China Pharmacy 2016;27(30):4212-4214
OBJECTIVE:To compare the efficacy and safety of Sodium hyaluronate versus Carbomer,Polyethylene glycol,To-bramycin and Dexamethasone in the treatment of glaucoma with cataract. METHODS:464 glaucoma patients with cataract were randomly divided into group A(116 cases),group B(116 cases),group C(116 cases)and group D(116 cases). All patients re-ceived intraocular lens implantation;1-1.5 cm to Tramycin and Dexamethasone eye ointment,3-5 times a day. Based on it,group A received 1-2 Sodium hyaluronate eye drops,4 times a day. Group B received 1 Carbomer eye drop,3-5 times a day. Group C re-ceived 1-2 Polyethylene glycol eye drops,3-5 times a day. Group D received Tobramycin and dexamethasone eye ointment,1-1.5 cm every times,1-3 times a day. They treated for 1 month. Clinical efficacy, recovery of visual acuity and ocular surface functions [dry eye symptom score, tear film break-up time(BUT),corneal fluorescein staining(CFS),basal tear secretion(SIt),ocular sur-face disease index(OSDI)] and the incidence of adverse reactions in all groups were observed. RESULTS:The total effective rate sorted, patients with visual acuity higher than 0.6,BUT,OSDI by group A>group B>group C>group D,with significant differenc-es (P<0.05). patients with visual acuity in 0.3-0.6,0.1-0.3 and lower than 0.1,drycye symptom score,CFS,SIt in group A was less than group B,which was less than group C and group D,with significant differences(P<0.05). The incidence of adverse re-actions in group A was lower than group B,C,D,with significant differences(P<0.05),but there was no significant difference in group B,C,D(P>0.05). CONCLUSIONS:Sodium hyaluronate shows superior efficacy and safety to Carbomer,Polyethylene glycol,Tobramycin and Dexamethasone in the treatment of glaucoma with cataract,it can significantly improve visual acuity and ocular surface function.

This article was aimed to study the action of antigout effects of compoundWeifu. Through the acute gouty arthritis models of rabbits and rats induced by micro-crystal sodium urate (MSU). The histopathological changes of synovial tissues among rabbits were observed. And the contents of TNF-α, IL-1β, IL-6 in rabbit serum were determined. The swelling degree of the rats’ feet and the content of prostaglandin E2 (PGE2) from the inflammatory exudate foot induced by MSU were also determined. The antigout effects of compoundWeifu were observed. The results showed that the grade of the vascular congestion, swelling and inflammatory cell infiltration in synovial tissues in the high-dose and medium-dose CompoundWeifu group had significantly improved (P <0.05). And the levels of TNF-α, IL-1β, IL-6 in serum were significantly decreased (P< 0.05, orP < 0.01). It can obviously reduce the swelling degree induced by MSU, and the level of PGE2 in the inflammatory exudate foot (P< 0.05, orP < 0.01). It was concluded that CompoundWeifu had obvious antigout effects in the study.

Objective To overcome the fact that SRV-NM virus can only multiple and amplify through partially pu-rified jaagsiekte retrovirus inoculated intratracheally in sheep but it cannot be augmented using in vitro cell culture, we con-structed JSRV-NM pseudovirions based on high efficiency packing system of lentivirus. Methods Lentivirus of three high efficiency packing plasmids system pMD.G, pCMV-HIV 8.2 and pHIV-eGFP was developed, and JSRV-NM-env coated plasmid pCMVJSRV-NM was used to substitute VSV-G virus coated plasmid pMD.G then co-transfected into 293T cells to replicate, package and produce restructured JSRV-NM pseudovirions. Gene expression of pseudovirion was determined through WPRE using real time PCR; Virus infectivity was detected through inoculating JSRV-NM pseudovirions into 24 pore plates. Results We construct JSRV-NM pseudovirions successfully based on the lentivirus system. JSRV-NM pseudo-virions can also be concentrated to higher titer (108 TU/mL detected by real time PCR by ultracentrifugation without signifi-cant loss of activity. JSRV-NM and VSV-G pseudovirions infected on Hela cells (both MOI= 3) respectively and no obvi-ous difference were shown on their infection efficiency detected by real time PCR. Conclusion Based on lentivirus system, JSRV-NM pseudovirions can be multipled and amplified in 293T cell culture in vitro. JSRV-NM pseudovirions is stable without loss its infection activity and the requirements of biological laboratory safety II was also met. JSRV-NM pseudoviri-ons will provide a useful tool for further study of JSRV-NM-env infection across species or its induction of lung adenocarci-noma.

AIM:The aim of this study is to investigate the relationship between the ratio of transcription factors T-bet/GATA-3 in peripheral blood mononuclear cells(PBMCs)and the imbalance of Th1/Th2 cytokines from patients with asthma, and to examine whether and how CpG ODN intervention would restore the imbalanced Th1/Th2 profile in asthma patients.METHODS:Three groups of subjects with asthma(asthma group),chronic obstructive pulmonary disease(COPD group),and normal controls(control group)were included in this study.T-bet,GATA-3 and TLR9 mRNA levels were measured by reverse transcription-polymerase chain reaction(RT-PCR).Th1 cytokine IFN-? and Th2 cytokines IL-4,IL-5 and IL-13 were measured by enzyme-linked immunoabsorbent assay(ELISA)in asthma group.Cultured PBMCs from asthma patients with CpG ODN incubation were collected for measuring T-bet,GATA-3 mRNA and TLR9 mRNA.RESULTS:The ratio of T-bet/GATA-3 in asthma group was significantly lower than that in the COPD group and control group. The serum levels of IL-4,IL-5 and IL-13 had positive correlation with the ratio of T-bet/GATA-3 in asthmatic patients, and the lower serum level of IFN-? had negative correlation with the ratio. The TLR9 mRNA expression in asthma was significantly attenuated than that in COPD group and control group.Consequently, the ratio of T-bet/GATA-3 in CpG group was significantly enhanced than that in control group.CONCLUSION:These results indicate that the ratio of T-bet/GATA-3 in asthma is decreased and could represent the imbalance of Th1/Th2 cells.CpG ODN influences the ratio of T-bet/GATA-3,upregulating the T-bet mRNA expression and downregulating the GATA-3 mRNA expression, which is able to reverse the imbalance of Th1/Th2 cells.CpG-ODN may be a promising therapeutic target for asthma.

Objective To understand patient pathway and clinical characteristics of chronic diseases in urban areas of Shanghai. Methods A total of 10 002 residents were enrolled and assigned to the chronic disease group (including hypertension, diabetes, coronary heart disease, myocardial infarction, and ischemic stroke) and the non-chronic disease group. Body mass index, fasting blood glucose, triglyceride, total cholesterol, high-density lipoprotein cholesterol and low-density lipoprotein cholesterol were tested.Difference of patient pathway and clinical characteristics of those chronic diseases was compared. Results Above chronic diseases were observed in 37.7% participants. About 2/3 diseases were confirmed and 80% patients were followed up in healthcare units not far away from home. Patients with coronary heart disease and myocardial infarction showed more outpatient visit to tertiary hospitals (P<0. 05 ). However, patients with ischemic stroke had health check, rehabilitation and pharmacy done mainly in local healthcare centers (P<0. 05 ). Diastolic blood pressure of patients visiting local doctors was significantly decreased (P<0. 05). Conclusion Some differences in patient pathway were found in this study. Communication and cooperation between medical institutions should be intensified for effective chronic disease control.

To analyze and evaluate the knowledge of Chinese Guidelines of Diabetes Prevention and Treatment in Shanghai medical staff. 175 medical staff working in endocrinology or community health were enrolled and evaluated by a questionnaire of guidelines about the state of professional, training, and related knowledge. Only 16. 6% medical staffwere trained about the guidelines( 46. 67% from the general hospitals, 14. 75% from secod-level hospital and 7. 14% persons from the community hospitals, P<0. 01 ). The total correct answer rate of the guidelines was 37. 36%. The correct rate of community hospitals was lower than others( P<0. 05 ). The rate of doctors' was higher than nurses'( P<0. 05 ). There were difference between doctors and nurses with the key point of diabetes care knowledge in different level hospitals. The effective method of clinical training in diabetes care should be explored. We still have to work hard to promote the effect of diabetes control and prevention. Effective training about the guidelines should be enhanced. The cooperation between general hospitals and community health institutions in diabetes prevention and treatment should be enhanced.
