1.Cryopreservation of in vitro shoot-tips of Platycodon grandiflorum by vitrification and plant regeneration

Objective To get a new approach to conserve the germplasm of Platycodon grandiflorum. Methods A method of vitrification was studied to cryopreserve in vitro shoot-tips of P. grandiflorum, and the regenerated plantlets were observed subsequently. Results Shoot-tips, 2—3 mm length, gotten from in vitro shoots of P. grandiflorum precultured on MS medium supplemented with 5% DMSO and 103 g/L sucrose for 3 d, were loaded with the 60% PVS_2 for 20 min at 20 ℃, and incubated in PVS_2 for 90 min at 0 ℃ prior to a direct plunging into liquid nitrogen (LN) and keeping for 1 d. After rapid thawing in water at 40 ℃, the shoot-tips were rinsed in the MS medium supplemented with 410 g/L sucrose for 20 min, and plated on the filter paper sustained by the MS regeneration medium supplemented with 0.6 mg/L KT, 0.2 mg/L BA, and 0.05 mg/L NAA for 1 d in dark and subcultured on the above regeneration medium for one week in dark prior to exposure to the light. The survival of shoot-tips was up to 80%, and they grew normally. Conclusion The method of vitrification to cryopreserve the germplasm of P. grandiflorum is simple in handling with high in survival and normal in regeneration and can be applied in practice.
2.Perceptual Priming in Brain Injury

Objective To investigate the perceptual priming in brain injured patients. Methods 41brain injured patients, including 24 brain injury and 17 stroke. 99 normal controls, including 39 young, 39 middle aged and 21elderly people. All of them were estimated by the Fragmented Picture Naming Task. Results The patient group had lower priming scores than the young (t=5.558, P<0.001), and middle and elderly-aged (t=3.100, P=0.003) control group in the Fragmented Picture Naming Task. Conclusion Perceptual priming is not only concerned with occipital lobe, but also with frontal and temporal lobes possibly.
3.An Evaluation of the Performance of Remifentanil Target-Controlled Infusion System Under Inhalation and Intravenous Anesthesia

Objective To evaluate the performance of the remifentanil target-controlled infusion(TCI) system(Minto model) during laparoscopic surgery under inhalation and intravenous anesthesia.Methods A total of 15 patients scheduled for laparoscopic surgery received remifenanil by TCI,and the anesthesia was maintained with inhalation of isoflurane and intermittent intravenous vecuronium.The target remifentanil plasma concentration(Cp) was increased step by step,with the concentration changed at intervals of 30 minutes.The beginning Cp was 3 ng/ml,then increased to 6 ng/ml,9 ng/ml.Blood samples were taken from radial arterial before anesthesia induction and 30 minutes after adjustment of Cp to determine the concentration of remifentanil using high performance liquid chromatography-tandem mass spectrometry(HPLC/MS/MS).The performance of the TCI system was evaluated by median performance error(MDPE),median absolute performance error(MDAPE) and wobble.Results The MDPE,MDAPE and wobble of the remifentanil TCI system were 8.78%,16.11% and 14.55%,respectively.The measured concentrations of remifentanil were positively correlated with Cp(r=0.891,P=0.000).The linear equation was Y∧ =1.1046X+0.1837.Conclusions The remifentanil TCI system with Minto pharmacokinetic model can meet the requirements of clinical anestehesia for Chinese patients within clinical treating concentration.

5.Application Analysis of IgM Antibody and DNA Test Results in Children Mycoplasma Pneumonia Infection

Objective To analyze the results of distribution of mycoplasma pneumoniae IgM antibody and DNA test,and ex-plore its application in diagnosis of Mycoplasma pneumoniae infection in children.Methods Retrospectively analysed myco-plasma pneumoniae antibody IgM (MP-IgM)and mycoplasma pneumoniae DNA (MP-DNA)results,114 374 passengers and 8 014 passengers,in 0 ~ 16 year-old children of Shengjing Hospital Affiliated to China Medical University from year 2013 to 2014.Analyzed of the relationship between gender,age,season and the positive rate of the MP-IgM and MP-DNA. Collected 5 106 patients with MP-IgM and MP-DNA results simultaneously and analyze the consistency between two posi-tive indexes in different ages.Results ①MP-IgM and MP-DNA positive rate varied in sex,age and season,the positive rate in female was higher than male.The positive rates of MP-IgM and MP-DNA at the age of 1~3 and 7~ 12 years old were higher than other age groups.The positive rate of MP-IgM in spring and autumn was higher,while MP-DNA was higher in summer and autumn and all differences were statistically significant (P < 0.001).②The positive rate of MP-DNA differed in specimens,the bronchoalveolar lavage fluid had the highest MP-DNA positive rate,49.95%.③The positive consistency of MP-IgM and MP-DNA was associated with age,with 4~6 and 7~12 year-olds the highest,respectively 35.2% and 45.8%. Conclusion ①Incidence of children pneumonia mycoplasma infection was mainly in preschool and school-aged children,more female than male,and autumn highest.② The detection of MP-DNA in bronchoalveolar lavage fluid was the best sample type.③For <1 and 1~3 year-olds,combined detection of MP-DNA and MP-IgM can improve diagnostic rates.
6.Mechanism of TWEAK on the synthesis of MMP-9 in fibroblast-like synoviocytes of rheumatoid arthritis

Objective To discuss the role of p38MAPK signal pathway in the process of TWEAK inducing rheumatoid arthritis(RA) fibroblast-like synoviocyte(FLS) to synthesize MMP-9 and look for a new target for RA treatment. Methods RA FLS were primarily cultured and stimulated with TWEAK. Western blot was used to detect the expression of p-p38MAPK and p65 in RA FLS. FLS were pretreated by SB203580 or not. ELISA was used to detect the concentration of MMP-9 in cell-cultured fluid. The mRNA expression of MMP-9 was measured by RT-PCR. Results TWEAK( 100 ng/ml) can make p38MAPK phosphorylated and increased the expression of p65 protein in the cell nucleus. SB203580 can partially inhibit the expression of MMP-9 and MMP-9 mRNA produced by RA FLS which is induced by TWEAK. Conclusion TWEAK induced RA FLS to synthesize MMP-9, in that process, the p38MAPK signal trausduction pathway was in active state, and induced the expression of NF-κB.

Lipoxygenase(LO) pathway had been implicated in the pathogenesis of such cardiovascular diseases as hypertension, atherosclerosis, restenosis, and palys and important role in the development of these disease. LO inhibitiors could suppress vascular contractile responses significantly, reduce blood pressure, inhibit migration of vascular smooth muscle cells(VSMC), attenuate neointimal thickening in the injuried arteries and generation reactive oxygen species(ROS), block monocyte binding to VSMC, etc. The effects of LO inhibitors were associated with marked inhibition of MAPK pathway. Therefore, inhibition of LO pathway may provide a new strategy for preventing and treating above diseases, suggesting that LO mat be a novel taget for such purposes.
9.Effects of dopamine alone or dopamine plus norepinephrine on hemodynamics, tissue oxygenation and renal function during orthotopic liver transplantation

Objective To evaluate the effects of dopamine (DA) alone or DA plus norepinephrine (NE) on hemodynamics, tissue oxygenation, acid-base balance and renal function during orthotopic liver transplantation (OLT).Methods Thirty ASA IE or IV patients undergoing OLT were randomly divided into 2 groups of 15 patients each. In group A DA was continuously infused during operation. The initial infusion rate was 1-3 ?g?kg-1 ?min-1 . The rate was then adjusted to MAP between 60-80 mmHg. While in group B DA and NE were continuously infused. The initial infusion rate of DA was 1-3 (?g?kg-1?min-1 and that of NE 0.03 ?g?kg-1?min-1 . Both rates were adjusted to MAP between 60-80 mm Hg but the maximal DA infusion rate was 5 ?g?kg-1?min-1 . Anesthesia was induced with midazolam, propofol, fentanyl and vecuronium and maintained with isoflurane inhalation, continuous propofol infusion and intermittent i.v. boluses of fentanyl, midazolam and pipecuronium. Radial artery was cannulated and S-G catheter was placed via right internal jugular vein. Hemodynamic variables such as MAP, CVP, PAP, PCWP, SVRI, PVRI, blood gases, mixed venous blood O2 saturation (SvO2), blood lactate, Cr and BUN were measured and O2 delivery (DO2 ) and O2 consumption (VO2 ) were calculated and recorded after induction and before surgery (T1 baseline), 60 min after operation was started (T2), at 60 min anhepatic phase (T3) and 60 min neohepatic phase (T4 ) and at the end of operation ( T5). Results There were no significant differences between the two groups with respect to age, sex, body weight, duration of operation and types of surgery. MAP and HR were quite stable in both groups.
