1.Research for early growth response gene-1 in ischemia-reperfusion injury
International Journal of Surgery 2012;39(4):267-270
Early growth response gene-1 is an immediate early response gene,a transcription factor which includ three zinc finger-like domains.Recentl studies demonstrate that early growth response gene-1 is involved in regulating cell cycle, proliferation and differentiation, and plays a critical role in injuring and repairing.Early growth response gene- 1 induces Target genes, and promotes extracellular environmental signal( such as oxygen deficit, oxidative stress, mechanical injury and cytokine)link with the complex biological responses.Intensive study to early growth respnse gene-1 will contribute to further illustrate the mechanism of the ischemia-reperfusion injury. This article summarizes from protein structure, signal transduction, biological functions and the relationship with ischemia- reperfusion injury.
2.A correlation between benign prostate hyperplasia and atherosclerosis
Chinese Journal of Geriatrics 2011;30(11):919-920
ObjectiveTo investigate the correlation between benign prostate hyperplasia (BPH) and atherosclerosis (AS).Methods 95 cases were divided into two groups based on prostate volume (PV):24 cases with non-BPH (PV≤20 ml) and 71 cases with BPH (PV>20 ml).Carotid intinal medial thickness (IMT) was determined by high resolution color Doppler sonography.ResultsLevels of triglyeeride (TG),hypersensitive C reactive protein (hs-CRP),ln fasting insulin [ln (FINS)],ln insulin resistance index [ln (HO)MA-IR)] and IMT in BPH group were significantly higher than in non-BPHgroup.The morbidity rate of coronary heart disease and cerebral arteriosclerosis in the BPH group enhanced as compared with non-BPH group.PV were positively correlated with ln(FINS),ln(HOMA-IR),TG and high density lipoprotein (HDL-C).Conclusions BPH is closely correlated with AS and their co-pathogenesis might be insulin resistance, inflammatory reaction and endothelial dysfunction.
3.The relation of peripheral leukocyte counts with urinary albumin in patients with type 2 diabetes
Chinese Journal of Diabetes 2008;16(1):12-14
Objective To investigate the relation of peripheral leukocyte total and its differential counts with urinary albumin(UAlb)in patients with type 2 diabetes. Methods The 564 subjects with type 2 diabetes were stratified according to UAER levels. The total and differential leukocyte counts of peripheral blood were measured.Demographic and potential metabolic confounding factors were analyzed with multiple correlation and logistic regressions to evaluate the effects of leukocyte counts on UAER.Results With the increase of UAER,the peripheral total leukocyte, neutrophil, monocyte, eosinophil and basophil counts increased. In contrast, the lymphocyte count decreased. The differences persisted even after adjusting the influence of age, sex, duration of diabetes and other known risk factors. Conclusions Our study shows that peripheral leukocyte counts, even within the normal range, are related with UAlb in subjects with type 2 diabetes in a dose-dependent manner.
4.Analysis of the different levels of serum high sensitivity C-reactive protein and its related factors in subjects with isolated IGT or IFG
Chinese Journal of Diabetes 2008;16(2):76-78
Objective To compare the levels of serum high sensitivity C-reactive protein(hsC-RP) among subjects with normal fasting glucose plus normal glucose tolerance(NFG/NGT),IFG,IGT and IFG plus IGT,and to analyze the related risk factors. Methods 443 nondiabetic subjects aged 39-65 years were seperated into four groups:NFG plus NGT,IFG,IGT and IFG plus IGT.The serum hsC-RP levels and related factors were analyzed. Results Compared with IFG subjects,IGT subjects had higher hsC-RP levels. No differences in hsC-RP levels were found between NFG plus NGT group and IFG group, as well as between IGT group and IFG plus IGT group.Stepwise multiple regression showed that BMI and HOMA-IR were independent predictors of hsC-RP. Conclusions IGT subjects have significantly higher serum hsC-RP level than IFG subjetcts. The serum level of hsC-RP is associated with insulin resistance and the components numbers of metabolic syndrome.

9.LncRNA-MEG3 mediated apoptosis of retinoblastoma by regulating P53 pathway
Recent Advances in Ophthalmology 2017;37(4):301-304
Objective To investigate whether MEG3 involved in the development of retinoblastoma by down-regulating the expression of P53 protein. Methods The MEG3 expression of retinoblastoma tissues and corresponding non-tumor tissues were detected by quantitative real-time PCR (qRT-PCR).Retinoblastoma cell lines SO-RB50 or HXO-RB44 were transfected with pcDNA-MEG3 or siRNA-MEG3,after which cell apoptosis was tested by flow cytometry and P53 protein expression was tested by Western blot. Results MEG3 expression of retinoblastoma tissues was significantly reduced compared with corresponding non-tumor tissues(P =0.014).MEG3 level was significantly increased in pcDNA-MEG3 transfected SO-RB50 cells (P =0.002) and significantly decreased in siRNA-MEG3 transfected HXO-RB44 cells (P =0.004).Flow cytometry showed that the SO-RB50 cells apoptosis was significantly increased with the MEG3 over-expression(P < 0.05),as well as the HXO-RB44 cells apoptosis was significantly decreased with the MEG3 knockdown(P < 0.05),compared with the control group, respectively.Furthermore,Western blot showed that P53 protein level was significantly increased after SO-RB50 transfected with pcDNA-MEG3 (P < 0.05),while significantly decreased after HXO-RB44 transfected with siRNA-MEG3 (P < 0.05),compared with the control group, respectively. Conclusion MEG3 is down-regulated in retinoblastoma, affect the development of retinoblastoma, and may induce the retinoblastoma cell apoptosis by promoting the expression of P53 protein.
10.The correlation between human leukocyte antigen-DRB1 alleles and anti-neutrophil cytoplasmatic antibodies in Han and Uygur ulcerative colitis patients in Xinjiang region
Chinese Journal of Digestion 2012;32(11):750-753
Objective To study the correlation between human leukocyte antigen (HLA)-DRB1 alleles and anti neutrophil cytoplasmatic antibodies (ANCA) in Han and Uygur ulcerative colitis (UC)patients in Xinjiang region. Methods The serum ANCA was determined by indirect immunofluorescence assay in 62 Uygur UC patients,58 Han UC patients,188 Uygur and 184 Han healthy control individuals.HLA-DRB1 typing was performed by polymerase chain reaction-sequence based typing (PCR-SBT).The allele frequency of HLA-DRB1 was compared in ANCA positive and negative Han and Uygur patients as well as healthy controls.Stratified analysis was performed according to UC clinical type, severity and involvement.SPSS 17.0 software was applied for x2 test.Once P<0.05,the odds ratio (OR) and 95% confidence intervals (95%CI) was calculated. Results The positive rate of ANCA in Uygur UC patients (53.2%,33/62) was significantly higher than that of Han patients (34.5%,20/58) and the difference was statistically significant (x2=4.269,P =0.045).In Uygur, the gene frequency of HLA-DRB1 * 13 in ANCA positive UC patients (0.202)was significantly higher than that of ANCA negative patients (0.017) (x2 =10.092,P=0.016,OR=16.000,95%CI:2.892 to 88.524) and healthy controls (0.075) (x2=9.351,P=0.040,OR=3.407,95%CI:1.666 to 6.971).The gene frequency of HLA-DRB1 * 13 in ANCA positive pancolitis type UC patients (9/15) was significantly higher than that of ANCA negative pancolitis type UC patients (1/14) and the difference was statistically significant (x2=8.955,P =0.040,OR =19.500,95%CI:2.787 to 136.461).However,in Han patients, there were no significant differences of HLA-DRB1 alleles frequencies among ANCA positive patients,ANCA negative patients and healthy controls (all P>0.05),and the results of stratified analysis were same. Conclusions In Uygur UC patients of Xinjiang region,HLA-DRB1 * 13 may correlated with ANCA and with ANCA of pancolitis type UC patients. There is no such correlation in Han patients of Xinjiang region.
