2.Value of ultrasonic elastic quantitative analysis technique of organization diffusion in adenomyosis

Objective To investigate the value of the ultrasonic elastic quantitative analysis technique of organization diffusion in adenomyosis.Methods Ninty-three patients with adenomyosis were divided into adenomyosis group (n =52) and adenomyoma group (n =41),and fourty healthy subjects were enrolled as control group.All subjects underwent conventional ultrasound and real-time tissue elastography.Eleven elastic characteristics of all the subjects were performed.Results Different characteristics of ultrasound elastography images appeared in different groups.The measured values of elastic characteristics were significantly different between case group and normal group (P < 0.05),but they were no significant difference between adenomyosis group and adenomyoma group(P > 0.05).The cutoff values of every index were analyzed by ROC curve.The strongest specificity index was SD(99%),the highest sensitivity index was ASM(100%),and the highest accuracy index was IDM(94%).Conclusions The value of ultrasonic elastic quantitative analysis of organization diffusion technique in adenomyosis can improve the ultrasound diagnosis of this diease.
3.The contrast research and application of electrochemiluminescence immuno-assay determination kits of human embryonic antigen

Objective To evaluate the comparability of test results of self-built human carcinoembryonic antigen(CEA)electro-chemiluminescence immunoassay(ECLIA)and imported reagent.Methods A total of different concentrations 77 fresh serum speci-mens were collected and detected CEA by two kinds of ECLIA kit.The results were analyzed with Excel2003 and SPSS1 9.0 soft-ware.Results The difference between each dose was significant (P <0.05),and the detection results between each had no signifi-cant difference (P >0.05);the sensitivity of the assay was 0.3 ng/mL,the intra coefficient of variation was 4.58%-5.83%,the inter coefficient of variation was 5.07%-5.97%,the analytical recovery was 99.13%-107.28%,the specificity of the assay had no cross reaction with CA1 99 and AFP.The correlation coefficient between two kinds of reagents determination results was greater than 0.95,with imported reagent as reference test,self-built carcinoembryonic antigen ECLIA clinical performance evaluation was acceptable.Conclusion The precision of the two kinds of ECLIA in detection of CEA accord to clinical requirement.Comparability exists in evaluating the acceptability of clinical.
4.Behavior, inflammatory response and cell autophagy of amyloid precursor protein / tau protein /presenilin-1 transgenic AD mice

Objective To explore the behavior characteristics ofamyloid precursor protein / tau protein / presenilin-1 (APP/Tau/PS1) transgenic mice in different months of age and study the progress of inflammation and the changes of autophagy level in brain of these APP/Tau/PS1 Alzheimer's disease(AD) mice.Methods Four-month-old wild-type (WT) mice (B6:129SF2/J) and Tau/APP/PS1 transgenic mice at 2,4,6,8 and 12 months old (n=8) were chosen in our study.Morris water maze was used to detect the spatial learning and memory abilities ofAPP/Tau/PS1 AD mice.After the paraffin section,immunohistochemical fluorescence technology was used to detect the expression and distribution of glial fibrillary acidic protein (GFAP) in the brain of mice in the above 6 groups.The expression changes of GFAP,CD45 (inflammation marker) and LC3 (autophagy marker) in the hippocampal tissues of 6 groups were detected by Western blotting.Results In the morris water maze test,the escape latency increased obviously following the increasing of months:the latency of 6 months old mice was significantly prolonged as compared with that of 2 months old mice (P<0.05),which was further significantly increased in 12-month-old mice (P<0.05).In the exploring experiment,with the increasing of months,the times of crossing the original table in mice showed a trend of decline,time ofresidencing in the original platform quadrant was significantly reduced.As compared with that in the WT mice,the GFAP expression in the hippocampus of 8 and 12 months old AD mice was significantly increased (P<0.05); meanwhile,the GFAP expression in 8-month-old mice was also significantly higher than that in 2 months old AD mice (P<0.05).The CD45 expression in 6 months old AD mice was obviously increased as compared with that of WT mice (P<0.05).As to the level of LC3-Ⅱ/Lc-Ⅰ,there was a significant increase in 2 months old AD mice as compared with WT mice (P<0.05),which was further increased in 4 months old AD mice (P<0.05),but decreased in 6 and 8 months old AD mice.Conclusion With the gradually loss of the leaming and memory abilities,inflammatory reaction is significantly increased,and the autophagy level increases at first,and then,decreases in the brain ofAPP/Tau/PS1 AD mice.
5.Research on the optimum reperfusion duration of liver ischemia-reperfusion injury model by occluding proper hepatic artery in rats

Objective To investigate the optimum reperfusion duration of liver ischemia-reperfusion injury (IRI) model by occluding the proper hepatic artery in rats. Methods Fourty-eight Sprague Dawley (SD) rats with mean weight of (200±25) g were randomly assigned to 8 groups by random number table method:IRI-0, 1, 3, 6, 12, 24, 48 h and sham operation (SO) group with 6 rats in each group. The proper hepatic arteries in rats of IRI groups were selectively occluded for 1 h and then blood lfow recovered. Samples of blood and liver tissues were collected at the time points of 0, 1, 3, 6, 12, 24, 48 h of reperfusion. In SO group, samples were collected after the proper hepatic artery was isolated and the ifrst portal was exposed for 1 h. The levels of serum alanine aminotransferase (ALT), aspartate aminotransferase (AST), pathological changes, DNA fragmentation rates, and levels of Ki-67 expression of liver tissues were observed in each group. Measurement data of multiple groups were compared using one-way analysis of variance and LSD-t test. Results The levels of serum ALT in IRI-0, 1, 3, 6, 12, 24, 48 h and SO group were (53±25), (85±20), (96±18), (411±96), (87±19), (81±15), (46±6), (60±14) U/L respectively. The levels of serum AST were (238±63), (364±111), (375±68), (1 291±511), (800±87), (854±218), (484±219), (248±94) U/L accordingly. The levels of serum ALT, AST in IRI 6 h group were the highest (F=36.015, 18.241;P<0.05). The damage of liver tissues in IRI 6 h group was the most serious. The DNA fragmentation rates of liver tissues were (7.5±1.5)%, (9.2±2.2)%, (9.3±2.3)%, (12.6±2.4)%, (6.3±1.0)%, (5.4±0.9)%, (4.5±0.8)%, (4.5±1.1)%accordingly, which was the highest in IRI 6 h group (F=15.992, P<0.05). The levels of Ki-67 expression of liver tissues were (3.5±1.4), (5.6±1.8), (8.7±2.3), (13.7±2.4), (15.2±1.2), (20.5±2.2), (31.8±2.5), (2.4±1.2)/high power ifeld accordingly, which was the highest in IRI 48 h group (F=261.707, P<0.05). Conclusions The liver IRI model can be successfully established by occluding the proper hepatic artery, and the optimum reperfusion duration of IRI is 6 h.
