Objective:After construction and identification of HBsAg gene recombinant backbone adenoviral vector,it is to prouduct HBsAg gene recombinant adenoviruses by packing PAd-Easy-1-HBs in 293 cells. Methods:The gene of interest was amplified from plasmid pEcob-6 PCR, the gene of interest which contained HBsAg gene was cloned into on adenoviral shuttle vector pAd-track-cmv. The pAd-track-cmv-HBs was linearized by digesting with restriction endonuclease Pme-1, and subsequently cotransformed into E.coli BJ-5183 cells with an adenoviral backbone vector pAd-Easy-1, Homologous recombinants were performed in bacterial cells. Finally, the linearized backbone adenoviral vector was transfected into adenoviruses packing cells lines,e.g. 293 cell by lipofectamine transfection. Transfections and viral productions can be minotored by green fluorescent protein(GFP). The expression of HBsAg in supermatant was investigated by ELISA. It was a certain HBsAg vaccine to amplify recombinant adenoviruses by repeating the infevtion cell to collect the viral supermatant. Results:GFP expression was visible by fluorescence microscopy after transfection. Adenoviral titer was monitored by GFP expression. GFP expression was visible after repeating the infection cell using the viral supernatant in more than 90 percent of the cells. The HBsAg also expressed in supermatant. Conclusion:HBsAg gene recombinant adenoviral backbone vector has been constructed successfully. HBsAg gene recombinant adenoviruses have been producted by packing in 293 cells. The study provides the possibility of further researches on the development of new anti-HBV vaccines.

BAcKGROUND:In recent years, the pedicle screws fixation technique, which is used in fixation for atlantoaxial instability associated with trauma, severe degeneration and tumorectomy, has been developed. However, this kind of technique easily causes several complications, including malpositional screws, vascular injuries, and even vertebral artery injury.Based on the biomechanical characteristics of memory alloy and determination of atlantoaxial data,a neotype shape memory alloy cervical hook was designed to treat atlantoaxial instability.OBJECTIVE:To investigate the biomechanieal characteristics of the neotype shape memory alloy cervical hook for atlantoaxial instability.DESIGN,TIME AND SETTING:Repeated measurement analysis of variance test was performed in the Laboratory of Clinical Anatomy and Medical Biomechanics,Southern Medical University between March and April 2008.MATERIALS:Eight fresh adult craniocervical specimens(C0-C4)were provided by Department of Clinical Anatomy,Southern Medical University.Atlantoaxial neotype shape memory alloy cervical hook(50.8%-51.8%nickel and the remaining part was titanium)was fabricated by Shanghai Xinchang Memory Alloy Co.,Ltd.METHODS:The included eight C0-C4 specimens were used to test three-dimension ranges of motion(ROM)by fixation and neotype shape memory alloy cervical hook fixation. Then, the positions of spine varying from no loading to the maximum loading status were scanned and analyzed using image processing software to determine the three-dimensional ROM under different statuses.MAIN OUTCOME MEASURES:Three-dimensional ROM of tested specimens.RESULTS:Neotype shape memory alloy cervical hook fixation and Germany AESCULAP SSE hanger fixation had similar flexion-extension range of motion(P=0.595).Lateral bending three-dimensional ROM was greater in the neotype shape memory alloy cervical hook fixation group than in the Germany AESCULAP AAE hanger fixation(P< 0.05).The rotatory three-dimensional ROM was smaller in the neotype shape memory alloy cervical hook fixation group than in the Germany AESCULAP AAE hanger fixation(P<0.05).CONCLUSION:Neotype shape memory alloy cervical hook fixation had comparative post-surgery immediate stability with the Germany AESCULAP AAE hanger fixation.Neotype shape memory alloy cervical hook fixation produced a little worse biomechanical lateral bending stability and a little better biomechanical rotatory stability than Germany AESCULAP AAE hanger fixation.

Objective To investigate the changes of excitatory amino acids (EAAs) of cerebrospinalfluid (CSF) in patients with delayed encephalopathy after acute carbon monoxide poisoning(DEACMP).Method The EAAs levels of CSF including glutamate (Glu) and aspartate (Asp) in 24 patients with DEACMP and 20 controls with migraine were measured by high performance liquid chromatography( HPLC ).Results Glu and Asp levels in patients with DEACMP were significantly higher than those of the controls [(3.76 ± 1.52) μmol/L vs ( 1.55 ± 1.03 ) μmol/L, (0.73 ± 0.44) μmol/L vs (0.38 ± 0.33 ) μmol/L, P all <0.01]. Glu and Asp levels in moderate and severe DEACMP patients were higher than those in mild DEACMP patients. Conclusion It suggests that EAAs participated in the pathogenesis of DEACMP. The Glu and Asp levels in CSF may be regarded as indicator of DEACMP.

Journal of Sun Yat-sen University(Medical Sciences) 2010;31(1):33-38
[Objective] The aim was to choose the best feeder layer by observing the effects of various human feeders supporting human embryonic stem cells (hESCs), and to probe the correlation between the levels of basic fibroblast growth factor (bFGF) secreted by feeders and the growth of the hESCs. [Methods] The primary cells from various tissues were cultured, including foreskin, stromal endometrium, villus, adult fallopian tubal, fetal skin, fetal muscle and mouse embryonic fibroblasts (MEFs). The hESCs were transferred to various feeders, and then the best condition was probed, which was based on the feeder density and the time of mitomycin-C acting on the feeder. Comparing the characteristics of the hESCs, the best feeder was chosen of all kinds of feeders from various tissues that support the hESCs. The level of bFGF secreted by various feeders was detected using ELISA. [Results] All of tested feeders could support the hESCs growth for over 10 passages in the culture, especially the foreskin and the adult fallopian tubal. The density of feeders was related with the morphology and the differentiation rate of the hESCs. According to the characteristics of feeder, the feeder ranking was as follows: foreskin, stromal endometrium, villus, adult fallopian tubal, fetal skin and fetal muscle. Based on the characteristics of the hESCs, the order of feeders was: foreskin, adult fallopian tubal, stromal endometrium, villus, fetal muscle and fetal skin. The levels of bFGF (pg·10~(-5)·mL~(-1)) secreted by various feeders were as follows: adult fallopian tubal (13.23±3.39), foreskin (1.99±0.17), villus (1.40±0.17), fetal muscle (2.02 ±1.59), stromal endometrium (0.38±0.28), and fetal skin (0.29±0.29). [Conclusion] The foreskin and the adult fallopian tubal could support the hESCs better than others though all of them could;do, especially the, foreskin. The bFGF that secreted by the adult fallopian tubal was the highest of all. The correlation was not obvious .between the levels of bFGF secreted by feeders and the growth of the hESCs.
