2.The New Healthcare Reform & the Reconstruction of Medical Ethics and Relevant Regulations——An Overall View of Current Chinese Medical Ethics Construction and Relevant Heated Issues
Chinese Medical Ethics 1994;0(06):-
The nature of medical science, especially the highly socialized characteristic of modern healthcare service determines that a larger ratio of medical ethics should be included in the new healthcare reform, with the construction of a new medical ethics taken as support and guidance. An overall view of medical ethics is crucial in the construction of medical ethics under the context of current Chinese healthcare reform, that is,the construction of medical ethics should be performed in an ultra-wide field.Medical ethics should take a complete participation in each and every field of healthcare reform, but not merely focuses on the particular field of medical professionals and the construction of a harmonious physician-patient relationship. In the systematic construction of new medical ethics, the current key issue lies in the reconstruction of a socialist medical ethics and related regulations, both for theoretical and practical reasons. The reconstruction is far more than releasing a couple of policies.Merely in a theoretical perspective, seven major issues are to be resolved during the reconstruction process.

Chinese Journal of Tissue Engineering Research 2014;(15):2345-2350
BACKGROUND:Runx2 is considered to the main regulatory factor of osteogenic gene expression and be necessary for osteoblast differentiation, it plays an extremely important role in the osteoblast development, differentiation, regulation, bone calcification formation and bone repair.
OBJECTIVE:To observe the biological properties of mesenchymal stem cells from human exfoliated deciduous teeth, explore the osteogenic differentiation potential of deciduous teeth stem cells, and observe the dynamic expression of Runx2 gene at varying time points.
METHODS:The stem cells from human exfoliated deciduous teeth were isolated and cultured in vitro. The cellsurface antigen was detected with flow cytometry. The third passage cells were cultured in the adipogenic medium for 4 weeks, and oil red O staining was conducted to test lipid droplets formation. The third passage cells were cultured in the osteogenic medium for 21 days, and mineralized nodules were detected by alizarin red staining. Runx2 mRNA dynamic expression was detected with semi-quantitative RT-PCR at different time points.
RESULTS AND CONCLUSION:The stem cells from human exfoliated deciduous teeth were obtained by enzyme digestion and limited dilution methods. Flow cytometry results showed that, CD146 and STRO-1 were expressed to varying degrees. Oil red O staining revealed salmon pink positive particles. Alizarin red staining showed positive expression. RT-PCR results showed that, Runx2 expression was found at day 0, up-regulated from day 0 to day 6, and subsequently dropped with an expression bottom at day 12, after that a second expression peak occurred at day 18, fol owed by a stably regulation. The stem cells from human exfoliated deciduous teeth can be isolated and cultured in vitro, express surface antigen of mesenchymal stem cells, and have the potentials of differentiating into adipocytes and ostetoblasts. Runx2 gene profiles are dynamical y expressed during osteoblastic differentiation. Runx2 express throughout every stage of osteoblastic differentiation. The expression is up-regulated during early and later stages, and down-regulated in metaphase.

Chinese Journal of Biochemical Pharmaceutics 2016;36(12):15-19
As a member of PARP superfamily, PARP3 shares a high homology with PARP1 and PARP2, which are all DNA-dependent enzymes that are catalytically activated by DNA strand breaks.Compared to PARP1 and PARP2, PARP3 exerted some special properties in tissue expression pattern and biological function. The evidence has shown that PARP3 could be activated by DNA double strand breaks and special DNA single strand breaks and synthesize mono(ADP-ribose) (MAR) covalently attached to target proteins including itself.PARP3 plays an important role in DNA double strand breaks, DNA single strand breaks, activation of PARP1 and development of nervous system. It has been reported that PARP3 is associated with glioma and breast cancers. In this review, PARP3 structure, activation mechanism, biological function and its relationship with diseases will be presented.

Chinese Journal of Microbiology and Immunology 2014;(5):337-342
Objective To investigate the effects of Kr üppel-like factor 2 ( KLF2 ) on oxidized low density lipoprotein (ox-LDL) induced expression of microRNA-146a (miR-146a) and proinflammatory cyto-kines (MCP-1 and IL-6) in human umbilical vein endothelial cells (HUVECs).Methods Human umbili-cal vein endothelial cells (HUVECs) were cultured and then stimulated with 50μg/ml ox-LDL for 24 hours. HUVECs were infected with adenovirus vectors over-expressing human KLF2 at an appropriate multiplicity of infection.KLF2-siRNA duplexes were transfected into HUVECs to silence the gene expression .HUVECs were collected at time points of 0 h, 3 h, 6 h, 12 h, 24 h and 48 h after infection.Real-time quantitative-PCR was performed to measure the expression of miR-146a at mRNA level.Silenced endogenous miR-146a using LNA-anti-miR-146 a was transfected into HUVECs with lipofectamine 2000 .The levels of MCP-1 and IL-6 in supernatants were detected by ELISA .Results KLF2 remarkably inhibited the expression of miR-146 a in unstimulated and ox-LDL-stimulated HUVECs in a time-dependent manner .The ox-LDL induced ex-pression of miR-146a, MCP-1 and IL-6 in HUVECs were significantly decreased by KLF2.Silenced expres-sion of miR-146a downr-egulated the ox -LDL induced expression of MCP-1 and IL-6 in HUVECs.Moreover, silenced miR-146 a could partly reverse the inhibitory effects of KLF 2 on ox-LDL induced expression of MCP-1 and IL-6 in HUVECs.Conclusion KLF2 inhibited ox-LDL induced expression of MCP-1 and IL-6 in HUVECs partly through down-regulating the expression of miR-146a.
