1.Investigation on Prevalence and Influencing Factors of Dental Fluorosis in School-age Children in Countryside with High Level of Fluoride in Drinking water in Ji'ning

Objective To explore the risk factors for school age chidren dental fluorosis in high fluoride rural areas.Methods Based on the fluoride screening results of rural drinking water in Shandong in 2006,three villages of Jiaxiang counties were selected as the survey sites,12 drinking water samples were collected and pH value,fluoride,hardness were determined,110 school-age children were chosen among high fluoride countryside in May,2008.Dental fluorosis was examined,and a questionnaire survey was performed.Results The average levels of pH value,fluoride content and hardness were 7.01,1.24,494.00 mg/L respectively,and the of dental fluorosis was 63.6%.Simple Logistic regression analysis showed that the prevalence of dental fluorosis was associated with Lower family income,lower education level of parents,lower nutrition knowledge score and few kinds of daily food.Multiple Logistic regression analysis showed that the prevalence of dental fluorosis was positively associated with family income,education level of father and consumption of meat,milk,eggs,fresh vegetables and fruit.Conclusion The rate of dental fluorosis among school-age children in the investigated area is higher.Family income,parents'education and nutritional deficiency are related to the prevalence of dental fluorosis.
2.Assessment and Analysis of Quality Control for Therapeutic Drug Monitoring of Cyclosporine A and Tacrolimus

OBJECTIVE:To assess the stability of the quality control of FPIA for monitoring of CsA and MEIA for monitoring of FK506.METHODS:The quality control on the blood samples of patients receiving therapeutic drug monitoring of CsA and FK506 from June to August in 2007 were subjected to a continual assessment and analysis to investigate the stability and accuracy of the quality control.RESULTS:The mean recoveries of the low,middle and high concentrations of CsA and FK506 were 93.14%~97.65% and 97.70%~103.09% respectively,with RSD at 4.41%~5.37% and 10.44%~14.23% respectively,which accorded with the requirement for biological sample determination stated in the Chinese Pharmacopoeia.CONCLUSION:The methods of FPIA for monitoring of CsA and MEIA for monitoring of FK506 were accurate,and worth to be applied in clinical to guide individualized medication.
3.ADR Caused by Quinolones:Analysis of 199 Cases in Our Hospital

OBJECTIVE:To investigate the characteristics of adverse drug reactions (ADR) caused by quinolones in our hospital and to promote their rational use in clinic. METHODS: By a retrospective review,a total of 199 quinolones-induced ADR cases collected from May 2004 to December 2007 in our hospital were analyzed statistically in respect of patients' gender and age,drug species involved,past ADR history,route of administration,clinic manifestations,etc. RESULTS: A total of 9 kinds of quinolones involved in the 199 ADR cases,of which,118 cases were induced by levofloxacin,16.1% had past ADR history,93.5% of the ADR cases were induced via intravenous drip,and most common presentations of the ADR were lesions of skin and its appendants as well as digestive system. There were 7 serious ADR cases. CONCLUSION: The indications of quinolones should be controlled strictly,which should be preferably administered orally and monitoring of its administration should be emphasized in clinical practice to decrease and avoid the occurrence of ADR.

5.Establishment of a real-time fluorescent quantitative PCR for the rapid detection of viral reservoir in the peripheral blood of SIV-infected rhesus monkeys

Objective To establish a TaqMan probe-based real-time fluorescent quantitative PCR ( real-time PCR) for the quantitative and rapid detection of viral reservoir in peripheral blood mononuclear cells (PBMCs) isolated from rhesus monkeys with simian immunodeficiency virus (SIV) infection and to evaluate its preliminary application. Methods A pair of primers and one TaqMan probe were designed ac-cording to the conserved sequence of SIVmac239 strain for real-time PCR amplification. A length of 2 090 bp of nucleotide fragment was digested from the plasmid p239SpSp5 containing 5′-end long segments of SIV-mac239 strain by restriction enzymes EcoRⅠand SpeⅠ. The standards used for quantitative detection of SIV DNA in peripheral blood samples were prepared by a 10-fold serial dilution and used for graphing the stand-ard curve. The numbers of SIV DNA ( copies per 106 PBMCs) in rhesus monkeys during acute and chronic phases of SIVmac239 infection were determined and the virological characteristics of SIV DNA at different phages of infection were analyzed. Results A linear positive correlation between cycle threshold ( Ct) val-ues and concentrations (10 copies/μl to 109 copies/μl) of the standards was found. High levels of SIV DNA were monitored in SIV-infected monkeys 14 to 22 days after acute infection. The levels of SIV DNA in the acute phase of infection were about 1 to 2 logs higher than those in the chronic phase of infection. The num-bers of SIV DNA ( copies per 106 PBMCs) were 1 log lower than the SIV viral load in peripheral blood of the same monkey. The ratios of SIV DNA load to SIV RNA load ( DNA/RNA) in chronic phase of infection were higher than those in the acute phase. Conclusion The established TaqMan probe-based real-time fluorescent quantitative PCR was a highly sensitive and specific assay for the detection of SIV DNA with an advantage of wide linear range. It could be used for the quantitative evaluation of latent reservoirs of SIV.

Objective To study the relationship of the plasma levels of high sensitive C-reactive protein(hs-CRP),adrenomedullin(ADM),brain natriuretic peptide(BNP)and matrix metalloproteinases (MMPs)with severity of heart falure, the left ventricular ejection fraction and the relationship among them in the elderly patients with congestive heart failure(CHF). Methods The blood samples were obtained from 50 patients with congestive heart failure and 24 healthy controls.The plasma level of hs-CRP was measured by immunoturbidometry.The plasma levels of ADM and BNP were determined by radioimmunoassay.The plasma levels of MMP-2 and MMP-9 were measured with ELISA.Left ventricular ejection fraction(LVEF)was measured by echocardiography. 7.Assay of Plasma (1→3)-?-D-glucan in Patients Suspected with Deep Fungal Infection

Objective To investigate the relationship between the concentration of plasma (1→3)-?-D-glucan (?-D-glucan) and deep fungal infection. Methods Thirteen patients were recruited in this study, who were suspected with deep fungal infection. G-test TE reagent for ?-D-glucan measurement was used to detect the plasma (1→3) ?-D-glucan in the patients by using UV-2450 spectrophotometer at 545 nm wavelength. The final concentrations were calculated according to concentration conversion formula. Results Nine of thirteen patients were confirmed as deep fungal infection by positive tissue culture, in whom high concentrations of ?-D-glucan were detected, the highest concentration was 352.94 pg/mL (mixed infection), with a mean value of 203.47 pg/mL. In the other four patients with negative culture, the ?-D-glucan concentration was over 54.40 pg/mL in three patients and 16.16 pg/mL in the another. Our results showed that the sensiti-vity, specificity, positive predictive value and negative predictive value of this test were 92.31%, 100%, 100% and 98.36%, respectively. Conclusion G-test TE method is a simple and rapid test and may be used for the diagnosis of patients with deep fungal infection.
8.Penicillium marneffei Infection in Guangdong Province China: Clinical Analysis of 15 Cases

Objective To analyze the incidence of the disease, clinical features, diagnostic criteria, therapy and prognosis of Penicilliosis marneffei found in Guangdong province. Methods To analyze patients data, clinical features, laboratory findings, response to therapy, and prognosis of 15 cases Penicilliosis marneffei found in Guangdong province of China. Results The male was predominant compared with the female (ratio 2 to 1) and without occupational preference, but the patients with AIDS as underlying disease were mostly drivers and the unemployed. Thirteen patients were immunocompromised such as AIDS, connective tissue disease, and kidney transplant. Clinical features showed different manifestations, such as high fever, loss of weight, skin lesion, and respiratory system symptoms. Biopsy of the skin lesion showed PAS stain positive yeast-like, or sausage-form spores. Four patients were localized infection of the skin, eleven patients were systemic infection. Nine patients died, five recovered, 1 patient refused to be treated. Fifteen isolates from different anatomic sites of the patients were identified to be Penicillium marneffei by morphology and dimorphism in the culture, and eleven isolates among these 15 isolates were also confirmed by DNA sequence analysis. Conclusion The incidence rate of Penicilliosis marneffei become higher in the recent years and many patients were accompanied with AIDS in Guangdong province. Attention should be paid to the disease.
9.The effect ofShaoyao-Gancao grain on serum prolactin level in female schizophrenia patients with high prolactin induced by risperidone

Objective To study the effect of Shaoyao Gancao Grain on serum prolactin level in female schizophrenia patients with high prolactin induced by risperidone.Methods 100 patients were randomly divided into the control group 50 cases,which were given risperidone. The study group 50 cases,which were given Shaoyao Gancao Grain on the basis of the control group. The two groups were treated for 12 weeks. Positive And Negative Syndrome Scale,serum prolactin and Estrogen level were measured by immunohistochemistry before and after 12 weeks of treatenmt.Results After 12 weeks of treatenmt,serum prolactin levels were decreased in the study group than control group (46.28 ± 14.06 ng/mlvs. 117.89 ± 22.11 ng/ml;t=10.242,P<0.01). PANSS score were decreased in the study group copare with before treatenmt (67.45 ± 7.00vs. 96.53 ± 11.88;t=7.125,P<0.01). PANSS score were decreased in the control group copare with before treatenmt (68.73 ± 7.71vs. 9 7.93 ± 12.65;t=6.541,P<0.01). But there were no difference in PANSS score between the two groups after treatenmt (t=0.682,P>0.05). ConclusionsShaoyao-Gancao grain can decrease serum prolactin level in female schizophrenia patients with high prolactin induced by risperidone,does not affect estrogen levels.
10.A novel role for tanshinoneⅡA in modulation of dendritic cells maturation and function

Objective:Bone marrow-derived dendritic cells ( DCs) were extracted and gave tanshinone ⅡA to intervene ,and we observed the change of DCs function ,which investigate the effects of tanshinone ⅡA in immune system.Methods:Extract bone mar-row-derived DCs,and cells were cultured in complete medium with 10 ng/ml GM-CSF and IL-4.On day 5,magnetic cell sorting was used to purify DCs,and the purify must be up to 90%.Then,a certain concentration of tanshinone ⅡA or LPS was gave to the cell cul-ture,and cells and supernatant were collected for following experiments .We used flow cytometry to detect phenotype , and enzyme-linked immunosorbent assay(ELISA) was used to detect cytokine production of TNF-αand IL-12.Also,allogeneic mixed lymphocyte reaction was used to detect the ability of DCs to induce T cell proliferation and polarization .Results:When the concentration of tanshi-none ⅡA was 500 ng/ml,the inhibition of secretion of TNF-αwas maximal.So we chose that concentration .Compared with the control group,DCs in experimental group had reduced expression of CD 80,CD86 and MHCⅡ(P<0.05).Compared with the control group ,DCs in experimental group displayed the reduced level of IL-12 and TNF-α(P<0.05).Compared with the control group,DCs in experimen-tal group had reduced ability to induce T cell proliferation (P<0.05).Compared with the control group ,DCs in experimental group in-duced T cell to secret increased level of IL-4,and reduced level of IFN-γ(P<0.05).Conclusion:TanshinoneⅡA can inhibit the den-dritic cells maturation induced by LPS ,which takes part in immune system and autoimmune diseases .
