1.Breast cancer stem cells:separation and resistance protein analysis

BACKGROUND:Breast cancer stem cels are relatively special cels in the body, which have the self-renewal and multi-differentiation ability to promote tumor formation and development, and maintain tumor growth for a long-term. Therefore, it is of great significance to analyze the expression of resistance proteins of breast cancer stem cels.
OBJECTIVE:To isolate breast cancer stem cels from human breast cancer tissues, to observe their differentiation and morphology characteristics and to analyze their resistance proteins.
METHODS:Thirty tumor samples of breast invasive ductal carcinoma were selected to separate single cel suspension using mechanical separation method, and breast cancer stem cels and differentiated cels were sorted with two-step immunomagnetic bead method. Two-step immunocytochemistry method was used to detect the expression of resistance proteins in breast cancer stem cels.
RESULTS AND CONCLUSION:Percentage of breast cancer stem cels had no significant correlations with age, long diameter of the tumor, lymph node metastasis and histological grading (P > 0.05). P-gp and GST-π positive rates in the breast cancer stem cels were significantly higher than those in the differentiated cels (P < 0.05); while TopoII and LRP positive rates in the breast cancer stem cels were significantly lower than those in the differentiated cels (P < 0.05). To conclude, breast cancer stem cels show stronger drug resistance than the differentiated cels by highly expressing P-gp and GST-π and lowly expressing TopoII and LRP, which may be the key reason for chemotherapy resistance in breast cancer.
2.Comparison of Therapeutic Efficacy of Lobaplatin or Cisplatin Combined with Tegafur, Gimeracil and Oteracil Potassium in the Treatment of Advanced Metastatic Breast Cancer

OBJECTIVE:To compare therapeutic efficacy and safety of lobaplatin or cisplatin combined with tegafur,gimeracil and oteracil potassium in the treatment of advanced metastatic breast cancer.METHODS:A total of 160 patients with advanced metastatic breast cancer were randomly divided into observation group and control group,with 80 cases in each group.Control group was given Cisplatin injection 30 mg/m2 intravenously,every 3 weeks Tegafur,Gimeracil and Oteracil Potassium capsules 50 mg orally after meal,twice a day,for consecutive 14 days.Observation group was given Lobaplatin for injection 30 mg/m2 intravenously,every 3 weeks+Tegafur,Gimeracil and Oteracil Potassium capsules (same usage and dosage as control group),every 3 weeks.A treatment course lasted for 3 weeks,and both groups received 2 courses.Short-term efficacies (ORR、DCR),chemotherapy effects of lymph node,lung,bone and liver,ADR and long-term efficacy were compared between 2 groups.RESULTS:After treatment,ORR(67.50% vs.46.25%),DCR(85.00% vs.66.25%),ORR of lymph node metastasis (71.43% vs.47.83%),ORR of lung metastasis (60.71% vs.40.00%),DCR of lung metastasis (78.57% vs.56.00%),ORR of bone metastasis (28.57% vs.16.67%),1-year survival rates (75.00% vs.52.50%) and 2-years survival rates (42.50% vs.17.50%) of observation group were significantly higher than control group;the incidence of chemotherapy ADR in observation group was significantly lower than control group (43.75% vs.70.00%),with statistical significance (P<0.05).There was no statistical significance in lymph node metastasis DCR,bone tissue metastasis DCR,liver metastasis ORR and DCR,or half year sarvival rate between 2 groups (P>0.05).CONCLUSIONS:Compared to cisplatin combined with tegafur,gimeracil and oteracil potassium,lobaplatin combined with tegafur,gimeracil and oteracil potassium show better short-term therapeutic efficacy,therapeutic efficacy of lymph node metastasis,bone metastasis and lung metastasis,more than 1-year long-term therapeutic efficacy and safety in the treatment of advanced metastatic breast cancer.
3.Effects of comprehensive schistosomiasis control in marshlands of Pukou District,Nanjing City

Objective To evaluate the effects of comprehensive schistosomiasis control in marshlands of Pukou District Nanjing City so as to provide the evidence for developing the strategy of the disease control. Methods The information of the prevention and control of schistosomiasis was collected in Pukou District from 2004 to 2013. The variations of the infection rates of human livestock and Oncomelania hupensis snails were analyzed. Results In 2013 the area with snails in the entire dis-trict was 384.09 hm2 which was 66.24%decrease compared to 1 137.61 hm2 in 2004. Moreover after 2008 no infected-snails and schistosomiasis patients were found and both the density of live snails and serum positive rate of schistosomiasis declined with years. Conclusions After the comprehensive prevention and control is conducted the schistosomiasis situation in Pukou District is stable. However in order to consolidate the results it is still essential to continue monitoring the previous infected snail environments and implementing the comprehensive prevention and control measures.
5.Expression of heranase and perlecan in the retina of mouse with oxygen-induced retinopathy

Background Heparanase degrade heparan sulfate side chains of heparan sulfate proteoglycans in the extracellular matrix.Heparanase induces angiogenesis and likely promotes the vascularization of tumor.ObjectiveThe present study is to investigate the expression of heparanase and perlecan in retinas with oxygen-induced retinopathy.Methods Sixty-five clean neonatal C57BL/6J mice were raised in a hyperbaric oxygen box with a volume percentage of 75%±2% for 5 days and then returned to the normal air room.Another 65 matched mice were raised in the normal environment as controls.Evans blue was infused by the superior vena cava in all the mice on postnatal days 12,13,17,21 and 30,afterwards fluorescein angiography was performed and then the mice were sacrificed.The retinas of mice were isolated and prepared and the retinal vessels were examined under a fluorescent microscope and optical microscope.Heranase and perlecan mRNA was detected using reverse transcription PCR (RT-PCR).Heranase and perlecan proteins were detected by Western blot.The analysis of variance was used to compare the mRNA and the protein levels of heranase and perlecan between the experimental and control groups.Results The expression of heparanase mRNA in the retinas of different ages of mice and the different groups showed significant differences (F_(group)=16.303,P=0.000;F_(time)=18.614,P=0.000;F_(interaction)=11.299,P=0.000),and the expression of heparanase mRNA was significantly enhanced in mice from postnatal days 12,13,17 and 21 compared with normal control mice (P=0.001,0.000,0.000,0.001,respectively).The expression of heparanase protein in the retinas of different ages of mice and the different groups followed the same tendency(F_(group)=458.134,P=0.000;F_(time)=78.466,P=0.000;F_(interaction)=71.398,P=0.000).The expression of perlecan mRNA in the retinas of different ages of mice and the different groups showed significant differences (F_(group)=7.351,P=0.013;F_(time)=9.098,P=0.000;F_(interaction)=3.349,P=0.000),and increase in differences also were clearly seen in mice from postnatal days 13,17 and 21 compared with normal control mice (P=0.048,0.000,0.003,respectively).Conclusion The expression of heparanase and perlecan is associated with the development and progression of retinal neovascularization,and perlecan and heparanase together produce a synergistic effect.Heparanase and perlecan may participate in the angiogenesis of oxygen-induced retinopathy.
6.Cytotoxicity and biocompatibility of nano-hydroxyapatite/bacterial cellulose composite scaffolds

BACKGROUND:Nano-hydroxyapatite/bacterial celulose (nHA/BC) composite scaffolds prepared by nano-bionics method exhibit a structure and performance similar to natural bone. OBJECTIVE:To study the cytotoxicity and biocompatibility of nHA/BC composite scaffolds. METHODS:(1) Acute systemic toxicity test: nHA/BC leach liquor and normal saline were respectively injected intraperitonealy into Kunming mice. After 24, 48, 72 hours, the body mass of mice was recorded. (2) Sensitization test: nHA/BC leach liquor and normal saline were respectively injected subcutaneously into the back of Japanese white rabbits. With 72 hours of injection, edema and erythema were observed, and the experiment was done again at an interval of 14 days. (3) Pyrogen test: nHA/BC leach liquor and normal saline were respectively injected into the ear vein of Japanese white rabbits, and after injection, the body temperature of rabbits were determined. (4) Hemolysis test: Diluted rabbit anti-coagulant was added into nHA/BC leach liquor, normal saline and distiled water, respectively. (5) Passage 3 bone marrow mesenchymal stem cels from rabbits were co-cultured with nHA/BC materials, and then, cel proliferation, growth and adhesion on the material surface were observed. RESULTS AND CONCLUSION: nHA/BC composite scaffold was of no acute systemic toxicity, non-alergenic, no pyrogenic and hemolytic reactions; nHA/BC had a three-dimensional porous structure that was better for growth, proliferation and adhesion of rabbit bone marrow mesenchymal stem cels. These findings indicate that the nHA/BC composite scaffold has good biocompatibility and cytocompatibility.
9.The Morphology of Adrenal veins

The morphology of the extraglandular and intraglandular portions of the adrenal veins and other veins concerned with the catheterization in adrenal venography was observed on 53 adult cadavers.The extraglandnlar portion of both adrenal veins is of single trunk type on all the specimens. The left adrenal vein consistently ends into the left renal vein while the right adrenal vein ends into the inferior vena cava in 83.02% of cases and into the right accessory hepatic vein in 9.43%.The length and diameter of the extraglandular portion of the left adrenal vein are 23.05?5.96 mm and 4.99?0.82 mm and those of the right adrenal vein are 8.73?5.13 mm and 2.93 ? 0.87 mm. The angle formed by the left adrenal and left renal veins is 117.35?11.95? and that formed by right adrenal vein and inferior vena cava is 47.5?14.58?.The distances from the femoral vein(lower margin of inguinal ligment)through the external and common iliac veins and inferior vena cava to the hila of both adrenal glands were measured. They may be used as references in performing adrenal venography.The intraglandular portion of the adrenal veins is of three types: single, double and triple trunk types. The single trunk type is the most common occurring in 86% of the left adrenal veins and 50% of the right veins. The triple trunk type exists only on the right side.In addition, the feasibilities and difficulties of the catheterization of the both adrenal veins were discussed according to the anatomy studied.
10.Effects of Low Dose Ultrashort Wave Therapy on Inflammation and Edema after Spinal Cord Injury in Rats

Objective To investigate the effect of ultrashort wave therapy on edema and inflammatory reaction after spinal cord injury (SCI) in rats. Methods 56 Sprague-Dawley rats were divided into sham-operated group (n=8), model group (n=24) and ultrashort wave group (n=24). The model was established with Allen's method. The sham-operated group was exposed endorhachis without hit. The ultra-short wave group was exposed to ultrashort wave radiation 7 minutes, once a day, 24 hours after modeling until the animals were sacrificed. Locomotors functional recovery was assessed every week post operation period by BBB score. Immunohistochemical staining was per-formed to observe the expression of the aquaporin-4(AQP-4) and ED-1. Results The BBB scores increased in the model group and the ultra-short wave group after treatment, and was higher in the ultrashort wave group than in the model group from 1 week after treatment (t>3.368, P<0.01). The expressions of AQP-4 and ED-1 were higher in the model group and ultrashort wave group than in the sham-operated group, and decreased as time went on. They were lower in the ultrashort wave group than in the model group(t>3.156, t>4.466, P<0.05). Conclu-sion Ultrashort wave therapy can alleviate the edema after SCI, reduce the activation and infiltration of inflammatory cells, and promote the recovery of neurological function.
