1.Real-time Detection of Ascorbic Acid in the Renal Cortex of Renal Artery and Vein blockage Model

Objective To establish a model of renal artery and vein blockage and detect the real time change of ascorbic acid in the renal cortex in vivo.Methods Sixteen rabbits were randomly divided into experiment and control groups(8 in each).Under anesthesia,the left kidney and artery-vein were dissociated and a microdialysis probe was inserted into the renal cortex.After being balanced for 60 minutes,the change of ascorbic acid in the renal cortex was detected.Ischemia was achieved by clamping the renal pedicle for 60 minutes followed by reperfusion for another 60 minutes in the experiment group.While in the control,the same procedure was performed during a same time interval without clamping the renal pedicle.Microdialysis samples were collected before,during,and after the ischemia in the rabbits,and then ascorbic acid in the microdialysates were real-time measured by an electrochemical method.Results The concentrations of ascorbic acid in the microdialysates were(18.9?7.5)?mol/L,(24.8?11.3)?mol/L,and(23.3?8.9)?mol/L respectively before,during,and after the renal warm ischemia.In the experiment group,the concentration of ascorbic acid was increased rapidly by 31.2% (24.8-18.9)/18.9] following ischemia(P0.05).Conclusions Ascorbic acid is a sensitive indicator of renal ischemia/reperfusion.The level of ischemia in renal context can be monitored in real time by detecting the concentration of ascorbic acid in vivo using microdialysis-electrochemical method.The method can be used for the further studies on ischemia/reperfusion.
2.Glucose Metabolism in the Warm Ischemia/Reperfusion Injured Kidney of Rabbits:In Vivo Microdialysis and Real-Time Electrochemical Detection

Objective To investigate the change of glucose metabolism in warm ischemia/reperfusion injured kidney of rabbits in vivo by microdialysis and real-time electrochemical detection.Methods A total of 20 New Zealand rabbits were divided into experiment and control groups(10 in each).After anesthesia by intravenous injection of barbitone,an oblique incision under the 12th rid was made in the animals to expose the right kidney and the right renal artery and vein.Afterwards,a microdialysis probe was placed into the dorsal renal cortex along the long axle of the kidney.After 60 minutes to equilibrate,an electrochemical detection system was used to continuously determine the concentration of glucose in the renal cortex before ischemia.Then,ischemia/reperfusion injury model was established by clamping the renal pedicle for 60 minutes followed by perfusion for 60 minutes.The control group received the same intervention without clamping the renal pedicle.The glucose concentration of the microdialysis samples were analyzed before,during,and after the ischemia.Results The reaction of the glucose electrode to the electric current was linearly correlated with the concentrations of glucose,and the rate of microdialysis probe recycling was(63.6?2.1)%.The concentration of glucose in the renal cortical interstitial samples were(1.89?0.37),(0.69?0.12),and(0.62?0.14)mmol/L respectively before,during,and after the warm ischemia.During the ischemia phase,the mean concentration of glucose decreased by(36.7?2.4)% from the base level(LSD test,P=0.000).The glucose concentration of the experimental group was significantly lower than that in the control at both ischemia and reperfusion phases(t=-11.975,P=0.000;t=-11.993,P=0.000,respectively).Conclusions In vivo microdialysis combined with electrochemical detection provides a sensitive and real-time method for measuring the glucose concentration in warm ischemia/reperfusion injured kidney,which can suggest the ischemic condition of the renal cortex.
3.Effects of body serum iron levels on female mice sexual maturity

Objective To study the influence of different body serum iron levels on follicle stimulating hormone (FSH),luteinizing hormone (LH) and uterine ovarian index,and explore the body serum iron level effects on female mice sexual maturity further.Methods 36 three weeks C57BL/6J female mice were randomly divided into the normal control group,strengthen-iron 1 group and strengthen-iron 2 group,12 cases in each group.The normal control group with normal diet(310mgFe/kg),strengthen-iron 1 group and strengthen-iron 2 group were given high -iron diets(respectively with 465mgFe/kg and 620mgFe/kg) after born 3 weeks.each group body,uterus and ovary mass after 2 weeks were checked;colorimetric method was used to detect the serum iron level;the serum follicle stimulating hormone (FSH) and luteinizing hormone(LH) were detected by chemiluminescent immunoassay.Results The body of mice serum iron levels,uterine ovarian index (uterine ovarian wet mass/body mass) and follicle stimulating hormone(FSH) level of strengthen-iron 1 group and strengthen-iron 2 group were higher than those of normal control group (F =9.11,11.11,7.09,all P < 0.01).Luteinizing hormone (LH) level of the strengthen-iron group was higher than those of normal control group (F =3.89,P < 0.05).Conclusion Uterine ovarian index,follicle stimulating hormone level and luteinizing hormone level were affected with increased serum iron level;It suggests that the increasing serum iron level can cause female mice sexual maturity in advance.
4.Influence of geniposide on collagen II synthesis of cultured chondrocytes in vitro

BACKGROUND:Chinese herb Cape Jasmine Fruit can activate the chondrocytes and geniposide is an important component in this herb.
OBJECTIVE:To investigate the effect of geniposide on col agen II synthesis in rat chondrocytes cultured in vitro. METHODS:Rat chondrocytes were separated and cultured in vitro. The chondrocytes were then interfered with 25, 50 and 100 mmol/L geniposide. Normal control group was also set. Col agen II mRNA and protein expression was detected with semi-quantitative RT-PCR and western blot analysis, respectively.
RESULTS AND CONCLUSION:RT-PCR and western blot analysis results showed that, geniposide at 25, 50 and 100 mmol/L increased the col agen II mRNA and protein expression (P<0.01). Geniposide can promote the synthesis of col agen II in rat chondrocytes cultured in vitro.
5.Effect of acupuncture on children with attention deficit hyperactivity disorder

ObjectiveTo observe the effect of acupuncture on children with attention deficit hyperactivity disorder (ADHD). MethodsFrom August to December, 2022, 54 children with ADHD from the outpatient clinic of the Fourth People's Affiliated Hospital of Jiangsu University were randomly divided into control group (n = 27) and observation group (n = 27). All the patients accepted electroencephalogram (EEG) neurofeedback training, while the observation group accepted acupuncture in addition, for twelve weeks. They were assessed with Integrated Visual and Auditory Continuous Performance Test (IVA-CPT), Conners Parent Symptom Questionnaire (PSQ), and EEG before and after treatment. ResultsThe comprehensive control quotient, comprehensive attention quotient and hyperactivity quotient of IVA-CPT increased in the observation group after treatment (t > 3.889, P < 0.001), and they were more in the observation group than in the control group (t > 2.040, P < 0.05); while all the factors of PSQ reduced in the observation group (t > 6.630, P < 0.001), and they were less in the observation group than in the control group (t > 3.871, P < 0.001); the mean frequency of β and sensorimotor rhythms increased in the observation group (t > 12.432, P < 0.001), and they were more in the observation group than in the control group (t > 7.561, P < 0.001); the mean frequency of θ reduced in the observation group (t = 9.966, P < 0.001), and it was less in the observation group than in the control group (t = 7.257, P < 0.001). ConclusionCombination of acupuncture is more effective on EEG in children with ADHD to improve attention, control and core symptoms.

Talents are the main force for the development of traditional Chinese medicine(TCM), and the construction of TCM talents and the reformation of talent evaluation system are essential to promote the inheritance and innovation of TCM. At present, we are still exploring and developing in the fields of the formulation, implementation and evaluation indicators of TCM talent evaluation system. However, there are shortcomings and difficulties. For instance, insufficient stratification in the evaluation, excessive emphasis on the quantity of achievements, neglecting the quality of the achievements and the actual contribution, imperfect assessment indicators, and the weak characteristics of TCM. Therefore, national ministries and commissions have jointly issued a document requesting to break the four only and set a new standard, in order to promote the construction of a scientific and technological talent evaluation system oriented by innovation value, ability and contribution. For the evaluation of TCM clinical talents, China Association for Science and Technology commissioned China Association of Chinese Medicine to build the China Clinical Cases Library of TCM(CCCL-TCM), which aims at collecting the most authoritative and representative TCM clinical cases and exploring the advantages of applying clinical cases as masterpiece of achievement in TCM clinical talents evaluation. CCCL-TCM can promote the construction of a talent evaluation system that is more in line with the development characteristics of TCM industry, and to carry out relevant pilot in TCM colleges and institutions across the country in order to promote the reformation of TCM talent evaluation system.
