1.Etudies of reducing poison and increasing effect of atractylodes macrocephala koidz on immune function of tumor-bearing mice after chemiotherapy

Objective To research the effect of atractylodes macrocephala koidz on immune function of tumor-bearing mice.Methods With S_ 180 bearing C_ 57 BL/6 mice as model.Effects of atractylodes macrocephala koidz on tumor weight and immunological function of spleen were cells systematically investigated.Results Atractylodes macrocephala koidz could obviously restore immune function of tumor-bearing mice reduced by chemiotherapy,promote proliferation of activated T cell and interleukin-2(IL-2) levels of tumor-bearing mice.Conclusion Atractylodes macrocephala koidz can reverse the immunological inhibition caused by tumor antigen and chemiotherapy.
2.Adjuvant Therapy of Sepsis-associated Thrombocytopenia with Intravenous Immunoglobulin

Objective To investigate the adjuvant effect of intravenous immunoglobulin on patients with sepsis-associated thrombocytopenia. Methods A total of 229 patients with sepsis with platelet count less than 3 × 109/L, were included in this prospective, randomized, controlled study. The patients were divided into the intervening group and the control group. Conventional treatments were applied in the two both groups , while in the intervening group, intravenous immunoglobulin with a dose of 0.4 g/(kg·d) for 5 consecutive days was administered. The end-points were the platelet counts on day 1,day 3,day 5,and day 7 post-intravenous immunoglobulin, patients’ in-ICU time and the 28-day in-hospital mortality. Results Compared with the control group, the platelet count recovered dramatically after 5-day intravenous immunoglobulin in the intervening group. Moreover , the 28-day in-hospital mortality and in-ICU time were also dramatically improved in the intervening group. Conclusion Intravenous immunoglobulin can enhance the recovery of platelet counts , shorten the in-ICU time and reduce the hospital mortality in patients with sepsis- associated thrombocytopenia (PLT count < 30 × 109/L).
3.The Standardization of Language Function EvaIuation for Hearing-impaired ChiIdren

Objective To revise the standards of the Language Function Evaluation for Hearing-impaired Children and to study the characteristics of language functions in hearing-impaired children.Methods A total of 741 normal hearing children at the age of 3~7 year old from 15 provinces in China were assessed using the Language Function Evaluation for Hearing-impaired Children.A total of 90 hearing-impaired children also completed the e-valuation.The scores of Representation Function,Tool Function,Coordination Function,Expression Function, Entertainment Function and Comprehensive Function of all the children were collected.ResuIts The regression e-quations of the language function were established:Representation Function score=3.34 +0.81 ×children's age, Tool Function score =6.00+0.51×children's age,Coordination Function score =5.72 +0.52 ×children's age,Ex-pression Function score =4.44+0.67×children's age,Entertainment Function score =6.34+0.47×children's age, Comprehensive Function score =4.96+0.62×children's age;Through the regression analysis,the reference stand-ards of the tool for children at the age of 3~6 year old were set down.Five-year-old hearing-impaired children's Tool Function and Expression Function were better than those of three or four-year-old ones(P<0.05).Five-year-old hearing-impaired children's Comprehensive Function were better than those of three-year-old ones(P<0.05).The language function scores had a big gap between the hearing-impaired children and the hearing-nor-mal children at the same ages.ConcIusion The new reference standards of the Language Function Evaluation for Hearing-impaired Children provides the Expectation values and Minimum values of language function from children at the age of 3 -6.They are rehabilitation criterions for hearing-impaired children.The older the hearing-im-paired children are,the higher their scores of language function evaluation.But their scores are much lower than those of the normal hearing children at the same age.
4.Influence of LOX downregulation by RNAi on hypoxic metastasis of human lung cancer cell and the underlying molecular mechanism

Objective To observe the influence of lysyl oxidase(LOX)downregulation via RNAi on hypoxic metastasis of human lung cancer cell 95D and stduy its molecular mechanism.Methods LOX siRNA was used to transfect 95D cell line in normoxia (19% O2 ).After 24-hour incubation,the cells were cultured in hypoxic incubator (0.5% O2 ) for 24h.Real-time PCR assay was applied to detect LOX mRNA and Snail mRNA expression.Levels of Src,phosphorylation of Src (P-Src Y418 ) and Snail protein were determined by Western blot assay.Transwell chamber was used to evaluate the cellular invasion potential.Results Compared with 95D cells under normoxic conditions,hypoixa treatment increased LOX mRNA expression by 14 times and invasion ability by 2.12 times respectively.Compared with siRNA control group,LOX siRNA transfection decreased LOX mRNA expression,the invasion ability of hypoxic cells,and the protein expression of P-Src Y418 and Snail by 70% - 75%,about 30%,and about 40% respectively (P < 0.05).However,it didn't affect the expression level of Src protein or Snail mRNA ( P > 0.05).Conclusions Impaired metastatic potential of hypoxic human lung cancer cell induced by LOX downregulation is associated with reduced expression level of Src activation and Snail protein.The present data provids experimental evidence for LOX as a potential target for prevention and treatment of lung cancer metastasis under hypoxia.
5.Application of language function assessment in children with hearing impairment

Objective To study the characteristics of language function development and establish the reference standards in children of4~6 years old. Methods 616 hearing normal children at the age of 3~7 from 13 provinces in China were assessed with Language FunctionEvaluation for Hearing-impaired Children. 583 usable data were analyzed via SPSS 17.0. Results The older the children were, the highertheir scores of language function evaluation were (P<0.001). The differences between boys and girls on coordination function and relaxationfunction were significant (P<0.01). The girls were better than the boys. The reference standards of children of 4~6 years old were set downbased on the regression analysis. Conclusion There are some laws on children's language function development. The reference standards ofchildren by 4~6 years old has been developed.
6.Study on quality evaluation of Panax ginseng based on effects for replenishing qi and preventing exhaustion.

Based on the effect for replenishing Qi and preventing exhaustion, this essay provides a method to rebuild the quality evaluation system of Panax ginseng. As an independent medical system, the traditional Chinese medicine (TCM) science lacks enough credible evidences to support its theory and thereby wants more relevant studies. Current, it is a misunderstanding for experimental studies on TCM to prove TCM drugs from the angle of western medicine. Therefore, following study modes are recommended to researchers focusing on either the theory of the unique importance of class origin or the theory of the unique importance of techniques. Under the guidance of TCM theories, a new TCM assessment mode, consisting of clinical efficacy, pharmacological effect and effective components, is established in line with clinical application of TCMs. Studies on Chemical fingerprint or active ingredient are made on effective components. In-depth studies are worth to the viewpoint to rebuild the TCM quality reassessment system. On the basis of analysis on ancient herbal literatures and experience in clinical application of TCMs, the author proves the irreplaceable effect of P. ginseng. (pure ginseng decoction) is to replenish Qi and prevent exhaustion. Based on the major effect, a pharmacological model is established in combination with modern clinical efficacy to screen effective components of ginseng herbs and make a quality assessment on safety and efficiency of clinical application of P. ginseng, in order to provide theoretical and experimental basis for cultivation, development and utilization of P. ginseng resources on Changbai mountain.

7.Study on human UDP-glucuronosyltransferase (UGT) isoforms involved in in vitro metabolism of trans-resveratrol.

OBJECTIVETo study major human UGT isoforms involved in trans-resveratrol (TR) phase II metabolism.
METHODtrans-resveratrol and 12 major human UGT isoforms were incubated in vitro and then glucuronic acid metabolites were determined by HPLC-MS, in order to preliminarily analyze the structure and observe the effect of different UGT isoforms on the generation rate of glucuronic acid metabolites.
RESULTIn in vitro metabolic system, two metabolites-4'-O-monoglucuronide resveratrol (M-1) and 3-0-monoglucuronide resveratrol (M-2)-were generated from trans-resveratrol after being catalyzed by UGT. During the cause, generation of M-1 and M-2 were catalyzed by UGT1A1, UGT1A3, 1A8, 1A9 andlA10, whereas only UGT1A6 and 1A7 contributed to the forma-tion of M-2. Both the formation rate of M-1 and M-2 catalyzed by UGT1A1, 1A10 and the formation of M-2 catalyzed by UGT1A8 slowed down with the increasing concentration of substrates, causing the phenomenon of "substrate inhibition".
CONCLUSIONUGT1A1, 1A8, 1A9, 1A10 get involved in the formation of M-1, and of them UGTIA9 is the most important contributor. UGT1A3 also makes small contribution to the formation of M-1 and M-2, while other UGT isoforms show hardly any reaction with the trans-resveratrol phase II metabolites.
