1.Optimization of transfection conditions for transfecting shRNA to endothelium of mouse angioma cells mediated by lentiviral vector

Objective To optimize the transfecfion parameters for transfecting small interfering RNA (siR-NA) to endothelium of mouse angioma (EOMA) cells mediated by lentiviral vector. Methods According to Poly-brene existing or not(8 μg/ml), series of multiplicity of infection (MOI) (1, 5, 10, 15, 20)and the time of infec-tion(4, 6, 12,24 h), EOMA cells were divided into 40 groups. 140 hours after transfection, cells were counted un-der fluorescent microscope to determine the percentage of the cells transfected under each condition. The cell viability was calculated by using Typan Blue. Results The percentage of positive cells was as high as 83% in the group with Polybrene (8 μg/ml), MOI(5) and the time of tranfectian (6 h), and the livability of EOMA in this group was 96%. After that, the transfection efficiency was not increased with the increasing of MOI, the time of transfection, but the mortality of EOMA was increased markedly. Conclusion The transfection efficiency and cell viability are dependent on the Polybrene(8 μg/ml),correct MOI (5)and the time of transfection (6 h).
2.STUDY OF THE AGE AND COMPARATIVE ANATOMY ABOUT THE ACTON OF LEVATOR AND TENSOR VELI PALATI MUSCLESON THE EUSTACHIAN TUBE

The attachments of eustachian tube, levator and tensor veli palati muscles on the base of the skull were measured from the skulls of human adults, infants, newborns ad well as monkeys, dogs and rabbits, respectively. Anatomical characteristics of the levator and tensor veli palati muscles that related to eustachian tubal ventilation and skull morphology were analysed. In addition, some head specimens were dissected for contrast. Levator veli palati muscle (LVPM) lies in the posterioinferior of the eustachian tube. The angles between LVPM and median saggital plane and the angles between LVPM and horizontal plane of palate are larger and their ability of raising the eustachian tube is more effective in the adult than in the newborn and animal. From the eustachian tube to the hamulus, tensor veli palati muscle (TVPM) descends almost vertically in the adult, but ventroward in the newborn and animals, which suggested their different actions upon eustachian tube. The skull morphology of the human adult differs from child and animal, which changes the position and function of LVPM and TVPM. In conclusion, eustachian tube was opened by LVPM chiefly in the adult while by TVPM in the newborn and animal. The incidence of otitis media with effusion is higher in the infant as it is the trasitional stage that the action of TVPM had been decreased while that of LVPM would not have been established.
4.Left ventricular diverticulum:case report and review of the literature

Objective To investigate the characteristic and diagnosis of left ventricular diverticulum. Meth-ods As one left ventricular diverticulum patient was diagnosed by echocardiography in our hospital,we reviewed and analyzed about published in China. Results Left ventricular diverticulum is a rare disease of congenital heart malfor-marion, Echocardiographic characteristics of the left ventricular diverticulum is the saccular evngination of the left ven-tricular wall with the neck of the diverticula being smaller than the body. Left ventricular diverticulum is classified into contractile muscular type and fibrous type ,The saccular wall of contractile muscular diverticulum remain a normal mo-tion and is thinner than the normal wall The saccular wall of the fibrous diverticulum is due to a loss of myocardial thickness, as thin as aneurysm with a dyskinetic motion. Conclusion Left ventricular diverticulum is a rare disease in clinic, echocardiography is very important for diagnosing classifying.
5.Expression and significance of the lysine-specific demethylase 1 in gastric cancer tissues

Objective To investigate the expressions and relationship between lysine-specific demethylase 1 (LSD1) and E-cadherin protein in gastric cancer tissues and adjacent normal tissues,and the correlation with the clinicopathological features and prognosis of patients with gastric cancer.Methods The case-control study was adopted.The gastric cancer tissues and adjacent normal tissues were collected by surgical resection from 80 patients with gastric cancer who were admitted to the Xiangya Hospital of Central South University from June 2008 to June 2009.Expressions of LSD1 and E-cadherin protein were detected by immunohistochemistry (IHC).The follow-up of telephone interview was performed to detect survival of patients till June 2015.Relationships between the expressions of LSD1 and E-cadherin protein and clinicopathological features or prognosis of patients were analyzed.Comparison of count data and correlation were analyzed by the chi-square test and Spearman rank correlation analysis.Survival curve was drawn using the Kaplan-Meier method,and survival analysis was done using the Log-rank test.Results Expressions of LSD1 in cancer tissues and adjacent normal tissues were located at the cell nucleus.The positive expression rate of LSD1 was 67.5% (54/80) and 43.8% (35/80) in cancer tissues and adjacent normal tissues,respectively,with a significant difference (x2=9.141,P < 0.05).Expressions of E-cadherin protein in cancer tissues and adjacent normal tissues were located at the cell membrane and cytoplasm.The positive expression rate of E-cadherin was 63.8% (51/80) and 81.3% (65/80) in cancer tissues and adjacent normal tissues,respectively,with a significant difference (x2 =6.140,P < 0.05).The positive expression rate of LSD1 was 83.3% (25/30),76.0% (19/25) and 40.0% (10/25) in the low-,moderate-and high-differentiated tumors,37.5% (6/16),72.7% (16/22),71.9% (23/32) and 90.0% (9/10) in the Ⅰ,Ⅱ,Ⅲ and Ⅳ stages of TNM stage,36.4% (4/11) and 72.5 % (50/69) in patients without and with lymph node metastasis,respectively,showing significant differences (x2=12.870,9.425,4.111,P < 0.05).The positive expression rate of E-cadherin protein was 53.3% (16/30),56.0% (14/25) and 84.0% (21/25) in the low-,moderate-and high-differentiated tumors,75.0% (12/16),63.6% (14/22),71.9% (23/32) and 20.0% (2/10) in the Ⅰ,Ⅱ,Ⅲ and Ⅳ stages of TNM stage,100.0% (11/11) and 58.0% (40/69) in patients without and with lymph node metastasis,70.0% (49/70) and 20.0% (2/10) in patients without and with distant metastasis,respectively,showing significant differences (/x2 =6.494,10.073,5.547,7.426,P < 0.05).There was a negative correlation in expressions between LSD1 and E-cadherin protein (r =-0.355,P < 0.05).The survival time and 5-year overall survival rate of patients with positive and negative expressions of LSD1 were (36.9 ± 2.5) months and 31.1%,(47.4 ± 3.4) months and 56.0%,respectively,showing a significant difference (x2=4.550,P <0.05).The survival time and 5-year overall survival rate of the patients with positive and negative expressions of E-cadherin protein were (44.0 ± 2.5) months and 46.7%,(32.6 ± 3.5) months and 24.9%,respectively,showing a significant difference (x2 =7.306,P < 0.05).Conclusions Positive expression of LSD1 in gastric cancer tissues is higher than that in adjacent normal tissues.There are increased expression of LSD1 and reduced expression of E-cadherin protein in low-differentiated gastric cancer tissues and high TNM stage,showing a negative correlation between them.Positive expression of LSD1 and negative expression of E-cadherin protein may indicate a poor prognosis.

Tagetes L.,the genus in the family Asteraceae,consists of about 30 species spread in South and Middle America as well as Mexico.More than one hundred secondary metabolites have been obtained in phytochemical investigation on the species,some of which have potent biological activities.The advances in phytochemical studies and biological activities of the plants in Tagetes L.from 1925 to 2011 are summarized in this paper.
7.Design of bridge type bone plate series and their clinical application

Objective To investigate the clinical effects of bridge type bone plate series designed by ourselves. Methods A sort of bridge type bone plate series were designed according to mechanics and structure of bridges. They were used in the internal fixation of 12 shinbone fractures and 41 fractures of middle and lower parts of thighbone. The treatment results were followed up and statistics was done to evaluate fracture healing at 8, 12, 16, 20, and 24 weeks respectively. Results Of the 53 fractures, 48 achieved clinical union and 27 solid union within 12 weeks, 51 reached clinical union and 47 solid union within 16 weeks, 53 reached clinical union and 52 solid union within 20 weeks, and all reached solid union within 24 weeks. Conclusion The self designed bridge type bone plate series are a nice internal fixation device for clinical treatment of fractures.
8.Bone grafts for scaphoid fracture nonunion: a review of research advances

In all the carpal fractures, scaphoid fracture is the most common in clinic (from 60% to 70% in proportion) and likely leads to nonunion. If nonunion is not treated in time, it probably causes instability of the scaphoid lunate joint, further leading to scapholunate advanced collapse. Its comprehensive manifestations include degenerative arthropathy of radial styloid process, radial scaphoid joint, capitulolunate joint and even total wrist joint, eventually leading to wrist joint dysfunction. Therefore, more and more attention has been paid to treatments of scaphoid fracture nonunion. Bone grafting is the most common practical treatment, but new surgical procedures have emerged in recent years. This article thus reviews the research advances in bone grafting for scaphoid fracture nonunion, commenting on the advantages and disadvantages of various techniques.
9.The correlation study between spatial memory and hippocampal long term potentiation in rats

ObjectiveTo investigate the correlation of hippocampal synaptic plasticity with spatial learning and memory under normal and pathological condition,and provide experimental evidence for the coincidence of hippocampal late-phase long-term potentiation (L-LTP) and behavioral experiments.Methods 38 SD rats were randomly divided into two groups,control and AD model.First,Morris water maze was used to test the ability of spatial learning and memory of rats.The escape latencies for rats to search for an underwater platform in 5 days of navigation tests and the swimming time percentage in target qtuadrant on the 6th day after withdrawing the platform in probe trails were recorded.Then,in vivo hippocampus L-LTP of field excitatory postsynaptic potential (fEPSP)in CA1 region was recorded after delivering high frequency stimulation (HFS).ResultsBilateral intrahippocampal injection of 4 nmol amyloid β peptide ( Aβ 25-35 ) significantly impaired spatial learning and memory of rats in water maze tests,as well as in vivo hippocampal L-LTP.In control group,there was a significant negative correlation between the amplitude of fEPSP and the escape latency ( r =-0.8306,P < 0.01 ) and a significant positive correlation between the amplitude of fEPSP and the swimming time percentage in target quadrant ( r=0.7709,P<0.01 ).In AD model group,similar correlations were found,with a correlation coefficient of r =-0.7675 (P <0.01 ) and r =0.8049 (P < 0.01 ),respectively.When putting all data from the two groups together,the hippocampal L-LTP was more correlated with escape latency ( r =-0.9124,P < 0.01 ) and swimming time percentage ( r=0.9745,P<0.01).ConclusionThere is very close correlation between the hippocampal L-LTP and the spatial learning and memory behavior in rats,suggesting that the hippocampal L-LTP may be involved in the electrophysiological mechanism of spatial learning and memory in rats,and the impairment of L-LTP could partly represent the deficits in cognitive function of animals.
10.Experience and procedure of the model of rat heterotopic heart transplantation

ObjectiveTo summarize the method and experience of the model of rat heterotopic heart transplantation and to improve the rate of success of the experiment.Methods120 heterotopic heart transplantations were performed by Ono's way,including 60 prepared transplantations and 60 formal transplantations.ResultsRate of success of prepared transplantations was 45%,and formal transplantation was 85%.Mean time of formal operation was (74.3±17.1)min,removal time of the donor heart was (7.9±4.2)min,ischemia time was (29.6±10.3) min and the survival time of allograft was (11.1±5.7)days.5 of the recipients had a long-term survival,and the maximum weight was 740 g.ConclusionsGood surgical technique and rigorous perioperative management is very important in setting up the rat model of heterotopic heart transplantation.
