1.Effects of total flavonoids of Epimedium sagittatum on the mRNA expression of the estrogen receptor α and β in hypothalamus and hippocampus in ovariectomized rats

Objective To explore the effect of total flavonoids of Epimedium sagittatum (TFE) on the mRNA expressions of the estrogen receptor alpha (ERα) and estrogen receptor beta(ERβ) in hippocampus and hypothalamus in ovariectomized (OVX) Sprague-Dawley(SD) rats, and the mechanism of TFE against postmenopausal osteoporosis. Methods Forty-eight female SD rats,aged 10-11 months old, were randomly divided into 4 groups: a sham group, an ovariectomy group (rats were bilaterally ovariectomized), a TFE group, and a 17β-estradiol group (rats were fed with TFE and 17β-estradiol for 4 months, respectively). The RT-PCR was performed to determine the mRNA expression of ERα and ERβ in hypothalamus and hippocampus. Results Serum estradiol level, bone mineral density (BMD) of vertebra,wet weight of uterus, and the mRNA expressions of ERα and ERβ in hypothalamus and hippocampus were markedly decreased in OVX rats, all of which were reversed by 17β-estradiol treatment except the mRNA expression of ERβ. Similar results were achieved by TFE treatment except the wet weight of uterus. Conclusion TFE can improve BMD of vertebra in the OVX rats without side effects on the uterus. The mechanism may be related to increasing the mRNA expressions of ERα and ERβ in hypothalamus and hippocampus.
2.Assay and clinical significance of serum tumor markers in diagnosis of pancreatic carcinoma

Objective To investigate the expression of serum tumor markers in pancreatic carcinoma and to assess the value of serum tumor markers in the diagnosis and prognosis of pancreatic carcinoma. Methods Five serum tumor markers including AFP, CEA, CA199, CA125 and CA242 were detected in 95 patients with pancreatic carcinoma by chemi-luminescence immunoassay (CLI) and enzyme linked immunosorbent assay (ELISA), 80 patients with other malignant tumor and 65 patients with benign disease served as controls. Five tumor markers in pancreatic carcinoma diagnosis for TNM staging were compared. The value of tumor markers for prognosis was analyzed. Results Among these 5 kinds of tumor marker, CA199, CAI25, CA242 were valuable in diagnosis of pancreatic carcinoma, and CA199 was the most valuable one, whose sensitivity and specificity were 76.2 % and 74.8 % respectively. CA199, CA125, CA242 were increased with progression of pancreatic carcinoma. Conclusion Serum CA199, CA125 and CA242 are the preferred tumor markers to be used in the diagnosis and prognosis of pancreatic carcinoma.
3.The surgical strategy of horseshoe kidney transplantation:a report of two cases and literature review

Objective To study the feasibility and safety of horseshoe kidney transplantation and avoid the waste of the donor kidney. Methods The horseshoe kidney was identified and confirmed during the organ procurement process. It was perfused in situ and procured en bloc. With an appropriate dissection and reconstruction on the beach table, the horseshoe kidney was carefully divided at the isthmus and transplanted into two separate recipients. The surgical strategies and postoperative outcomes of transplanting the cadaveric horseshoe kidneys were evaluated. Results Two recipients had a immediate return of renal function after the blood vessels were opened. One recipient had a normal renal function presented as lower post-transplant serum creatinine values with a follow-up of 12 months. There was no complications related to the horseshoe kidney. Another case died of infection 1.5 month later after the transplantation. Conclusions Cadaveric horseshoe kidney may be transplanted successfully using various individual technical strategies based on the specific renal anatomy. Considering the lack of donor horseshoe kidney transplantation is feasible and safe.

The effect of pH on the extracellularpolysaccharide synthesis by Aureobasidium pullulans was studied by addding CaCO3and HCI. Cultivated in P2 liquid medium for 24h,pH dropped to 3. 6 because of strongly producing acid. Under this low pH environment, further fermentation for 120h,only 5. 9g/L of polysacharide was obtained. When grown in MP2 medium contaning0. 5 % CaCOs,the pH was kept above 5.0 during 144 hours,production of polysaccharide increased to 31g/L. The detailed information of effects of controlled pH on polysaccharide production showed an optimal pH value 5. 0 must be maintained through the fermentative period.
5.Prognostic factors in 339 patients with T2N0M0 colorectal cancer

Objective To find T2N0M0 colorectal cancer patients at high risk for relapse or metastasis.Methods From January 1993 to December 2014,339 patients with histologically confirmed stage T2N0M0 primary colorectal cancer treated by radical surgery with complete clinical follow-up data were enrolled into this study.Survival rates were calculated using Kaplan-Meier method,and survival cures were compared using the Log-rank test.Cox proportional hazards model was used to analyze the significant factors defined in univariate test.Results The 5-year and 10-year overall survival rates were 83.0% and 68.9%,respectively.Male gender,old age,lymphovascular involverment,perineural invasion,poor differentiation and invasive micropapillary carcinoma were associated with low cancer-specific survival rates in Kaplan-Meier analysis.Multivariate analyses revealed male gender,old age,lymphovascular involverment,poor differentiation and invasive micropapillary carcinoma as significant independent factors predicting poor prognosis.Conclusions Male gender,old age,lymphovascular involvement,poor differentiation and invasive micropapillary carcinoma are risk factors predicting poor prognosis for T2N0M0 colorectal patients.
6.Distribution and antimicrobial resistance of pathogens isolated from hospi-talized patients with bloodstream infections

Objective To investigate the species and drug resistance of pathogens causing bloodstream infections in hospitalized patients,and provide scientific evidence for antimicrobial use and control of healthcare-associated blood-stream infection.Methods From January 1 to December 31,2012,16 428 blood specimens were performed blood culture,pathogens were isolated and performed antimicrobial susceptibility testing.Results Of 16 428 blood speci-mens from 5 546 patients,384 (6.92%)were positive for blood culture,398 pathogenic isolates were detected,of which gram-positive bacteria,gram-negative bacteria,and fungi accounted for 23.62% (n=94),68.34% (n=272),and 8.04% (n=32)respectively,positive rate of blood culture were highest in 61-80 age group(8.26%), the top five departments of positive rate of blood culture were departments of burn,traditional Chinese medicine, cardiac intensive care unit,transplantation and traumatology;gram-positive cocci were highly susceptible to vanco-mycin,teicoplanin and linezolid,one Enterococcus faecium strain was found to be resistant to vancomycin;Among gram-negative bacilli,Enterobacteriaceae were highly susceptible to amikacin and carbapenems;drug resistance rates of Acinetobacterbaumannii and Pseudomonasaeruginosa to carbapenems was 70.97% and 35.90% respective-ly.Conclusion Gram-negative bacteria are the major pathogens causing bloodstream infection,positive rate of blood culture of elderly people is high.It is necessary to conduct regular surveillance on distribution and drug resistance of pathogens.

Measuring the innovation capacity and competitive strength of the department mainly depends on the quality and quantity of its independent intellectual property.Here we discussed intellectual property issues occurring in the different stages of the application,approval,implementation and concluding,including the theses written at the application stage,the novelty retrieval of patent documentation,periodic achievement protection during implementation,the association between the patent application and paper publication,etc.And the intellectual property ownership was expounded according to Contract Law of PR China.The strategy of intellectual property protection was put forward from the view points of the mechanism,regulation,and personnel.The helpful tips are expected to the scientific researcher and the administrative staff.
8.Kidney transplantation in patients with severe abnormal lower urinary tract (report of 3 cases)

Objective To evaluate the surgical strategy and methods of kidney transplantation in patients with abnormal lower urinary tract. Methods Kidney transplantation was performed in 3 patients with abnormal lower urinary tract.Of them a 45 year old male recipient with tuberculous contracted bladder underwent cutaneous ureterostomy as kidney transplantation was conducted and then received immunosuppressive therapy of zenapax + tacrolimus (FK506) + mycophenolate mofetil (MMF) + steroid (Pred).The other 2 male patients with neurogenic bladder who were 24 and 34 years experienced kidney transplantation plus ileal conduit by one stage and by two stages,respectively.These 2 patients received immunosuppressive therapy of zenapax + FK506 + MMF + Pred and zenapax + CsA + MMF + Pred,respectively after transplantation. Results The patient who received ileal conduit by one stage experienced hematochezia and urinary fistula,which vanished after effective treatment.The rest 2 patients recovered smoothly.The functions of transplanted kidneys were satisfactory with no severe complications during follow up period. Conclusions The kidney transplantation and urinary diversion should be completed simultaneously or by stages depending on the conditions of the patients with abnormal lower urinary tract. The function of the transplanted kidney and the drainage of the ureter should be protected as much as possible.

10.T-2 toxin inhibits mitochondrial function of differentiated murine embryonic stem cells

OBJECTIVE ToexplorethepossiblemechanismoractiontargetsofT-2toxinembryo toxicity by observing the effect of T-2 toxin on mitochondrial function of differentiated murine e mbryonic stemcells(mESCs).METHODS Duringdifferentiationat24,72and120h,ESCswereexposedto T-2 toxin 0.5 μg·L-1 .Meanwhile,mESCs were pre-treated with antioxidant Trolox (200 μmol·L-1 )for 30 min and exposed to T-2 toxin (0.5 μg·L-1 )for 72 h.The mitochondrial ultrasture of differentiated mESCs was observed under a transi mission electrical microscope (TEM).The differentiated ESC mito-chondrial function,including respiratory control ratio (RCR),ATP synthase activity and mitochondrial membranepotential(MMP),wasmeasuredat144hafterdifferentiation.RESULTS Significant decrease of the mitochondrial number,deformation of mitochondrial structure,and lack of complete mito-chodrial crest were observed through TEM in the groups of T-2 toxin exposed for 72 and 1 20 h,respec-tively.Compared with the normal control group,RCR declined by 49.5% and 55.1%,ATP synthase activity decreased by 84.9% and 89.3%,and MMP decreased by 23.2% and 35.2% in T-2 toxin 0.5 μg·L-1 exposure 72 and 1 20 h group,respectively.However,the inhibition of mitochondrial function by T-2 toxin in differentiated mESCs recovered significantly in the presence of the antioxidant Trolox. CONCLUSION T-2toxininducesoxidativestressandinhibitsmESCsmitochondrialfunctionindifferenti-ated mESCs,and ROS-induced mitochondrial malfunction plays an i mportant role in T-2 toxin e mbryonic toxicity mechanis m.
