Objective:To develop a method for the determination of natural benzoic acid in Mesona chinensis Benth. with different production places and storage time. Methods:HPLC was adopted, the column was an Agilent ZORBAX SB-C18 (250 mm × 4. 6 mm, 5 μm) column, the mobile phase was methanol-0. 02 mol·L-1 ammonium acetate (5∶95) at the flow rate of 1. 0 ml·min-1 , the col-umn temperature was 35℃, the detection wavelength was 230 nm, and the injection vocume was 10μl. Results:Benzoic acid showed good linearity over the concentration range of 0. 1-10. 0 μg·ml-1(r=0. 999 9). The average recovery was 99. 59%(RSD=1. 57%, n=9). Natural benzoic acid in powered Mesona chinensis Benth. was different because of various production places. The maximum content of natural benzoic acid in Mesona chinensis Benth. was 141. 8 mg·kg-1, while the minimum content was 73. 8 mg·kg-1. The content of natural benzoic acid in Mesona chinensis Benth. stored for two years was declined obviously. Conclusion: The method with simple operation is accurate and reproducible. It can provide a basis for the further study of Mesona chinensis Benth. resources, and provide a reference for the content determination of natural benzoic acid in other plants.

Objective To investigate the influence of postoperative cognitive dysfunction (POCD) to clinical outcome in adult patients and the risk factor. Methods 122 patients after operation on chest were surveyed with Wechsler Adult Intelligence Scale-Revised China (WAIS-RC) within 3 d before operation and on the 7th day after operation. Their pulmonary functions were measured before discharged. Their time on bed, total time in hospital, time after operation in hospital, time in intensive care unit and spending for therapy were investigated. Results 45 patients were finally diagnosed as POCD (POCD group) 7 d after operation. Before operation, the age, the body weight index, the smoking index and the number of risk factor of angiosclerosis in the POCD group were all higher than those whose cognitive function were normal (normal group) (P<0.05). The sores of insight, count, digit scope, vocabulary, figure collocation, figure patch of WAIS-RC in POCD group were obviously lower than those in normal group (P<0.05). Every index of pulmonary function in POCD group was lower than those in normal group(P<0.05). The time keeping on bed, total time in hospital, time after operation in hospital, time in intensive care unit and spending for therapy in POCD group all exceeded those in normal group (P<0.05).Conclusion The adult patients with POCD may spend more time and money for therapy in hospital, and their pulmonary functions may be recovered poorly.

Mesenchymal stem cells(MSCs), which can be introduced in brain tissue by the strategies of brain parenchyma, blood and neurolymph transplantation, to repair tissue injury and promote functional recovery of neural system, have been considered as the ideal seed cells for the treatment of central nervous system diseases. However, as the special structure exists in the blood-brain barrier, study on correlation between MSCs and blood-brain barrier permeability will make it possible for more MSCs to pass the barrier to perform the treatment. This paper gives a review on the latest research development in the field.

Objective To raise the vigilance for pregnancy complicated with acute abdominal diseases and to reduce the maternal mortality rate. Methods A retrospective study was done. Results (1) The maternal mortality of pregnancy complicated with acute abdominal diseases was 1 31/10 5 in the period of 1989～1993, which was significantly higher than that in the period of 1994～1998 (0 71/10 5). (2) The main cause of maternal deaths was misdiagnosis and the rate of misdiagnosis was 72 7%.(3) Among the acute abdominal diseases, pregnancy complicated with pancreatitis ranked the first that accounted for 54 6%. Conclusions For the purpose of reducing the mortality rate of pregnancy complicated with acute abdominal diseases, the correct diagnosis and proper treatment especially for those with acute pancreatitis are important.

Objective We detected the expression of the low risk HPV6/11 and the high risk HPV16/18 in cervical lesions in order to investigate the correlativity,mechanism and clinical values between them.Methods We chosed 150 cases of cervical disease tissues that included chronic cervicitis (30 cases).cervieal condyloma without atypia(30 cases),CIN Ⅰ(30 cases),CIN Ⅲ(30 cases)and cervical squamous cell carcinoma (30 cases).And all of them were constructed into tissue microarrays(TMA)that include 300 spots.Then,the expression of HPV6/11 and HPV16/18 were determined by in situ hybridization.Results The positive expression rates of HPV6/11 were 13.33%,90%,33.33%,0 and 0 respectively in chronic cervicitis,cervical condyloma without atypia,CIN Ⅰ,CIN Ⅲ and cervical squamous cell carcinoma.Condyloma showed significantly higher HPV6/11 infection rates than the other groups(P＜0.001).And there were obvious dliference between CIN Ⅰ and CIN Ⅲ & carcinoma (P=0.001.＜0.05).The spearman correlation analysis suggested that the expression of HPV6/11 in the five cervical deseases was negative correlation (rs=-0.037,P＜0.001).The positive rates of HPV16/18 were 0,6.67%,10%.56.67%.76.67% respectively in chronic cervicitis,cervical condyloma without atypia,CIN Ⅰ,CIN Ⅲ and cervical squamous cell carcinoma.Comparing CIN Ⅲ and carcinoma with the other groups,the positive rates were obvious defrerence between them (P＜0.001).But there was not significant defference between CIN Ⅲ and carcinoma (P=0.10).The spearman correlation analysis suggested that the expression of HPV16/18 in the five cervical deseases was positive correlation (rs=0.628,P＜0.001).Conclusion Low risk HPV6/11 is a main reason for the cervical condyloma and CIN Ⅰ.High risk HPV16/18 is closely related with CIN Ⅲ and carsinoma.and also a main reason for CIN Ⅲ and carcinoma.Detecting and differentiating HPV type will help for the diagnosis and monitor of cervical lesions,especially,it will be very important to prophylaxis,early diagnosis and early therapy of cervical carcinoma.

Objective To investigate the present situation of elective course,find out the problems,explain the causes and offer some suggestions.Methods Using the information collected from self-made questionnaires,and analysis data.Results The survey shows that there are the problems of elective courses in the installation,time,management,contents,teachers,exam,and so on.Conclusion The system of elective course should be made perfect in construction,management and operation.

Objective To establish an easier and better method for primary culture of rabbit fibroblast-like synoviocytes(FLS) by modified tissue cell culture.Method Healthy rabbit joint synovial layers were cut into small pieces and siphoned off water attached on the tissue with sterile filter paper and then cultured.The growth status and morphology were observed.The growth curve of the 3rd passage cells was measured by CCK-8 assay, and the expression of vimentin was detected by immunocytochemistry.Results The FLS cultured in vitro exhibited a spindle-shaped appearance and could rapidly expand.The cell growth curve was typical of S type, and the cells highly expressed vimentin.Conclusions Primary culture of rabbit FLS by the improved explant culture method is successfully established.The method is simple and highly efficient.It provideds a new method for isolation of FLS.

Aim To investigate the the effect of ginsenosides of stem and leaf(GSL) on mouse fatty liver and its mechanism.Methods Mice were randomly divided into normal control group(NC,fed with normal diet) and model group(fed with high-fat diet).Twelve weeks later,when the establishment of experimental model of fatty liver was confirmed,the model mice were subdivided into 4 subgroups: HF group(fed continuously with high-fat deit),NT group(fed with normal deit),GSL1 group(fed with normal deit and treated intragastrically with GSL 50 mg?kg-1?d-1),GSL2 group(fed with normal deit and treated intragastrically with GSL 100 mg?kg-1?d-1).Mice in NC,HF and HT groups were given distilled water by gastric perfusion.Two weeks later,all mice were killed,and blood was collected for measuring serum TC,TG,HDL-C,LDL-C,ALT contents,liver tissue for determining hepatic TC,TG,MDA SOD levels.In addition,liver index and pathology were also observed.Expression of PPAR? and CYP2E1 mRNA in liver was examined by RT-PCR.Results In GSL2 group,GSL significantly reduced liver index,serum lipid,hepatic lipid and MDA contents,and elevated SOD activity.Moreover,GSL obviously improved pathological change,increased PPAR? mRNA and suppressed CYP2E1 mRNA levels in the liver.In NT group,hepatic lipid and MDA contents remained high and SOD activity is low,although liver histology somewhat improved.In GSL1 group the results were similar to those in NT group.Conclusions These results suggest that GSL might be effective in the treatment of hepatic adipose infiltration of mice.It may be associated with increasing PPAR? mRNA,then decreasing serum lipid and hepatic lipid;and inhibiting CYP2E1 mRNA expression in the liver,thus suppressing lipid peroxidation.

Objective To explore the role of MMP-9 in blood-brain barrier (BBB) disruption and to promote rMSCs migration.Methods To investigate the effect of MMP-9 on the permeability of BBB,an in vitro model of BBB was established and performed Transwell experiments.To observe the pathologic changes of the cerebral cortex,rat brains from Sham treated group,HIBD group,MMP-9 treated group and TIMP-1 (intervention) treated group at different time points (0.5,1,3,7,14 d) were prepared and stained with Hematoxylin-Eosin and quantified the brain water content.Permeability of BBB examined by EB values measurement.MMP-9 protein level of rat cortex was detected by Western Blot.The number of Brdu labeled rMSCs in the rat cerebral cortex of each group was quantified by Immunohistochemistry (IHC).Results Transwell experiments results showed that migration of rMSCs increased remarkably in hypoxic condition compared to that of normal control (P＜0.01).The number of rMSCs migrated in the MMP-9 treated group was much more than that of negative control group and TIMP-1 group (P＜0.01).The results of pathology showed that compared to HIBD group,brain water content and the permeability of the BBB were increased in MMP-9 treated group but reduced in TIMP-1 treated one.The expression of MMP-9 protein in MMP-9 treated group was reached the peak at 3 d point,which was higher than that of HIBD group and TIMP-1 group (P＜0.05,P＜0.01).The quantity of Brdu labeled rMSCs crossed BBB in MMP-9 treated group was extremely higher than that of HIBD group (P＜0.05,P＜0.01).Conclusions The expression level of MMp-9 protein was improved after hypoxic-ischemic.MMP-9 could facilitate the migration of rMSCs through vitro BBB and control the opening of BBB,which indicate that MMP-9 may facilitate the migration of rMSCs through BBB into brain.
