1.Clinical analysis on laparoscopic improved Soave operation for treating neonatal congenital megacolon

Objective To study the clinical effect of laparoscopy-assisted improved Soave operation for treating neonatal congeni-tal megacolon .Methods 68 cases of neonatal congenital megacolon treated in our hospital from March 2010 to July 2013 were di-vided into 2 groups .The treatment group(34 cases) was performed the laparoscopy-assisted improved Soave operation after the sys-tematic examinations ,while the control group(34 cases) was given the conventional operation therapy .The postoperative curative effects were observed and compared between the two groups .Results The effects of intraoperative bleeding amount ,operative time and the pain score in the treatment group were superior to those in the control group with statistical difference between the two group(P<0 .05);the postoperative hospital stay ,postoperative intestinal peristalsis recovery time and fasting time in the treatment group were superior to those in the control group with statistical difference between the two group (P<0 .05) .Conclusion Laparo-scope-assisted improved Soave operation has better effect for treating neonatal congenital megacolon and should be widely used in clinic .
2.Compliance of hand hxgiene among health care workers

Objective To evaluate the intervention efficacy of hand hygiene of health care workers(HCWs)in a hospital before and after the comprehensive interventions were taken. Methods The baseline investigation stage was in April-June,2013 ,intervention was performed from July,2013,post-intervention efficacy evaluation stage was in October-December ,2013. Hand hygiene compliance was observed by investigators ,and questionnaires were filled out. Results After multiple interventions were taken,hand hygiene compliance rate of HCWs increased from 38.02% to 53.86% (χ2= 181.82,P<0.05),post-intervention compliance rates of hand hygiene improved significantly in departments of internal medicine,surgery and intensive care unit (all P<0.05).After intervention,the overall hand hy-giene compliance rate at different work opportunities rose from 41.87% (1 740/4 156)to 53.45% (2 044/3 824)(χ2=107.2,P<0.05);except sterile manipulation,the other 4 manipulation opportunities were all higher than pre-intervention (all P<0.05);HCWs of different professions were all higher than pre-intervention(all P<0.05).Conclusion Active hand hygiene intervention can effectively improve hand hygiene compliance of HCWs.
3.Effect of down-regulation of integrin β1 on the migrative and adhesive capacity of non-small lung cancer cells

Objective:To construct RNA interference plasmid specific for integrin β1 gene, and to explore the effects of inhibition of integrin β1 protein expression on the biological behavior of non-small cell lung carcinoma(NSCLC) cells. Methods:Genomic sequences of integrin β1 gene was retrieved from GenBank, and cDNA encoding small hairpin RNA(shRNA) for integrin β1 was designed and named as 17-2. A non-specific sequence was designed as negative control and named as N. The cDNA was synthesized and inserted into plasmids pUC19. Recombinant plasmids were then transformed into competent E.coli. The positive clones were selected and recombinant plasmids were extracted.The two shRNA vectors were transfected into NSCLC cell line PC-9/AB2 mediated by lipofectAMINE 2000. The stably transfec-ted cell clones were selected by G418 screening. Fluorescence microscope, real-time fluorescent quantitative (RFQ)-PCR, and Western blotting were performed to examine integrin β1 mRNA and protein expressions.Cell scratch test and adhesion test were used to detect the influences of silencing integrin β1 on cell migration and adhesion capacity. Results:The stably transfected 17.2 cell clones and N cell clones were screened by G418. The green fluorescence-staining cells were observed in full-field under fluorescence microscope. The level of integrin β1 was significantly down-regulated in 17-2 group compared with N group and primary PC-9/AB2 cells.Scratch test and adhesion test showed that the migration and adhesion capacity of PC-9/AB2 cells was significantly reduced after silencing integrin β1. Conclusion:This study successfully constructed integrin β1 specific shRNA expression vector. The expression of integrin β1 was significantly silenced in NSCLC cells transfected with that vecton. Silencing integrin β1 can significantly reduce the migration and adhesion capacity of NSCLC cells.
4.Best formulation and clinical effects of marzulene-S grranuls membrane for oral ulcer

AIM:To choose the best formulation of marzulene-S grranuls oral membrane and to evaluate the clinical effects.METHODS: Orthogonal method was used for studying.Take the appearance,viscidity and dissolubility of the oral membrane as the indexes of scoring 108 cases of patients were chosen and divided into two groups randomly.58 case were treated with marzulene-S grranuls oral membrane and 50 cases were treated with gargarisma chlorhexidine as the control group.RESULTS: The best prescription is sodium carboxymethyl cellulose 4%,polyvinyl alcohol_(1750) 1% and polyvinyl alcohol_(17-88) 3%.CONCLUSION: The design of the formulation is reasonable and the clinical effect was satisfactory.
5.Analysis of the results of multifocal visual evoked potential examination in patients with anterior ischemic opticneuropathy before and after treatment

Objective To observe the results of multifocal visual evoked potential (mfVEP) examination in patients with anterior ischemic optic neuropathy (AION) before and ater treatment,and to probe its clinical significance.Methods A total of 90 patients (90 eyes) with AION were examined by mfVEP;the second-order reaction of mfVEP was analyzed.The reaction was divided into upper and lower hemi-field of visual field,or 1 / 4 quadrant visual field (superior nasal,inferior nasal,superior temporal,and inferior temporal).The sum of waves of each response was analyzed and the results in various regions were compared.The features of wave configuration was compared between the AION eyes and the contralateral eye,and between the AION eyes before and after treatment.Results The amplitude and latency of P-wave of mfVEP was 0.198±0.033 and 100.197±7.354 respectively in AION eyes before treatment,and was 0.271±0.024 and 98.567±6.794 in the contralateral eyes;the difference was significant (t=16.556,18.330;P<0.01).The amplitude and latency of P-wave of mfVEP was 0.229±0.016 and 100.104±10.603 respectively in AION eyes after treatment,which differed much from that before the treatment (t=13.649,8.858;P<0.01) and also from that of the contralateral eyes (t=13.649,8.858;P<0.01).Conclusions The amplitude and latency of P-wave of mfVEP may accurately reflect the recovery of local optic nerve damage in AION eyes before and after treatment with good repeatability.AION can be used as a new method for AION diagnosis and detection of the prognosis.
6.Nursing of patients with mandibular fractures treated with micro titanium plate rigid internal fixation

Objective To summarize the experience of nursing patients with mandibular fractures treated by micro rigid internal fixation.Method The following nursing methods including psychological interventions,pain intervention,dietary intervention, rehabilitation,intervention and prevention of complications were applied in the nursing care to 56 patients with mandibular fractures treated by micro rigid internal fixation.Results The surgical procedures of micro rigid internal fixation were successful,and all of them were cured and discharged.One case contracted gingivitis and another one oral ulcers,both cured by rinse with mouthwash. Conclusion Nursing intervention can improve the rehabilitative effect on patients with titanium mandibular fracture fixation,reduce the complications and improve clinical outcomes.
7.PEDF Inhibits Non?small Cell Lung Cancer Proliferation by Down?regulating VEGF Expression

Objective To analyze the mechanism of pigment epithelium?derived factor(PEDF)regulation in lung cancer cell proliferation and migration by vascular endothelial growth factor(VEGF). Methods VEGF expression was observed after overexpression or silencing of PEDF. Proliferation and migration were analyzed by MTT and transwell assays. Real?time PCR and Western blotting were performed to investigate the mechanism underlying PEDF regulation of the VEGF/SRC/FAK pathways. Results PEDF could inhibit the proliferation and migration of A549 cells by VEGF. Conclusion PEDF can be considered as a potential therapeutic target for lung cancer.
8.Determination of Main Component and the Related Substances in Pirenoxinum Natricum Eye Drops by HPLC

OBJECTIVE:To establish a HPLC method for the determination of main component and the related substances in pirenoxinum natricum eye drops. METHODS: A Inertsil C8-3 column was used with pH2.2 perchloric acid-methanol (35∶65) as mobile phase at a flow of 1.0 mL?min-1. The detection wavelength was 230 nm and the sample size was 10 ?L. RESULTS: Pirenoxinum natricum and the related substances could be baseline separated. The calibration curve was linear in the range of 49.5~990 ng(r=0.999 9). The detection limits was 89.3 pg, and average recovery was 99.1%, RSD was 0.42%. CONCLUSIONS: This method is rapid, accurate sensitive and can be used for the quality control of pirenoxinum natricum eye drops.

Diabetic nephropathy (DN) is one of the most important microvascular complications of diabetes, which is one of the most important causes of death in diabetic patients. Because of its high efficiency and few adverse reactions, TCM has been widely studied and used in the treatment of DN, with unique advantages. The recent research showed that a variety of TCM polysaccharides have the effects of reduce blood sugar and protect kidney. This article reviewed the research on TCM polysaccharides on the prevention and treatment of DN, which can provide references for further study and application.
10.Study of the bone marrow mononuclear cell apheresis procedure and clinical applications

Objective To study the apheresis procedure of collecting mononuclear cells from bone marrow for clinical treatment.Methods Bone marrow mononuclear cells were separated from bone marrow using COBE Spectra apheresis system.Cell morphology,cell counts,positive expression of CD34+,CD133+,CD271+ were detected,and the recovery rate of nucleated cell and mononuclear cell were also calculated.And the main parameters of the procedure with upper efficiency were set via data analysis.Results The efficiency of collection line with colourimetric card of 3% was higher than that of 5% and of 7%.The recovery rate of mononuclear cell is 75% when the cycle times ranged from 7 to 9.The positive expression of CD34+,CD133+ and CD271+ were 1.17%,0.63%,and 0.53%,respectively.Conclution This mononuclear apheresis procedure is safety,pollution-free and efficient,and remaming the bone marrow could be retransfused to the donor.
