OBJECTIVE:To provide references for hospital pharmacy's culture construction. METHODS: To analyze and pondered over the practice of culture construction in pharmacy department of our hospital. RESULTS: With the organizational culture construction, cohesive force and innovative spirit of our staffs had been improved significantly. CONCLUSION: Organizational culture construction is indispensable for the development of hospital pharmacy.
2.Exploration of functional experiment evaluation system to cultivate practical medical talents

Functional experiment plays an important role in basic theory and clinical practice. For training high-quality medical talents in medical colleges,many improvements have been made in the functional experiment teaching and certain effects have been obtained. But a comprehensive evalu-ation system was still needed. Clinical medical students of 2010 grade were taken as the appraisal objects. Assessment content,methods and standards were designed according to the basic requirements of functional experiment. Students were evaluated by ordinary performance,test papers and experiment competition. Practice showed that the system was of significant importance in improving the experi-mental teaching quality and developing students' practical ability and comprehensive quality.
3.Study of the ubiquitination of RNF6 on insulin receptor substrate-1

Objective To construct an eukaryotic expression vetor of ring finger protein 6 (RNF6)and to investigate its ubiqu-itylation on insulin receptor substrate-1(IRS-1).Methods Human RNF6 coding sequence was amplified by polymerase chain reac-tion (PCR)method with human cDNA as template.The pcDNA3.1-CHA-RNF6 was constructed with routine molecular methods and transfected into hepatocarcinoma cell HepG2.Relative quantification of IRS-1 mRNA was preformed by real-time reverse tran-scription PCR.Western blot was applied to detect the expression levels of IRS-1.Results 48 h after the plasmid carring RNF6 gene transfected HepG2 cells,the mRNA level of ISR-1 gene reduced to 69% of control group.The expression of IRS-1 was markedly lower than control group (P <0.01).Conclusion The expression of IRS-1 is markedly down-regulated in HepG2 and enhancement of the ubiquitylation level of IRS-1 contribute to the disorder in insulin signal transferring.
4.Study of NO and FOXP3 in patients with pulmonary tuberculosis

Objective To Study the expressions and significance of nitric oxide (NO) and forkhead box protein 3 (FOXP3) in patients with pulmonary tuberculosis .Methods Serum NO levels were measured by Griess colorimetric reaction .Flow cytometry was used to determine the number of CD4+CD25+ T cells in peripheral blood .The expression of FOXP3 mRNA was measured by real‐time polymerase chain reaction .Results Serum NO level in patients was (15 .71 ± 1 .26)μmol/L ,higher than the (5 .45 ± 0 .98)μmol/L of healthy controls (P<0 .05) .CD4+ CD25+ T cells comprised (4 .57 ± 0 .85)% of CD4+ T cells in patients ,higher than the (1 .83 ± 0 .49)% in healthy controls (P<0 .05) .CD4+ CD25+ T cells in the peripheral blood of patients with pulmonary tuberculosis highly expressed FOXP3 .Conclusion Patients with pulmonary tuberculosis could be with an increased level of NO and FOXP3 ,which might have important role in the pathogenesis of pulmonary tuberculosis .
5.The application value of hysteroscopy in recurrent implantation failure

Objective To explore the application value of hysteroscopy in recurrent implantation failure ,and discuss what was the best time to transfer embryos after the operation .Methods The patients who had undergone two or more failed IVF-ET cycles were separated into two groups .The patients of group A ( n=210) received hysteroscopy , and the patients of group B ( n=71) did not receive hysteroscopy evaluation .Intrauterine lesions diagnosis were trea-ted during the procedure .Patients without intrauterine pathologies were given slight curettage .The recurrent IVF-ET were performed after the procedure .Then we analyzed the clinical pregnancy rate of the two groups and determined the best time to transfer embryos after the operation.Results In group A,57 patients were pregnant.In group B, 8 patients were pregnant .There was a significant difference in the clinical pregnancy rates between the two groups (χ2 =7.52,P<0.01).In the pregnant patients of group A ,about 94.7%were pregnant in 6 months after hysterosco-py.Conclusion The patients with recurrent IVF -ET transfer failure should be evaluated prior to commencing IVF-ET cycle to improve the clinical pregnancy rate .It is better to transfer embryos within 6 months after hysteroscopy operation.
6.Role of IL-6 in the contraceptive mechanism of intrauterine device

Objective:To discuss the relationship between the serum level of interleukin-6 (IL-6) and the presence of an intrauterine contraceptive device (IUCD). Methods:The experimentalgroup was subdivided into the bleeding and the non-bleeding groups which consisted of 64 womenwith IUCD. There were 60 healthy women in the control group. The serum levels of IL-6 and C-reactive protein (CRP) were detected by enzyme-linked immunosorbent assay (ELISA) in bothgroups. Results :In the experimental group,the serum levels of IL-6 in 3 cases were increased,andthere was a significant difference in comparison with those of the control group (P<0. 01),butthere were no statistical differences between the bleeding group and the non-bleeding group withIUCD (P>0. 05). Serum CRP was negative both in the experimental groups and in the controlgroup. Conclusion:The non-bacterial endometrial inflammation induced by IUCD causes the in-crease of serum level of IL-6 in some women. It is suggested that IL-6 may play an important rolein the contraceptive mechanism of IUCD.
7.Study progress on antitumor activities of artemisinin and its derivatives

The antitumor activities of artemisinin and its derivatives has become a hot topic in recent years.Artemisinin and its derivatives have been reported to exert antitumor effects mainly by blocking the cell cycle, promoting tumor cell apoptosis, inhibiting angiogenesis and so on.Also, their influence on signaling pathways has attracted considerable interest of many scholars.Currently, in order to discover some compounds with better therapeutic potentials, many studies dedicated to the synthesis and development of new artemisinin derivatives.In addition, the effects of artemisinin and its derivatives on multi-drug resistantace in tumor cells also become a trend of the future research.In this paper, we review the research advances in the antitumor activities of artemisinin and its derivatives during the past five years to provide a reference for future study .
8.3-MA combined with TSA inhibits growth of triple-negative breast cancer cells

AIM: To investigate the effect of 3-methyladenine (3-MA) combined with trichostatin A (TSA) on triple-negative breast cancer cells and the mechanism involved.METHODS: The viability of MDA-MB-231 cells was detected by MTT assay, the migration ability was determined by scratch assay, and the expression of autophagy-related proteins was detected by Western blot.RESULTS: TSA significantly inhibited the viability of MDA-MB-231 cells in a time-and dose-dependent manner.The results of scratch assay showed that TSA inhibited cell migration ability.Western blot data indicated that TSA resulted in a moderate increase in LC3-Ⅱ expression.Moreover, 3-MA inhibited cell autophagy induced by TSA, and combination of 3-MA and TSA further inhibited the viability of MDA-MB-231 cells.CONCLUSION: Combination of 3-MA and TSA may effectively inhibit the growth of triple-negative breast cancer cells.
9.Establishment of a real-time fluorescent quantitative PCR for the rapid detection of viral reservoir in the peripheral blood of SIV-infected rhesus monkeys

Objective To establish a TaqMan probe-based real-time fluorescent quantitative PCR ( real-time PCR) for the quantitative and rapid detection of viral reservoir in peripheral blood mononuclear cells (PBMCs) isolated from rhesus monkeys with simian immunodeficiency virus (SIV) infection and to evaluate its preliminary application. Methods A pair of primers and one TaqMan probe were designed ac-cording to the conserved sequence of SIVmac239 strain for real-time PCR amplification. A length of 2 090 bp of nucleotide fragment was digested from the plasmid p239SpSp5 containing 5′-end long segments of SIV-mac239 strain by restriction enzymes EcoRⅠand SpeⅠ. The standards used for quantitative detection of SIV DNA in peripheral blood samples were prepared by a 10-fold serial dilution and used for graphing the stand-ard curve. The numbers of SIV DNA ( copies per 106 PBMCs) in rhesus monkeys during acute and chronic phases of SIVmac239 infection were determined and the virological characteristics of SIV DNA at different phages of infection were analyzed. Results A linear positive correlation between cycle threshold ( Ct) val-ues and concentrations (10 copies/μl to 109 copies/μl) of the standards was found. High levels of SIV DNA were monitored in SIV-infected monkeys 14 to 22 days after acute infection. The levels of SIV DNA in the acute phase of infection were about 1 to 2 logs higher than those in the chronic phase of infection. The num-bers of SIV DNA ( copies per 106 PBMCs) were 1 log lower than the SIV viral load in peripheral blood of the same monkey. The ratios of SIV DNA load to SIV RNA load ( DNA/RNA) in chronic phase of infection were higher than those in the acute phase. Conclusion The established TaqMan probe-based real-time fluorescent quantitative PCR was a highly sensitive and specific assay for the detection of SIV DNA with an advantage of wide linear range. It could be used for the quantitative evaluation of latent reservoirs of SIV.
10.Anti-oxygen free radicals and anti-lipoperoxidation of nicotinic acid on hyperlipodemia quail

Aim To study the oxygen free radical scavenging activities and antilipid peroxidation of nicotinic acid. Methods High lipid models of quail were induced by high fat diet. Five groups were divided, namely control group, high lipid group, NA group(150 mg?kg-1 and 75 mg?kg-1), and Vit E group (100 mg?kg-1). The contents of TC,TG,MDA and activities of SOD were examined for 3 times in the plasma and liver after administering NA for 9 weeks. Results The results showed the increase of TC,TG and MDA contents and decrease of SOD activities in high lipid groups by 3 weeks high fat diet. After administering NA for 3 weeks, NA reduced TC,TG and MDA contents and increased SOD activities, revealing the relationship to dose-dependence. Conclusion NA is most likely to possess the capabilities of anti-oxygen free radicals and anti-lipoperoxidation to hyperlipodemia quail.
