1.Efficacy Observation of A Clotrimazole Combined with Ultrasonic Atomization of Ozone in the Treatment of Candidal Vaginitis

Objective:To observe the efficacy of A clotrimazole combined with ultrasonic atomization of ozone in the treatment of candidal vaginitis. Methods:Totally 200 cases of candidal vaginitis were divided into the observation group and the control group with 100 cases in each. The control group was given A clotrimazole creams through vaginal administration, qd, while the observation group was treated by ultrasonic atomization of ozone additionally, qd, 15min per time with five times. The clinical efficacy and the recurrence rate after 6 months of the two groups were compared. Before the treatment and in the first and fourth week after the treatment, vaginal secretion bacterial culture was performed and pH value was detected. Results:The cure rate and the total effective rate of the observa-tion group was 88% and 98%, respectively, which was significantly higher than that of the control group (69% and 82%, respective-ly, P<0. 05). After the 6-month follow-up, the recurrence rate was 1. 0% in the observation group (1/98), which was significantly lower than that in the control group [7. 3%(6/82),P<0. 05]. After the treatment, the vaginal pH value in the observation group was reduced notably and the number of lactobacillus was increased significantly(P<0. 05), while those in the control group were without significant changes (P>0. 05). Conclusion:A clotrimazole combined with ultrasonic atomization of ozone in the treatment of candidal vaginitis can improve the therapeutic effect.
2.Comparison of FACS analysis indicators and traditional infection indicators in early diagnosis of infection in premature infants

Objective To compare the clinical application of FACS analysis indicators and traditional infection indicators in early diagnosis of infection in premature infants .Methods 60 premature infants were divided into the infected group (n=29) and non-in-fected group(n=31) according to their clinical symptoms and laboratory results .BD FACSCanto Flow Cytometry was employed to detect CD11b ,CD64 and CD45RO ,BACTEC 9120 Automated Blood Culture System was used to conduct body fluids and secretions culture .Sysmex XE-5000 Automated Hematology Analyzers and i-CHROMA Reader Immunofluorescence Analyzer were adopted to conduct the complete blood count test and hypersensitive C-reactive protein (hs-CRP) detection ,respectively .Receiver operator characteristic(ROC) curve was used to analyze the diagnostic value of indexes above in preterm infants with infection ,and their sen-sitivity ,specificity ,positive predictive value and negative predictive value were calculated .Results On the first day after birth ,neu-trophil CD11b ,CD64 ,monocyte CD64 and hs-CRP levels of preterm infants in infection group were obviously higher than those in non-infection group(P<0 .05) .Differences of CD45RO ,WBC ,neutrophil absolute and percentage between the two groups showed no statistical significance(P>0 .05) .ROC area under the curve(AUC)>0 .7 was found in Neutrophil CD64 ,monocyte CD64 and hs-CRP ,which had higher value in early diagnosis of infection in premature infants .The highest sensitivity ,specificity ,positive pre-dictive value and negative predictive value were 79 .31% ,96 .78% ,83 .34% and 75 .00% ,respectively .Conclusion FACS analysis indicators has better clinical value in the early diagnosis of infection in premature infants .
3.Effect of Yiqi Huatan Decoction on Aquaporin 5, Tumor Necrosis Factor-alpha and MUC5AC in Bronchoalveolar Lavage Fluid of Chronic Obstructive Pulmonary Diseases Rats

Objective To investigate Yiqi Huatan Decoction(YHD), a compound recipe with the actions of tonifying Qi and resolving phlegm, on aquaporin 5(AQP5), tumor necrosis factor-alpha(TNF-α)and mucin 5AC(MUC5AC)in bronchoalveolar lavage fluid(BALF)of chronic obstructive pulmonary diseases(COPD)rats. Methods SD rats were randomized into blank control group, model group, and low-, middle- and high-dose YHD groups(in the dosage of 7.398, 36.99, 73.98 g·kg-1·d-1 respectively). The rat model of COPD was induced by cigarette smoking combined with intratracheal dripping of lipopolysaccharide(LPS). After COPD rats were treated with YHD for 30 days, the histological features of lung tissues were observed after hematoxylin-eosin (HE) staining, the expression of AQP5, TNF -α and MUC5AC in the lung tissue was detected by immunohistochemistry, the concentrations of AQP5, TNF-α and MUC5AC in BALF of rats were measured by enzyme-linked immunosorbent assay(ELISA). Results Compared with the blank control group, the concentration of AQP5 in BALF of the model group was decreased significantly(P<0.01), while the concentrations of TNF-αand MUC5AC were significantly increased(P<0.01). Compared with the model group, the pathological features of the lung tissue were relieved, and the concentration of AQP5 was increased significantly in low-, middle-, high-dose YHD groups (P<0.01), but the concentrations of TNF-α and MUC5AC were significantly decreased (P<0.01), and the effect of high-dose group was superior to low-and middle-dose groups(P<0.01). Conclusion The therapeutic mechanism of YHD for COPD is probably related with the regulation of fluid transport in aquaporin water channels of rats.
4.Down-regulation of ubiquitin specific peptidase 18 suppresses the invasion and proliferation of gastric cancer cells

Objective To analyze the expression and role of ubiquitin specific peptidase 18 (USP18) in gastric cancer cells,and to investigate the relationship between the development of gastric cancer and USP18.Methods The levels of USP18 protein and mRNA expression in immortalized gastric mucosa epithelial GSE cell lines and gastric cancer cell lines (AGS,MKN45,MKN25,BGC823,BGC803,SGC7901) were detected by using reverse transcription-polymerase chain reaction (RT-PCR) and Western blot,respectively.The role of USP18 in the invasion and proliferation of gastric cancer cells was analyzed by using CCK8 and Transwell assays.Results The mRNA level of USP18 was lower in GSE cell lines than that in gastric cancer cells (F =794.052,P < 0.000 1).In six gastric cancer cell lines,mRNA level of USP18 was relatively high in BGC823 (17.62±0.55) and BGC803 (13.52±0.50) cell lines,and low in MKN28 (1.40±0.17) and MKN45 cell lines (4.23±0.26).As for the protein level,the expression of USP18 was lowest in GSE cell line.In six gastric cancer cell lines,the expression of USP18 was the highest in more aggressive SGC7901 and BGC803 cell lines and the lowest in AGS and MKN45 cells.Compared with the control group,interference of USP18 decreased the invasion and proliferation abilities of SGC7901 and BGC803 cell lines (P < 0.01).Conclusion USP18 is highly expressed in more invasive gastric cancer cells,and the downregulation of USP18 can suppress the invasion and proliferation of gastric cancer cells.
5.High field magnetic resonance background suppression diffusion imaging in the diagnosis of liver foci of space occupying lesion.

OBJECTIVE:
To explore the diagnostic value of the 3.0T magnetic resonance liver diffusion weighted imaging with background suppression (DWIBS) in liver foci of space occupying lesion.
METHODS:
A total of 43 cases of liver bureau stove perch pathological change were included: 15 were hepatocellular carcinoma (HCC) with 24 lesions; 7 were liver metastatic tumor with 13 lesions; 10 were liver hemangioma with 12 lesions; and 11 liver cyst with 20 lesions. After taking the conventional T1WI and T2WI sequence, the magnetic resonance background suppression diffusion imaging technology (diffusion weighted imaging with background suppression, DWIBS) was applied, following the dynamic enhanced scan. With the MRI to DWIBS workstation for classifying positron emission computed tomography (PET) processing, the T2WI diagram and dynamic enhanced diagram were compared respectively for the 3 sequences of lesion detection rate, T2WI, and DWIBS, to enhance the delay time between the two joint and combined lesion detection rate. With the MRI workstation software scanning image generation ADC diagram, the ADC values were measured for liver cancer, liver metastatic tumor, liver hemangioma and liver cysts.
RESULTS:
The 3 sequences of detection rates of the T2WI, DWIBS and enhanced delay period T1WI were 91.3%, 94.2%, and 95.6%. The detection rate of DWIBS plus T2WI was 92.7%; that of T2WI plus enhanced delay time was 94.9%, and that of DWIBS plus enhanced delay time was 96.3%, with the rate of DWIBS plus enhanced delay period obviously higher than that of the DWIBS plus T2WI (P<0.05). The ADC value of the benign liver tumor was obviously higher than that of the malignant tumors: hepatic cyst (2.614 ± 0.57)×10⁻³ mm²/s, liver hemangioma (2.055 ± 0.21)×10⁻³ mm²/s, metastatic carcinoma (1.374 ± 0.32)× 10⁻³ mm²/s, and liver cancer (1.287 ± 0.14)×10⁻³ mm²/s. Except for the liver cancer and the liver metastatic tumor, there was significant difference between the other groups (P<0.05).
CONCLUSION
Combing the DWIBS technology, the PET-like images and the ADC value acquired, the combined enhanced sequences could further facilitate the demonstration of the liver foci of space occupying lesion, the accuracy of identification and diagnosis of the liver foci of space occupying lesion.

6.Effect of electroacupuncture on the expression of P53 and P21 in articular cartilage and subchondral bone of aged rats with knee osteoarthritis

BACKGROUND:There are many treatment methods for knee osteoarthritis,among which electroacupuncture,as an important non-drug treatment,is effective in the treatment of knee osteoarthritis,but its exact mechanism is not clear. OBJECTIVE:Effect of electroacupuncture on the expression of p53 and P21 in articular cartilage and subchondral bone of aged rats with knee osteoarthritis. METHODS:Eight 6-month-old male Sprague-Dawley rats were included in the young group and sixteen 24-month-old male Sprague-Dawley rats were randomly divided into old group(n=8)and electroacupuncture group(n=8).The rats in the electroacupuncture group received electroacupuncture stimulation once a day,5 days a week,for 8 continuous weeks,and the other two groups did not do any treatment.Eight weeks later,the level of type Ⅱ collagen C-terminal peptide in peripheral blood was detected by ELISA,the morphology of left knee cartilage and subchondral bone was observed by safranin O-fast green staining,the degree of knee cartilage degeneration was evaluated by modified Mankin's score,the microstructure of left knee cartilage and subchondral bone was detected by micro-CT,and the expression levels of matrix metalloproteinase 13,P53,P21 Mrna and protein were detected by RT-PCR and western blot respectively. RESULTS AND CONCLUSION:Compared with the young group,the level of C-terminal peptide of type Ⅱ collagen in the peripheral blood was increased in the old group(P<0.05).The micro-CT results showed that the bone volume fraction,bone mineral density and the number of bone trabeculae were decreased in the old group compared with the young group(P<0.05),while the trabecular separation increased(P<0.05).Safranin O-fast green staining showed that in the old group,the surface layer of cartilage was uneven with fissures,the morphology of chondrocytes was irregular and stained unevenly,the boundary between the cartilage and subchondral bone was blurred,and the matrix loss was serious.The Mankin's score was higher in the old group than the young group(P<0.05).The expression of matrix metalloproteinase 13,P53,P21 at Mrna and protein levels increased in the old group compared with the young group(P<0.05).Compared with the old group,electroacupuncture decreased the level of C-terminal peptide of type Ⅱ collagen(P<0.05),increased the bone volume fraction,bone mineral density and the number of bone trabeculae(P<0.05),and decreased the trabecular separation(P<0.05).Safranin O-fast green staining showed that in the electroacupuncture group,the surface of cartilage was smooth and red staining was uniform,and the cell morphology and structure were between the young group and the old group.Following electroacupuncture treatment,the Mankin's score(P<0.05),matrix metalloproteinase 13 and P21 Mrna expression(P<0.05),and matrix metalloproteinase 13 and P53 protein expression decreased(P<0.05),while there was a decreasing trend of P53 Mrna and P21 protein expression,but with no statistical significance(P>0.05).To conclude,electroacupuncture may delay articular cartilage degeneration and subchondral osteoporosis in aged rats by inhibiting the expression of P53 and P21,so as to protect joints and delay joint aging.

"The Expert Group on Tumor Ablation Therapy of Chinese Medical Doctor Association, The Tumor Ablation Committee of Chinese College of Interventionalists, The Society of Tumor Ablation Therapy of Chinese Anti-Cancer Association and The Ablation Expert Committee of the Chinese Society of Clinical Oncology" have organized multidisciplinary experts to formulate the consensus for thermal ablation of pulmonary subsolid nodules or ground-glass nodule (GGN). The expert consensus reviews current literatures and provides clinical practices for thermal ablation of GGN. The main contents include: (1) clinical evaluation of GGN, (2) procedures, indications, contraindications, outcomes evaluation and related complications of thermal ablation for GGN and (3) future development directions.
