1.Expression of surfactant protein A in lung carcinoma and its clinical significance

Objective To investigate the expression of surfactant protein A(SP-A)in adenocarcinoma and squamous cell carcinoma of lung and its clinical significance.Methods The expression of SP-A was determined by immunohistochemical assay (EnVision) in 27 cases of adenocarcinoma,15 cases of squamous cell carcinoma of lung.Then the association of SP-A expression with clinicopathological parameters was analyzed.Results The expression rate of SP-A was significantly higher in adenocarcinoma of lung than that in squamous cell carcinoma of lung(51.85% vs 6.67%,P<0.01).The expression rate of SP-A in papillary adenocarcinoma,bronchioloalveolar carcinoma,acinar adenocarcinoma and musinous adenocarcinoma of lung was respectively 85.71%,50%,40%and 25%.There was no correlation between the expression of SP-A and gender,age,tumor size and lymph node metastasia.Conclusion The expression of SP-A may be associated with histological types of the lung carcinoma,which may be used as important marker in differential diagnosis of adenocarcinoma and squamous cell carcinoma of lung.
2.The slatus quo of personality dysfunction of the secondary vocational students and the correlation to resilience

ObjectiveTo study the correlation of personality dysfunction with resilience in secondary vocational students.MethodsBy adopting the questionnaire of Personality Dysfunction Inventory for Middle School Students (MPDI) and Healthy Kids Resilience Assessment scale (HKRA) to assess 358 secondary vocational students.ResultsThe secondary vocational student's scored lower on relationship dysfunction (4.15 ±2.67) than ordinary high school students'( 5.18 ± 3.37 ),and higher on behavioral deviation (6.07 ± 3.20) than ordinary high school students (5.18 ± 3.37 ).There existed great capable difference of total and every fact of personality dysfunction according to grade (P < 0.05,P < 0.01 ),and the quo of personality dysfunction on second year in high vocational school students was better than high and senior high vocational school students.Both parents of secondary vocational student's scored lower on relationship dysfunction( 3.96 ± 2.36),emotional deviation (3.96 ± 2.36 )and personality dysfunction( 15.50 ± 6.27 ) than Single-parent family of secondary vocational students ( 5.73 ±4.12,6.51 ± 3.72,19.78 ± 10.44).There existed great capable difference of total and every fact of personality dysfunction according to relationship with parents (P < 0.05,P < 0.01 ),and the more harmonious in parental relationship,the better in personality.There also existed great capable difference of total and every fact of personality dysfunction according to educational background of parents (P < 0.05,P < 0.01 ),and the higher degree,the better in personality.Personality dysfunction of Students of secondary vocational was negatives correlated with their resilience (Pearson correlation coefficient was between -0.064 and -0.345 for each pair).The linear regression showed thatthepeerrelationshippredictedpersonalitydysfunctionofsecondaryvocationalstudents.ConclusionThe personality dysfunction of secondary vocational students is affected by their parental relationships,educational background of parents,grade and resilience.
3.Expression and diagnostic significance of DOG1, CD117 and CD34 in gastrointestinal stromal tumors

Objective To investigate the expression and diagnostic significance of DOG1,CD117and CD34 in gastrointestinal stromal tumors.Methods Expressions of DOG1,CD117 and CD34 were determined by immunohistochemical techniques (EnVision) in 60 cases of gastrointestinal stromal tumors (GIST) and 14 cases of other mesenchymal tumors.Results Expression of DOG1 in GIST was significantly higher than in other mesenchymal tumors (P =0.000).Expression of DOG1 in GIST from stomach was significantly higher than that from intestines.The expression rates of DOG1,CD117 and CD34 in GIST were 95%,95%,and 83.3%,respectively.Expression of DOG1 was significantly higher than CD34 (P =0.040),but no significant difference between DOG1 and CD117 (P =1.000).The expression rates of DOG1 in CD117-positive,CD34-positive tumors were 94.7% and 94.1%,respectively.All the CD117-negative,CD34-negative tumors expressed DOG1,3 cases which were negative for CD117 and CD34 expressed DOG1.Conclusions DOG1 is a novel marker for GIST,It is necessary to the combined detection of the expression of DOG1,CD117 and CD34,which will further improve the diagnostic accuracy of GIST.
4.A study of post-traumatic stress disorder symptoms and its drug intervention among middle age acute leukemia patients

Objective To investigate and understand the characteristics and drug intervention of post-traumatic stress symptoms(PTSD) among middle age acute leukemia patients.Methods The Essen Trauma-Inventory (ETI) was administrated to 100 middle age acute leukemia patients who were admitted in our department and study the PSTD symptoms.Results The positive rate of PTSD is 21.21% in sample of the patients.Total PTSD symptoms scores were (18.10 ± 9.42),there is no significant different between the 3 main PTSD symptoms in these patients(P > 0.05).Type of leukemia,sexuality,levels of education and marital status are influencing factors of PSTD symptoms,there is significant different between the subgroups (P <0.01).Drug intervention can significantly relieve PTSD symptoms in these patients (Z =13.56,P < 0.01).Conclusions Middle age acute leukemia patients are generally found PTSD symptoms and with a high incidence rate of PTSD,so it needs timely diagnosis and treatment.
5.Application of fallopian tube embolization before in vitro fertilization and embryo transfer dealing with the hydrosalpinx

Objective To explore the effectiveness of the fallopian tubes embolization for the hydrosalpinx before in vitro fertilization and embryo transfer(IVF-ET).Methods The fallopian tubes embolization was performed on 46 hydrosalpinx patients-Fimt the hysterosalpingography was performed to show the positions and shape of the hydrosalpinx.Then the hydrops was drawn into the celiac cavity in order to be absorbed after performing fallopian tube recanalization. Finally,transv(o)ihal therapy was performed,putting the embolization microcoils into the fallopian tube through a micro-catheter.One month after the fallopian tubes embolization,hystemsalpingography was conducted to check for the effectiveness of the embolization.After 3 months.all the 46 patients received the treatment of IVF-ET,Results The interventional treatment of 82 fallopian tubes obtained one time success among 46 cases of fallopian tubes embolization.Among them,obvious results were achieved in 72 fallopian tubes,taking up 88%0f tIle total;effective results were seen in 10 fallopian tubes.accounting for 12%0f the total.No one was invalid.In the same period,compared with the 91 cases of non-hydrosalpinx as the control group.the embolization group of patients achieved a higher fertilization rate(69%),and clinical pregnancy rate(41%),compared with the control group(63%and 39%respectively),but without a significant difference(P>0.05).However,the ectopic pregnancy rate(O)and the abortion rate(8%)were significantly lower than the control group(8%,16%respectively;P<0.05).Conclusions Fallopian tubes embolization used in hydrosalpinx treatment before IVF-ET is an innovative approach,simple,safe,economical,with no negative impact on ovarian functiorr It can significantly increase the clinical pregnancy rate and prevent the occurrence of tubal pregnancy.It is a feasible and effective method.
6.Embryonic stem cell transplantation for treating acute myocardial infarction in the central and peripheral infarct regions

BACKGROUND: Present therapeutic tool cannot supplement infarct myocardium. Studies have shown that stem cell transplantation can promote regeneration of myocardium and vessels and improve heart function and prognosis.OBJECTIVE: To observe changes in morphology and hemodynamics in myocardium following embryonic stem cell transplantation in and surrounding the acute myocardial infarct site.DESIGN, TIME AND SETTING: The randomized, controlled, animal study was performed at the Laboratory of Neurobiology,Department of Human Anatomy and Neurobiology, Xiangya Medical College, Central South University from March 2007 to October 2008.MATERIALS: A total of 40 SPF grade Wistar rats were equally randomized into 4 groups, normal control, infarct model,central transplantation and peripheral transplantation groups. Embryonic stem cells-D3 (ES-D3) and Buffalo rat hepatocytas were supplied by Shanghai Cell Institute, Chinese Academy of Sciences.METHODS: Following resuscitation, ES-D3 cells at (2.0-5.0)×107/L were incubated in a flask, and induced to in vitro differentiate in conditioned medium containing Buffalo rat hepatocytes. Except normal control group, rat models of acute myocardial infarction were established by ligating left anterior descending coronary artery in the infarct model, central transplantation and peripheral transplantation groups. At 1 week following model induction, ES-D3 cells were labeled by BrdU for 1 day, and implanted at 1×109/L. Three sites were selected in the infarct site in the central transplantation group. 10 μ L cell suspension (104 cells) was implanted in the ventricular wall through each site. In the peripheral transplantation group, an equal volume of cell suspension was separately implanted in three peripheral infarct sites by the same method.MAIN OUTCOME MEASURES: Results of immunohistochemistry and hemodynamics were measured.RESULTS: ES-D3 cells in buffalo rat hepatocyte conditioned medium presented regular colony-shaped. At 8 days following differentiation, some embryo proper had spontaneous rhythmic contraction, showed positive reaction of cardiac troponin T after immunostaining. Under the electron microscope, myotube and muscle fiber appeared, which verified the differentiation of cardiomyocytes. Cells were positive for BrdU in the peripheral transplantation group, but negative in the central transplantation group. Cells were also positive for cardiac troponin T. 4 weeks following transplantation, left ventricular systolic pressure,minimum/maximum rate of ventricular pressure (±dp/dtmax) were significantly reduced (P < 0.01), but left ventricular end diastolic pressure was significantly increased (P < 0.01), left ventricular mass and left ventricular mass index were significantly increased (P < 0.01 ) in the infarct model group compared with the normal control group. Compared with the infarct model group, no significant changes in hemodynamics indices were found in the central transplantation group (P > 0.05); left ventricular systolic pressure, ±dp/dtmax were significantly increased (P < 0.01), left ventricular end diastolic pressure was significantly decreased (P < 0.01 ), left ventricular mass, left ventricular mass index and infarct area were significantly reduced(P < 0.01) in the peripheral transplantation group.
7.Pancreatic cell apoptosis and endocrine alterations in rats with acute obstructive pancreatitis

Objective To investigate the occurrence of apoptosis and its relationship with endocrine hormones altemtions in rats with acute obstructive pancreatitis(AOP). Methods The model of AOP wag establisbed by ligation of pancreaticobiliary duct.8,12 hrs after operation,the serum insulin,glucagons and amvlase were determined;pancreatic tissues were harvested and apoptotic rate wag evaluated by TUNEL and flow cytometry(Annexin V-FITC/PI assay).Results 8 and 12 hrs after AOP induction,serum amylase levels wefe(1198±687)U/L and(1698±1103)U/L respectively;serum insulin levels were(8.1±5.8)ng/ml and (12.7 ±6.9)ng/ml respectively;sertlm glucagon levels were(6.8±4.6)ng/ml and(7.3±2.9)ng/ml respectively;all these parameters were significantly high than(404±222)U/L,(5.6±2.7)ng/ml and(2.6±2.1)ng/ml in the sham operation group(P<0.05).AnnemnV FITC/PI assay confirmed apoptosis occurred both in exocrine acinus cells and endocrine panclreas islet;and the apoptotic rate wag(20.5±11.2)%and (15.5±8.9)%at 8 and 12 hrs after AOP induction,which wag significantly high than(4.2±1.6)%in the sham operation group(P<0.05).Conclusions Cell apoptosis occurred in both acinar and islet in the model of AOP,and this may be the pathophysiological basis of endocnne hormones alterations in the model of AOP.
8.Inhibitory molecules expressed in T lymphocytes in patients with lung cancer before and after chemotherapy

Objective:The tumorigenesis, progression, and metastasis of lung cancer are mostly governed by the immunosuppres-sive profile. This study aimed to explore the levels of various immunosuppressive inhibitory molecules in lung-cancer patients subject-ed to different chemotherapy cycles. Methods:Thirty-three patients with advanced lung cancer (ALC;stages III-IV) without receiving prior chemotherapy and 23 healthy subjects were enrolled in our study. Peripheral blood samples were collected from the patients be-fore each chemotherapy cycle. The inhibitory markers expressed in T cells such as TIM3, PD-1, and CTLA4 were analyzed by flow cy-tometry. Results:The percentages of CD4+TIM3+, CD8+TIM3+, CD4+PD-1+, CD8+PD-1+, CD4+CTLA-4+, and CD8+CTLA-4+T cells in the peripheral blood of the ALC patients were significantly higher compared to the controls. The percentage of CD4+TIM3+, CD8+TIM3+, CD4+PD-1+, and CD8+PD-1+T lymphocytes in the peripheral blood of patients (n=19) who achieved PR or SD significantly de-creased after five cycles of chemotherapy (P<0.05). Similarly, the percentages of CD4+CTLA-4+and CD8+CTLA-4+T cells in the pa-tients also decreased after five cycles of treatment. Conclusion:The immune status of ALC patients was evidently suppressed. Effec-tive chemotherapy successfully potentiated effective immune responses by downregulating inhibitory molecules in T cells.
9.Magnetic-activated cell sorting combined with multiplex ligation-dependent probe amplification for detecting molecular cytogenetic abnormalities in multiple myeloma.

OBJECTIVETo investigate the reliability of magnetic-activated cell sorting (MACS) combined with multiplex ligation-dependent probe amplification (MLPA) in the detection of the molecular cytogenetic abnormalities in multiple myeloma.
METHODSThe bone marrow cells were collected from 29 patients with multiple myeloma. The immuno magnetically sorted and unsorted cells were detected for TP53 and RB1 expressions using MLPA probes and the results were compared with fluorescent in situ hybridization (FISH).
RESULTSThe detection success rate was 100% for MLPA, which yielded results with an concordance rate of 99.1% with the FISH results. The positivity rates of MLPA and FISH were both increased after immunomagnetic sorting of the bone marrow cells.
CONCLUSIONMLPA can well suit the clinical needs for detecting molecular cytogenetic abnormalities in multiple myeloma, and the samples should be immuno magnetically sorted before the assay.

10.The mediating effect of emotion regulation strategy between inappropriate parenting styles and self-injury in medical students

Objective To explore the relationships between inappropriate parenting styles,emotional regulation strategies and self-injury.Methods A total of 1 096 medical students anonymously completed adolescents self-harm scale,emotion regulation scale (ERS) and the rejection and over-protection subscales of parental bonding instrument (PBI).Results The scores of paternal rejection,maternal rejection,paternal over-protection,maternal over-protection,cognitive reappraisal,expression suppression and self-injury were (4.3±3.3),(3.9±3.0),(5.7±3.3),(6.1±3.5),(36.1±6.7),(25.7±7.4) and (1.0±2.9),respectively.The scores of self-injury were positively correlated with the scores of paternal rejection,maternal rejection,paternal over-protection,maternal over-protection and expression suppression (r =0.08-0.20,P< 0.01),meanwhile negatively correlated with the scores of cognitive reappraisal(r=-0.10,P<0.01).Cognitive reappraisal mediated the relationship between parental rejection and self-injury,which accounted for 9.3%.Expression suppression mediated the relationship between parental rejection,parental over-protection and self-injury,which accounted for 14.5% and 17.9%,respectively.Conclusion The emotion regulation strategy act as a mediator between the inappropriate parental styles and self-injury in medical students.In addition,different emotion regulation strategies (cognitive reappraisal and expression suppression) play different roles in such relationship.
