Objective:To elucidate the characterization of antigen-specific memory T cells from PPD~+ individuals after stimulation with BCG in vitro.Methods:PBMCs were isolated from PPD~ -/+ normal human peripheral blood and stimulated with BCG. The level of IFN-? in the culture supernatants was assayed by ELISA and the frequency of IFN-?-producinging cells was detected by ELISPOT. 3.MRI diagnosis of epithelioid angiomyolipoma of liver

Objective To retrospectively analyze the MRI findings of epithelioid angiomyolipoma (EAML) of the liver.Methods The MR findings in a series of 8 patients with pathologically proven liver EAML were retrospectively analyzed. Distribution of lesions, signal intensity characteristics, tumor vascularity,feeding arteries and draining veins, fat, hemorrhage, cystic necrosis and pseudocapsule formation were evaluated and correlated with pathology.Results A single lesion was found in each of the 8 patients.All eight lesions showed long T1,long T2 inhomogeneous signal,and high DWI signal. There was only 1 lesion presented with fat,3 lesions presented with hemorrhage. On the dynamic contrastenhanced MRI scan,all 8 lesions manifested as hypervascular with tortuous vasculature in 7 patients.Dilated feeding arteries and draining veins were seen in 5 patients. Conclusion Liver EAML has unique MR imaging features,exhibiting hyperwascular lesions with dialated,tortuous feeding arteries,and draining veins on DCE MRI,which could be helpful in the differential diagnosis of liver EAML.

5.Mutation analysis of a Chinese family with Alport syndrome and genetic diagnosis before embryo implantation

Objective To study the gene mutation of a Chinese family with Alport syndrome and to perform preimplantation genetic diagnosis before embryo implantation.Methods Next generation sequence analysis was done for checking COL4A3,COL4A4 and COL4A5 genes in the Alport syndrome family members.Array comparative genomic hybridization(CGH) was used to detect the embryos.Results A mutation c.2605G > A was found and identified in COL4A5 gene of all of the Alport syndrome patients in the family,but COL4A3 and COL4A3 genes were normal in all of the detected people.After searching for the mutation database,the mutation c.2605G > A of COL4A5 gene was related to the X-linked dominant Alport syndrome.Three embryos were detected by using the preimplantation genetic diagnosis.Among these embryos,there were two male and one female.One of the male embryos was chromosomal aneuploidy,which was 45,XY,-16 and the other was normal.This normal embryo was implanted,and after 20 weeks the prenatal amniocentesis diagnosis approved that the fetus was normal.Conclusions The mutation of COL4A5 gene (c.2605 G > A) is the cause of Alport syndrome in this family,which indicates that next generation sequence analysis proves to be an accurate and rapid method to detect Alport syndrome disease.Meanwhile array CGH can be used to reduce birth rates as a useful preimplantation genetic diagnosis method.
6.Clinical Application of Balloon Bionic Midwifery in 832 Cases

Objective To investigate the effect of the balloon Bionic midwifery on the delivery outcomes and to analyze the clinical value.Methods 1 683 parturients from June 2014 to May 2015 were selected.They were randomly divided into observation group (832 cases,applied balloon Bionic Midwifery) and control group 851 cases.We compared the labor,the postpartum hemorrhage,the outcomes of pregnancy and the rate of survival of neonates of the two groups.Results The first and second stage as well as the total stage of labors of the observation group were lower than the control group (P<0.01);Also,the rate of cesarean delivery and the hemorrhage together with the asphyxia of neonates were lower than the control group (P<0.01).However,the rate of vaginal delivery was higher than the control group (P <0.05).The differences between them had a great statistical significance.Conclusions The balloon bionic midwifery technology has an advantage in reducing the rate of cesarean delivery and the maternal pain of pregnant women as well as the maternal complications.It is an effective and safe midwifery technology.So it has a great value of spreading in clinical trials.
7.Non-motor Symptoms of Parkinson's disease Associated with Quality of Life

Objective To investigate the status about non-motor symptoms (NMS) in Parkinson's disease (PD) patients and the effect on quality of life. Methods 50 PD patients and 50 normal controls were investigated with the NMS questionnaire and the Healthy Survey short form (SF-36), and analysed with Spearman correlation. Results 98% of PD patients presented NMS, and recent memory loss, restless legs,constipation were the most frequent. All the NMS were more frequent than in the controls (P<0.05) except sexual desire, sexual difficulties and nausea or vomiting. All the dimensions of SF-36 were significantly different between the groups (P<0.05). The symptoms of constipation,decline in recent memory, decreased attention, restless legs and sleep disorders correlated with the scores of SF-36 (P<0.05). Conclusion NMS is common in PD patients, and impacts their quality of life.

Objective To evaluate the efficacy and safety of solifenacin in patients with overactive bladder (OAB). Methods A multicenter clinical trial was conduced. 216 patients with OAB were enrolled. All the patients received solifenacin(5 mg once daily). With 5 weeks'treatment, all the patients recorded the diary and the adverse events as well. The symptoms of urgency, frequency, nocturia, urine volume, incontinence were evaluated. The results of the efficacy and safety were analyzed by using SPSS 13. 0. Results After 5 week treatments, all the index obviously improved(P<0.05). 187 cases (86.7%)were cured and 43 cases recovered normal voiding, 29 cases improved obviously. 11cases(5.0 %)reported adverse effect as dry mouth, dry eye. Conclusion Solifenacin could be the safe and effective drug in the treatment of OAB patients.
9.Logistic regression analysis of combining transvaginal conventional ultrasonography and 3D power Doppler ultrasonography(3D-PDUS) in differential diagnosis of ovarian tumor

Objective To develop a logistic regression model for differential diagnosis of the malignant ovarian tumor by combining transvaginal conventional ultrasonography and 3D power Doppler ultrasound(3D-PDUS) techniques. Methods The transvaginal ultrasonography and 3D-PDUS data were collected from 291 patients with ovarian tumors received clinical pathological diagnosis.According to the pathological resuts,the 291 patients were divided in to benign group(GB) and borderline/malignant group (GM). Univariate and multivariate logistic regression analyses were applied to establish models for predicting malignant tumor. Results ① Within the 291 ovarian tumor patients,175 (60.14% ) were classified as the GB and 116 (39.86% ) were considered as the GM,the number for postmenopausal cases and the serum CA125 level in GB group were significantly lower than those in GM group ( P <0.001); ②The tansvaginal conventional ultrasound analysis suggested that borderline/malignant tumor often presented as larger volume,more irregular shape,and higher incidence in the blood flow within a solid papillary projection and ascites (P< 0.05,compared with GB group);In 3D-PDSU data,the vascularization index (VI) was lower than that in GB group( P <0.001,compared with GM group),but there was no significant difference between GB group and GM group in flow index (FI) and vascularization-flow index (VFI) ( P =0.559,0.454); ③ Multivariate logistic regression analysis showed that postmenopausal status,serum CA125 levels,tumor echo,papillary with blood flow,ascites and 3D-PDUS VI parameters were independent risk factors for the progression of borderline/malignant tumor.The ROC curve showed that the established regression model accuracy was 92.0%,sensitivity was 86.2%,specificity was 95.7%,positive predictive value was 92.6% and negative predictive value was 91.7%. Conclusions Combining multi-model transvaginal conventional ultrasound and 3D-PDUS analyses is a useful non-invasive technique for the differential diagnosis of ovarian tumor.
10.Phenotypic and genetic analysis of a child featuring multiple malformations due to chromosome 14q deletion.

OBJECTIVETo analyze a child with mental retardation, growth retardation and language development disorders.
METHODSConventional G-banding analysis was performed on chromosomes cultivated from peripheral blood samples derived from the child and her parents. Array-comparative genomic hybridization (aCGH) was performed to detect minor structural chromosomal abnormalities, and the result was confirmed by short tandem repeats (STR) analysis.
RESULTSFor the child and her parents, no karyotypic abnormality was detected. However, aCGH analysis has identified a 14q22.1 deletion in the child. The microdeletion, with a size of 2.9 Mb was confirmed by STR analysis.
CONCLUSIONThe 2.9 Mb chromosomal microdeletion probably underlies the mental retardation, growth retardation and language development disorders in the child.
