Orthopedic Journal of China 2006;0(22):-
[Objective]To assess whether calcitonin exerts an chondroprotective action in unstable knees which induce experimental osteoarthritis and its effect to MMP-1.[Method]Twenty 6-month New Zealand rabbits underwent anterior cruciate ligament transaction(ACLT) and were divided into two groups (ACLT group and ACLT+CT group) randomly after transaction on day 7 postsurgery. ACLT+CT group rabbits received a daily subcutaneous injection of salmon calcitonin at a dose of 5 IU per kg of body weight for six weeks.ACLT group were treated with the normal saline at the same dose. All animals were sacrificed 7 weeks postsurgery. The articular cartilage degeneration of medial femoral condyle was evaluated at mascroscopically and microscopically. The mRNA expression and tissue level of MMP 1 was detected by RT-PCR and immunohistochemistry.[Result]Cartilage degradation in ACLT group was significantly more severe than that in ACLT+CT group both in microscopic grading scale and in Mankin's grading scale (P
5.Intestinal absorption and renal excretion mediated by transporters and the relationship with drug-drug interaction.
Acta Pharmaceutica Sinica 2010;45(9):1089-94
Drug-drug interaction (DDI) is referred as the changes of physical and chemical properties, as well as the pharmacokinetics or pharmacodynamics of drugs administered simultaneously or consecutively. The clinical results for drug-drug interaction could be divided into good clinical efficacy and adverse interaction. With the kinds of drugs increasing every year, new drug resistances spring up frequently. This phenomenon makes drug combination increased so that the drug interaction, especially the adverse interaction emerged. The mechanisms of in vivo drug-drug interaction are relevant to a number of factors, including drug-metabolizing enzyme systems and membrane transporters. Recent studies have revealed the important role played by transporters in drug absorption, distribution, metabolism and elimination. In order to avoid severe side effects mediated by transporters and to promote rational combination in clinics, the mechanisms of intestinal absorption and renal excretion mediated by transporters are reviewed.
6.Establishment of a rat chronic asthma model and its evaluation
Acta Pharmaceutica Sinica 2010;45(6):718-23
This study is to establish a rat chronic asthma model. Sensitive SD rats were selected through histamine challenge. The asthmatic groups were sensitized by ih and ip with OVA, aluminium hydroxide gel and inactivated bacillus pertussis on day 1 and 14. From day 21, acute asthmatic group was aerosolized 1% OVA for 1 week, chronic asthmatic group was aerosolized 0.1% OVA for 12 weeks. The control groups received saline as the substitution of OVA. Twenty four hours after the last provocation, physiological monitoring equipment was used to detect the pulmonary function, then the rats were sacrificed. Bronchoalveolar lavage fluid (BALF) was collected to calculate the ratio of different inflammatory cells. ELISA was used to detect total IgE and OVA-specific IgE in serum. Microscopy was conducted to observe the histopathology of lung stained with haematoxylin and eosin staining. Collagen fibers were detected using Picric acid-Sirius red staining technique. The optical density at 610 nm of extractive from locus caeruleus was detected by passive cutaneous anaphylaxis (PCA). The results showed that the asthmatic characteristics were significantly developed in model groups, but not in control groups. Chronic asthmatic group had significantly higher indexes than acute asthmatic group, including the thickness of airway smooth muscle and bronchial basement membrane, and goblet cell hyperplasia, the area of collagen in airways, A610 of extractive from locus caeruleus, the concentration of total IgE and OVA-specific IgE in serum. However, inflammatory cell infiltrate in lungs and the percentage of eosinophils of white blood cells in BALF were lower in chronic asthmatic group than those in acute asthmatic group. Respiratory rate and respiratory flow showed no significant difference in both model groups. In conclusion, the rat chronic asthma model is established by the way in this study, which is comparable to the physiopathologic characteristics of human asthma.
7.Community-acquired Legionella pneumonia in elderly patients: clinical characteristics and outcome
Chinese Journal of Geriatrics 2010;29(8):621-624
Objective To analyze and compare the clinical features of community-acquired pneumonia caused by Legionella in elderly (age≥60 years) and younger patients. Methods The epidemiological, clinical, laboratory data and the outcomes of 80 cases with community-acquired Legionella pneumonia (CALP) from January 2001 to October 2009 were studied retrospectively, and the differences between elderly and younger group were compared using univariate and multivariate analysis. Results (1) Underlying diseases, such as cerebrovascular diseases, chronic obstructive pulmonary diseases, diabetes mellitus and immunosuppressive therapy were significantly more frequent in elderly group than in younger group (all P<0. 05). (2) No significant differences were observed between the two groups in legionella serotype. (3) Malaise, hyponatremia and hypophosphatemia were all more frequent in elderly group than in younger group (x2=5. 300, 5. 520, 4. 470; P=0. 021,0. 019, 0. 034, respectively). And elderly versus younger group had higher creatinine levels [(108.55±56.57) μmmol/L vs. (75.42±17.62) μmmol/L, t= -3. 062, P=0. 002], lower PaO2/FiO2 (<300) (57.7% vs. 29.2%, x2=4. 120, P=0.042), less leucocyte counts [(8.34±3.65)×109/Lvs. (10.63±5.02)×109/L, t=-2.287, P=0.022]. (4) Elderly patients more likely complicated with congestive heart failure (P=0.037) and their length of staying in hospital was also longer (Z=-2.194, P=0.028). No significant differences were observed between the two groups in mortality, intensive care unit admission, and the need for corticosteroids or mechanical ventilation. Conclusions Elderly patients with CALP have a higher frequency of underlying comorbidities and more organs involvement than younger patients, although greater severity of illness at onset and outcome are not significantly different between the two groups.
8.MMP-9 expression in breast cancer metastasis and its clinical significance
International Journal of Biomedical Engineering 2010;33(6):381-384
Matrix metallopmteinase 9(MMP-9) is one kind of matrix metalloproteinases and plays a role in the process of breast cancer metastasis. It can not only degrade the basement membrane and the extracellular matrix around tumor tissue but also promote angiogenesis, even create conditions for the growth of tumors, local invasion and distant metastasis. Current study shows that the over-expression of MMP-9 can significantly speed up the breast lymph node metastasis and the relevant research will provide guidance for clinical treatment and prognosis of breast cancer.

10.Attenuated Salmonella as tumor vaccine vectors
Journal of International Oncology 2010;37(9):649-651
Attenuated Salmonella can be used not only as live vaccines but also as vectors for exogenous genes. As vectors, attenuated bacteria can deliver genes directly to tumor tissues, leading to the expression of the cloned genes and induction of specific anti-tumor immunologic responses to inhibit tumor growth. Using attenuated Salmonella as a vector offers a new approach for vaccine development.
