1.EXPRESSION OF HYPOXIA INDUCIBLE FACTOR-1? AND p53 IN LUNG OF MICE DURING HYPOXIA

Objective To explore the expression of hypoxia inducible factor-1? and p53 in lung during hypoxia. Methods The mice were divided into two group (n=36,male,hypoxia group=36,control group=9).The hypoxia group mice were placed in a hypoxia chamber with 10%,7%,or 5% O 2 respectively.Immunofluorescent histochemical staining technique was used.The staining results were observed under the laser confocal scanning microscope. Results HIF-1? proteins didn't express and p53 proteins expressed weakly in control group mice; Compared with the controls,the expression of HIF-1? and p53 increased dramadically.There was significant difference in expression of HIF-1? and p53 between the hypoxia groups and the control group; Positive relationship was found between the expression of HIF-1? and p53. Conclusion The expression of HIF-1? and p53 increased in lung during hypoxia,which indicates that HIF-1? may play an important role in the expression of p53 and the regulation of apoptosis.;
2.A MORPHOLOGICAL STUDY ON TWO TYPES OF ALVEOLAR MACROPHAGES FROM NORMAL RAT LUNG

Alveolar macrophages (AM) were studied by means of phase contrast microscope, differential interference contrast microscope, SEM and TEM. The content of AcP, LDH, SDH of the AM was measured by MPV3-TSA microspectrophotometerimage analytical instrument. Under phase contrast microscope, two different types of AM can be distinguished, i. e, the light cells and the dark cells. The two types of cells are spherical and flat in shape under differential interference contrast microscope. The observation with SEM and TEM showed that the spherical cells possess more filopodia and more lysosomes in cytoplasm, but the flat cells possess more lamellar podia. Quantitative cytochemistry demonstrated that the content of AcP, LDH, SDH in spherical cells are much more than that of flat cells. The results suggest that there are two types of alveolar macrophages exist in normal rat lung.
3.A STUDY OF INCUBATION AND INITIAL IDENTIFICATION OF MURINE YOLK SAC MESENCHYMAL STEM CELLS

Objective To investigate the isolation,incubation and identification of murine yolk sac mesenchymal stem cells(YS-MSCs). Methods Murine yolk sacs were harvested on the 10th day of postcoitus microisolation.Yolk sac cells were obtained after the yolk sacs were digested by type Ⅰ collagenase of 0.1% for 1 hour.Adherent cells were cultured in DMEM containing 10% FBS and 1% SP,and passaged when they became subconfluent.The ultrastructure of YS-MSCs was observed under transmission electron microscope;the surface markers of YS-MSCs were detected by flow cytometry;the alkaline phosphatase(AKP) expression of YS-MSCs was tested;and cytochemical changes of YS-MSCs were observed with cytochemical detection.Adipogenic differentiation of YS-MSCs was induced by 1?mol/L dexamethasone and 10mg/L insulin,and oil red O was used for fat staining. Results YS-MSCs were obtained in vitro,and most of them were of spindle shape;under transmission electron microscope,there were microvilli on the surface of YS-MSCs,there were abundant mitochondria,rough endoplasmic reticulum and golgi complex in the endochylema.The cell nuclei of YS-MSCs were big and the shape of cell nucleus was irregular;the analysis of flow cytometry suggested that the primary and first generation of YS-MSCs were positive for CD44,CD105,and the expression of CD34 was small;Cytochemistry showed that YS-MSCs were positive for glycogen,and negative for SB and AKP;Adipogenic differentiation of YS-MSCs was induced by 1?mol/L dexamethasone and 10mg/L insulin;accumulation of lipidrich vacuoles appeared in the cells,positive for oil red O staining.Conclusion The biological characteristics of murine yolk sac mesenchymal stem cells are similar to,and more primitive than,those of adult mesenchymal stem cells.It suggests that YS-MSCs may be used as an ideal resource of seed cells of tissue engineer.
4.CYTOCHEMICAL CHANGES OF HEPATOCYTES IN MICE EXPOSED TO HYPOXIA

Objective To study the cytochemical changes of hepatocytes in mice exposed to hypoxia. Methods The mice were divided into two groups(n=45, all males, hypoxia group=36, control group=9). The mice of the hypoxia group lived in hypoxia (O 2:10%). Using imaging analytical instrument LEICA-500IW, We observed cytochemical changes of the hepatocytes. The PAS, LDH, cytochrome oxidase, Mg 2+ dependent ATPase activity of hepatocytes was assayed. Results Compared with the controls, the LDH activity of hepatocytes increased dramatically, the longer the hypoxic time, the higher the LDH activity of the hepatocytes; the cytochrome oxidase activity of hepatocytes decreased sharply, the longer the hypoxic time, the lower the cytochrome oxidase activity of the hepotocytes.Conclusion Hypoxia can injury the hepatocytes of the mice and result in sharp changes of the hepatic cytochemistry.;
