Objective To investigate the effects of short-term sleep deprivation(SD)at different time during night on serum cortisol level and emotion state of healthy men,and to find out the influence of work shift onto human circadian rhythm.Methods Two periods of sleep-time were deprived in the present study:the late-night SD(permitted to sleep from 0:00 to 3:00 each day)and the early-night SD(permitted to sleep from 3:00 to 6:00 each day).Ten healthy male adults were chosen to undergo the late-night SD first,followed by normal sleeping for eight nights for washout,and then underwent early-night SD.Each SD period lasted 6 days,i.e.1 day before SD,4 days of SD and 1 day after SD for recovery.Fasting blood samples,for detection of serum cortisol,were collected at 7:00 am every SD day,and the state anxiety inventory(S-AI)scoring was done at the same time.Results The cortisol levels were lowered after 2 types of SD(P0.05).Dependability analysis showed that the cortisol level during the late-night SD period was negatively related to the SD days(r=-0.954 7,P0.05).Both types of SD could elevate anxiety scores,which positively correlated with the SD days(late-night SD:r=0.990,P

Objective The methods for determination of Chlorogenic Acid(CA) in Eucommia ulmodies Oliv were established and determinated the content of CA. Extract solvent, method, time and times were studied and HPLC method for determination was used. Methods The HPLC system consisted of the column of Lichrospher C18 (4.6 mm?250 mm, 5 ?m) and acetonitrtle-water-phosphoric acid (13∶86.5∶ 0.5) as the mobile phase. The temperature was 25℃. The detective wavelength was 327nm. The flow rate was 0.8 mL/min. External standard method was used. Results The powder of eucommia was extracted twice with 60%methanol-water solution at 100 ℃, the first time for 1.5 h,the second for 0.5 h was the optimum one. The method was linear within the range of 2.22?10-2～55.40?10-2 ?g, the average recovery was 99.55%, and the RSD was 1.98% (n =6). Conclusion The method is simple, sensitive and highly reproductive, and it is suitable for ananlysis of CA in Eucommia ulmodies Oliv.

Objective To study and the succession of sarcosaphagous insects and their regular activity on carcass in Xinxiang area. Methods Six rats and five rabbits were sacrificed and placed at the remote grassland of Xinxiang Medical University in July 2015. The main insect species and the decomposition process were observed. The different decomposition stages and the community succession of insects were recorded. Results Seven main species could be identified belonging to 3 families from Diptera, main including Musca domestica, Chrysomya megacephala, Lucilia sericata, Boettcherisca peregrina, Helicophagella melanura. Muscina stabulans and Fannia prisca. And showed regulations of the succession of sarcosaphagous flies community. Conclusion It may be used for estimating postmortem interval in Xinxiang area.

Objective To identify the gene locus and the mutation of DSRAD (double-stranded RNA adenosine deaminase) in a Chinese dyschromatosis symmetrica hereditaria(DSH) family. Methods After confirming the diagnosis of the DSH proband, the genomic DNA was extracted from the whole blood samples of every members of the pedigree. The DSRAD gene intervals were localized by linkage analysis and haplotype reconstruction. The mutation of DSRAD was detected by direct sequencing. Results The candidate gene was localized at the 1q region, consistent with the reported region. The direct sequencing results showed that there was a CAA→TAA transition at exon 2 of DSRAD in all affected family members, which consequently led to a nonsense mutation of Gln517Ter. Conclusion A nonsense mutation is found in the Chinese DSH family.

Objective To study the acute toxicity of ginkgolic acids from Ginkgo biloba sarcotestas against fish.Methods The ginkgolic acid mixture was obtained from Ginkgo biloba sarcotestas.The acute toxicities of the ginkgolic acid mixture against the fry of Cyprinus carpio var.and fry of Carassius auratus were detected.Results LC50 and LC90 of the ginkgolic acid mixture against the fry of Cyprinus carpio var.were 1.805 mg/L and 2.191 mg/L,respectively.LC50 and LC90 of the ginkgolic acid mixture against the fry of Carassius auratus were 1.930 mg/L and 2.217 mg/L,respectively.Conclusions The toxicity of ginkgolic acids against the fry of Cyprinus carpio var.and fry of Carassius auratus is medium.

Objective To investigate the analgesic efficacy and safety of parecoxib-sodium in patients undergoing laparoscopic cholecystectomy. Methods Ninety patients received laparoscopic cholecystectomy were divided into parecoxib-sodium group,flurbiprofen group and control group by random digits table, 30 cases in each group, all patients received the drug by vein 30 minutes before surgery. The postoperative pain intensity (VAS score), satisfaction of patients, changes of blood coagulation function and maximum agglutination rate (MAR) of platelet agglutination test before and after treatment and the incidence of postoperative adverse reaction were observed and compared. Results Pestoperative VAS scores and psychiatric symptoms at different time in parecoxib-sodium group and flurbiprofen group were significantly lower than those in control group (P ＜ 0.05), analgesic satisfaction rate were 93.3% (28/30) in parecoxibsodium group and 90.0% (27/30) in flurbiprofen group, which were significantly higher than that in control group [20.0%(6/30)] (P＜0.05). There were no significant difference in blood coagulation function and MAR before and after treatment among three groups (P ＞ 0.05). Conclusion Parecoxib-sodium displays significant analgesic effects and low rate of adverse events for the patients undergoing laparoscopic cholecystectomy, it is worthy of clinical application.

Objective To observe changes in the working memory and brain functional imaging on functional magnetic resonance imaging(fMRI) after 36 hours sleep deprivation (SD) in healthy volunteers and to explore the possible mechanism of the changes.Methods FMRI scannings were performed in ten male healthy young volunteers before and after 36 hours SD and results were analyzed using SPM2 software.Subjects were also tested LTR and PLUS task to measure the persistence and operation of working memory before and after 36 hours SD.Results The reaction time of LTR task after 36 hours SD ( (866 ± 102) ms)was significantly longer than that before SD ( (754 ± 91 ) ms, t = 2.59, P < 0.01 ).The reaction time of PLUS task after SD ( (848 ± 94) ms) was significantly longer ( t = 2.37, P < 0.05 ) than that before SD ( (756 ± 79) ms).The error rate of LTR task after SD (95.3% ± 3.56% ) was significantly higher (t=3.52,P < 0.01 ) than that before SD (84.8% ± 8.71% ).The error rate of PLUS task after SD (95.7% ±4.72% ) was significantly higher (t =3.38 ,P <0.01 ) than that before SD (84.2% ±9.66% ).There were no significant differences between the two tasks.The frontal and parietal lobes, anterior cingulate gyrus and thalamus were activated during memory tasks testing before SD.Brain activation was broader and stronger in PLUS task than in LTR task.After SD, activation in parietal lobe was decreased and activation in prefrontal and thalamus was increased significantly.Conclusions The working memory performance decreased after SD.Both LTR and PLUS tasks of working memory activate frontal and parietal lobes, anterior cingulate gyrus and thalamus.The activation of parietal lobe decreased and the activation of prefrontal lobe and thalamus increased after 36 hours SD.This is the possible mechanism of SD to causes the cognition decline.

In this study, daphnetin and its major metabolites in the intestinal wall of rats were identified by liquid chromatography and quatrupole-time of flight mass spectrometry. Perfusion fluid of duodenum, jejunum, ileum and colon were collected separately for 2 hours from the rat intestine following perfusion with daphnetin. The metabolites of daphnetin in the perfusion fluid of different intestine segments were analyzed by the liquid chromatography and quatrupole-time of flight mass spectrometry. It is shown that the parent drug daphnetin and four metabolites were found in the perfusion fluid of duodenum, jejunum and ileum. However, no metabolites were found in the colon. Among the four metabolites, two daphnetin sulfates (m/z 257) were first discovered as the phase II metabolites of daphnetin in rats, which revealed a new way of daphnetin metabolism in rats.
