Objective To evaluate left ventricular function in patients with acute myocardial infarction and to investigate the clinical value by two-dimensional strain imaging of ultrasound.Methods Study population was consisted of 28 patients with acute myocardial infarction of wall anterior and 30 healthy subjects.Images were recorded from the left ventricular the apical 4-chamber view,apical 3-chamber view,apical 2-chamber view respectively.Two-dimensional strain and strain rate were measured in the left ventricular segments using two-dimensional strain software.We analyzed that relationship between two-dimensional the global peak systolic strain,global strain rate and left ventricular end systolic volume(LVESV),left ventricular end diastolic volume(LVEDV),wall motion score index(WMSI),left ventricular ejection fraction(LVEF).Results ①Infarct segments in myocardial infarction group showed significant reduction of peak systolic strain as compared with normal group(P

3.Analysis on Utilization of Chinese Patent Medicines in Our Hospital During 2005~2009

OBJECTIVE: To analyze the utilization of Chinese patent medicines in our hospital to provide scientific basis of rational use of drugs in clinic.METHODS: Data about annual consumption sum and consumption amount of Chinese patent medicines in our hospital during 2005~2009 were collected to analyze annual consumption sum,DDDs end daily cost of Chinese patent medicine statistically.RESULTS: The annual consumption sum of Chinese patent medicines showed a upward trend in our hospital during 2005~2009.The top 15 Chinese patent medicine in the list of consumption sum which took up a large proportion affected the total consumption sum.Injection was used more and more so that the ratio of its consumption sum to rank of DDDs was lower than one.CONCLUSION: Cheap and effective Chinese patent medicines play an important part in the clinic.DDDs order should be emphasized in the drug supervision and Chinese traditional medicines injection should be managed strictly.
4.The impact of oncology nurses' compassion fatigue on doctor-patient relationship satisfaction

Objective To examine the impact of oncology nurses' compassion satisfaction and compassion fatigue on doctor-patient relationship satisfaction.Methods Questionnaire survey was conducted by 388 oncology nurses,the questionnaires include Professional Quality of Life Scale and DoctorPatient Relationship Satisfaction Scale.Results The total scores of compassion satisfaction was 34.22±7.19,the total scores of secondary traumatic was 26.32±6.12,the total scores of job burnout was 25.13±6.08,the total scores of doctor-patient relationship satisfaction was 4.24±0.75.Pearson correlation analysis showed that doctor-patient relationship satisfaction score was positively correlated with oncology nurses' compassion satisfaction (r=0.468,P < 0.05),negatioely correlated with nurses' compassion fatigue factor scores (r=-0.182,-0.531).Regression analysis showed that compassion satisfaction,secondary traumatic,job burnout and night shift frequency were influencing factors of doctor-patient relationship satisfaction,which could explain29.2% of the variance.Conclusions Improvement of the oncology nurses' compassion satisfaction and compassion fatigue can improve the doctor-patient relationship satisfaction and the quality of nursing service,and create a harmonious medical environment.
5.Effects of KLF2 on ox-LDL induced expression of microRNA-146a and proinflammatory cytokines in human umbilical vein endothelial cells

Objective To investigate the effects of Kr üppel-like factor 2 ( KLF2 ) on oxidized low density lipoprotein (ox-LDL) induced expression of microRNA-146a (miR-146a) and proinflammatory cyto-kines (MCP-1 and IL-6) in human umbilical vein endothelial cells (HUVECs).Methods Human umbili-cal vein endothelial cells (HUVECs) were cultured and then stimulated with 50μg/ml ox-LDL for 24 hours. HUVECs were infected with adenovirus vectors over-expressing human KLF2 at an appropriate multiplicity of infection.KLF2-siRNA duplexes were transfected into HUVECs to silence the gene expression .HUVECs were collected at time points of 0 h, 3 h, 6 h, 12 h, 24 h and 48 h after infection.Real-time quantitative-PCR was performed to measure the expression of miR-146a at mRNA level.Silenced endogenous miR-146a using LNA-anti-miR-146 a was transfected into HUVECs with lipofectamine 2000 .The levels of MCP-1 and IL-6 in supernatants were detected by ELISA .Results KLF2 remarkably inhibited the expression of miR-146 a in unstimulated and ox-LDL-stimulated HUVECs in a time-dependent manner .The ox-LDL induced ex-pression of miR-146a, MCP-1 and IL-6 in HUVECs were significantly decreased by KLF2.Silenced expres-sion of miR-146a downr-egulated the ox -LDL induced expression of MCP-1 and IL-6 in HUVECs.Moreover, silenced miR-146 a could partly reverse the inhibitory effects of KLF 2 on ox-LDL induced expression of MCP-1 and IL-6 in HUVECs.Conclusion KLF2 inhibited ox-LDL induced expression of MCP-1 and IL-6 in HUVECs partly through down-regulating the expression of miR-146a.
6.Prokaryotic expression and purification of Spink12,a mouse epididymal secretary protein

Objective To study the effect of immunocontraception of Spink12,the fusion protein 6?His-Spink12 was induced to express in E.coli BL21,and purified.MethodsThe total mouse epididymal RNA was extracted,and the Spink 12 protein coding exon without signal peptide sequence was amplified by RT-PCR,digested by restrictive enzyme Nde Ⅰ and EcoR Ⅰ,and then cloned into the prokaryotic expression vector pET28 (a) which contained the 6?His tag in N terminal.The recombinant plasmid pET28a-Spink12,after being confirmed by enzyme digestion and sequencing,was transformed into the competent cells of E.coli BL21.The fusion protein 6?His-Spink12 was expressed by IPTG induction and analyzed by SDS-PAGE and Western blotting.The products were purified by Ni-NTA under native condition with different imidazole gradients and characterized by Western blotting with anti-His monoclonal antibody.ResultsThe results of digestion and sequencing indicated that the recombinant prokaryotic expression vector pET28a-Spink12 was successfully constructed.After E.coli BL21 was transformed with pET28a-Spink12 and induced with IPTG,the recombinant target protein of about 9 kDa was obtained by SDS-PAGE analysis,which was coincident with the theoretical value.The fusion protein was of 23.5% of the total E.coli cell proteins.The Western blotting analysis with anti-His monoclonal antibody showed that the band was fusion protein 6?His-Spink12.After purified with Ni-NTA,a higher quality of fusion protein 6?His-Spink12 was obtained in a purity of 91.1%.ConclusionsThe fusion protein 6?His-Spink12 has been successfully expressed in E.coli BL21 and purified.It may provide a base for the study of the effect of Spink12 on immunocontraception.

Objective To study the chemical constituents of Paeonia veitchii. Methods Isolation and purification were carried out on repeated silica gel column chromatography. The structures of the compounds were identified by physico-chemical properties and spectral analyses. Results Nine compounds were isolated and identified as paeoniflorin (Ⅰ), hydroxypaeoniflorin (Ⅱ), benzoylpaeoniflorin (Ⅲ), benzoylhydroxypaeoniflorin (Ⅳ), albiflorin (Ⅴ), paeonisothujone (Ⅵ), mudanpinoic acid A (Ⅶ), 2-hydroxybenzyl alcohol (Ⅷ), bis (2-hydroxybenzyl) ether (Ⅸ). Conclusion Compound Ⅸ is obtained from natural products for the first time. Compounds Ⅳ, Ⅵ-Ⅸ are isolated from this plant for the first time.
8.Continuous resting energy expenditure measurement in ventilated critically ill children

Objective To monitor the changes of resting energy expenditure in ventilated critically ill children,to compare the results of standard equations and indirect calorimetry (IC) in predicting energy expenditure,and to investigate the possible influence factors of the metabolic status of the critically ill children.Methods From September 2012 to September 2013,56 critically ill children on assisted ventilation and fitting the requirements of IC in pediatric intensive care unit of Shanghai Children's Medical Center were enrolled in this prospective study.IC measurements were performed using metabolic cart on day 1,4,7,10 after trachea intubation.General clinical data of these children were recorded.Results 130 IC measurements were performed in the 56 children.The measured resting energy expenditure (MREE) did not exhibit significant differences among day 1,4,7,and 10 (P =0.379).Although there were no significant differences between MREE and energy expenditure predicted with Schofield and WHO equations (P =0.917,P =0.995),the agreement was poor between the measured and predicted values (R2 =0.185,R2 =0.322).The metabolic status of the children on day 1 of ventilation was only correlated with age (P =0.000) and height (P =0.027),not with severity of underlying diseases or clinical outcomes.Conclusions MREE of IC method in ventilated critically ill children did not significantly change over time in this study.A poor agreement was observed between equationpredicted energy expenditure and MREE.IC measurement of resting energy expenditure is recommended for guiding individual nutritional support among critically ill children so as to improve clinical outcome.
9.Effect of lentinan polysaccharides on hippocampus in Alzheimer 's disease rats

Objective To evaluate the effect of lentinan polysaccharide (LPS) on Alzheimer's disease (AD) rats, and providing a new idea for prevention and treatment of Alzheimer's disease (AD).Methods 96 SPF rats were randomly divided into control group, model group, LPS low dose-treated group, LPS middle dose-treated group, LPS high dose-treated group and positive control group(piracetam treated), each had 16 rats.The model of AD was induced by injection of Aβ25-35(80 pmol/μL), rats in four treatment groups were treated with LPS (200 mg/kg), LPS (400 mg/kg), LPS (600 mg/kg) and piracetam(600 mg/kg) orally once a day for 60 days while control group and model group were given equal-dose saline.After treatment, the ability of navigation and memory were evaluated by water maze test (MWM), and then rats in all groups were sacrificed to obtain the hippocampus.HE staining was used to observe the histopathological changes, the expressions of TACE,Aβ,BACE1 and PP2A protein in hippocampus were detected by Western blot.Results Compared with the model group, high dose LPS could significantly improve the ability of learning and memory (P<0.01) and improve the ability of locating navigation (P<0.01).HE staining results showed that LPS treatment could repair the rat hippocampus injury induced by Aβ25-35 .Western blot analysis showed that the levels of Aβ, and BACE1 were down-regulated in LPS group compared with the model group, while the protein level of PP2A and TACE was up-regulated.Conclusion LPS has a protective effect on Aβ25 -35 induced AD model, and it is expected to be a strategy for treatment of AD .
10.LC-MS/MS assay of methylphenidate: stability and pharmacokinetics in human.

The study aims to solve the instability problem of methylphenidate (MPH) in plasma, and establish a LC-MS/MS method for simultaneous determining of MPH in human plasma. The stabilities of MPH in different media were studied, and the degradation characteristics of MPH in these media were also investigated by HPLC and LC-MS/MS. To a 200 microL aliquot of freshly collected plasma sample, 10 microL 2% formic acid was added immediately to prevent the hydrolysis of MPH in human plasma samples. Chromatographic separation was performed on a Sapphire C18 column using the mobile phase of methanol - 5 mmol.L-1 ammonium acetate buffer solution containing 0.1% formic acid (46 : 54). MPH was quantified by tandem mass spectrometry operating in positive electrospray ionization mode with multiple reaction monitoring. The detection used the transitions of protonated molecules at m/z 234.2-->84.1 for MPH and m/z 260.3-->183.1 for propranolol (IS), separately. The intra- and inter-assay precisions were all below 5.0%. The accuracies were all in standard ranges. The linear calibration curve was obtained in the concentration range of 0.035-40 ng.mL-1. The methods fulfilled the demand. The method was used to determine the concentration of MPH in human plasma after a single dose of 36 mg MPH tablet to 6 healthy Chinese volunteers. The method is suitable for the precisely determination of MPH and for pharmacokinetic study of MPH in human plasma.
