Purpose　To investigate the pathomorphological and immunohistochemical features of gastrointestinal stromal tumors (GIST). Methods　Light microscopy was used to study the morphologic characteristics of 77 cases of GIST. The expression of c-kit(CD117), CD34, vimentin, SMA and S-100 protein were detected in all cases with S-P immunohistochemical method. Results　GIST tended to have a higher degree of cellularity and lesser degree of cytoplasmic eosinophilia than observed in classic leiomyomas. The neoplastic cells of GIST were spindle, epithelioid or both in different proportion. Focal to prominent cytoplasmic vacuolization was often seen. The histological spectrum included interlacing fascicles, diffuse sheets, storiform and palisading pattern. The formation of cell clusters was rather special. Stromal and vascular hyalinization was common. GIST primary in the mesentery tended to have a higher potentiality of malignancy. The positivity of CD117 and CD34 were 90%, 92%, respectively. Conclusion　The characteristic histological features exist in GIST. The detection of CD117 and CD34 is helpful to differential diagnosis.

Objective To establish a model of isoniazid induced necrosis and apoptosis in HepG2 cell and to observe the expressions of Fas/Fas ligand (FasL) in this model.Methods HepG2 cells were treated with different dosages of isoniazid (0,1,2,4,6 and 8 mg/mL) or blank control for 24 hours.Flow cytometer was used to observe the cellular morphology of the HepG2 cell.Annexin V/propidium iodide double staining and flow cytometry were employed to detect the necrosis and apoptosis of HepG2 cells.The expressions of Fas/FasL on the cells were also determined by flow cytometry.The data were analyzed by one-way ANOVA.The comparisons between the drug groups and the control group were performed by using Dunnett t test. Results The higher the dose of isoniazid (4,6,8 mg/mL) was,the more necrosis and apoptosis were observed.In the 4,6 and 8mg/mL isoniazid arms,the total mortality rates were all higher than the control group [(32.1 ±7.5)％,(34.9±8.1)％,(38.2±9.4)％ vs (7.2±1.5)％ respectively](t=4.62,5.14 and 5.75,respectively; all P＜0.01 ).The expression levels of Fas increased along with the dose of isoniazid increasing [(8.7±2.2)％,(11.5±2.8)％,(12.3±3.0)％ and (10.6±2.9)％ in isoniazid 2,4,6and 8 mg/mL arms,respectively],which were all higher than that in control arm [(3.1 ±0.8) ％](t=2.97,P＜0.05; t=4.46,P＜0.01; t=4.88,P＜0.01; t=3.98,P＜0.05).Furthermore,the expressions of FasL increased as well when the dose of isoniazid increased.The expression levels of FasL were (16.2±3.5)％,(21.7±4.8)％ and (18.7±4.9)％,respectively in isoniazid 4,6 and 8 mg/mL arms,which were all higher than that in the control group [(7.4±1.4)％](t=3.11,P＜0.01; t=5.06,P＜0.01; t=3.99,P＜0.05).HepG2 cell necrosis increased with isoniazid of 8 mg/mL.However,the increase of apoptosis was not observed.Conclusion Isoniazid can induce HepG2 cell necrosis and apoptosis,and the apoptosis may be related with the increased expressions of the Fas/FasL on the cells.

Objective To evaluate the best experimental technique with high-sensitivity and specificity for enhancing positive rate of syphilis screen test and preventing the blood dissemination of this disease.Methods Comparison of the results of TP-ELISA,TRUST and TPPA methods of syphilis examination was performed.Results Using three methods to exam 43,323 samples,299 samples were for positive syphilis.Among them, 294 positive samples with the ELISA method,the positive rate of syphilis test was 98.33%(294/299):92 positive samples with TRUST method,the positive rate only 30.77%(92/299);239 positive samples with TPPA method,the positive rate 79.93%(239/299).Conclusion TP-ELISA method with high sensitivity is suitable for the instrument standardization and data preservation,which is an ideal method for blood screen test of syphilis.For guarantee of blood safety and conventience and economic benefit of blood syphilis screen test, it is the best way to use both TP-ELISA and TRUST methods.The specificity of TPPA method is perfect, which is suitable for confirmation test of syphilis positive samples.

Objective: To observe the change of blood cell,marker of vascular endothelium and lung of rabbits during experimental severe decompression sickness(DCS) and to compare each parameter between survival group and dead group.Methods: 14 rabbits were put into the chamber.They stayed for 35mins under 0.55 MPa,and another 35 mins under 0.7 MPa,then decompressed to 0.1 MPa in 4 minutes.Samples were drew before the compression and after decompression for testing count of white blood cell(WBC),neutrophilic leukocyte,lymphocyte and platelet(PLT) and the concentration of ET-1 and the vWF.The tissue of lung of all the rabbits was observed under light and electron microscope.The changes of above parameters of survival rabbits were compared with that of the dead.Results: After rapid decompression,there were 8 rabbits died within 30 minutes,while the other 6 rabbits survived without any symptom of decompression sickness after 24 hours observation.Before the compression,the total count of WBC of dead rabbits was?(11.13?2.37)?10~(9)/L,which was higher the that of survival rabbits,(8.87?2.11)?10~(9)/L.After decompression,the decrease of PLT of dead group,(273?(63)?)10~(9)/L,was more than that of survival group,(148?63)?10~(9)/L.Ttissuedamage of lungs of both dead and survived rabbits was observed as partial alveoli collapse,interstitial edema,local emphysema,various emboli in microcirculation and thickening of respiratory membrane.The damage of lung of dead rabbits was much worse than that of the survival.Conclusion:Experimental DCS induced the increasing of circulation tension,WBC obturation,various emboli,thickening of respiratory membrane,consumption of surface active substance in the body of rabbits.Those rabbits who reacted too severe and had bad compensation ability would die,on the contrary,those rabbits who had a good compensation could survive.

[Objective] To observe the effect of Qi-strengthening and blood-activating method for chronic nephritis (CN). [Methods] One hundred and ten patients with CN were randomized into 2 groups. Group A (n = 60) was treated with symptomatic treatment combined with Qi-strengthening and blood-activating herbs including Radix Astragali, Radix Codonopsis, Poria, Rhizoma Dioscorease, Fructus Corni, Rhizoma Curculiginis, Herba Lycopi, Radix Salviae Miltiorrhizae, Semen Persicae and Flos Carthami. Group B (n = 50) was treated with symptomatic treatment combined with oral use of Bailing Capsule, the principal ingredient of which is fermented Cordyceps powder with the actions of tonifying lung and kidney and invigorating essence and Qi. The treatment course lasted 8 weeks. Total therapeutic effect, and the changes of symptoms, immune function, hemorrheology indexes and urine routine indexes before and after treatment were observed. [Objective] This study was to investigate the effect of TMP on function of EPCs in normal or hydrogen peroxide(H2O2)inducing injury model cell culture,and to approach a therapy to coronary heart disease(CHD).[Method] Total mononuclear cells were isolated from the cord blood by ficoll density gradient centrifugation,cultured in vitro.After the cells cultured for 7 days,identified them by cellmarkers and the ability of intaking the ac-LDL,and made sure they were EPCs.To divide the cells into three groups,TMP(5,25,100,200mg),H2O2(500??)model and TMP together with H2O2,cultured for 24h and then detected the function and apoptosis of EPCs.[Result] Versus to control group,the TMP(5,25,100mg)groups had no important effect on the proliferation and adhesion of EPCs,but TMP(200mg)group had negative effect on EPCs.Versus to H2O2 model group,TMP(5,25,100mg)preincubated groups had important protective effect on EPCs in proliferation,adhesion and apoptosis.[Conclusion] TMP could protect EPCs from oxidative stress injury,but had little benefit to EPCs in normal incubation.

[Objective]To discuss the midterm effects of bipolar artificial femoral head arthroplasty in treatment of elder femoral neck fracture.[Method]From September 1995 to Febuary 2005,73 elderly patients with femoral neck fracture were operated with domestic bipolar artificial femoral head arthroplasty (average 74 years old).Forty-seven cases had been followed up for average 65 months.Harris score system and X-ray had been used for evaluation.[Result]There was no vessel and nerve injury,no infection and dislocation,no perioperative death.The complications included one greater trochanteric fracture,3 cases with pneumonitis,2 urinary infection,1 cerebral infarction,1 dysfunction in central nerve system and 1 case with deep venous thrombosis.Of all the 36 X-rays there were 3 cases with prosthesis subsidence,lucent line lesser 2mm in 2 cases,narrow joint space in 21 cases,protrusion in 1 case.According to the Harris scoring system,there were excellent in 12,good in 26,fair in 7,poor in 2.[Conclusion]Bipolar artificial femoral head arthroplasty are reasonable choice for treatment of femoral neck fracture in the elderly.Proper indication and perioperative management is critical to get good results.
