3.Significance of ambulatory blood pressure monitoring in the prevention and early intervention of hypertension
Chinese Journal of Tissue Engineering Research 2005;9(27):248-249
OBJECTIVE: The application of ambulatory blood pressure monitoring (ABPM) has been a great impetus to clinical hypertension research in respect of the liability, differentiation between high-risk and low-risk patients and the therapeutic effect of antihypertensive drugs. Therefore, in this study we reviewed recent research on ABPM in the prevention and early intervention of hypertension. DATA SOURCF: Related articles were retrieved by computer in the Medline between January 1993 and January 2002, with the key words of 'ambulatory blood pressure monitoring', 'hypertension', 'life satisfaction', 'effect factors'and 'blood pressure monitoring' and the language confined to English. STUDY SELECTION: After the first trial, literature containing research randomized controlled clinical trial and review on the confirmed primary, allel control group. Repetitive studies were excluded. DATA EXTRACTION: We selected a total of 20 articles on the association of ABPM with the prevention and early intervention of hypertension, 7of which accorded with the inclusion criteria. The other 13 articles were excluded because 10 of them had little relevance to the objective of our study and 13 were repetitive studies.DATA SYNTHESIS: ABPM was used to assess general BP profile, the peak level of drug action and lasting time under various states, such as resting, instable emotion, as well as movement and sleep. Then the safe dosage and medication time were draw up according to the peak-effect (the maximum decrement of diastolic pressure and systolic pressure after a dosage administration) and the valley effect (the decreasing amplitude when completing a dosage medication), as well as the relationship between them, aiming to keep the antihypertensive peak-to-valley ratio above 60%,which guaranteed the stable antihypertensive effect within 24 hours and was beneficial to the individualized treatment. CONCLUSION: ABPM can decide whether patients with hypertension need earlier intervention, and has been used in the assessment of therapeutic effect. It is also considered as an important independent predictor for the occurrence of hypertension complications. Twenty-four hours ABPM can not only determine the type of hypertension, but also keep the patients informed of their BP fluctuation profile at any time in order to avoid hypotension when sleeping, thus helping prevent severe hypertension complications such as stroke. Furthermore, ABPM can predict the severity of hypertension, and BP variation and day-night fluctuation, which provides guidance for the early intervention, correct medication and regular life.

7.Putative cell wall-binding proteins of Streptococcus suis serotype 2 isolate 98HAH33 identified by bioinformatic genome analysis
Chinese Journal of Zoonoses 2010;(1):72-75,80
To identify the cell wall-binding proteins of Streptococcus suis serotype 2(SS2), according to their structure feature, the substrates of sortase and I type secreted proteins with LysM domain, WxL domain, choline-binding domain,GW domain or S layer homologous domain in the recently published genome of SS2 strain 98HAH33 were firstly identified and then the putative functions were attributed to individual proteins by reference to the identification of conserved domains of InterPro and BlastP servers. Homologous proteins were identified by unfiltered BlastP homology searches (including conserved domain detection). Among the 23 putative proteins with a C-terminal LPXTG recognition signal for covalent attachment to peptidoglycan by sortase, 9 with I signal peptide were identified as sortase substrates. Among 9 substrates , YP_001201232, YP_001201531 and YP_001201656 had been experimentally verified to anchor to bacterial cell wall , and YP_001201232 known as the opacity factor of S. suis (OFS) was proved to be the virulence factors. According to function analysis, YP_001201484, YP_001201544, YP_001199825, YP_001197640, YP_001197840 and YP_001199755 appeared to be involved in SS2 pathogenesis. and YP_001200959, YP_001201233 and two proteins with LysM domain( YP_001199784 and YP_001201729) were the hypothetical proteins. These data suggest the majority of putative sortase substrates may implicate in the virulence of SS2 and could serve as a basis for targeted experimental studies into the function of these proteins.

9.Effects of vitamin E on the expression of hyaluronic acid synthetase-2 gene in human dermal fibroblasts in vitro
Chinese Journal of Medical Aesthetics and Cosmetology 2009;15(4):217-219
Objective To explore the mechanism of vitamin E on delaying skin aging by observ-ing the expression of hyaluronic acid synthetase-2 (HAS-2) in human dermal fibroblasts in vitro. Methods Human skin fibroblasts were cultured in vitro, and these fibroblast cells were then divided into 3 groups: different concentration of vitamin E (0, 0.1 × 10-10, 1 ×109mol/L) was added in the medium in the different group. 24 hours later, the fibroblasts were collected, RNAs extracted, and then amplified by RT-PCR. The PCR product was determined by agarose gel electrophoresis, to analyze the level of HAS-2 mRNA expression. Results RT-PCR showed the lever of HAS-2 mRNA was higher in the low-dose group than the control group, with significant difference (P<0.05) ; the lever of HAS-2 mRNA was higher in the high-dose group than control group, with significant difference (P<0.05). There was no significant difference in the lever of HAS-2 mRNA between the low-dose group and the high-dose group. Conclusions Vitamin E can enhance the expression of hyaluronic acid synthetase-2 mRNA, may increase the synthesis of HAS in skin fibroblasts and increase water content in the skin, so that it might reverse or delay the skin aging.
10.Improvement of phencynonate hydrochloride on posterior circulation vertigo in rats and positive vertigo in mice
Chinese Journal of Pharmacology and Toxicology 2017;31(3):231-236
OBJECTIVE To evaluate the anti-vertigo effect of phencynonate hydrochloride. METHODS To detect the improvement of phencynonate hydrochloride on cerebral blood flow, a rat model wa s es?tablished with bilateral common carotid arteries occlusion. Phencynonate hydrochloride 0.1-4.0 mg·kg-1 was ig given, twice a day for three consecutive days and the alteration of cerebral blood flow was measured with laser Doppler flowmetry. Rotating acceleration equipment was used to provocate mouse vertigo for 30 min, and the spontaneous locomotor activities were tested for occurrence of vertigo in mice. Phencynonate hydrochloride 1.4-5.6 mg·kg-1 was ig given before rotating acceleration. Gastric phenol red emptying rate was used to determine the anti-nausea effect of the test drug in mice 30 min after phencynonate hydrochloride 1.4-8.4 mg·kg-1 was ig given. RESULTS The cerebral blood flow of the rat model with bilateral common carotid arteries occlusion was reduced significantly after 24 h (P<0.01). Compared with the model group, phencynonate hydrochloride (0.5, 2.0 and 4.0 mg · kg-1) increased the cerebral blood flow in a dose-dependent manner in rats with cerebral ischemia (P<0.01). The spontaneous locomotor activities were significantly reduced after vertigo stimulation in mice (P<0.05). Compared with the model group, phencynonate hydrochloride (2.8 and 5.6 mg · kg-1) increased the movement distance and speed of vertigo mice (P<0.05). Phencynonate hydrochloride (2.8, 5.6 and 8.4 mg·kg-1) inhibited the gastric emptying of mice (P<0.05). CONCLUSION Phecynonate hydrochloride can improve the cerebral blood flow and locomotor activities in vertigo rats, while inhibiting gastric emptying, which points to the therapeutic potential of phencynonate hydrochloride for vertigo in clinic.
