2.Effect of Mild Hypothermia on NSE and SOD in Serum of Patients with Large-area Cerebral Infarction

ObjectiveTo investigate the effect of mild hypothermia on neuron specific enolase (NSE) and superoxide dismutase (SOD) in serum of patients with large-area cerebral infarction.Methods160 cases with large-area cerebral infarction were divided into the treatment group and control group with 80 cases in each group. Patients of the control group were treated with routine therapy. Those of the treatment group were added with mild hypothermia therapy (MHT). The scores of NIHSS were assessed and NSE and SOD in serum were examined before treatment and 7, 14 and 30 days after treatment in two groups.ResultsThe NIHSS score of the treatment group was significantly lower than that of the control group ( P<0.05), and NSE level decreased, SOD vitality increased in the treatment group. Other indexes such as respiratory, pulse, serum kalium and etc of two groups were not different ( P>0.05).ConclusionMHT can improve he nerve function of patients with large-area cerebral infarction recovering and improve prognosis.
3.Rapid Genomic DNA Fragments Extraction From Serum Denatured With High Temperature

Objective To develop an approach for a rapid extraction of genomic DNA fragments from serum.Methods Serum specimen from human was processed using high temperature denaturation and was centrifuged, then using various primer sets to amplify the DNA fragment extracted, so as to verify the efficiency of this DNA extraction method.Results The templates preparing from this DNA extraction method were able to guarantee the amplification of fragments shorter than 400 base pairs.Conclusion This method is simple, rapid and inexpensive.These genomic DNA fragment extracted can be utilized to amplify definite DNA template for polymerase chain reaction.
4.Clinical observation of auricular point sticking combined with western medicine for preventing and treating postoperative complications of external excision and internal ligation on mixed hemorrhoid.

OBJECTIVETo explore the effect of auricular point sticking before operation for postoperative complications of external excision and internal ligation on mixed hemorrhoid.
METHODSEighty patients with mixed hemorrhoid were randomly divided into an observation group and a control group, 40 cases in each group. In the control group, paracetamol and dihydrocodeine tartrate tablets were applied with oral administration 1 h after operation, 510 mg each time, twice a day; at the same time, diosmin tablets were treated with oral administration, 0.9 g each time, twice a day. Three days' treatments were required successively. In the observation group, auricular point sticking was used before operation based on the treatment in the control group. The auricular points of Shen (Co10), Pangguang (CO9), Shenmen (TF4), Pizhixia (AT4), Jiaogan (AH6a) and Gangmen (HX5) were selected. The patients were asked to press the points 3-6 times per day, 3-5 min each time, 3 days' treatment in total. The scores of the postoperative complications in the 1st and the 2nd days were compared between the two groups such as pain, edema, hematochezia, retention of urine, etc.
RESULTSAfter operation, the scores of pain, edema, hematochezia, retention of urine in the 2nd day were all decreased obviously than those in the 1st day in the two groups (all P<0.05); and the scores of pain, edema, hematochezia, retention of urine in the 1st and the 2nd days of the observation group were lower than those in the control group (all P<0.05).
CONCLUSIONAuricular point sticking before operation combined with conventional western medicine with oral administration for preventing and treating postoperative complications of external excision and internal ligation on mixed hemorrhoid achieves positive and reliable efficacy.

6.Treatment of 92 Cases of Allergic Rhinitis by Combination of Acupuncture and Medicine

Objective: To investigate the therapeutic effect of combination of acupuncture and medicine in treating allergic rhinitis. Methods: Acupuncture, moxibustion, nasal inhalation and oral administration of Chinese materia medica were used as a composite treatment for different constitutions. The relations of the patient's age, syndrome type and course of disease to the therapeutic effect were investigated. Results: Among the treated 92 cases, marked effectiveness occurred in 68, effectiveness in 16 and ineffectiveness in 8, with a total effective rate of 91.3%. The therapeutic effect was not good in the patients who belonged to dry heat in lung meridian type and had the course of disease for more than 20 years. Conclusion: A combination of acupuncture and medicine has an exact therapeutic effect on allergic rhinitis.
7.Effect of electric vagal stimulation on myocardial ischemia-reperfusion injury in rats

Objective To evaluate the effect of electric vagal stimulation on myocardial ischemia-reperfu-sion (I/R) injury in rats.Methods Forty-five male Sprague-Dawley rats,aged 4-5 months,weighing 180-220 g,were randomly divided into 3 groups (n =15 each):sham operation group (S group),I/R group,electric vagal stimulation group (EVS group).Myocardial I/R was induced by occlusion of left anterior descending branch of coronary artery for 30 min followed by 120 min reperfusion in I/R and EVS groups.In group EVS,the left cervical vagus nerve trank was stimulated for 30 min with continuous electric pulses (5 V,2 ms,10 Hz) starting from 10 min before reperfusion until 10 min of reperfusion.The animals were sacrificed at the end of 120 min reperfusion and the hearts were removed for determination of the infarct size,TNF-α and IL-6 contents and myeloperoxidase (MPO) activity in myocardial tissues.Results Compared with group S,the infarct size was significantly enlarged,and TNF-α and IL-6 contents and MPO activity were increased in I/R and EVS groups (P < 0.05).The infarct size was significantly smaller,TNF-α and IL-6 contents and MPO activity were lower in EVS group than in I/R group (P < 0.05).Conclusion Electric vagal stimulation can reduce myocardial I/R injury in rats and activation of cholinergic anti-inflammatory pathway is involved in the mechanism.
8.Advances in research on oncolytic adenoviruses in tumor therapy.

Oncolytic adenoviruses (Ads), which are live, replication-competent viruses that can selectively replicate in tumor cells and lead to cell lysis, have been used in tumor therapy. But due to the complexity and high mutability of human tumors, it becomes a major strategy to improve the selectivity, efficacy, and safety of oncolytic Ads. The oncolytic Ads that can express short hairpin RNA, cytokines, suicide gene, and matrix-modulating proteins have higher antitumor activity than the wild type. Tumor-specific promoters, especially hTERT and HRE promoters, increase the selectivity of oncolytic Ads for tumor cells. Moreover, oncolytic Ads surface-modified by polyethylene glycol (PEG), liposomes, biodegradable nanoparticles, and polypeptides have reduced immunogenicity and hepatotoxicity and improved antitumor activity when systemically administered, and the selectivity of oncolytic Ads can be significantly increased when linking PEG to antibodies, small peptides, cytokines, and ligands. Therefore, engineered oncolytic Ads combining the advantages of viral and non-viral vectors, as well as immunotherapy, are a promising strategy for improving the efficacy of targeted virotherapy.

9.Expression and its Clinical Pathological Correlation of Heat Shocking Protein 40 in Epithelial Ovarian Cancer

Objective:The expression of Hsp40 in epithelial ovarian cancer and its correlation with clinical pathology were investigated. Methods:The expression of Hsp40 was detected by immunohistochemistry in 45 cases of epithelial ovarian cancer and 20 cases of benign ovarian tumor and 20 cases of normal ovary. Sta-tistical analysis was done to correlate the expression of Hsp40 with pathological characteristics. Results:The expression of Hsp40 in epithelial ovarian cancer was significantly higher than benign ovarian tumor and nor-mal ovary, the positive rates were 62.2% ,25.0% and 20.0% respectively ( P <0.01 ). There was no signifi-cant correlation between the expression of Hsp40 and clinical stages, histological grades,with or without me-testasis, with or without residual tumor. Kaplan-Meier survival curves and Log - rank analysis showed that the survival time in Hsp40 expression negative patients was significantly longer than those positive expression patients( P <0.01 ). Cox analysis showed residual tumor size was the only one covariant factor, the expres-sion of Hsp40 was not significantly relevant to prognosis. Conclusions. Hsp40 may play an important role in the occurrence and development of epithelial ovarian cancer.
10.Application of Response Surface Methodology in Optimization of Precursors for Taxol Production by Fusarium mairei K178

The concentration of precursors for the production of taxol with Fusarium mairei K178 was optimized by response surface methodology.Firstly,Plackett-Burman design was undertaken to evaluate the effects of eight factors.With statistic regression analysis,the significant factors affecting taxol production were determined as follows: phenylalanine,sodium acetate and sodium benzoate.Box-Behnken design was used to optimize the three critical internal factors mentioned above,and the optimum concentration levels and the relationships among these factors was found out.By quadratic regression model equation with Design-Expert statistic methods,the optimal concentration of the variables were determined as: phenylalanine 2.8mg/L,sodium acetate 3.3g/L,sodium benzoate 31.8mg/L.Under such conditions,the taxol production was increased to 242.6?g/L,which was 15.6% higher than the maximum value in the single factor tests.The experiment values under the optimal conditions agreed with the predicted values,which indicated that the model was proper and effective.
