1.Cloning of Fab Gene of an Anti-Human Bladder Cancer Monoclonal Antibody and Its Expression in E. coli

Objective: To clone the Fab gene of a monoclonal antibody (mAb) BDI against human bladder cancer and its expression in E. coli. Methods: Fd and K genes of mAb BDI were cloned by RT-PCR and inserted into an Fab expression vector. Phage displaying Fab and soluble Fab were expressed in E. coli. The N-terminal sequence of VH region was corrected by PCR mediated mutagenesis. The antigen-binding characteristics of the Fab were tested by ELISA and immu-nohistochemistry. Results: Fd and K genes were cloned into the expressing vector p3MH and the phage displaying antibody and soluble Fab were expressed in E. coli, which showed weak binding activity to bladder cancer cells. Correction of the N-terminal sequence of the VHimproved the biding activity dramatically. The feasibility of the application of the Fab in phage antibody library screening was confirmed by a simulated panning procedure. Conclusion: The Fab gene of an anti-human bladder cancer mAb was expressed in E. coli. The importance of the N-terminal sequence on antibody binding activity was suggested.
2.Quality standard for Naolingsu Tablets

Objective:To establish the quality standard for Naolingsu Tablets.Methods: Applying the technique of TLC, Radix Ginseng and Herba Epimedii were identified. The content of icariin was determined by HPLC. Results: Radix Ginseng and Herba Epimedii can be determined by TLC. Icariin showed a good linear relationship in the range of 0.011 ~ 0.424 ?g and the average recovery is 102.1% , RSD is 1.0% . Conclusion: The methods are simple, sensitive and accurate with good reproducibility. The methods may be used for quality control of Naolingsu Tablets.
3.Application of Roy adaptation model in patients with adolescent insanity

Objective To investigate the effect of Roy Adaptation Model(RAM)in patients with adolescent insanity.Methods One hundred patients with adolescent insanity during October 2011 to March 2012 were randomized in equal number into two groups by random digit table:the study group and the control group.The former were intervened with RAM and the latter received routine care and health education.Seven weeks after intervention,Hamilton Depression Rating Scale(HAMD),Hamilton Anxiety Scale(HAMA) and Observation Scale(NOSIE)were used for the assessment. Results After intervention,the scores on HAMD and HAMA in the study group were significantly lower,compared to the control group(P<0.01).The scores on social function,social interest,subjective support and use of social support were all significantly higher than those of the control group(all P<0.01). Conclusions RAM can improve their ability of the patients with adolescent insanity to adapt to the environment.It may improve their mental state and their quality of life.
