To characterize the ultra-fine particles of Atractylodes lancea (Thunb.) DC., Phellodendron chinense Schneid and the ERMIAO PILL*, a compounded preparation of the above two drugs, for the purpose to observe a deeper view on their physico-chemical properties. Methods By comparing their particle size, porosity and specific surface area with light and scanning electron microscope. Results The size of the ultra-fine particles were more uniform in size, 90% of which were under 20 μm, their specific area were increased by 60% ~ 190% and bulk density were around 0.42 g/cm3, and the great majority of the plant cells were broken as compared with the conventional coarse powder. Conclusion Both A. lancea and P. chinense together with their combined preparation became smaller in particle size, more uniformly distributed with increased specific surface area and broken cell wall.
2.Effect of Individual Rehabilitation Nursing Guidance on the Initiative of Rehabilitation Therapy of Patients with Spinal Cord Injury

Objective To explore the effect of individual rehabilitation nursing guidance on the initiative of rehabilitation therapy of spinalcord injury patients. Methods 50 spinal cord injured patients were divided into conventional group (group A) and individualized group(group B). Group A was given conventional rehabilitation nursing guidance. Group B was given special rehabilitation nursing guidance consideringthe patients' and their families' requirement. Initiative of rehabilitation therapy was assessed between group A and group B beforeand 2 months after intervention. Results Group A had no significant increasing of initiative assessment score (P<0.05) 2 months, while theinitiative assessment score of group B increased significantly (P<0.01), even group B was better than group A (P<0.05) after the intervention.Conclusion Individual rehabilitation nursing guidance may facilitate to improve the initiative of SCI patients.

Object To investigate the effects of ultra-fine powder technique and granularity of pellets on dissolution rate in vitro. Methods The dissolution rate of ultra-fine ERMIAO PILL with different granule diameters in vitro was measured and compared with the index of berberine by UV spectro-photometry. Results The dissolution parameters T 50 and T d of four kinds of ultra-fine ERMIAO PILL are 61.60, 19.48, 17.84, 8.97 min and 102.3, 33.29, 26.98, 14.77 min, respectively. Those of general powder ERMIAO PILL with granule diameter of 2.4 mm are 89.61 and 155.68 min. Conclusion The dissolution rate of ultra-fine powder is quicker than that of general powder, and the rate increases with the granularity of PILL decreasing.

Objective To explore the relationship between myocardial glucose metabolism in the right heart and cardiac function in patients with idiopathic dilated cardiomyopathy (IDCM).Methods From October 2010 to May 2013,64 consecutive patients (45 males,19 females; mean age:(53 ± 12) years) with IDCM were enrolled in this study.All patients underwent 18F-FDG myocardial PET imaging,late-gadolinium enhancement cardiac MRI (cMRI-LGE) and UCG.TrueD software was used to determine the SUVmax in right (SUV-R) and left (SUV-L) ventricular free wall,and the ratio of SUV-R to SUV-L (SUV-R/L) was calculated.The cMRI-LGE was used to evaluate LVEF,RVEF and LGE in left heart.Pulmonary artery systolic pressure (PASP) was measured by UCG.Two-sample t test and Pearson correlation analysis were used to analyze the data.Results Both SUV-R and SUV-R/L had negative correlation with LVEF (r=-0.48,-0.33; both P<0.01) and also with RVEF (r=-0.45,-0.44; both P<0.01).SUV-R/L positively correlated with PASP (r =0.58,P<0.01).SUV-R/L of patients with and without LGE were statistically different (0.45±0.18 vs 0.36±0.12; t=2.50,P<0.05).SUV-R and SUV-L of patients with LGE were 2.0±0.9 and 4.7±1.6,and those of patients without LGE were 1.7±0.9 and 5.0±2.8.Both differences of SUV-R and SUV-L between the two groups were not statistically significant (t=0.54 and 0.63,both P>0.05).Conclusions Myocardial glucose metabolism in right heart is closely correlated with left and right ventricular function.The assessment of 18F-FDG uptake in right heart by PET imaging is potentially useful for evaluation of the severity and prognosis in IDCM patients.
6.Acute T-B cell biphenotypic leukemia:report of four cases with literature review

Objective To strengh the awareness of acute T-B cell biphenotypic leukemia.Methods Four new cases of acute T-B cell biphenotypic leukemia were reported and the related literature were reviewed.Results Fourteen patients with acute T-B cell biphenotypic leukemia.including 4 diagnosed at our hospital and other 10 cases reported in literature,were retrospectively analyzed.Similar clinical fleatures as the typical acute lymphocytic leukemia(ALL) were presented.Ten of 14 cases were male and were within 17 to 46 years old.The disease were refactory to the conventional ALL chemotherapy regimens and deteriorated progressively.Six patients died within 12 months after their diagnosed with the median survival time of ten months. Conclusion Acute T-B cell biphenotypic leukemia is one of the most rare type of leukemia.A better understanding of the clinical and hematological features of this type of leukemia and new therapeutic strategies are needed.
7.Study of dopamine-producing cells derived from human umbilical cord mesenchymal stem cells in treatment in rats with Parkinson’s disease

Objective To investigate the efficacy of dopamine producing cells (DPCs) derived from human umbilical cord-derived mesenchymal stem cells (hUC-MSCs) in treatment in model rats with Parkinson’s disease (PD). Methods The cultured hUC-MSCs were induced into DPCs in vitro. The dopamine (DA) and glial cell line-derived neurotrophic factor (GDNF) expression levels were detected by ELISA to identify the DPCs. The PD rat model was established by injecting 6-OHDA into the right substantia nigra (SN). A total of 60 successfulled PD model rats were randomized into hUC-MSCs-DPCs group (5 × 105 hUC-MSCs-DPCs were transplanted into right striatum, n=20), hUC-MSCs group (5 × 105 hUC-MSCs were transplanted, n=20) and control group (same volume PBS, n=20). All the transplanted cells were labeled with CM-Dil. The apomorphine induced rotation behavior was assessed at 4, 8 and 12 weeks after cell transplantation. The rats were executed after 12 weeks. The immunofluorescence staining was used to detect the tyrosine hydroxylase (TH) expression level, and FLUORO-JADE? C staining was used to test the apoptotic neurons in brain of rats. Results The hUC-MSCs-DPCs were induced successfully in vitro, which showed a high expression level of DA and GDNF. Furthermore, at 4, 8 and 12 weeks after cell transplantation, the rotation behavior was improved, and expression levels of GDNF were significantly higher in hUC-MSCs-DPCs group than those of hUC-MSCs group and control group (P<0.05). In addition, we found that most of the transplanted TH+hUC-MSCs-DPCs at the right striatum and a few cells around both the left and the right substantia nigra at 12 weeks after transplantation. The apoptotic neurons were decreased after cell transplantation in hUC-MSCs-DPCs group than that of control group (P<0.05). Conclusion The hUC-MSCs-DPCs can improve the rotational behavior induced by apomorphine in PD model rats, which may be involved in improving levels of DA and GDNF in damaged striatum and protecting neurons.

9.Relationship between high-expressed TL1A and level of IFN-γ secreted by T cells in acute stage of Guillain-Barr(e) syndrome

Objective To probe the relationship between the expression of TL1A and the level of IFN-γ secreted by T cells in the acute stage of Guillain-Barre syndrome (GBS). Methods ① Six-week female Bal b/c mice were immunized by purified recombinant human soluble TNF-like molecular 1A (rhsTL1A) protein. The polyclonal antibody against rhsTL1A was identified by immunofluorescence using human umbilical vein epithelial cells (HUVEC). ② To detect the biologic activity of rhsTL1A, the peripheral blood mononuclear cells (PBMC) from the healthy donors were separated by Ficoll gradient centrifugation and were seeded on 96-well plates with medium containing 2 μg/ml PHA (control group), 2 μg/ml PHA + 25 ng/ml rhsTL1 A, 2 μg/ml PHA + 100 ng/ml rhsTL1A and 2 μg/ml PHA + 400 ng/ml rhsTLlA respectively. T cell proliferation assay was carried out using ~3H-TdR. ③ IFN-γ productions in the sera of the children with GBS in the acute stage were detected by ELISA. ④ The ratio of CD_3~+ TL1A~+ T cells to CD_3~+ T cells in the peripheral blood of the children with GBS in acute stage was detected with flow, cytometry. ⑤PBMC from the children in acute GBS were separated and cultured in the environment adding 2 μg/ml PHA and 400 ng/ml rhsTL1A in vitro. Then, the IFN-γ in the supernatant was determined by ELISA kit after 72 hours. Results ① hTL1A A expressed by eukaryotic HUVECs was recognized by rhsTL1 A polyclonal antiserum. ② The result of T cell proliferation assay showed that SI of 25 ng/ml rhTL1A, 100 ng/ml rhTL1A A and 400 ng/ml rhTL1A group was increased compared with control group. The SI of 2 μg/ml PHA +400 ng/ml rhsTL1 A group was the highest (2. 65) among them. ③ IFN-γ productions in the sera of the children with GBS in the acute stage ((102. 25±22. 17) pg/ml) were increased significantly compared with healthy control ((28.75 ± 1.31) pg/ml, t = 3. 309, P < 0. 05). ④ The ratio of CD_3~+ TL1A~+ T cells to CD_3~+ T cells in the peripheral blood of the children with GBS in acute stage (18.22%± 1.83%) was enhanced significantly compared with healthy control (5. 17% ±0. 48%, t = 6. 884, P < 0. 01). ⑤ PBMC both in healthy control and the acute GBS secreted more IFN-γ markedly ((43.56± 4.41) pg/ml and (180.64 ± 38.39) pg/ml) after being incubated in 2 μg/ml PHA and 400 ng/ml rhsTL1A (t =4. 523 and 2. 600, P <0. 01 and 0. 05 respectively). Moreover, PBMC in acute GBS secreted more IFN-γ, than that of the healthy group markedly (t = 3. 545, P < 0. 05). Conclusions ① The mouse antiserum recognizing rhsTL1A is successfully obtained. ② In this study, 400 ng/ml rhsTL1A promotes the proliferation of T cells activated by 2 μg/ml PHA, indicating that rhsTL1A has biological activity. ③ The expression of hTL1A of activated T cells in the peripheral blood of the children with acute GBS is up-regulated. These TL1A proteins promote the secretion of IFN-γ through binding to their receptors DR_3.
10.Correlation between the deletion of phosphatase and tensin homolog gene on chromosome 10, syncytin-1 and the damage of maternal endothelium and trophoblast in preeclampsia placenta

Objective:To study the correlation between phosphatase and tensin homolog deleted on chromosome ten(PTEN), Syncytin-1 in preeclamptic placenta and maternal endothelial injury, trophoblast injury.Methods:From February 2015 to December 2017, 46 patients diagnosed as preeclampsia and delivered in Xiaolan Hospital Affiliated to Southern Medical University and 80 pregnant women with normal delivery in the same period were selected.Before delivery, maternal peripheral blood was collected and the content of endothelial injury markers was determined; after delivery, the placenta was retained and the expression of PTEN, Syncytin-1, apoptosis gene and the contents of oxidative stress markers were measured.Results:The mRNA expression of PTEN in placental tissue of preeclampsia was significantly higher than that of normal placental tissue( t=32.797, P<0.000), and the mRNA expression of Syncytin-1 was significantly lower than that of normal placental tissue( t=28.506, P<0.001); the content of ET-1 and sFlt-1 in the blood circulation of the maternal body of preeclampsia was significantly higher than thsoe of normal pregnant women( t=39.884, 40.243; all P<0.000) and positively correlated with the mRNA expression of PTEN in placenta(correlation coefficien t=0.674, 0.596; P=0.004, 0.007), negatively correlated with the mRNA expression of Syncytin-1 in placenta(correlation coefficien t=-0.615, -0.637; P<0.009, 0.008); the content of NO, PGI2, VEGF and PLGF were significantly lower than thse of normal pregnant women( t=30.944, 27.404, 32.827, 26.644; all P<0.001) and negatively correlated with the mRNA expression of PTEN in placenta(correlation coefficien t=-0.648, -0.592, -0.537, -0.613; P=0.006, 0.009, 0.013, 0.007), positively correlated with the mRNA expression of Syncytin-1 in placenta(correlation coefficien t=0.632, 0.572, 0.537, 0.654; P<0.010, 0.013, 0.015, 0.009); The contents of NOX4, MDA, 8-OHdG and the mRNA expression of Fas, FasL, Bax, GRP78, CHOP, Caspase-3 in placental tissue of preeclampsia was significantly higher than that of normal placental tissue( t=27.714, 27.704, 26.786, 27.964, 22.622, 25.496, 25.893, 30.837, 28.975; all P<0.001) and positively correlated with the mRNA expression of PTEN in placenta(correlation coefficien t=0.703, 0.651, 0.593, 0.628, 0.449, 0.552, 0.612, 0.569, 0.502; P<0.003, 0.007, 0.004, 0.006, 0.016, 0.010, 0.006, 0.012, 0.018), negatively correlated with the mRNA expression of Syncytin-1 in placenta(correlation coefficien t=-0.615, -0.714, -0.577, -0.548, -0.512, -0.585, -0.619, -0.495, -0.662; P<0.009, 0.001, 0.013, 0.015, 0.018, 0.011, 0.009, 0.023, 0.003); The contents of PrxII, PrxIII, SOD and mRNA expression of Bcl-2 in placental tissue of preeclampsia was significantly lower than that of normal placental tissue( t=26.115, 23.502, 17.194; all P<0.000) and negatively correlated with the mRNA expression of PTEN in placenta(correlation coefficien t=-0.713, -0.657, -0.591, -0.642; P<0.001, 0.006, 0.009, 0.007), positively correlated with the mRNA expression of Syncytin-1 in placenta(correlation coefficien t=0.562, 0.711, 0.682, 0.608, P<0.013, <0.001, 0.002, 0.008). Conclusion:The high expression of PTEN and the low expression of syncytin-1 in placenta of preeclampsia can aggravate the damage of endothelium and trophoblast.
