2.Update on targeting EGFR in advanced non-small-cell lung cancer

The results of chemotherapy in the treatment of advanced non-small-cell lung cancer (NSCLC)had reached a plateau.Targeted therapy is the key to improving efficacy.In the last 5 years the epidermal growth factor receptor(EGFR)has emerged as one of the most important targets for drug development in oncology.Small molecules that inhibit the tyrosine kinase(TK)domain of EGFR have become new critical weapons in the treatment of NSCLC.Monoclonal antibodies targeting the external domain of EGFR have been shown to have clinical benefit when combined with chemotherapy.Recent data suggest that multitargeted inhibitors might have greater activity owing to their ability to simultaneously block key signaling pathways.Preliminary results of such agents in clinical trials shows that they have a well-tolerated toxicity profile,and might have a promising value in the treatment of advanced non-small-cell lung cancer.
3.Cryopreservation of in vitro shoot-tips of Platycodon grandiflorum by vitrification and plant regeneration

Objective To get a new approach to conserve the germplasm of Platycodon grandiflorum. Methods A method of vitrification was studied to cryopreserve in vitro shoot-tips of P. grandiflorum, and the regenerated plantlets were observed subsequently. Results Shoot-tips, 2—3 mm length, gotten from in vitro shoots of P. grandiflorum precultured on MS medium supplemented with 5% DMSO and 103 g/L sucrose for 3 d, were loaded with the 60% PVS_2 for 20 min at 20 ℃, and incubated in PVS_2 for 90 min at 0 ℃ prior to a direct plunging into liquid nitrogen (LN) and keeping for 1 d. After rapid thawing in water at 40 ℃, the shoot-tips were rinsed in the MS medium supplemented with 410 g/L sucrose for 20 min, and plated on the filter paper sustained by the MS regeneration medium supplemented with 0.6 mg/L KT, 0.2 mg/L BA, and 0.05 mg/L NAA for 1 d in dark and subcultured on the above regeneration medium for one week in dark prior to exposure to the light. The survival of shoot-tips was up to 80%, and they grew normally. Conclusion The method of vitrification to cryopreserve the germplasm of P. grandiflorum is simple in handling with high in survival and normal in regeneration and can be applied in practice.

8.AN EXPERIMENTAL STUDY OF THE SURVIVAL MECHANISM OF AN AXIAL FLAP WITH EXTRA-TERRITORIAL BLOOD SUPPLY

Because of massive defects requiring repair, a flap with blood supply of extraterritorial blood vessels is often needed clinically, which is a little bigger than that with blood supply of axial blood vessels. In order to provide theoretical evidence for the survival of such a flap, we designed a 11cm ? 15 cm flap involving the thoracodorsalic and the lateral thoracic arteries on a rat's lateral thorax and abdomen. The flap was stained by methylene blue and filled with emulsion. The diameters of the anastomotic branches within the flap were measured at different times. The volumes of blood flow on different points of the flap were observed quantitatively by means of ECT scanning. The vitality of the flap was also observed.Our experiment proved that anastomotic branches between blood vessels are the anatomic bases for extra-territorial flap. Blood flows from one blood vessel supplying zone to another through the anastomotic branches between the blood vessels. After the flap formed, The diameters of its anastomotic branches would grow larger.Since 1988, we have designed 33 extra-territorial flaps on cervicothorac area. The flaps have survived well. Our present experiment provided the flap with theoretical evidence that an extraterritorial flap can be devised provided that there is abundant anastomosis between the two axial blood vessels.
9.Effects of intermittent conventional and continuous pressure-controlled tissue expansion on cell proliferation and apoptosis

Objective To investigate the cell dynamic changes of conventional intermittent tissue expansion (CITE) and continuous pressure controlled tissue expansion (CPTE). Methods Domestic pigs were chosen for CITE and CPTE models. Immunohistochemical technique was used to detect proliferative cells, DNA fragments in situ labeling for apoptotic cells, and H E stain for total fibroblast counting. Results Proliferative index of basal cell increased during expansion. The peak value of 78.5% reached at the ninth day in the CITE group versus 84.2 % at fourth day in the CPTE group. Proliferation index in both groups decreased after expansion. Fibroblast proliferation, apoptosis and total fibroblast density increased slowly in both groups during and after expansion. At the sixth day, total fibroblast densities in CITE group (38.1 cells/H) and in CPTE group (40.9 cells/H) were significantly increased, compared with 34.93 cells/H in the normal skin. There were obvious proliferation and apoptosis phenomena in epidermal cells and skin adnexa. Conclusion Tissue expansion has both effects of proliferation and apoptosis on cell dynamics. Continuous tissue expansion can induce tissues growing much effectively.
10.The influence of aspirin on apoptosis and Fas/FasL expression of Caco-2 cell

Purpose: To study the mechanism of chemoprevention of colorectal cancer by aspirin. Methods: Different concentrations of aspirin have been used in the culture of colonic cell-Caco-2, its proliferation and apoptosis was observed during the culture and the expression of Fas/FasL mRNA was assayed by RT-PCR. Results: Major portion of cells died in the culture medium that was added high concentration of aspirin, but the lower concentration promoted apoptosis. The Fas mRNA was expressed in cells added with 25 mmol/L aspirin, but not in that of 1640. The FasL mRNA expressed in 25mmol/L aspirin was lower than in 1640. Conclusions: The lower concentration of aspirin can promote apoptosis of coloniccell, the mechanism has correlation with induction of the expression of Fas, and down-regulation of the expression of FasL.
