1.Application of Cytological CT Technique in Anti-tumor Research in TCM Pharmacology.

By applying advanced technique such as cytological CT the action of 926, a Chinese drug, on ascitis cervical cancer cells was observed. Results revealed that it destroys the DNA and RNA of cancer cell, prevent it from synthesis of nucleic acid and cellular proliferation. These has upgraded the experimental research to a new level.
2.Study on Protective Effect of Astragaloside on Nerve of in Vitro and in Vivo Model of Parkinson Disease

OBJECTIVE: To study the protective effect of astragaloside on nerve of in vitro and in vivo model of Parkinson disease (PD).METHODS: MPP+ induced PC12 cell injury to establish in vitro PD model.PD mice model was induced by MPTP.MTT assay was used to determine the effect of astragaloside on survival rate of PC12 cell induced by MPP+ and the content of LDH and MDA.The effect of astragaloside on spontaneous behavior and the content of striatal DA and HVA were also detected.RESULTS: 25 ?mol?L-1,50 ?mol?L-1,100 ?mol?L-1 dose of astragaloside inhibited the decrease of survival rate of PC12 cell induced by MPP+ in dose dependent manner.10 ?mol?L-1,20 ?mol?L-1,40 ?mol?L-1 dose of astragaloside can obviously enhanced spontaneous behavior of model mice,and reduced the content of striatal DA and HVA.CONCLUSION: Astragaloside can protect nerve of in vivo PD and in vitro PD model.
3.Optimization of image process parameters of digital radiography

ObjectiveTo reduce the effective dose and maintain the image quality by adjusting the image processing parameters in the lumbar spine examinations.Methods This study investigated the influence of image processing parameters on image quality of Philips DR system by evaluating image quality of CDRAD 2.0 phantom.The parameters include detail contrast enhancement,noise compensation,unsharp masking and unsharp masking kernel.The entrance surface dose of phantom was measured by solidose meter.A synthetical parameters optimization project was proposed by analyzing the results of the investigation.This project was also testified by phantoms.ResultsImportant effects were the main effects of DCE,unsharp masking and kernel at the clinically used tube potential of 70 kVp( F =91.45,373.79,429.88,P < 0.05).These effects indicated an increase of the 1QF about 20 units with increasing unsharp masking,while an increase of DCE and kernel led to decrease of IQF about 10 and 21 units.When the tube potential was increased to 85 kV,keeping the settings of the process parameters unchanged,the IQF increased.The results showed statically significant difference ( t =5.31,P < 0.05 ).ConclusionIt is possible to lower the effective dose to the patient by the use of a higher tube potential and maintain a good image quality,and it will have little influence on clinic diagnosis through the most optimal setting of the process parameters.

Taxol could induce tumor cell apoptosis throug h activation of Fas/FasL and caspases. bcl-2 also played an important role in t axol-induced cell apoptosis. Furthermore, taxol had the similar action with lip opolysaccharide on inducing the release of bioactive substances including tumor necrosis factor, interleukin and nitric oxide. There were as well some progresse s in taxol-resistance of tumor cells.
5.Scatter filtration method in digital radiography:The imaging quality and irradiation dose evaluation for scatter filtering template performance

Objective The imaging quality and irradiation dose were compared between scatter filtering template performance and conventional grid in digital radiography.MethodsImages of contrast-detail phantom and ROC statistic phantom corrected with scatter filtering template method and grid were obtained,respectively.The image quality factors(IQF)and sihnal detection probability values(Pdet)and imaging dose were analyzed by ANOVA,SNK and t test statistics for the two methods of scatter removing.ResultsThe difference of IQF and signal detection probability of images corrected by scatter filtering template technique and conventional snd Was statistically significant.ConclusionsIn digital radiography,STFF can effectively filter scattering,improve image quality without increasing the exposure.With the same image quality.it can reduce the entrance surface dose by about 30% than filter grid.In order to effectively filter scattering,improve the quality of X-ray image,and reduce the patient dose.STFF is an alternative method.
6.The effects of simulated family nursing on the cognitive function and activity of daily living in patients with Alzheimer's disease

Objective To evaluate the effects of simulated family nursing on the cognitive function and activity of daily living in patients with Alzheimer's disease.Methods Sixty-eight patients with Alzheimer's disease were assigned to routine nursing condition and simulated family nursing condition.The patients were assessed with Mini-Mental State Examination (MMSE) and Activity of Daily Living Scale (ADL) before intervention,and 3 months and 6 months after intervention.Results Compared with the control group,the patients' cognitive function (t=2.31,P=0.026) and activity of daily living (t=2.59,P=0.012) were improved significantly in the experimental group.Conclusion The simulated family nursing can improve the cognitive function and activity of daily living in patients with Alzheimer's disease.
7.Correlation Analysis of Maternal and Neonatal HBV Test Results

Objective To investigate the risk factors of intrauterine hepatitis B virus (HBV)infection and its preventive measures.Methods 254 cases of pregnant women with positive HBsAg and their neonatuses from Shaanxi People’s Hospi-tal were selected as the research object.The serum of pregnant women were tested HBV markers and HBV-DNA,their neo-natal umbilical cord blood were only detected HBV markers.Results In 254 samples,the positive rates of HBsAg and HBeAg within their neonatal umbilical cord blood were 5.12% and 13.78% respectively;the positive rate of HBV infection in neonatal umbilical cord blood among maternal HBeAg-positive group was 53.33%,far higher than 3.61% in HBeAg-neg-ative group (χ2 =99.44,P <0.001);the positive rates of HBsAg and HBeAg in neonatal umbilical cord blood were elevated along with the increase of maternal HBV-DNA copy (Hc= 13.50,66.70;P < 0.001).The positive rate of HBV-DNA in pregnant women with HBeAg-positive group was 100%,far more than 19.59% in HBeAg-negative group (P <0.001).Con-clusion Both maternal positive HBeAg and HBV-DNA are risk factors for neonatal intrauterine HBV infection.Prenatal HBeAg screening is predominant,especially in some hospitals without real-time quantitative assay.An effective measure to reduce intrauterine HBV infection is to be pregnant when HBV DNA and HBeAg are turned into negative after HBV inter-vention and treatment for women in reproductive age.

Second, this article discusses the teaching results and reflection so as to provide reference for the continuous localization reformation of PBL and further medical curriculum integration in college and university.
9.Ethacrynic acid promotes apoptosis in lung cancer A549 cells when combined with cisplatin chemotherapy

Objective To investigate the killing effect of ethacrynic acid (EA) on lung cancer A549 cells derived spheres and explore the underlying mechanism.Methods A549 spheres were cultured in serum-free medium,and the protein expression of CD133,SOX2,EpCAM and ABCG2 was detected by Western blotting.MTT assay was used to evaluate the cell viability of A549 spheres and A549 cells after treated by 1,2,5,10 and 20 mg/mL cisplatin (DDP) for 48 h.The activity of glutathione S-transferase (GST) was measured by colorimetric method after A549 spheres were treated with 10,50,100 and 200 μmol/L EA,respectively.Flow cytometry,Western blotting,real-time PCR and luciferase assay were used to analyze the levels of cellular reactive oxygen species (ROS),formation of A549 spheres,mRNA and protein expression levels of β-catenin,Sox2 and ABCG2,and promoter activity of β-catenin upon 200 μmol/L EA treated cells for 48 h.A549 sphere was infected with β-catenin adenovirus for 24 h,followed by 200 μmol/L EA treatment (in presence or absence of 5 mg/mL DDP) for 24 h.The expression of β-catenin,Sox2 and ABCG2 at mRNA and protein levels was detected by real-time PCR and Western blotting,and cell growth of A549 spheres was evaluated by MTT assay.Results The A549 spheres,with high expression of tumor stem cells markers CD133,SOX2,EpCAM and drug resistance related molecule ABCG2,and resistance to DDP at different doses,were successfully derived.After 200 μmol/L EA had treated A549 sphere for 48 h,the levels of ROS were significantly increased (P < 0.05),and the mRNA and protein levels of β-catenin,Sox2 and ABCG2,and promoter activity of β-catenin were notably decreased (P < 0.05).The treatment of 200 μmol/L EA enhanced the inhibitory effect on proliferation and the promoting effect on apoptosis in A549 spheres induced by 5 mg/mL DDP (P < 0.05).Up-regulation of β-catenin by adenoviral infection partly reversed the effects of 200 μmol/L EA on suppressing the expression levels of β-catenin,Sox2 and ABCG2,compared to the spheres infected with blank adenovirus.Additionally,β-catenin over-expression significantly remitted the inhibitory effect of 200 μmol/L EA and 5 mg/mL DDP on the proliferation in A549 spheres.Conclusion EA exerts inhibitory effect on the proliferation and stemness of A549 spheres through suppressing GST activity and β-catenin expression,and then promotes cell apoptosis.EA might be a novel drug in treatment of lung cancer and cancer stem cells.
10.A pilot study of the ERCC1 and XPF genes in forensic age estimation

Objective The aim of this study is to detect the mRNA and protein expression levels of ERCC1 and XPF genes among different age groups of healthy Chinese Han individuals,and to analyze the correlation between the mRNA and protein expression levels andthe age of individuals in order to find new molecular markers for forensic age estimation.Methods Peripheral blood samples were obtained from 150 unrelated healthy Chinese Han individuals.The plasma was centrifuged from the whole blood by gradient centrifugation,and the totalRNA was extractedwithTrizol fromperipheral blood mononuclear cells(PBMCs).Real-time quantitative reverse transcription polymerase chain reaction (RT-qPCR) was used to quantitatively analyze the mRNA relative expression levels of ERCC1and XPF in PBMCs.Enzyme linked immunosorbent assay (ELISA) was used to quantitatively analyze the protein expression levels of ERCC1and XPF in plasma.Results There were no significant differences in the mRNA relative expression levels of ERCC1 and XPF in PBMCs between males and females(P>0.05).Significant differences were found in the mRNA relative expression levels of ERCC1 and XPF between different age groups (P<0.05).Regression analysis showed thatthe mRNA relative expression levels of ERCC1 and XPF were both negatively correlated with age.The correlation coefficients(r) were-0.578 and-0.844,respectively.When the age was used as independent variable(x) and the mRNA expression relative level as dependent variable (y),the fitting curveswere Y=3.3E-5X2-0.0261X+1.9175 (R2=0.3244,P<0.01),Y=0.0003X2-0.0459X+2.0439 R2=0.729,P<0.01),respectively.There were no significant differences inthe protein expression levels of ERCC1 and XPF in plasma between different age groups or genders (P>0.05).Conclusion The mRNA relative expression levels of ERCC1 and XPF in PBMCsdeclined with the increase of age,however,the protein expression levels in plasma were unrelated to age.ERCC 1 and XPF genes can be used asnew molecular markers for forensic age estimation,so as to providetheoretical basis for establishing the mathematical model of ERCC1/XPF genesin concern ofindividual ages.
