Chinese Journal of Diabetes 2006;14(3):224-226
Objective To investigate the effects of fluvastatin on expression of connective tissue growth factor (CTGF)in the myocardium of diabetic rats. Methods Thirty-two Sprague-Dawley rats were randomized into four groups: normal control, untreated and STZ-induced diabetic rats, and diabetic rats treated with low-dose and high-dose fluvastatin. After 12 weeks′ intervention, body weight as well as the weights of both whole heart and its left ventricle were measured to calculate the ratio of heart weight to body weight (H/B) and the left ventricle mass index (LVMI).The mRNA expression of CTGF, transforming growth factor β1 (TGF-β1), fibronectin (FN),collagen type Ⅲ (ColⅢ) in the myocardium were detected by RT-PCR. Results Compared with the normal control group, the untreated diabetic group showed the increased (all P<0.05) H/B, LVMI and mRNA expressions of CTGF, TGF-β1, FN, and Col Ⅲ in the myocardium. In addition to the effective regulation of lipid metabolism, fluvastatin treatment could obviously reduce the increment of H/B and LVMI(P<0.05~0.01), and suppress the mRNA expressions of CTGF, TGF-β1, FN, and Col Ⅲ in the myocardium of diabetic rats(P<0.01).The down-regulation of CTGF was more significant than that of TGF-β1.All these effects were in dose dependent way. Conclusions Fluvastatin inhibits extracellular matrix accumulation in the myocardium of diabetic rat.Down-regulating the overexpression of CTGF in diabetics is an underlying mechanism of fluvastatin in the cardiac protection.

Objective:To explore the clinical characteristics of patients with pituitary thyrotropin-secreting adenoma and evaluate the effect of preoperative short-acting octreotide treatment on hyperthyroidism. Methods:A retrospective analysis was performed in 40 patients with pituitary thyrotropin adenoma diagnosed in Beijing Tiantan Hospital from January 2008 to January 2018. The general data, laboratory examinations and imaging findings were reviewed and analyzed. The clinical effect of preoperative octreotide on hyperthyroidism was evaluated. Results:The age of onset year of the 40 patients (male: female = 24∶16) was (30.5±5.1) years. Among them, 35 patients (87.5%) were with macroadenoma. The most common symptoms were thyroid hypermetabolism syndrome, followed by headache, dizziness, visual field damage and hypogonadism. The thyroid function of 30 patients (75%) recovered to normal within 3-5 days after the octreotide treatment. The total effective rate of the octreotide was 90.0%. The level of free thyroxine (FT 4) before treatment in patients with more than 10 times of effective cumulative dose was significantly higher than that in patients with less than 10 doses. Conclusions:Thyroid hypermetabolism syndrome and pituitary occupying effect are the most common clinical manifestations of thyrotropin-secreting adenoma. Preoperative octreotide treatment can effectively control hyperthyroidism. The level of FT 4 is a crucial factor affecting the efficacy of octreotide.

Objective To investigate the effect of nitric oxide synthase(NOS) inhibitors and L-arginine(L-Arg) on the prognosis of traumatic shock in rats. Methods Traumatic shock models of Sprague-Daulay rats were made and randomly devided into control group (n=24),L-NAME treatment group (n=24),AG treatment group (n=24) and L-Arg treatment group (n=24). Serum nitric oxide(NO) levels and oxygen partial pressure in tissues include skeletal muscles, liver and small intestine were detected at 1h,3h,5h after resuscitation. Meanwhile, hemodynamic data of the rats and their survival rates of 12h and 24h were monitored and recorded. Results Serum NO concentration was statistically lower after resuscitation in L-NAME group than that in control group, while there were no statistical significance of tissues oxygen partial pressure and survival rate in 12、24h between the two groups. AG could decrease serum NO levels only at late stage of traumatic shock, but no effect on the synthesis and relase of NO at early stage of traumati shock.AG could improve tissues oxygen partial pressure of the liver and small intestine, and prolonged the mean survival time. L-Arg could increase serum NO levels, and improve oxygen partial pressure of intestine and significantly increase the survival rate at 12h and 24h in rats with traumatic shock. Conclusions Treatment with AG and L-Arg can improve the prognosis of traumatic shock rats much better than that with L-NAME.
