Objective To investigate the effect and mechanisms of the Berberine on the human breast adenocarci-noma cell line MCF-7 . Methods MTT method was used to evaluate the proliferation effect of MCF-7 and the per-centage of apoptotic cells were determined by flow cytometric analysis. The expressions of JAK2, p-JAK2, p-STAT3, STAT3, Bax, Bcl-2, Cleavage-PARP and Cleavage-Caspase3 were detected by Western blotting. Results The results showed that BBR treatment decreased the activation of JAK2/STAT3 signal pathway and up-regulated the expression of Bax, Caspase3 and PARP activation with the decrease of the expression of Bcl-2. Wherefore, ex-pression of the constitutively active form of STAT3 could attenuate the effect of BBR on the MCF-7 cell. Conclusion Berberine can induces apoptosis and proliferation inhibition of breast adenocarcinoma cell lines of MCF-7 through inhibition of the JAK2/STAT3 signaling pathway.

Objective:To investigate the influence of Mannan Peptide combined with 5-Fu on the proliferation and apoptosis of HepG2 cells and to explore the related mechanism.Methods:HepG2 cells were divided into four groups:the control group,5-Fu group,Mannan Peptide group and combination group.Cell proliferation was evaluated by MTT assay.Flow cytometry was used to examine the cell cycle, proliferation index(PI)and apoptosis ratio. The expression of Bcl-2 protein in each group was analyzed by immunocytochemical method. Results:The inhibitive effect of 5-Fu plus Mannan Peptide was significantly stronger than that of Mannan Peptide alone,in a time-and dose-dependent manner. Cells were treated with medicine for 48 hours.Compared with cells in the control group and 5-Fu group, the percentage of cells in G_0/G_1 stage and the apoptosis ratio in the combination group were increased(P<0.01).In immunocytochemical analysis, the combination of Mannan Peptide and 5-Fu could significantly decrease Bcl-2 protein level(P<0.01).Conclusion:The combination of Mannan Peptide and 5-Fu can synergistically suppress the proliferation and induce apoptosis of human hepatoma HepG2 cells. The mechanism might be associated with down-regulation of Bcl-2 protein expression.

Journal of International Oncology 2013;40(12):902-904
Silent information regulator 1 (SIRT1) is a kind of histone deacetylase which dependents on nicotinamide adenine dinucleotide. It is involved in the occurrence and development of tumors.Data show that SIRT1 is highly expressed in many tumors, such as gastric cancer, breast cancer, prostate cancer and so on.On the other hand, the expression of SIRT1 is lower in many other types of tumors, including glioblastomas, bladder cancer, ovarian cancer and so on.Therefore, the role of SIRT1 in tumor formation process is still uncertain and controversial.

HCV-related decompensated cirrhos, hypersplenism, thrombocytopenia, which not only affect the standard antiviral therapy, fail to achieve the sustained virological response(SVR),but also increase the risk of infection and bleeding. The only successful option is liver transplantation (LT),but the recurrence of HCV after LT remains to be resolved. The patients of HCV genotype 2 are suitable for splenectomy and antiviral therapy following splenectomy, which can achieved a higher SVR and reversed cirrhosis. As an effective alternative to splenectomy, the partial splenic embolization (PSE) can improve the changes of portal hemodynamics and reduce the sequelae of portal hypertension. The appearance of direct antiviral drugs (DAAs)has bring hope for those with decompensated cirrhos and whom IFN is contraindicated or tolerated poorly, those who are waiting for LT or with recurrence of hepatitis C after LT.The treatment of patients with decompensated cirrhos is as follows.

Chinese Journal of Medical Imaging Technology 2010;26(2):231-233
Objective To explore the pulsed wave Doppler (PWD) manifestations of acute experimental incomplete testicular torsion. Methods Eight healthy dogs were surgically modeled to obtain unilateral acute testis torsion from 180° to 630°, respectively. The peak systole velocity (PSV) and resistance index (RI) of intratesticular artery, capsular artery and arteria spermatica interna were measured with PWD before torsion, 2 h, 4 h and 6 h after torsion. Testes were examined pathologically after the experiment. Results PSV and RI of capsular artery and intratesticular artery decreased gradually with the extension of torsional time (P<0.05). PSV and RI of superior, torsional and inferior segments of arteria spermatica interna varied greatly and showed no strong regularity (P>0.05) . The pathological results of testes showed no distinct changes or just mild interstitial hyperemia and edema. Conclusion The decrease of PSV and RI of capsular artery and intratesticular artery, especially RI have referential value in the judgement of acute experimental incomplete testicular torsion, whereas changes of PSV and RI of superior, torsional and inferior segments of arteria spermatica interna are not reliable.

BACKGROUND: Diabetes mel itus is one of the most common systemic diseases, which often leads to the changes of the jaw and other bone structure, as wel as the abnormal changes of mineral metabolism. OBJECTIVE: To observe the three-dimensional structure and histopathological changes of the mandible in type 1 diabetes mel itus mice. METHODS: The mice were randomly divided into control group and diabetes mel itus group. The diabetes mel itus group received intraperitoneal injection of 50 mg/kg streptozotocin for 5 days to establish a type 1 diabetes mel itus model, and the control group received intraperitoneal injection of citrate buffer. RESULTS AND CONCLUSION: At 3 weeks after modeling, the micro-CT technique was used to observe the three-dimensional structure of the mandibles in the two groups. The quantitative analysis on the microstructure of cancel ous bone and cortical bone showed that the bone mineral density, bone volume fraction, trabecular number and trabecular thickness of cancel ous bone in the interest region in the mandible of type 1 diabetes mel itus mice were significantly decreased when compared with that in the control group (P < 0.01, P < 0.05), while the structure model index was increased significantly (P < 0.05); the mineral density and area of cortical bone were decreased in the diabetes mel itus group (P < 0.05). Hematoxylin-eosin staining showed that the number and volume of mandibular trabeculae of type 1 diabetes mel itus mice were decreased. The results suggest that the three-dimensional structure of the cancel ous bone and cortical bone in the streptozotocin-induced type 1 diabetes mel itus mice are changed significantly, and the microstructure change of the cancel ous bone is more obvious.
