1.Efficacy comparison between saccharomyces boulardii sachets and tetragenous viable bifidobacterium tablets in the treatment of rotaviral gastroenteritis

Objective To compare the effect between Saccharomyces boulardii sachets and tetragenous viable bifidobacterium tablets in the treatment of rotaviral gastroenteritis of children .Methods A total of 191 patients (below 2 months to 8 years old) with rotaviral gastroenteritis admitted to this hospital from January 2012 to May 2014 were divided randomly into control groups (smectite ,conventional treatment ,number=62) ,tetragenous viable bifidobacterium tablets group (conventional treatment combined with tetragenous viable bifidobacterium tablets ,number= 64) ,saccharomyces boulardii group (conventional treatment combined with saccharomyces boulardii ,number=65) .Therapeutic effects were compared between 3 groups after 3 days and 5 days of treat‐ment .Results The obvious curative rate ,effective ratio and total rate in the groups of saccharomyces boulardii and tetragenous via‐ble bifidobacterium tablets were higher than those in the control group(P<0 .05) .However ,no significant difference was found in the two cases among the groups (P>0 .05) .There was significant difference in 3 days and 5 days therapeutic effect among 3 groups (P<0 .05) .The duration of diarrhea in the group of Saccharomyces boulardii was lower than this in the group of tetragenous viable bifidobacterium tablets (P<0 .05) .Conclusion The therapeutic effect of Saccharomyces boulardii is similar to that of tetragenous viable bifidobacterium tablets in treatment of children with Rotaviral gastroenteritis .The duration of diarrhea in the group of Sac‐charomyces boulardii was lower than that of the group of tetragenous viable bifidobacterium tablets .

3.Clinical study of pemetrexed-based chemotherapy in the treatment of elderly patients with advanced non-squamous non-small-cell lung cancer

Objective To assess the clinical efficacy and adverse effect of pemetrexed-based chemotherapy as first-line treatment in elderly patients with advanced non-squamous non-small-cell lung cancer (NSCLC).Methods A total of 40 elderly patients with advanced non-squamous NSCLC confirmed by pathology were retrospective analyzed in the study,who received pemetrexed plus cisplatin (group A,n =18) or pemetrexed plus carboplatin (group B,n =22) as the first-line treatment.RECIST was used to assess the efficacy of the treatment and NCI-CTC AE version was used to describe adverse events.Results In the 40 patients,no one received complete response (CR),17 patients showed partial response (PR),16 had stable disease (SD),7 had progress disease (PD),the objective response rate (ORR) was 42.5 % (17/40) and the disease control rate (DCR) was 82.5 % (33/40).The median progression-free survival (PFS) time was 5.3 months.1 year suvival rate was 63.2 % (24/38).In further subgroup analyses,the ORR,DCR,PFS and 1 year suvival rate in group A were higher than those in group B,but they had no statistically significant difference (P > 0.05).The drug-related adverse events were myelosuppression,gastrointestinal response,most of which were grade 1 to 2.Conclusion Pemetrexed combined with cisplatin or carboplatin may be recommended as first-line chemotherapy for elderly patients with advanced non-squamous NSCLC because of its safety and efficacy.

Objective To investigate the values of serum vascular endothelial growth factor (VEGF) ,carcinoembryonic antigen (CEA) ,carbohydrate antigen 19-9 (CA19-9) and ferritin (SF) determination in assessing the efficacy of chemotherapy in patients with advanced gastric cancer .Methods Wedetermined the concentrations of serum VEGF ,CEA ,CA19-9 and SF for 85 patients with advanced gastric cancer ,before and after 1 cycles of chemotherapy and evaluated the the effect of chemotherapy after 2 cycle of chemotherapy .Results In effective group ,serum concentrations of VEGF ,CEA ,CA19-9 and SF significantly decreased after chem-otherapy cycle(P< 0 .05) ,serum VEGF and CEA concentrations significantly increased in progress group (P< 0 .05) ,serum VEGF ,CEA ,CA19-9 and SF concentrations in stable group and serum CA19-9 and SF concentrations in progress group did not change significantly(P> 0 .05) ,compared with those before chemotherapy cycle .Multiple regression analysis showed that serum CEA ,VEGF ,CA19-9 concentration changes correlated with chemotherapy efficiency ,while serum SF didn′t(t=1 .58 ,P=0 .119) . Conclusion Serum CEA ,VEGF ,CA19-9 concentration changes associated with gastric cancer chemotherapy efficiency ,which help predict the efficiency of chemotherapy .
6.Protective and therapeutic effects of Shugan Lifei Recipe on rats with asthma under stress and the mechanisms

OBJECTIVE: To explore the influence of psychological factor on asthma, the protective and therapeutic effects of Shugan Lifei Recipe on rats with asthma under stress and the mechanisms. METHODS: Allergy and stress-induced asthma models were established in rats by giving ovalbumin and restraining stress. Radioimmunoassay, enzyme linked immunosorbent assay, in situ hybridization and immunohistochemical method were used to detect plasma corticosterone, IL-4, IFN-gamma, expression of glucocorticoid receptor (GR) mRNA in hippocampus and adrenocorticotropic hormone corticoliberin (CRH) positive neuron in hypothalamus. Optical and electron microscopes were used to observe the morphological changes of pulmonary hilar and hippocampal tissues. RESULTS: Shugan Lifei Recipe (SGLFR) could reduce plasma corticosterone, decrease CRH positive neurons in paraventricular nucleus of hypothalamus, up-regulate the expression of GR mRNA in hippocampus and reduce the injury of hippocampal neuron. SGLFR had certain inhibitive effect on hyperfunction of hypothalamo-pituitary-adrenal cortex (HPA) axis in rats with asthma under stress and could also relieve the pathological changes of the pulmonary hilar tissue. The level of plasma IFN-gamma was increased while the level of plasma IL-4 was decreased in SGLFR-treated group. CONCLUSION: The mechanism of SGLFR in treating the rats with asthma under stress is probably to regulate the hyperfunction of HPA axis and the disorder of immuo-system.
7.Expression of miRNA-148a in Bladder Carcinoma Tissues and Its Bioinformatics Analysis

Objective To explore the role of miRNA-148a in bladder tumorous development and progression.Methods Ex-pression of miRNA-148a was assessed in 35 bladder carcinoma tissues and 16 non-carcinoma tissues by fluorescence quanti-tative real time PCR,and correlation with clinical features was evaluated.Target genes and transcription factors of miRNA-148a were predicted using bioinformatic analysis,then TF-miRNA-148a-target genes network diagram was built and the tar-get genes was analyzed of gene ontology enrichment and KEGG pathway.Results Expression of miRNA-148a was lower in bladder carcinoma tissues than in non-carcinoma tissues(0.000 8±0.000 2 vs 0.002 1±0.000 5)(t=2.46,P <0.05),but its expression was no statistical significance in different groups of gender,age,pathological classification,clinical stage, lymph node metastasis (P >0.05).268 target genes of miRNA-148a were predicted by three softwares at the same time,60 transcription factors were predicted and the binding sites with combination scroes above 80 was 657.The target genes of miRNA-148a was enriched in many biological processes,such as neuron differentiation,generation of neurons,neuron projec-tion development,cytoplasmic mRNA processing body,cytoplasm(P <0.001).They also participated in p53 signaling path-way,proteoglycans in cancer-homo sapiens,pathways in cancer,prostate cancer,protein processing in endoplasmic reticulum, focal adhesion and so on(P <0.05).According to TF-miRNA-148a-target genes network diagram,miRNA-148a was regula-ted by SP1,ESR1,AP1,MYC and BRCA1,genes of IGF1,P27kip1 ,NCOA1,PTEN,SERPINE1 might be regulated by miR-NA-148a.Conclusion miRNA-148a which was significantly down-regulation in bladder carcinoma tissues may be participate in bladder tumorous development and progression,bioinformatics analysis provides some ideas for further research.
8.Preparation of canine bone marrow stromal stem cell sheets and investigation on their osteoblastic differentiation

BACKGROUND:Conventional methods,including trypsin digestion and cells transfer using single call suspension,have many drawbacks,which limit the development of bone tissue engineering.OBJECTIVE:To culture bone marrow stromal stem calls,induce osteoblastic differentiation,and prepare cell sheets.METHODS:Canine bone marrow stromal calls were isolated by density gradient centrifugation technique,inoculated into DMEM medium,and induced to differentiate into osteoblasts.Complete call sheets were harvested by call sheet engineering based on the temperature change of temperature-responsive medium.RESULTS AND CONCLUSION:Immediately after inoculation,primary calls were scattered on the bottom of culture flask,presenting a transparent spherical body with a good refractive capacity.At 12 hours,calls exhibited a long shuttle shape,reached complete confluency,and grew in a whirlpool-like fashion.After osteoblastic induction,the majority of bone marrow stromal stem calls appeared tetragonal,polygonal,and squamose.At 21-28 days,round or oval-shaped calcified nodules formed.When the bone marrow stromal stem calls in the temperature-responsive culture dishes were cooled below the critical temperature 32℃,cells were gradually detached from the bottom of culture flask and formed complete bone marrow stromal stem call sheets.These findings indicate that density gradient cantrifugation technique can be used to successfully isolate and culture canine bone marrow stromal stem cells to differentiate into osteoblasts and call sheet engineering enables to harvest complete bone marrow stromal stem call sheets.
9.Culture and preparation of dog bone marrow mesenchymal stem cell sheet in vitro

BACKGROUND:There are some disadvantages in harvesting and transferring cells in the traditional tissue engineering technique,and it is difficult to form dense tissues,which significantly limits the development of tissue engineering.OBJECTIVE:To explore the culture and fabrication of dog bone marrow mesenchymal stem cell(BMSC)sheet in vitro.METHODS:Bone marrow was extracted from dogs following anesthesia.BMSCs were isolated with the method of density gradient centrifugation in vitro.BMSCs at passage 4 at a density of 1×10~9/L were incubated in the temperature-responsive culture dishes with a diameter of 3.5 cm,and cultured in an incubator at 37 ℃,5% CO_2 and saturated humidity.The temperature of the incubator was changed from to 37 V to 20 ℃ to prepare BMSCs cell sheet for 20 minutes.Cell morphological changes and cell sheet formation were observed under an inverted microscope.RESULTS AND CONCLUSION:Dog BMSCs following 24 hours of primary culture presented ellipse or polygonal shape.Most cells adhered at hour 72,and cell colonies were visible at day 7.Cells showed long spindle and completely confluence at day 12,with unclear boundary.BMSCs in the temperature-responsive culture dishes presented short spindle shape,and gradually separated from the dish bottom,forming entire cell sheet containing extracellular matrix at 20 V.These verified that dog BMSCs can be effectively obtained with method of density gradient centrifugation.Complete cell sheet layer can be fabricated with temperature-responsive culture dishes.
10.Preparation and releasing behavior of chitosan microspheres/nano-hydroxyapatite/PLGA scaffolds: Compared to nano-hydroxyapatite/PLGA scaffolds and chitosan microspheres

BACKGROUND: How to make growth factor plays a role persistently and efficiently is a key in constructing bone tissue engineered bone. Currently, varied microspheres or scaffolds were used as release carriers, however, the delayed release effects needs elevating.OBJECTIVE: To prepare chitosan microosPheres/nano-hydroxyapatite/poly (lactic-co-glycolic acid) (CMs/nHA/PLGA) scaffolds, and to measure its characteristics of delayed release of bovine serum albumin (BSA).METHODS: CMs were prepared by an emulsifying cross linking method with BSA as a model protein. Using ice particulates as porogen, composite CMs/nHA/PLGA scaffolds were prepared by freeze-drying. The characteristic and morphology of the composite were observed by scanning electron microscope, later particle size analyzer, mercury porosimeter and universal testing machine, and the release behavior of BSA was investigated in vitro.RESULTS AND CONCLUSION: The CMs were spherical shape with a regular surface, with diameters of 20-40 μm. The encapsulation efficiency of the CMs was 86.5%, and the loading capacity was 0.8%. With the increase of initial BSA dosage, the loading capacity increased to 2.6%, while the encapsulation efficiency decreased to 74.1%. The CMs can be uniformly distributed in PLGA scaffolds to form CMs/nHA/PLGA scaffolds, which had 100-400 μm pore diameter and over 80% porosity, with 1.1-2.3 pMPa compressive strength, and 26.5% cumulative degradation at 10 weeks. The cumulative release of BSA from nHA/PLGA scaffolds was above 85% at 36 hours, which from CMs was 33.6% at 10 days, and that from CMs/nHA/PLGA scaffolds was 81.5% at 40 days. The results demonstrated that CMs/nHA/PLGA scaffolds have an excellent releasing efficiency for protein drugs with suitable compressive strength and degradation, which would be used as delivery system and tissue engineering scaffolds.
