OBJECTIVE:To set up data management system for therapeutic drug monitoring(TDM),and to realize the remote database access by means of network.METHODS:The TDM data management system was set up by means of com?puter network technology and database technology.RESULTS&CONCLUSION:A satisfactory management interface and remote database access were available through TDM Data management system,which were sure to benefit clinic and patients.

Objective To explore the role of endothelial progenitor cells (EPCs)in improving functional recovery and promoting neurogenesis in damaged area of ischemic rat brain by MRI,neurological severity score (NSS),and pathological examinations.Meth-ods EPCs wereseparated,cultured,and identified,ultrasmall superparamagnetic iron oxide(USPIO)particles was used to label EPCs in vitro.Transient middle cerebral arterial occlusion(tMCAO)was successfully performed in 30 adult SD rats.Magnetically labeled cells(the experimental group)and normal saline (the control group)were injected intravenously into the tMCAO rats through the tail vein respectively,then MR imaging and NSS was performed 0 day,2 days,4 days,6 days and 8 days later.At last the paraffin-embedded rat brain tissues were obtained and examed by Prussian blue staining and Measurement of microvessel density.Re-sults EPCs wereseparated,cultured,and identified successfully.MR imaging showed significant low signal intensity at the ischemic area on T2 WI sequence.Prussian blue staining images were corresponding to the EPCs staining in vitro.The NSS and microvessel density in experimental group were significantly higher than in control group.Conclusion EPCs can migrate to the damage zone, improve functional recovery and promote neurogenesis in damaged area of ischemic rat brain,and may be a new source of multipoten-tial stem cells for stroke treatment.

Test on coagulation time,prothrombin time and platelet aggregation of mice and rats showed that all four species of Puhuang had effects on blood coagulation, in which Typha angustata Bory et Chaub. was the strongest followed by T. davidiana Hand. -Mazz., T. latifolia L,and T. angustifolia L. Their mechanism of coagulation were not the same,in which T. angustata Bory et Chaub. mainly acted on thrombogen,while T.davidiana Hand. -Mazz. mainly acted on platelet aggregation.

OBJECTIVE:To study the effects of Astragalus granules on the expression of Caveolin-3(Cav-3)and Smad family member 3 (Smad3) in the myocardial cells of rats with viral myocarditis. METHODS:90 rats were randomly divided into a nor-mal group,a model group,a Shenmai injection group [positive drug,0.2 g/(kg·d)] and the groups of low,medium and high-dose Astragalus granules [0.42,0.84,1.68 g/(kg·d)],with 15 rats in each group. The rats in all groups except for the normal group were given CVB3 ip for the establishment of viral myocarditis model. Meanwhile,the rats in the drug administration groups were given corresponding drugs ig,while those in the normal group and the model group were given normal saline ig,for 15 consecu-tive days. 5 rats were selected from each group respectively on the 3rd,9th and 15th days of drug use to take an experiment. For the rats,the pathological change of the cardiac muscle tissue was observed and scored,and the mRNA and protein expression of Cav-3 and Smad3 in the myocardial cells were detected. RESULTS:After 15 days of drug use,compared to the normal group,the rats of the model group had hyperplasia of a large number of cardiac muscle fibers,obvious lesions at cardiac muscle fibers, and significantly higher pathological score and levels of the mRNA and protein expression of Cav-3 and Smad3 in the myocardial cells (P<0.05). Compared to the model group,the rats in the drug administration groups had cardiac muscle tissue lesions improved and had obviously lower pathological score and levels of the mRNA and protein expression of Cav-3 and Smad3 in the myocardial cells(P<0.05). CONCLUSIONS:Astragalus granules can markedly downregulate the gene expression of Cav-3 and Smad3 in the myocardial cells of rats with viral myocarditis,which is inferred as a prevention and treatment mechanism of viral myocarditis.

Objective To establish the HPLC fingerprint of cultured Cordyceps militaris. MethodsHPLC Method was used for the determination of C. militaris on Agilent ZOBAX SB-Aq C18 column (250 mm?4.0 mm, 5 ?m), and measuring with methanol 5%—60% in 30 min as elution detective wavelength at 260 nm, injection sample of 1.0 ?L and flow rate of 1.0 mL/min. The systematic optimized design of the chromatographic conditions, such as supersonic extraction, mobile phase, and detective wavelength as well was carried out. Results Fingerprint consisted of 11 common peaks and the result of methodology determination fitted to the related standards. The evaluation of cultured C. militaris by systematically comparing chromatograms with a professional analytical software recommended by National Institute for the Control of Pharmaceutical and Biological Products has been established. Conclusion The method is accurate, simple, and useful for the quality control of cultured C. militaris.

Objective:To evaluate the effects of Er,Cr:YSGG laser combined with NaF treatment on the acid resistance of human dental hard tissue.Methods:The enamel or dentin samples were randomly divided into four groups and prepared by different methods as follows: ①without treatment(control group),②laser irradiation(Laser group),③ treated with 20 g/L NaF solution(NaF group),④laser irradiation+ NaF solution treatment(Laser and NaF group).The samples of each group were respectively demineralized in 0.1 mol/L of lactic acid for 24 h and the calcium ion in the demineralization solution in each group was measured by atomic absorption spectrophotometry.The fluoride on the surface of the samples was examined by SED-X and the ultrastructure was investigated by SEM.Results:The concentrations of Ca~(2+) dissolved from the enamel or dentin samples in NaF,laser and NaF and laser groups were significantly lower than that in the control group(P(0.05)).Only in the NaF group,the F peak appeared.SEM observation showed the lased surface showed various patterns of microirregulation with a scaly appearance,the openings of dentinal tubules were visible,but no melting or carbonization;there were lot of particles attached to the surface of NaF treated samples;the ultrastructure of the NaF and laser-treated sample surfaces was similar to that of the laser treated.Conclusion:The combined use of laser and NaF dose not have synergetic effects on acid resistance of human dental hard tissue.

[Objective] To understand the application effect of aromatherapy massage by using plant extracts and its impact factors.[Methods] 94 pregnancy women whose Edinburgh Postnatal Depression Scale(EPDS)assessment score ranged between 10～20 points were selected in this study.They were implemented the aromatherapy massage 24 hours after delivery.49 cases treated with implementa-tion of the aromatherapy massage from maternal to puerperium were set as the observation group,45 cases whose aromatherapy massage was interrupted by various reasons after discharge were named as the control group.The EPDS score and growth and development indexes were compared 7,14,28,42 days postpartum.[Results] The EPDS scores in the observation group were lower than those in the control group 28,42 days postpartum.The growth and development indexes if infants were better than in the control group.Family-related factor was a direct impact on continuous implementation of aromatherapy massage postpartum.[Conclusions] Continuous implementation of aromatherapy massage by using plant extracts in the whole puerperium combined with psychological counseling by the massage nurses can regulate the maternal emotion,alleviate depression of these women.The puerpera with higher EPDS scores can't take good care of the infants,and can affect the development of the infants.

Objective To study the effect of apelin-13 on myosin light chain (MLC)phosphorylation in vascular smooth muscle cells under the condition that the permeability of endothelial layer is changed by ADMA.Methods Human umbilical vein endothelial cell (HUVECs) were cultured in bottom of upper well of Transwell system to establish the endothelial monolayer barrier.The cells were cultured normally or incubated with ADMA.Some FITC-apelin-13was added to the upper well.After a period of leakage time,the amount of FITC-apelin-13 in the lower well was measured to evaluate Pa value,which reflected the permeability of endothelial monolayer. Thechangesofcytoskeletonandintercellularjunctionwereobservedby immunofluorescence.Subsequently,Transwell dishes were used to establish a co-culture system with HUVECs in the upper well and human umbilical vein smooth muscle cells(HUVSMCs) in the lower well.There were four groups: blank control group; ADMA group,stimulated by ADMA only;apelin group,apelin-13 used only; ADMA+apelin group,apelin-13 used after HUVECs stimulated by ADMA.Immunofluorescence and Western blotting were used to detect phosphorylation of MLC in HUVSMCs.Results ADMA induced changes of cytoskeleton and intercellular junction and increased endothelial permeability of apelin-13.The percentage of Pa between ADMA-stimulated and control groups varied during the period of apelin-13 pass and peaked at 20 minutes,which was significantly higher than that at 0 minutes [(176.3±9.2)％ vs (100.3±1.5)％,P＜0.05].In the co-culture system,Western blotting showed that p-MLC expression in ADMA +apelin group was significantly higher than that in apelin group(P＜0.05).In ADMA group,a few p-MLC expression was found also.Phosphorylation of MLC in above 3 groups was significantly higher as compared to blank control group(P＜0.05).Conclusions ADMA can increase endothelial permeability of apelin-13 by inducing changes of cytoskeleton and intercellular junction.Apelin-13 passes through the damaged endothelial barrier and directly acts on vascular smooth muscle cells to elevate phosphorylation of MLC,which may play an important role in hypertension of uremic patients.

Objective To explore effect of isoorientin on gastric cancer cell autophagy.Methods Isoorientin and autophagy activator and effect of inhibitors on proliferation of human gastric cancer cell line SGC-7901 by MTT assay.Cell apoptosis was detected by flow cytometry.Production of autophagy lysosomal in gastric cancer SGC-7901 cells was obseroved by AO and MDC staining.Characteristic expression of autophagy protein was analysed by Western blot.Results MTT assay indicated that isoorientin could inhibit gastric cancer cell growth, RAP has the same effects, but 3-MA inhibition of apoptosis.Flow cytometry was used to detect the apoptosis rate of isoorientin and RAP could promote cell apoptosis , while 3-MA had the opposite effect.In AO staining, the red light appeared in the cells, and the green fluorescence appeared in the cells of MDC staining, which showed that there was an autophagy in the cells.Western blot test found the isoorientin was cell autophagy specific protein LC-3II, Beclin-1 expression increased, 3-MA suppressed the expression of the two proteins, and RAP had the opposite effect.Conclusion Isoorientin could induce apoptosis in gastric cancer SGC-7901 cells, autophagy is involved in the process of death.
