1.On Thought of Globus Hystericus Treated by Basing on Differentiation of Signs

[Objective]To discuss the treatment based on syndrome differentiation of globus hystericus.[Methods]We combine consulting historical literatures with clinical experience and completely summarize the syndrome differentiation of globus hystericus.[Results]More pathogenesis can lead to globus hysteri-cus from clinical practice, such as insufficiency of heart yang ,insufficient kidney yang and exogenous pathogenic factors. In the principle of syndrome dif-ferentiation, we must base on the different pathogenesis to treat globus hystericus. [Conclusion]Because of its complexity, we can not rapidly draw a con-clusion without a careful analysis when facing globus hystericus and its like, which leads us to neglect the potential dangerous factors.
2.Efficacy and safety of 3-dimensional conformal radiotherapy combined with temozolomide for glioma.

OBJECTIVE:
To study the efficacy and safety of 3-dimensional conformal radiotherapy combined with temozolomide (TMZ) for gliomas.
METHODS:
A total of 78 patients with pathologically confirmed glioma ( from September 2005 to March 2007) were postoperatively divided into 3 groups: a chemotherapy group (n=24), a radiotherapy group (n=25), and a comprehensive therapy group(n=29). The patients received temozolomide alone,3-dimensional conformal radiotherapy alone,3-dimensional conformal radiotherapy combined with temozolomide in the chemotherapy group,the radiotherapy group and the comprehensive therapy group respectively. The survival rate, progression-free survival, overall survival time and adverse reactions were observed.
RESULTS:
The 3-year survival rate in the comprehensive therapy group was significantly higher than that in the other two groups. The 3-year survival rates were 20.83%, 20.00%, and 41.38% in the chemotherapy group, the radiotherapy group and the comprehensive therapy group respectively. The progression-free survival time was 17.68,17.94, and 23.29 months and the average overall survival time was 20.28, 21.54, and 25.75 months in the chemotherapy group, the radiotherapy group and the comprehensive therapy group, respectively.The adverse reactions were mild and tolerable.
CONCLUSION
Three-dimensional conformal radiotherapy combined with temozolomide is more effective for gliomas than the simple 3-dimensional conformal radiotherapy and the temozolomide chemotherapy alone.

3.Effect of Chinese herbal compound "Jisuikang" on engulfment of neuron debris by microglia

Objective To investigate the effect of Chinese herbal compound"Jisuikang"on the phagocyto-sis of neuronal debris by microglial cells. Methods To prepare serum containing drugs of JSK and divide them into the low,middle and high dose groups,the blank serum group and LPS+blank serum group. BV2 was labeled by lentiviral vectors containing the green fluorescent protein gene (GFP). To establish the damage neuron model and mix injured neurons with the transfected microglia. To observe the situation of microglia which was affected by serum containing drugs devour the neuronal debris. Results The middle and high dose of JSK showed greater phagocytic percentage and phagocytic index than those of the control group(P<0.001). In comparison of LPS+blank serum group,no significant difference was found in the middle and high dose of JSK. However,to the phagocytic index, which was better than that of LPS+blank serum group(P<0.05). Conclusion JSK may enhance the engulfment of neuron debris by BV2,which could provide a better living environment for the growth of neurons.
4.Effect of CDK2-AP1 gene over-expression on proliferation and cell cycle regulation of breast cancer cell line MCF-7.

OBJECTIVE:
To over-express cyclin-dependent kinase 2-associated protein 1 (CDK2-AP1) gene, and investigate its effect on the proliferation and cell cycle regulation in breast cancer cell line MCF-7.
METHODS:
CDK2-AP1 gene coding region was cloned into lentivirus vector. Lentivirus particles were infected into MCF-7 cells to upregulate the expression of CDK2-AP1 gene. The expression level of CDK2-AP1 was detected at both mRNA and protein levels by real-time PCR and Western blot. MTT assay, colony formatting assay, and flow cytometry were performed to detect the change of proliferation and cell cycle in MCF-7 cells. We examined the expression of cell cycle associated genes (CDK2, CDK4, P16Ink4A, and P21Cip1/Waf1) followed by CDK2-AP1 over-expression by Western blot.
RESULTS:
CDK2-AP1 gene was up-regulated significantly at both mRNA (6.94 folds) and protein level. MTT based growth curve, colony formatting assay and flow cytometry showed that CDK2-AP1 over-expression lentivirus inhibited the proliferation of MCF-7 cells with statistical difference (P<0.05). In addition, with CDK2-AP1 over-expression, MCF-7 cells were arrested in G1 phase accompanied by apoptosis. Western blot showed that the expression level of P21Cip1/Waf1 and P16 Ink4A was upregulated, while the expression level of CDK2 and CDK4, members of the CDK family, was downregulated.
CONCLUSION
CDK2-AP1 gene plays a cancer suppressor role in breast cancer. Its function includes inhibiting the proliferation of MCF-7 cells and arresting the cell cycle in G1 phase.

5.Clinical significance of tumor markers in the diagnosis of lung cancer.

BACKGROUNDTo determine the clinical value of serum TPA, Cyfra 21.1 and NSE in the diagnosis of lung cancer.
METHODSThe serum samples of 136 patients were measured: including 15 adenocarcinomas, 39 squamous cell carcinomas, 8 small-cell lung cancers (SCLC), 34 unclassified lung cancers, and 40 benign pulmonary diseases. Serum TPA and Cyfra 21.1 were detected by ELISA and the normal value was ≤0.9μg/l and ≤3.6μg/l respectively. Serum NSE was detected by radioimmunological method and the normal value was ≤20μg/l. All data were dealed with t and Chi-square test.
RESULTSThe level of TPA in each lung cancer group was significantly higher than the normal value except for that in SCLC group. The level of Cyfra 21.1 in adenocarcinoma, squamous cell carcinoma, and unclassified lung cancer groups was significantly higher than the normal value; The level of NSE in SCLC group was significantly higher than the normal value. In the benign pulmonary disease group the level of TPA was significantly higher than the normal value. TPA was sensitive for the diagnosis of lung cancer and the sensitivity was from 69.2% to 87.5%, but the specificity was low (29.3%); the specificity of Cyfra 21.1 was 97.6%, but the sensitivity was low (12.5%-35.9%); the sensitivity (75.0%) and specificity (82.9%) of NSE were high for SCLC.
CONCLUSIONSTPA shouldn't be a qualified tumor marker for lung cancer because of low specificity; the specificity of Cyfra 21.1 is higher, but the sensitivity is lower; NSE may be a satisfactory tumor marker for SCLC.
6.Recent advance in neuroprotection effect of salvia miltiorrhiza on central nervous system and related toxicology

Recently, salvia miltiorrhiza has made a great progress in research of central nervous system (CNS) injury and neurodegeneration. Salvia miltiorrhiza and its active ingredients can exert multiple effects involving reduction of cell loss, attenuation of oxidative stress, improvement of micro-circulation and promotion of neuroregeneration, and show a protective effect on CNS diseases. Regulation of oxidative stress and removing accumulated metabolite may be the important mechanisms by which salvia miltiorrhiza exerts neuroprotective effects. This study will systematically discuss the pharmacological effects and possible mechanisms of salvia miltiorrhiza based on the core pathological changes in CSN diseases, and evaluate its drug safety through combing the related toxicology researches to provide a reference for clinical transformation of Chinese medicine in threatment of CNS diseases.
7.Preliminary Study on the Improvement Effects of Astragaloside Ⅳ on Allergic Rhinitis Model Mice

OBJECTIVE:To preliminarily s tudy the potential mechanism of astragaloside Ⅳ on allergic rhinitis (AR)model mice. METHODS :C57/BL6 mice were randomly divided into blank group ,model group and astragaloside Ⅳ group,with 10 mice in each group. Except for blank group ,AR model was prepared by sensitization and challenge with ovalbumin on day 0,7,14 and 21-27. Astragaloside Ⅳ group was given astragaloside Ⅳ 40 mg/kg intraperitoneally at the dose of 0.02 mL/g on the 15th to 27th day of modeling (given the drug 1 h before challenge sensitization on the 21st to 27th day ). Blank group and model group were given constant volume of normal saline intraperitoneally ,once a day. Twenty-four hours after sensitization from the last challenge , the infiltration of inflammatory cells in the nasal mucosa of each group was observed ,and the contents of interleukin 4(IL-4), IL-5 and interferon gamma (IFN-γ)in the nasal lavage fluid were measured. The levels of reactive oxygen species (ROS),and the count of phosphorylated Janus kinase 2(p-JAK2)and phosphorylation signal transduction and activation of transcription protein 6 (p-STAT6)positive cells in the nasal mucosa and spleen as well as the phosphorylation levels of JAK 2 and STAT 6 proteins in spleen tissue (i.e. p-JAK 2/JAK2 ratio,p-STAT6/STAT6 ratio)were also determined. RESULTS :Compared with blank group ,the number of inflammatory cells in the nasal mucosa (eosinophils and mast cells )in the model group ,the contents of IL- 4 and IL- 5 in the nasal lavage fluid ,and the levels of ROS in the nasal mucosa and spleen tissues in the model group ,the count of p-JAK 2 and p-STAT 6 positive cells increased significantly ,the p-JAK2/JAK2 ratio,p-STAT6/STAT6 ratio in the spleen tissue were significantly increased (P<0.05),and the content of INF-γ in the nasal lavage fluid was significantly decreased(P<0.05). Compared with model group ,the count of inflammatory cells infiltrated in the nasal mucosa ,the contents of IL- 4 and IL- 5 in the nasal cavity lavage fluid ,the level of ROS and the number of p-JAK 2 and p-STAT 6 positive cellsin the nasal mucosa and spleen tissue as well as the p-JAK2/JAK2 ratio and p-STAT 6/STAT6 ratio in spleen tissue were decreased significantly (P<0.05),and the content of INF-γ in nasal lavage fluid was significantly increased(P<0.05). CO NCLUSIONS:Astragaloside Ⅳ can effectively improve the inflammatory response in AR model mice ,the mechanism of which may be related to down-regulation of JAK2/STAT6 signaling pathway and ROS level.
8. Experimental study on the effect of Yiqi Wenyang Decoction on nasal mucosa infiltration of NK cells in mice with allergic rhinitis

Objective:
To observe the effect of Yiqi Wenyang Decoction on the infiltration and activation of NK cells in nasal mucosa of mouse model with allergic rhinitis (AR), and to explore the potential mechanism for effective intervention of AR with Yiqi Wenyang Decoction.
Methods:
Fourty-eight mice were randomly divided into blank group, model group, low, medium and high dose of Yiqi Wenyang Decoction group and Cetirizine group, with 8 rats in each group. After modeling of AR, the model group was filled with 0.9% sodium chloride solution. Yiqi Wenyang Decoction groups of each dose were given different concentrations of Yiqi Wenyang Decoction water extract, while the Cetirizine group was given aqueous solution of Cetirizine. The behavior, morphological changes of nasal mucosa and infiltration of NK cells in nasal mucosa were observed. The levels of IL-4 and INF-γ in nasal lavage fluid were measured. Besides, the drug safety was observed by acute toxicity test.
