1.Parenteral organ injury and Immune status in children with rotavirus enteritis

Objective To evaluate the parenteral complications and their risk factors and to study the immune status by detecting immunoglobulin and lymphocyte subsets in children with rotavirus enteritis. Methods Sixty-four children with rotavirus enteritis who were treated in Shengjing Hospital of China Medical University between January 2007 and December 2008(observation group) and 18 healthy chiIdren for health screening at pediatric health care center (normal control group) were reeruimd.Clinical manifestations were collected,stool specimens were detected for rotavirus using ELISA,lymphocyte subsets were detected using flow cytometer,and immunoglobulin.liver enzyme and myocardial enzyme were detected.Results Rotavirus enteritis may be complicated by parenteral complications(liver,myocardium,respiratory and nervous system).The activity of ALT,AST,LDH,CK and CK-mB were higher in observation group than that in normal control group(P<0.05).In observation group,liver injury rate in children younger than 12 months was higher than that in children older than 12 months,and the activity of CK and CK-mB were highcr in severe diarrhea cases[CK(324.5±995.5)U/dl,CK-mB(93.8 4±61.5)U/dl]than that in mild diarrhea cases [CK(252.8±130.4)U/dl,CK.mB(59.6±32.6)U/dl](P<0.05).The level of IgG was lower in observation group[(4.46±1.56)g/L]than that in control group[(5.80±1.67)g/L](P<0.05).Lymphocytes subsets study revealed that the activity of CD4+ in observation group[(29.0±4.18)%]was lower than that in control group[(38.6±3.97)%](P<0.05),the activity of CDl9+ [(38.8±3.94)%]was higher than that in control group[(23.1±7.70)%](P<0.05)and the CD4+/CD8+ ratio was reverse in observafion group.Conclusion Rotavirus enteritis may be complicated by parenteral injuries which get liver,myocardium,respiratory and nervous system involved.Children with rotavirus enteritis Call lead to low immune function.Determination of liver enzyme,myocardial enzyme,immunoglobulin and lymphocyte subsets has important clinical significance to monitoring the change of condition and guiding treatment.
2.Injured effects of electromagnetic pulse on hippocampal neurons and [Ca2+]i

BACKGROUND: Electromagnetic pulse (EMP) radiation can affect the learning and memory function of experimental rats and induce injury of hippocampal issues and change of ultrastructure of rats.OBJECTIVE: To observe the effects of EMP on injury of hippocampal neurons cultured in vitro and [Ca2+]i, and analyze deeply possible mechanism of cerebral injury induced by EMP.DESIGN: Randomized controlled animal experiment.SETTING: Department of Pathology, Chengde Medical College.MATERIALS: Several Wistar neonate rats, of either sex (half and half),were selected. Source of EMP radiation was high intensity EMP dummy source.METHODS: The experiment was performed from March to December 2004 at the Academy of Military Medical Science and Chengde Medical College, respectively. Several Wistar neonate rats were decapitated to take out the brains under narcotization. Hippocampal tissues were isolated. The cell suspension was adjusted to 5×108 L-1 for inoculation. Grouping: ①Cultured cells were assigned into control group and radiation group. Cells were collected immediately after radiation to perform observation of morphology and determination of free calcium ion concentration. ②Other cultured cells were divided into control group, 0-hour radiation group and 12-hour radiation group. Cell apoptosis rate and necrosis rate were determined. (Dosage of cultured cells: one culture flask of each group was checked in each item for 3 times). EMP radiation was in 6×104 V/m, with pulse rise time of 20 ns,pulse width of 30 μs, frequency of 2.5 pulses/min, totally for 2 minutes.EMP radiation was performed in primary cultured hippocampal neurons,and then morphological change of neurons was observed under inverted phase contrast microscope before and after radiation. Cell apoptosis and necrosis were measured with FACS method; Free [Ca2+]i concentration in neurons was measured with Fluo-3-AM fluorescent probe loading and laser confocal microscopy scanning.MAIN OUTCOME MEASURES: Morphological change of neuron, cell apoptosis rate and necrosis rate and free [Ca2+]i concentration.RESULTS: ①Immediately after EMP radiation, the onset of colliquation appeared in nerval cells gradually, and neurite was recovery and degeneration. ②Apoptosis rate after 12-hour EMP radiation recovered as compared with that at hour 0 after radiation, but significantly increased as compared with the control group [(59.27±1.27)%, (72.17±6.21)%, (17.45±5.63)%,P<0.05]. ③Necrosis rate at hour 0 and hour 12 after radiation increased as compared with the control group, but there was no statistical significant difference [(13.71±2.31)%, ( 11.96±1.04)%, (8.45±0.67)% ,P > 0.05].④[Ca2+]i fluorescence intensity at hour 0 after EMP radiation was higher obviously than that in the control group (107.34±26.14,54.93±16.08,P < 0.05).CONCLUSION: EMP induces morphological injury, necrosis and increase of apoptosis rate in hippocampal neurons, and Ca2+ fluorescence intensity increases markedly in neurons.
3.Epidemiological survey of urinary Cadmium in Shengyang urban children

Objective To evaluate the body burden of Cadmium(Cd) in normal children from Shenyang city,and to analyze the effect of Cd on kidney.Furthermore,to investigate the influences of living factors on the level of Cd in children,so as to make recommendations for children's health.Methods The subjects was composed of 1 170 healthy children recruited from 0 to 17 years old by cluster random sampling method.They were divided into three groups(0-5 years old group,6-11 years old group,12-17 years old group).Questionnaires were used to obtain essential information about age,gender,weight,socioeconomic status,medication,and so on.Second morning urine samples were collected to make routine analyses,urinary Cd(Cdob),urine microalbumin(MALB) and α1-microglobulin(α1-MG).Results 1.A total of 1 070 children including 544 males and 526 females were enrolled.2.The concentration of Cdob increased with age,in agreement with the level of urinary Cd corrected by urine specific gravity (Cdsg).However,the level of Cd in urine corrected by urinary creatinine(Cdcr) presented the opposite status with the age increasing.3.Cdcr and urine MALB adjusted by urinary creatinin(MALBcr) revealed a positive correlation(rs =0.45,P < 0.01).4.Preference values for Cdob and Cdsg,Cdcr respectively were:0-5 years old <0.56 μg/L,<0.83 μg/L and <2.17 μg/g Cr;6-11 years old <0.65 μg/L,<1.01 μg/L and <1.23 μg/g Cr;12-17 years old <0.74 μg/L,<1.15 μg/L and < 1.25 μg/g Cr.Conclusions There is an age-dependent cumulative increases in Cdob.Although renal damage was not found in this study,the uptake of Cd should be reduced as much as possible and it is necessary to strengthen the follow-up study of Cdob in body burden.
4.Analysis of data from surveillanc e of iodine deficiency disorders in Futian District of Shenzhen City from 2011 to 2015

Objective To investigate the situation of iodized salt practicing measures, the nutritional status of iodine and the trend of iodine deficiency disorders in Futian District of Shenzhen City, and to evaluate the effectiveness of preventive measures. Methods Every year from 2011 to 2015 in Futian District, five sub-districts were selected according to their position of the east, the west, the south, the north and the center, one school was selected in each sub-district, then sixty students were selected from each school to detect iodine content of their household salt samples;over twenty children (half males and half females) aged 8-10 of those sixty students were selected randomly to measure their thyroid volumes and detect their urinary iodine content;one or two drinking water samples were collected in each sub-district to test iodine content. Thyroid volume was examined by ultrasound method; the salt iodine was tested by the method of direct titration; iodine content in urine and drinking water samples was tested by arsenic cerium catalytic spectrophotometry. Results Between 2011 and 2015, totally one thousand four hundred and eighty-eight edible salt samples were tested. The iodized salt coverage rate, the qualification rate of iodized salt and the consumption rate of qualified iodized salt was 96.1%(1 430/1 488), 99.2%(1 419/1 430), and 95.4%(1 419/1 488), respectively . Four hundred and sixty-one children aged 8-10 were examined;nine children were diagnosed goiter, and the goiter rate was 1.95%. A total of 661 children urine samples were collected. The median urinary iodine was 229.5 μg/L; urinary iodine in 100 - 199 μg/L accounted for 29.8%(197/661), and in 200 - 299 μg/L accounted for 31.0%(205/661). Forty-eight drinking water samples were tested, the iodine content of the drinking water was 10.3 - 22.1 μg/L, and the median water iodine was 16.8 μg/L. Conclusions The iodine deficiency disorders surveillance indicators in Futian District of Shenzhen City all meet the national standards for elimination of the disease. Yet, the level of iodine nutrition needs to be further observed, and both the iodized salt market supervision and the health education should be strengthened.
5.Flow cytometry analysis of unusual peripheral monocyte population

Objective To analyze a population of cells on the right lower lateral of monocyte population in forward scatter/side scatter(FSC/SSC)(X-axis/Y-axis)scatterplot of peripheral blood leucocyte by flow cytometry(FCM)and its influencing factors.Methods The type of cells were identified based on cluster of differentiation antigen(CD)by FCM.The impact of temperature,hemolysin concentration,and incubation time was evaluated.Blood lipid tests were performed to observe the relation between them by statistical methods.Results (1) Phenotypo of this population of cells on the right lower lateral of monocytes in FSc/SSC scatterplot is CD+45 CD+13 CD+14 CD3- CD-19 ,which was the same as monocyte cells:(2)The monocytes in FSC/SSC scatterplot shifted to left side after using haemolysin;(3)The monocytes showed less resistance to antihemolysin in 37℃ than that in 220C:There were more monocytes shifting to left side with the increase of haemolysis time:(4)The swarming ratio of monocytes in patients (31.5%,40/126) Was higher than it in normal controls (5.1%,5/98)(x2=22.74,P<0.01);(5)The levels of total serum cholesterol(TC),triglyeride(TG),low density lipoprotein cholesterol(LDL-C), apoprotein B100(Apo B100) in patients with swarming monocytes were lower than that in the patients without swarming monocytes,(P<0.05).There was no statistical significance between the two groups with respect to levels of total bilirubin(TBIL),albumin(Alb),hish density lipoprotein cholesterol(HDL-C),apoprotein A I(Apo A I),lipoprotein(Lpa).Conclusions Peripheral blood monocytes can be divided in two groups in FSC/SSC scatterplot when analyzed with FCM.The presence of this population of cell Was related to resistance to hemolysin.It can be influenced by haemolysis time and incubation temperature.Therefore,the effect of swarming monocytes and abnormal cell membrane should be taken into consideration when the markers and function of monocytes are detected by FCM.
6.Chloride currents activated by cisplatin in poorly differentiated naso-pharyngeal carcinoma cells are not Ca2+-activated chloride currents

AIM:To investigate the type of chloride channel activated by cisplatin in poorly differentiated na -sopharyngeal carcinoma cells (CNE-2Z cells).METHODS:The technique of whole-cell patch-clamp was used to investi-gate the role of Ca 2+in the activation of cisplatin-activated chloride currents and to analyze the effect of hypertonic stress on these currents in CNE-2Z cells.RESULTS:Chloride currents were induced when the cells were exposed to the calcium -free cisplatin solution , showing the similar density to the currents induced by cisplatin with the presence of extracellular cal -cium.However , the latency and the peak time of cisplatin-activated currents in the absence of extracellular calcium were prolonged.The activation of cisplatin-activated chloride currents was insensitive to the depletion of intra-and extracellular calcium.Calcium channel antagonist nifedipine had no effect on the cisplatin -activated chloride currents , while hypertonic solution completely inhibited those currents .CONCLUSION:The cisplatin-activated chloride currents are independent on intra/extracellular calcium .The chloride channels activated by cisplatin are not calcium-activated chloride channels , but are probably volume-sensitive chloride channels .
7.Influence of epidermal growth factor and gonadotrophin on the in vitro maturation of human oocytes.

OBJECTIVETo study the effect of epidermal growth factor(EGF) and different concentrations of gonadotrophin (Gn) on the in vitro maturation of human oocytes.
METHODSEGF was added to the vitro culture medium in order to observe the effect of Gn combined with or without EGF on the result of in vitro maturation. The concentrations of hCG and FSH were changed respectively to observe the difference between the results.
RESULTSAdding EGF to the culture medium improved the maturation rate of oocyte significantly (P < 0.05). There was no difference between the results with different concentrations of hCG and FSH in the culture medium.
CONCLUSIONEGF can improve the results of the in vitro maturation of human oocytes by increasing the maturation rate significantly. Increasing the concentration of Gn does not influence the results of in vitro maturation.

8.Roles of chloride channels in apoptosis induced by adriamycin in nasopharyngeal carcinoma cells

Aim To investigate the roles of chloride channels in the apoptosis and apoptotic volume de-crease (AVD)induced by adriamycin in nasopharyn-geal carcinoma CNE-2Z cells.Methods Apoptotic rates were detected by flow cytometry,and the volume changes were measured by the time-lapse live cell ima-ging technique.The patch clamp technique was used to record whole-cell chloride currents.Results Adria-mycin induced apoptosis of CNE-2Z cells.An early ap-optotic volume decrease was observed in the cell trea-ted with adriamycin.The cell volume was decreased by about 10% in 2 h.Adriamycin activated a chloride current which showed outward rectification.The chlo-ride channel blocker 5-nitro-2-(3-phenylpropylamino)-benzoate (NPPB ) could inhibit the adriamycin-in-duced chloride currents,apoptosis and prevent cell shrinkage.Conclusions Our findings suggest that ad-riamycin causes cell apoptosis by activation of chloride channels.Chloride channels may be involved in the apoptosis and apoptotic volume decrease induced by adriamycin in CNE-2Z cells.
9.Sphingosine 1-phosphate signaling pathway:a novel target for regulation of epithelial and endothelial barrier function

Sphingosine 1-phosphate(S1P)is a pleiotropic sphingolipid metabolite that has been shown to regulate important physiological function by activation of its G-protein-coupled receptors S1PRs. S1P has been identified as an important signaling molecule in maintaining epithelial and endothelial barrier function. S1P signaling pathway is involved in epithelial and endothelial barrier function by regulation of adherens junction and tight junction assembly, cytoskeletal reorganization, and focal adhesion formation. Thus, S1P signaling pathway may become a novel therapeutic target for cell barrier dysfunction during some illnesses such as acute lung injury, inflammatory bowel disease and sepsis. In this review, the research progress of S1P signaling pathway in regulating epithelial and endothelial barrier function and the application of S1P in barrier dysfunction-related diseases were summarized, so as to provide references for future research.
10.Effect of acupuncture on cholinergic anti-inflammatory pathway in hippocampus of aged rats during global cerebral ischemia-reperfusion

Objective To investigate the effect of acupuncture on cholinergic anti-inflammatory pathway in the hippocampus of aged rats during global cerebral ischemia-reperfusion ( I∕R ) . Methods Ninety-six clean-grade healthy male Sprague-Dawley rats, aged 17-22 months, weighing 440-580 g, were divided into 3 groups ( n=32 each) using a random number table method: sham operation group ( group S), global cerebral I∕R group (group I∕R) and acupuncture group (group AP). Global cerebral I∕R was induced by 4-vessel occlusion method described by Pulsinelli in group I∕R and group AP. Baihui and Feng-chi were stimulated for 14 consecutive days before ischemia in group AP. Four rats were sacrificed at 1, 3, 5 and 7 days of reperfusion, and brains were removed for determination of neuronal apoptosis by TUNEL. Four rats were sacrificed at 1, 3, 5 and 7 days of reperfusion, and brains were removed for determination of the expression of α7 nicotinic acetylcholine receptor (α7nAChR), choline acetyltransferase (ChAT), tumor necrosis factor-α ( TNF-α) and interleukin-1β ( IL-1β) in the hippocampal CA1 region by Western blot. The apoptosis rate was calculated. Results Compared with group S, the apoptosis rate of hippocam-pal neurons was significantly increased, and the expression of α7nAChR, ChAT, TNF-α and IL-1β was up-regulated at each time point of reperfusion in I∕R and AP groups ( P<0. 05) . Compared with group I∕R, the apoptosis rate of hippocampal neurons was significantly decreased, the expression of α7nAChR and ChAT was up-regulated, and the expression of TNF-α and IL-1β was down-regulated at each time point ofreperfusion in group AP (P<0. 05). Conclusion The mechanism by which acupuncture mitigates global cerebral I∕R injury may be related to activating cholinergic anti-inflammatory pathway in the hippocampus of aged rats.
