2.Effect of Herpes Simplex Virus-2 Infection on the HLA Class II Antigen Expression of Human Monocytes

Objective To investigate the role of HLA class II antigen of human monocytes in pathogenesis of herpes simplex virus-2 (HSV-2) infection. Methods Human monocytes were infected by HSV-2 (333 Strain). The cultured cells were collected at the first, third, 5th, and 7th days of post-infection. The expression of HLA class II antigen of the infected monocytes was measured by alkaline phosphotase anti-alkaline phosphotase method (APAAP). Results The expression of HLA class II antigen on the monocytes significantly decreased at the first day of post-infection,and then gradually increased and reached to the normal control level at the 7th day of post-infection. Conclusion HLA class Ⅱ antigen expression of monocytes was inhibited by HSV-2 infection at early stage, which may be one of means of virus escaping host immunity. The expression of HLA class Ⅱ antigen may play an important role in HSV-2 pathogenicity and immunity.
3.The Induction of Cytokine-induced Killer and its Cytotoxic Effect on Hepatic Carcinoma Cells

Objective To prepare cytokine-induced killer(CIK) in vitro, and study the biological activities of CIK. Methods Lymphocytes were isolated from peripheral blood and umbilical blood, and induced by cytokine.The cell surface markers CD_3 and CD_~56 of CIK were analyzed by FACS. The proliferation of CIK was tested using 3H -TdR release method, and the cytotoxic effect of CIK on hepatic carcinoma cells was measured by MTT. Results CIK cells quickly proliferated from the second week, and expanded more than 60-fold at the 31th day after induction. The CD_3+ and CD_~56 + cells expanded more than 800 folds. The cytotoxic effect of CIK on hepatic carcinoma cells was obviously stronger than that of LAK cells, and CIK had not obvious cytotoxic effect on fetal liver cells. Conclusion CIK was a highly efficient cytotoxic cells against tumors, and had clinical application potentials.
4.Expression of Insulin-like Growth Factor 2 and Ki-67 in Hemangioma and Vascular Malformation

Objective To study the role of insulin-like growth factor 2 (IGF- Ⅱ ) in the proliferation and degeneration of hemangiomas. Methods The expression of IGF- Ⅱ and Ki-67 was detected with SP im-munohistochemical method in 52 specimens of hemangiomas, 27 of vascular malformation and 5 of normal skin. The expression of IGF-Ⅱ mRNA was examined by reverse transcription polymerase chain reaction (RT-PCR) in specimens from 9 cases of hemangiomas and 5 of vascular malformation. This paper studies tile regulation of cytokines such as human tumor necrosis(Hu-TNF) and recombinant human interferon - (rHu -IFN - ) on activity of human polymorphonuclear neutrophils (PMN) against Candida albicans by determing numbers of colony - forming unit of Candida albicans. We detected that Hu - TNF and rHu IFN st did not interfere directly with fungal growth. These two cytokines enhanced PMN to inhibit Candida albicans growth. When preincubating PMN with Hu --TNF and rHu-IFN -togather, the activity of PMN against Candidaalbicans was synergically enhanced. The degree of enhancement of the activity of PMN againstCandida albicans was highly dependent on these two cytokines - PMN preincubation time. Thus,Hu -- TNF and rHu -- IFN -- have the ability to activate PMN, and the synergistic action of thetwo cytokines may prove clinically effective for increasing the ability of immuncompromised hosts against opportunistic fungal infectons.
6.Study of the Levels of TNF ?,IL 6,IL 8 in Patients with Condyloma Acuminatum

Objective To study the role of tumor necrosis factor alpha(TNF ?), interleukin(IL) 6 and IL 8 in the pathogenesis of condyloma acuminatum(CA). Methods Double antibody sandwich ELISA was used to study the levels of TNF ?,IL 6 and IL 8 in 21 cases of CA. Results The results showed that the levels of TNF ?,IL 6 and IL 8 of the plasma in the patients were significantly lower than those of normal controls. The levels of TNF ?,IL 6 and IL 8 produced by peripheral blood mononuclear cells in response to LPS mitogen in vitro significantly decreased in comparison with those of normal controls. The levels of IL 6 and IL 8 of the wart tissues excised from 17 patients were significantly lower than those of normal tissue. No TNF ? was detected in the tissues of both patient and normal groups. Conclusion These findings suggest that the monocyte macrophage function of CA patients might be decreased systemically and locally which may contribute to the disorder of lymphocyte function. Improvement of the monocyte macrocyte function might have some significant effect on the prevention and treatment of CA.
7.Cone-shaped Moxa Moxibustion Treating Herpes Zoster and Its Mechanism

The therapeutic effect and mechanism of moxibustion in the treatment of Herpes Zoster were studied. It was found that the level of T lymphocyte subsets of the patients with Herpes Zoster was within the normal range before treatment, but slightly lower than in the health group. Th cells and NK cells were significantly decreased in the patients with Herpes Zoster as compared with those in the health group (P<0. 05),suggesting the cellular immunity of the patients with Herpes Zoster was decreased. After treatment with moxibustion, the level of all the T lymphocyte subsets was increased, close to the normal. Among them the levels of Th cells and NK cells were significantly different from those before the treatment (P<0. 05). The disappearance of exanthema and relief of neuralgia in the patients was also quicker than in the controls (P<0. 01). It was indicated that moxibustion had a good effect in the treatment of Herpes Zoster, which was contributed to the enhancement of the body antiviral ability by the improvement of immunity, especially the cellular immunity.
8.Secretion of Tumor Necrotic Factor of Peripheral Blood Leukocytes from Patients with Condylomata Acuminata: An In Vitro Study

Objective To investigate the correlation between the relapse of condyloma acuminatum(CA)and the potential capability of tumor necrotic factor (TNF) production of the host′s peripheral blood leukocytes. Methods Forty-two CA patients and 58 normal controls were enrolled in this study. CA relapse was diagnosed clinically. EB virus-transformed B lymphoblastoid cell line(LCL)were used as TNF producing cells. The TNF producing capability of LCL was measured by bioassay using L929 (a TNF sensitive tumor cell line) as target cells. The LCL were stimulated with LPS to produce TNF. Results The average level of TNF production of LCL from all CA patients (including recurrent and non-recurrent CA patients) was similar to that of normal controls (30.14% ? 12.27 vs 34.06% ? 12.06,P = 0.1136). However, the level of TNF production of LCs from recurrent CA patients was significantly less than that from non-recurrent CA patients (24.75% ? 7.51 vs 36.62% ? 10.96,P = 0.00016). Compared with that of normal controls, recurrent CA patients showed a lower capability to produce TNF (24.75% ? 7.51 vs 34.06% ? 12.06,P = 0.00054), whereas non-recurrent CA patients showed a similar capability to normal controls (36.62% ? 10.96 vs 34.06% ? 12.06,P = 0.3517). Conclusions These results indicate that the cellular immune mechanism might play an important role in the clearance of the residual HPV from the host, in which TNF is involved.
9.Study on the application of high-frequency ultrasound to diagnosis of early rheumatoid arthritis

Objective To study the application value of high frequency ultrasound to the diagnosis of early rheumatoid arthri‐tis .Methods 40 cases were selected ,who were diagnosed with early rheumatoid arthritis ,as the RA group ,another 40 healthy per‐sons were selected as the normal control group .Objects in the 2 groups were examined with high frequency ultrasound and X‐ray . Comparison of the examination results was made ,at the same time ,the results of suprapatellar bursa effusion ,synovial hyperplasia and bone erosion of the positive detection rate in group RA from the analysis of high‐frequency ultrasound and X‐ray examination were also compared .In addition ,the relativity between the related index and ESR index and CRP index in group RA was also ana‐lysed .Results The suprapatellar bursa effusion ,synovial hyperplasia ,bone erosion of color flow rate and bone erosion of the posi‐tive detection rate were higher than those in normal control group(P<0 .05);Compared with the control group ,the artery resist‐ance index was less ,the femoral condyle and lateral condylar cartilage thickness of RA group increased more(P<0 .05) .High fre‐quency ultrasound positive detection rate suprapatellar bursa effusion ,synovial hyperplasia ,bone erosion in patients of group RA were higher than those of X‐ray(P<0 .05) .The knee joint suprapatellar bursa effusion ,synovial hyperplasia of synovial membrane thickenss ,the thickness of the color blood flow grade and CRP ,ESR of patients in RA group were correlated(P<0 .01) .Conclusion High frequency ultrasound can respond to the situation of early rheumatoid arthritis patients ,and the lesion detection rates are higher ,there is a certain correlation between ultrasonographic indexes and CRP ,ESR .
10.A comparative study on the effect of BCG-PSN and thymopeptides on T-lymphocyte subsets of normal and immunosuppressed mice.

To compare the effects of polysaccharide nucleic acid fraction of bacillus calmette guerin (BCG-PSN) and thymopeptides on T-lymphocytes of normal and immunosuppressed mice, CD4+ and CD8+ T-lymphocyte subsets of single nucleic cell in thymus, spleen and peripheral blood were detected successively by flow cytometry after application of BCG-PSN and thymopeptides. Meanwhile, CD4+/CD8+ ratio was also calculated. The results showed that both BCG-PSN and thymopeptides could decrease the proportion of CD4+ CD8+ T-lymphocyte subsets in the thymus, at the same time increase CD4+ T-lymphocyte, CD8+ T-lymphocyte proportion in the three tissues. The fluctuation in amplitude was greater in thymopeptides group than that in BCG-PSN group. It is concluded that acting location of thymopeptides is in thymus, its stimulating action is stronger than that of BCG-PSN, while BCG-PSN not only accelerates the differentiation in thymus, but also has some direct stimulation to peripheral CD4+ T-lymphocytes, and can maintain CD4+/CD8+ ratio within normal range. So, BCG-PSN is safer.
