ESR and CRP, but no positive relationship was found in number of swollen joints; ③ The serum level of OPN was significantly higher in RA-ILD patients(median, 20.0 ng/ml) than that in non-lLD (median, 17.0 ng/ml, P<0.05). And there was remarkable negative correlation between the concentration of serum OPN and the value of PaO2, but no association was found with pulmonary function %VC and %DLCO. ④ Compared with the non-ILD group, the ILD group had more active disease in terms of tender joint counts and swollen joint counts, ESR, CRP(P<0.01) and the serum titer of RF-IgM,(P<0.05). ConclusionOPN plays a role in the pathogenesis of RA and is related to the disease activity. It may serve as an active disease inflammatory marker of RA . OPN may be involved in the pathogenesis of RA related ILD and is associated with the severity of pulmonary damage.

Objective To study the effects of DMF on the quality of spermatozoa and sex hormone levels in male mice,and the possible mechanism. Methods Forty male KM mice were randomly divided into four groups and then treated with DMF dissolved in distilled water by gavage (10 ml/kg) at the doses of 0,0.5,1 and 2 g/kg,once a day for 30 consecutive days. The mice were weighted and then sacrificed on day 31. The epididymises were collected for determination of sperm motility,sperm counting and determination of abnormal sperm rate. 7.Isolation,culture and characterization of mesenchymal stem cells derived from Wharton's jelly of human umbilical cord

Objective To investigate the bionomics of mesenchymal stem cells (MSCs) derived from Wharton's jelly of human umbilical cord. Methods Umbilical arteries, vein and umbilical cord tunica externa were removed, and the remaining tissue (Wharton's jelly) was harvested. To gather MSCs, the umbilical cord was cut into small fragments and digested with 0.075% type Ⅰ collagenase, or the small fragments were cultured with DMEM. The cells derived from Wharton's jelly of umbilical cord were serially subcultivated, the growth curve was drawn, and the identification of living cells was made for studying the cell growth kinetics. The surface antigens were detected with flow cytometry, the cartilage markers were detected by histochemistry and immunohistochemistry, and the expressions of Sox-9 and Col-2A1 mRNA were analyzed by RT-PCR. Results The confluence time of primary culture cells was 3-5d in cells harvested with enzyme digestion, and 10-15d in cells harvested with micro mass. The results of flow cytometry showed that the MSCs derived from umbilical cord expressed CD44 and CD105; the expression of HLA-ABC was positive, while HLA-DPDQDR was negative. There was no significant change on the immunophenotype of umbilical cord MSCs after cryopreservation and thawing. The findings of histochemistry and immunohistochemistry showed that the expression of chondrocyte markers in Wharton's jelly MSCs was weakly positive. The results of RT-PCR showed the chondrocyte markers Sox-9 and Col-2A1 were positively expressed in Wharton's jelly MSCs. Conclusion The MSCs derived from Wharton jelly of human umbilical cord do not differentiate into hematopoietic cells, may express the chondrocyte markers (they are suggested to have characteristics of pre-chondrocytes), and is expected to be a new type of stem cells of tissue engineering cartilage.
8.Application of asthma journal and corresponding monitoring of pulmonary ventilation function in treating bronchial asthma

Objective To explore the value of asthma journal and corresponding monitoring of pulmonary ventilation function in treating bronchial asthma with the existing classification of asthma severity as control.Methods After 137 patients with bronchial asthma were evaluated according to the existing grades,they kept asthma diaries and received corresponding monitoring of pulmonary ventilation function at least 5 times per day,and all of the above clinical data were analyzed by relevant statistical methods.Results Among the 126 patients who completed monitoring,only 61(48.41%) patients could be graded by the Guide for Prevention and Treatment of Bronchial Asthma;27(21.43%) patients felt sensitive while 33(26.19%) ones felt insensitive.Conclusion The corresponding monitoring can not only accurately evaluate the severity of patients' bronchial asthma condition,but also detect those patients with deviated symptom perception.
9.REPEATED OPERATIVE TREATMENT FOR LUMBAR PROLAPSED DISC WITH RECURRENCE OR FAILURE AFTER INITIAL OPERATION

18 out of 779 operated cases for lumbar prolapsed disc at our department were reoperated with excellent or good results obtained in 17 and improvement in 1. 14 out of these were initially treated at our department with satisfactory results for from 2 months to 12 years before symptoms recurred while 4 were first operated upon at other hospitals with good results in 2 and failure for the others after the primary operation. The recurrence rate is 14/779 in our service, i. e. 1.8%, lower than those previously reported.Reoperation for recurrent cases is generally considered extremely difficult and result uncertain because of extensive scar and dense adhesions about the nerve coot and dura. More often than not the results reported in literature were not good enough to warrant reoperation. Consequently those recurrent patients in our series so badly incapacitated would have to suffer or remain crippled for lifetime were they not reoperated. However our patients were salvaged by reoperation with meticulous performance after careful clinical evaluation as to diagnosis, indication and tyfe of operation adopted. Operative procedure for exploration and resolution of coexisting spinal stenosis is discussed in detail.
10.THE PROTECTIVE EFFECTS OF DRUGS ON APOPTOSIC NEURAL CELLS AFTER SPINAL CORD INJURY

To study the protective effects and the mechanism of drugs on apoptosic neural cells after spinal cord injury and the mechanism of their effects on severe degree compression injury to the lower thoracic spinal cord (T 8.9). Rats were divided into four groups randomly . The first group of rats were treated with saline solution(20?l) intrathecally after injury, the second group with cyceoheximide (CH)(30?g/20?l) intrathecally, the third group with monosialicganglioside(GM)(100?g/20?l),and the fourth group methylprednisolone(MP)(30mg/kg). The changes in weight, spinal cord blood flow(SCBF), terminal deoxynucleotidyl transferase-mediated dUTP nick end labeling (TUNEL),neurological function and quantitative histological changes were observed.The results indicated that drugs had little effects on the SCBF. But drugs could improve the neurological function and area of spared normal tissue, and obviously reduce the number of TUNEL-positive cells. The results suggested that drugs had neuroprotective effects on the injuried spinal cord. The mechanism of the beneficial effects might be a reduction in neuronal cells apoptosis.
