1.Effect of ursolicacid on contractile activity of uterine smooth muscle strips of virginal rats

Objecitve To observe the effects of ursolicacid on contractile activity of uterine smooth muscle strips of virginal rats.Methods 20 Wistar virginal rats,weight 200-300g were selected,to record the effects of ursolicacid(1.0?10~(-8),2.0?10~(-8),3.0?10~(-8),4.0?10~(-8)g/L) on contractile activity of uterine smooth muscle strips with Biolap 410 Biological System.Diphenhydramine(2?10~(-6)mol/L),Phentolamine(2?10~(-6)mol/L),Verapamil(2?10~(-7)mol/L),Indomethacin(2?10~(-5)mol/L) were used as antagonists to study the mechanisms of ursolicacid on uterine smooth muscle strips.Results Ursolicacid could significantly potentiate uterine contractile activity of virginal rats.It could increase the frequencies,prolong the duration and enlarge the amplitude of contractile activity.The effect of which could be inhibited by the Indomethacin;but could not restrained by other antagonists.Conclusion The results of Ursolicacid may be mainly primarily with prostaglandin synthese or release,but not mediated by H_(1) receptor,? receptor and L-voltage-dependant calcium channels.
2.Effect of nutgrass galingale rhizome decotion on contractile activity of uterine smooth muscle strips of non-pregnant rats in vitro

Objective To study the effects of nutgrass galingale rhizome decotion on contractile activity of uterine smooth muscle strips of non-pregnant rats.Methods We selected 60 Wistar non-pregnant rats of weight 250-300 g to record the effects of nutgrass galingale rhizome decotion(1.0 g/L,2.0 g/L,4.0 g/L,8.0 g/L) on contractile activity of uterine smooth muscle strips of non-pregnant rats with Biolap 410 Biological System.Four antagonists,diphenhydramine(2?10-6mol/L),phentolamine(2?10-6mol/L), verapamil(2?10-7mol/L),and indomethacin(2?10-5mol/L) were used to study their mechanisms respectively.Results Water extract of nutgrass galingale rhizome(4?10-3kg/L) could significantly weaken uterine contractile activity of non-pregnant rats,lessen the frequencies,shorten the duration and lower the amplitude of contractile activity.Conclusion The effect of nutgrass galingale rhizome on contractile activity of uterine smooth muscle strips in rats may be mainly associated with prostaglandin synthese or release nothing with L-voltage-dependant calcium channels,H1 receptor or ? receptor.
3.A survey of oral health status of population in Gansu Province

Objective:To explore the oral health status of population in Gansu Province and get scientific information for oral diseases prevention. Methods: An equal-sized stratified multi-stage random sampling design was applied in the investigation. According to standard and method used in the third national oral health epidemiological investigation, the oral health investigation was made in 3 158 people aged 5 years, 12 years, 35-44 years and 65-74 years with a gender ratio of half to half in Gansu. Results: The caries prevalence rate was 55.80%, 19.09%, 61.95% and 78.63% and the caries-filling rate was 3.11%, 10.76%,10.34% and 4.60% in the four groups respectively. Detection rates of gingival bleeding and dental calculus were 82.81%, 93.55%, 85.34% and 69.66%, 99.37%, 92.54% in 12, 35-44 and 65-74 groups respectively. Detection rates of periodontal pocket and diseases of oral mucosa were 60.94%, 62.20% and 6.32%, 7.71% in 35-44 and 65-74 groups respectively. The most denture-prosthesis was removable partial dentures and irregular fixed bridge. Conclusion: The prevalence of caries is serious but better than the average results of the nation in 2005 and the results of Gansu in 1995. The prevalence of periodontal disease is more serious than the results of the nation in 2005. It is essential to strengthen oral health education and oral health service, especially in rural area in Gansu, so as to ensure the oral diseases be treated in time and effectively.
4.Comparison of minimally invasive percutaneous iliosacral screw and reconstruction plate fixation in treating unstable pelvic fractures

ObjectiveTo compare the clinical effect of percutaneous iliosacral screws osteosynthesis (PISO) and open reduction internal reconstruction plate fixation in treating unstable pelvic fractures combined with sacroiliac joint dislocation,and evaluate their safety and practicality.MethodsFrom March 2004 to October 2010,37 patients with vertical unstable pelvic fractures were admitted to our department.Twenty cases were treated with percutaneous sacroiliac screw fixation and 17 cases were performed opened reduction and internal reconstruction plate fixation under C-arm X-ray's guide.The perioperative parameters and postoperative imaging indexes were compared and analyzed.ResultsAll patients were followed up for 6 months to 26 months,with an average of 15 months.There were statistical significances between the PISO group and open reduction internal fixation group in operation time,blood loss,postoperative pain,mean fever time and hospital stay.The two groups showed no significant difference on postoperative X-evaluation of reduction effect.The average healing time was 3.2 months and the difference was not statistically significant between two groups.PISO group had no complications such as infection,bent nails or broken nails.ConclusionThrough compared and analyzed the two groups in treating unstable pelvic fractures,the percutaneous sacroiliac screw fixation has been proved for a kind of ideal minimally invasive surgery method because of locating exactly,less damage and blood loss,milder pain and quicker recovery.But it demands higher operation techniques.Adequate preoperative preparation and postoperative patients' cooperation can reduce complications incidence.The second group of anterior reconstruction plate or T-shape plate to fix vertically unstable pelvic fractures at same time shows a good result of stabilization.
5.A study of association between the interleukin-1 single nucleotide polymorphism and risk of chronic periodontitis among the Hui and Dongxiang minorities in Gansu province.

OBJECTIVETo examine and analyze the interleukin (IL)-1B gene single nucleotide polymorphism (SNP) at positions +3954, and explore the association between SNP and risk of chronic periodontitis (CP) among the Hui and Dongxiang minorities in Gansu province.
METHODSCP group consisted of 215 patients from Hui and Dongxiang and healthy control group consisted of 219 subjects. Anti-coagulated peripheral blood sample was obtained from each subject and genomic DNA was extracted from each sample. SNP at IL-1B+3954 were analyzed by standard polymerase chain reaction-restriction fragment length polymorphism (PCR-RFLP) assay.
RESULTSStatistically significant differences were found in genotype and allele frequencies between CP group and healthy control group. The exist of position alleles C increased the incidence of CP. The trend of frequency distribution of gene polymorphism above in Dongxiang was the same as that of Hui.
CONCLUSIONThe present study reveals that IL-1B+3954C/T genotypes are significantly associated with the risk of CP. These results indicate that genetic polymorphism is significantly associated with the risk of CP among the Hui and Dongxiang minorities in Gansu province.

7.Oral health status and its correlation with oral health knowledge among middle-aged people in Dongxiang, Bonan, and Yugur.

OBJECTIVETo provide basic data for the prevention of oral diseases in minorities by investigating the oral health status and behavior related to oral health knowledge of individuals aged 35 to 44 years in Dongxiang, Bonan, and Yugur.
METHODSThe caries and periodontal health of 445 individuals aged 35 to 44 years were examined according to the method and criterion prescribed by the World Health Organization and the Third National Oral Health Epidemiologic Investigation. A questionnaire survey on related oral health knowledge and behavior was conducted.
RESULTSThe crown caries prevalence rate in Dongxiang, Baoan, and Yugur were 48.28%, 79.47%, and 67.11%, respectively; the root caries prevalence rates were 38.62%, 69.54%, and 42.95%, respectively. The rates of gum bleeding in Dongxiang, Bonan, and Yugur were 86.90%, 90.07%, and 65.77%, respectively. The rates of dental calculus in Dongxiang, Bonan, and Yugur were 99.31%, 100.00%, 99.33%, respectively, and the rates of periodontal bags were 68.97%, 67.55%, and 43.62%, respectively. Only 69.84% of the respondents brush their teeth every day; 94.90% of the respondents do not floss. Only 20.19% of the respondents contact a doctor for a toothache, and 42.23% of the respondents have never seen a dentist.
CONCLUSIONCaries morbidity is high among the respondents aged 35 to 44 years from Dongxiang, Bonan, and Yugur. The periodontal health status and oral hygiene of the respondents are poor, and behavior related to oral health knowledge is insufficient. Thus, more attention must be provided to the prevention and control of caries and periodontal diseases among middle-aged people in the area.

8.Value of highly sensitive nucleic acid detection and aminotransferase level in evaluating precise antiviral therapy for liver cirrhosis patients with a low viral load

ObjectiveTo investigate the clinical significance of highly sensitive nucleic acid detection in precise antiviral therapy for patients with liver cirrhosis and its association with aminotransferase level. MethodsA total of 377 patients with hepatitis B cirrhosis who were hospitalized or attended the outpatient service from May 2013 to April 2019 were enrolled and tested by both domestic HBV DNA detection and highly sensitive Cobas HBV DNA detection. All patients underwent biochemical examination, four blood coagulation tests, routine blood test, and upper abdominal computed tomography or ultrasound. Sensitivity of different HBV DNA detection reagents was compared in liver cirrhosis patients with a low viral load, and the correlation between alanine aminotransferase (ALT) level and viral load was analyzed. The paired t-test was used for comparison of continuous data before and after treatment. The receiver operating characteristic (ROC) curve was used to screen out the optimal predictive values of ALT at different cut-off values of HBV DNA. ResultsAmong the 377 patients with hepatitis B cirrhosis, 215 tested positive and 162 tested negative by domestic HBV DNA, and among these 162 patients, 104 (64.2%) tested positive by Cobas HBV DNA detection, with a mean level of 267.5±42.3 IU/ml. After 24 weeks of antiviral therapy, the 104 patients with hepatitis B cirrhosis had significant improvements in viral replication level, ALT, and Child-Pugh score for liver function; HBV DNA decreased from 267.5±32.2 IU/ml before treatment to 59.6±7.7 IU/ml after treatment (t=3.486, P=0.002), ALT decreased from 871±10.8 U/L before treatment to 36.5±7.6 U/L after treatment (t=3.235, P=0.020), and the Child-Pugh score decreased from 6.5±0.7 before treatment to 5.7±0.5 after treatment (t=2.928, P=0.041). The ROC curve analysis of ALT in predicting HBV DNA decision point showed that an ALT level of 29 IU/L was the most sensitive cut-off value for predicting HBV DNA <20 IU/ml, with an area under the ROC curve of 0.904, a sensitivity of 1.0, and a specificity of 0.237. ConclusionPrecise detection helps to guarantee the precise clinical treatment of patients with hepatitis B cirrhosis and improve their treatment outcome and prognosis. An ALT level of 29 IU/L is a sensitive indicator for predicting patients with negative Cobas HBV DNA, so as to achieve individualized precise screening and treatment.
9.Expression of the transmembrane emp24 domain-containing protein 4 in liver tissues of patients with hepatocellular carcinoma and its effects on biological behavior of hepatocellular carcinoma cells

Objective:To examine the expression of transmembrane emp24 domain-containing protein 4(TMED4) in liver tissue of patients with hepatocellular carcinoma, and to investigate the effects of TMED4 gene on the proliferation and migration of hepatocellular carcinoma cells and related molecular mechanisms. Methods:The expression of TMED4 at protein level in liver cancer tissue and paracancerous tissue of patients with hepatocellular carcinoma were detected by Western blotting and immunohistochemical stainning, and the correlation between its expression and clinicopathological features was analyzed. The effects of TMED4 overexpression or knockdown on proliferation, migration and healing ability of hepatocellular carcinoma cells in vitro and in vivo were determined by cell proliferation test, Transwell test, wound healing test and subcutaneous tumor formation in nude mice. The molecular mechanism of TMED4 in regulating the biological behavior of hepatocellular carcinoma cells was preliminarily explored by pathway analysis. Independent sample t test, Mann-Whitney U test and chi-square test were used for statistical analysis. Results:The results of Western blotting showed that the expression of TMED4 at protein level in hepatocellular carcinoma tissue was lower than that in paracancerous tissue(0.52±0.29 vs. 0.83±0.22), and the difference was statistically significant( t=2.54, P=0.022). The results of immunohistochemical examination indicated that the expression of TMED4 at protein level in liver cancer tissue was lower than that in paracancerous tissue(5.46±3.37 vs. 7.58±3.08), and the difference was statistically significant( t=3.49, P<0.001). The expression of TMED4 at protein level was significantly correlated with vascular invasion and Barcelona clinic liver cancer stage( χ2=6.83 and 4.20, P=0.009 and 0.040). The results of cell proliferation assay showed that the absorbance value of SMMC-7721 cells in TMED4 overexpression group was lower than that in control group(1.38±0.05 vs. 2.37±0.08), while the optical density value of HepG2 in TMED4 knockdown group was higher than that in control group(0.76±0.04 vs. 0.54±0.01), and the differences were statistically significant( t=18.23 and 8.85, both P<0.001). The results of Transwell test showed that the number of migrated SMMC-7721 cells in TMED4 overexpression group was less than that in control group(286.30±13.01 vs. 439.70±12.34), while the number of migrated HepG2 cells in TMED4 knockdown group was higher than that in control group(249.00±6.00 vs. 160.00±6.56), and the differences were statistically significant( t=14.81 and 17.34, both P<0.001). The wound healing test showed that the healing rate of SMMC-7721 cells in TMED4 overexpression group was lower than that in control group((0.21±0.01)% vs.(0.45±0.01)%), the healing rate of HepG2 cells in TMED4 knockdown group was higher than that in control group((0.46±0.01)% vs.(0.20±0.01)%), and the differences were statistically significant( t=200.10 and 30.46, both P<0.001). The results of subcutaneous tumor formation assay in nude mice showed that the growth rate of cells in TMED4 overexpression group was slower than that in control group. After cell inoculation for 6 weeks, the subcutaneous tumor volume of mice in TMED4 overexpression group was smaller than that in control group(27.36 mm 3(138.70 mm 3) vs. 1 741.62 mm 3(1 783.39 mm 3)), the tumor weight was lower than that in control group(0.06 g(0.14 g) vs. 1.46 g(1.09 g)), and the differences were statistically significant(both Z=-2.31, both P<0.001). The results of Western blotting showed that the expression of Snail at protein level in SMMC-7721 cells of the TMED4 overexpression group was lower than that of the control group(0.32±0.01 vs. 0.90±0.03), the protein level of Snail in HepG2 cells of TMED4 knockdown group was higher than that of control group(1.03±0.01 vs. 0.97±0.01), and the differences were statistically significant( t=28.49 and 12.31, both P<0.001). The results of real time fluorescent quantitative polymerase chain reaction showed that the expression of Snail at mRNA level in SMMC-7721 cells of TMED4 overexpression group was lower than that of control group(0.13±0.05 vs. 1.00±0.15), the expression of Snail at mRNA level in HepG2 cells of TMED4 knockdown group was higher than that of control group(1.25±0.32 vs. 0.21±0.14), and the differences were statistically significant( t=9.62 and 5.10, P<0.001 and P=0.007). Conclusion:TMED4 may affect the proliferation and migration of hepatocarcinoma cells by regulating the expression of Snail, and which is expected to become a potentially therapeutic target for hepatocellular carcinoma.
10.Association between depressive symptoms and risk of coronary heart disease in middle-aged and elderly Chinese community: An empirical analysis based on CHARLS database

【Objective】 To investigate the association of depressive symptoms with the predicted risk of coronary heart disease in middle-aged and elderly Chinese based on a large community study. 【Methods】 A total of 2532 cases in the group without depression and 2758 cases in the group with depression were included. We compared the two groups in general demographics, information related to coronary heart disease risk, and physical function and ability to perform daily living. We also analyzed the factors associated with coronary heart disease risk by linear regression. 【Results】 ① Demographic information: The group with depression had a higher mean age, a higher proportion of women, more people with poor marital status, and a higher number of comorbid chronic diseases compared with the group without depression (all P<0.05). ② Risk indicators related to coronary heart disease: The group with depression had more people with diabetes and a significantly higher systolic blood pressure compared with the group without depression (P<0.05). The two groups did not significantly differ in the proportion of smokers, diastolic blood pressure, LDL-C, or HDL-C (all P>0.05). The risk of coronary heart disease was significantly higher in the group with depression than in the group without depression (P<0.05). ③ Physical function and ability of daily living: The physical function score, physical self-care score, and instrumental daily living ability were significantly higher in the group with depression than in the group without depression (all P<0.001). ④ Linear regression showed that except for gender, age, marital status, comorbid diabetes, smoking, systolic and diastolic blood pressure, HDL-C and LDL-C were associated with risk of coronary heart disease (P<0.05); CESD was the only factor associated with the risk of coronary heart disease [B=0.019, 95% CI: (0.015, 0.032), P=0.032]. 【Conclusion】 The risk of coronary heart disease is higher in middle-aged and elderly people with depressive symptoms than in those without depressive symptoms. Having depressive symptoms is one of the risk factors for coronary heart disease in middle-aged and elderly people.
