1.Extraction of 5 Volatile Oils from Rutongxiaojie Granule and Preparation of ?-cyclodextrin Inclusion Compound

OBJECTIVE: To optimize the extraction condition of volatile oil from five medicinal materials in Rutongxiaojie granules and to optimize the inclusion process of the extracted volatile oil. METHODS: The extraction condition was evaluated by orthogonal test with the recovery of volatile oil as indicator; and the inclusion condition was evaluated with the comprehensive scores of recovery ratio of inclusion compound, the content of volatile oil in inclusion compound and the utilization ratio of the volatile oil as criteria. RESULTS: The optimum extraction condition was established as follows: the medicinal materials were soaked in 10 times volume of water for 1hour and distilled for 9h by steam distillation. The optimum inclusion condition was as follows: the proportion of volatile oil to ?-CD was 1∶8; the inclusion temperature was 30℃ and including time was 20min. CONCLUSION: Both the extraction condition and the inclusion condition were reasonable and feasible.
2.Differentiation efficiency of human umbilical cord mesenchymal stem cells into hepatocytes under two kinds of liver homogenate supernatants: a comparative study

BACKGROUND:Previous studies have demonstrated that normal rat liver homogenate supernatant can induce human umbilical cord mesenchymal stem cels to differentiate into hepatocyte-like cels with partial hepatocyte functions. However, whether fibrotic liver homogenate supernatant can work or how the inducing effect is remains unclear. OBJECTIVE:To investigate the differentiation potential of human umbilical cord mesenchymal stem cels into hepatocytes under the normal liver and fibrotic liver microenvironment in vitro. METHODS:Liver fibrosis was induced in the SD rats by repeated intraperitoneal injections of 3% thioacetamide at a dose of 200 mg/kg body mass, twice a week for 4 weeks, and then fibrotic liver tissues and normal liver tissues were used to prepare liver homogenate supernatants. Passage 3 human umbilical cord mesenchymal stem cels were used and divided into standard control group (cels were cultured in DMEM/F12 with 10% fetal bovine serum), fibrotic liver homogenate supernatants group (cels were cultured in DMEM/F12 with 10% fetal bovine serum and 50 g/L fibrotic liver homogenate supernatants), normal liver homogenate supernatants group (cels were cultured in DMEM/F12 with 10% fetal bovine serum and 100 g/L normal liver homogenate supernatants). The morphological changes of the cels in each group were recorded under inverted microscope; the protein levels of CK18, AFP, CYP3A4, CYP2E1, CYP2D6 and TPH2 were evaluated using western blot assay. Furthermore, the concentration of albumin in the cels was measured. RESULTS AND CONCLUSION:After a 7-day inducement, the stem cels in liver homogenate supernatants groups lost their fusiform shape and changed into hepatocyte-like cels with the morphous of round shape. Compared with the standard control group, the hepatocyte-like cels in the two liver homogenate supernatants groups exhibited human hepatocyte biomarkers, CK18 and AFP. The standard control group cels could express a little amount of CYP2E1, while cels in the two liver homogenate supernatants groups could express CYP3A4, CYP2E1, CYP2D6, TPH2. Compared with the standard control group, the expression level of CYP2E1 in the two liver homogenate supernatants groups increased significantly (P < 0.01), and however, the relative levels of CYP3A4, CYP2E1, CYP2D6, TPH2 in the two liver homogenate supernatants groups showed no statistical significance (P > 0.05). At the same time, compared with the standard control group, the concentration of albumin in the two liver homogenate supernatants groups markedly increased (P < 0.01), but there was no difference between the two liver homogenate supernatants groups (P > 0.05). Experimental findings demonstrated that both of normal liver tissue and fibrotic liver tissue microenvironments could induce human umbilical cord mesenchymal stem cels to differentiate into hepatocyte-like cels. To achieve the same effect, compared with normal liver tissue, fibrotic liver tissue required lower concentrations, suggesting that fibrotic liver tissue microenvironment may be more conducive to differentiation of umbilical cord mesenchymal stem cels into hepatocytes.
3.Development of luminescent oxygen channeling immunoassay for pregnancy associated plasm a protein A

Objective:To develop a luminescent oxygen channeling immunoassay for pregnancy associated plasm a protein A.Methods:The monoclonal antibody of PAPP-A was labeled with biotin ,the polyclonal antibody of PAPP-A was coated on receptor particles.The LOCI reagents also contained sensitizer particles coated with streptavidin.The optimal test conditions and analytical per-formance of the method were studied.Results:The within-run and the between-run coefficients of variation were 5.91%-7.94% and 6.14%-9.69%,respectively;the analytical sensitivity was 2.8 mU/L and the function sensitivity was 4.6 mU/L,good linear in 2.8 mU/L-8 000 mU/L range;the recovery rate was 96.7%-100.3%.The interference rate of hemolysis , icterus and triglycerides were less than 10%; there is no Hook effect of PAPP-A concentrations up to 8 000 mU/L;the correlation coefficient between clinical samples detection results and Time resolved fluoroimmunoassay analysis results was 0.974.Conclusion:This LOCI can be used for the quantitative of serum PAPP-A,and detection performance in line with the requirements of clinical diagnostic reagents .
4.Resting state functional magnetic resonance imaging in patients with first-episode major depression disorder

Objective To explore the abnormally functional brain region in resting state in first-episode major depression disorder patients with function magnetic resonance imaging.Methods 51 patients diagnosed with first-episode major depression disorder according to DSM-Ⅳ and 50 gender-,age-,and education-matched healthy controls completed resting state fMRI scan.The severity of depression,and unpredicted homodynamic responses across the whole brain were analyzed using Hamilton depression scale,regional homogeneity and amplitude of low-frequency fluctuation,respectively.Results Compared with control group,the right medial frontal gyms (BA6,MNI:3,-3,63,K=34) and left medial frontal gyrus(BA9,MNI:-9,36,30,K=10) (P<0.001,uncorrected) in the case group showed higher regional homogeneity,with statistical significance.Compared with control group,right medial frontal gyrus (BA6,MNI:3,-3,63,K =35) and right posterior cingulated gyrus (BA31,MNI:3,-36,36,K =11) (P< 0.001,uncorrected)in the case group showed higher amplitude of low-frequency fluctuation,with statistical significance.Conclusion First-episode major depression disorder patients in resting state had several abnormally functional brain regions,which might be related to the pathological mechanism of depression disorder.

6.Effect of Baogan Ning on Hepatic Leptin Receptor and Its JAK2/STAT3 Message Pathway in Rats with Hepatic Fibrosis

Objective To explore the anti-hepatic-fibrosis mechanism of Baogan Ning(BN).Methods Fifty Wistar rats were randomized into 5 groups: normal group,model group,high-and low-dosage BN groups(35.4 and 17.7 g/kg respectively),and colchicine group(0.11mg/kg).Rat models of hepatic fibrosis were induced by multiplex factors.Hepatic protein was extracted,and the expression of hepatic leptic receptors of OB-Rb,JAK2 and STAT3 was detected with Western Blot method.Results The protein expression was not obvious in the normal group,and the color of protein band in the medication groups was light but dark in the model group.Compared with the normal group,the expression of OB-Rb,JAK2 and STAT3 increased in the model group.However,the expression of OB-Rb,JAK2 and STAT3 decreased in the medication groups as compared with the model group.And the expression of OB-Rb,JAK2 and STAT3 decreased in the BN groups as compared with the colchicines group.Conclusion The anti-hepatic-fibrosis mechanism of Baogan Ning is probably related with the inhibition of OB-Rb expression,thus inhibit the JAK2/STAT3 message pathway.
7.Hemocyanin is an important high-molecular-weight allergen of Eriocheir sinensis

Objective:To prove hemocyanin is an important high-molecular-weight allergen from Eriocheir sinensis.Methods:The proteins were extracted from Eriocheir sinensis tissue with lysate extraction method .The protein components were analyzed by SDS-PAGE,two-dimensional electrophoresis and Western blot.Serum IgE from allergic donors identified a candidate protein,which was char-acterized by MALDI-TOF/TOF.The immune property of the candidate protein was tested using Dot-blot.Results: By SDS-PAGE and two-dimensional electrophoresis analysis,we prove that the native protein components were completely.According to the Western blot result,at least 18 components could react with the positive serum.Among them, two 70-80 kD proteins had the same isoelectric point.So,may be they were the same protein.MALDI-TOF/TOF analysis results showed that the suspected proteins were hemocyanin.A sensitization frequency of 61%was observed in Eriocheir sinensis patients by Dot-blot.Conclusion:Hemocyanin was identified as an important high-molecular-weight allergen from Eriocheir sinensis.
8.Effect of Chinese medicine Baoganning on lipid and serum leptin of liver fibrosis model rats

Objective:To discuss the mechanism of Baoganning anti-liver fibrosis.Methods:liver fibrosis rat models were established by complex factors.Automatic biochemical analyzer was used to detect TC、TG and RIA isotope analysis was used to detect serum leptin.To observe the influence of Baoganning on TC、TG and leptin level of liver fibrosis model rats. Results:Compared with the normal group,levels of serum TC,TG decreased apparently in model and every drug groups(P0.05).Conclusion:Baoganning can effectively prevent and treat liver fibrosis in rats,which may be closely related to the improvement of liver function,adjustment of lipid and reduction of serum leptin.
9.Construction of expression plasmid in human WAF1 gene

Objective :Humna WAF1 gene was cloned,and WAF1-pcDNA3 recombinant plasmid was con-structed. Methods :Human WAF1 gene was amplified by RT-PCR from Hela cell,then cloned into pcDNA3vector. Results :Human WAF1-pcDNA3 recombinant plasmid was constructed,and DNA direct sequencingindicated that was correct. Conclusion:Our study laid the foundation for studying WAF1 gene expressionand it's biological effects.
10.Effects of Dioscornin on levels of adiponectin, tumour necrosis factor alpha and S100B in serum of elderly patients with rheumatoid arthritis and cerebral infarct

Objective To investigate the change of adiponectin (AD),tumor necrosis factor-alpha (TNF-α) and S100 levels in serum elderly patients with rheumatoid arthritis and cerebral infarct in order to evaluate the therapeutic effect of dioscornin.Methods One hundred patients with rheumatoid arthritis and cerebral infarct were selected as our subjects,who were hospitalized in the Department of Neurology of Recovery Changzhi Municipal People's Hospital and the Department of Internal Medicine of the Affiliated Hospital of Shaoxing Medical College,between 2006 September and 2010 September.All subjects (male 55,female 45,average age of 57 years old) were randomly divided into regular and dioscomin groups,50 for per group.Patients in regular group were treated with routine therapy and patients in dioscornin groups were treated with dioscornin 80mg,three daily plus regular treatment drug.Meanwhile 40 middle patients with single rheumatoid arthritis subjects were severed as controls.The changes of BMI,fasting plasma glucose,lipid factors,insulin sensitivity index (ISI),serum adiponectin,TNF-α and serum S100B were determined at treatment before and 6 months after treatments.Results The level of TNF-α,serum S100 in ERA patients were significantly higher andAdiponectin significantly lower than that of the control group,(TNF-α:[(89.0 ± 25.3) ng/L,(88.0 ± 24.2)ng/L vs(74.0 ±21.0) ng/L,F =3.292,P <0.05],[S100B:(0.102 ±0.051) μg/L,(0.101 ±0.045) μg/L vs(0.092 ± 0.031) μg/L,F =2.792,P < 0.05],and AD and BMI were lower [AD:(7.2 ± 1.4) μg/L,(7.3 ±1.4) μg/L vs (18.1 ± 3.5) μg/L,F =17.057,P < 0.01],[BMI:(18.9 ± 2.4) kg/m2,(19.0 ± 1.9) kg/m2 vs (21.8 ± 1.8) kg/m2,F =6.147,P < 0.01].There was a negative correlation between adiponect and TNF-α,S100B (r =-0.46,-0.52,P < 0.01) and positive correlation between adiponect and BMI (r =0.44,P <0.01).The adiponectin level was significantly increased in patients for six months after dioscornin treatment than that of control group.[AD:(12.2±2.9) μg/L,(7.8 ±1.8) μg/L vs (18.0 ±4.3) μg/L,F=6.480,P<0.01].The level of TNF-α and S100B significantly decreased than that of the control group,TNF-α:[(72.0 ±21.0) ng/L,(82.0±23.0)ng/L vs (68.0 ±20.0) ng/L,F =3.065,P <0.05],[S100B:(0.092 ±0.021)μg/L,(0.099 ±0.031) μg/L vs (0.091 ±0.029) μg/L,F=3.030,P<0.05].Conclusion Dioscornin could ameliorate the prognosis through decreasing the levels of TNF-α and S100B,and increasing adiponectin level in patients with rheumatoid arthritis and cerebral infarct.
