Objective To explore the effect of endogenous H_2S in idiopathic pulmonary fibrosis(IPF) patients, and its relationship with the severity of IPF. Methods Plasma H_2S in 27 patients with IPF (IPF group) and 28 healthy people (control group) were tested. PaO_2, ESR, CRP, LDH, HRCT and pulmonary function in IPF group were tested to evaluate the severity of the disease at the same time. Then the concentrations of H_2S in aggravating and palliative period in IPF group were observed. Results (1) Compared with the control group, the concentration of plasma H_2S in IPF group was significantly reduced(P <0. 01). (2) The concentration of H_2S in aggravating period was higher than that during palliative period(P <0. 01) . (3) With the progress of HRCT,there was a descending trend of plasma H_2S(P >0. 05). Conclusion Endogenous H_2S was probably involved in the course of IPF, and was associated with the progress of the disease.
3.Inductive effect of copper 8-quinolinolate on oxidative DNA damage in HepG2 cells

OBJECTIVETo assess the DNA damage of copper 8-quinolinolate (CuQ) and to elucidate the plausible mechanisms.METHODSHepG2 cells were treated with CuQ0-4 μmol·L-1 for different time,DNA damage was measured by Comet assay.Catalase (CAT) activity,glutathione(GSH) level and thiobarbituric acid reactive substances (TBARS) were measured.NF-κB was examined using Western blotting.8-Hydroxydeoxyguanosine (8-OHdG) was measured by immunoperoxidase staining analysis.RESULTSCuQ 0.5 -4 μmol·L-1 caused significant increase of DNA migration in HepG2 cells.CuQ significantly decreased levels of GSH and activity of CAT in HepG2 cells (P <0.05).Moreover,CuQ significantly increased accumulation of the p65 subunit of NF-κB into nucleus,levels of lipid peroxidation product TBARS and the formation of 8-OHdG (P <0.05).The intracellular GSH level was modulated by pre-treatment with buthionine-(S,R)-sulfoximine (BSO),depletion of GSH in HepG2 cells pre-treated with BSO dramatically increased susceptibility of HepG2 cells to CuQ-induced DNA damage.CONCLUSIONCuQ exerts DNA damage by oxidative stress and increases accumulation of p65 subunit of NF-κB into nucleus in HepG2 cells.
4.Docetaxel induced human uterine cervical carcinoma cell apoptosis and it's enhancement effect on radiosensitization

Objective To investigate therapeutic mechanism of radiation therapy associated with Doeetaxel on human uterine cervi-cal carcinoma. Method Hela cells were incubated with 0. 5 μmol/L, 1.01 μmol/Docetaxel for 24 hours, and were radiated with 0, 2, 4, 6, 8Gy X ray. The proliferation activity was detected with MTT method. Apoptosis was evaluated with TUNEL method. Expression of caspase-3 protein and the degradation fragment was measured with Western blot. Result Inhibitory effect of radiation therapy associated Docetaxel on Hela cells was significantly stronger than that of radiation group. Treated with 2 Gy and 6MV X ray radiation, inhibitory rate of Hela cells increased with the increment of associated Docetaxel doses (P<0.05). Inhibitory rate of Hela cells in radiation associated Docetaxel group was significantly increased compared with radiation group. Apoptosis index in radiation associated Docetaxel group 48 hours after treatment was significantly increased compared with radiation group or Docetaxel group with TUNEL method (P<0.05). Western blot results indies-ted that caspase-3 protein was degraded and small active fragments were produced. Conclusion The reason that Docetaxel enhanced Hela cell radiation sensitivity may probably he correlated to easpase-3 protein activation.
5.Combination therapy of Docetaxel with radiation increases apoptosis of human Hela cell line and expressions of TRAIL and its receptors

Objective: Docetaxel as a newly semi-synthesized anti-tumor drug,exhibits significant cyrotoxic activity and enhanced sensitivity for radiotherapy.The aim of this study was to investigate the correlation of the apoptosis of human Hela cell line with the expressions of TRAIL and its receptors induced by the combination therapy of Docetaxel with radiation.Methods: We treated the Hela line by Docetaxel,radiation,and combination therapy of Docetaxel with radiation,respectively,detected the apoptosis of the Hela cells using Hoechst 33342 staining and the TUNEL method,and observed the changes in the mRNA expressions of TRAIL and its receptors TRAIL-R in the three groups by RT-PCR.Results: Compared with the Docetaxel and the radiation groups,the apoptosis of the Hela cells was increased significantly in the combination therapy group,and so were the mRNA expressions of TRAIL and its receptors TRIAL-R1 and TRAIL-R2.Conclusion: The combination therapy of Docetaxel with radiation increased the apoptosis of the Hela cell line,which might be mediated by the up-regulation of mRNA expressions of TRAIL and its receptors TRAIL-R1 and TRAIL-R2.

7.Effect of Shenlongjiannao Capsule on Learning and Memory and Reactive Nitrogen Intermediates in Cerebral Cortex of Cerebral Ischaemia Rats

ObjectiveTo explore the effects of Shenlongjiannao capsule on the learning and memory of the cerebral ischaemia model rat as well as reactive nitrogen intermediates in the cerebral cortex.MethodsThe cerebral ischaemia animal model was established by two-vessel occlusion. 50 rats were randomly divided into 5 groups: control group with sham operation, model group, positive control group with pisacetam, Shenlongjiannao capsule groups (low and high dosage). After four weeks, the learning and memory function was tested with Morris water maze, and the contents of nitric oxide(NO) and inducible nitric-oxide synthsae(iNOS) in cerebral cortex were measured.ResultsAfter treatment, the learning and memory ability improved significently and the contents of NO and iNOS in cerebral cortex decreased in Shenlongjiannao capsule groups.ConclusionShenlonjiannao capsule can improve the learning and memory of rats with cerebral ischaemia by lowering the activity of NO and iNOS in the cerebral cortex.
8.Application of video laryngoscope in diagnosis and treatment of laryngeal diseases

Objective:To investigate the value of video laryngoscope in diagnosis of laryngeal diseases. To study the effect of video laryngoscope in treatment of laryngeal diseases.Method:Pentax VNL-1530T and Olympus BF-240T video laryngoscope were used in diagnosis of 3628 cases of laryngeal diseases and in treatment of 1221 cases of laryngeal diseases.Result:3628 cases were examined by video laryngoscope,and correct diagnosis was made.768 cases of vocal cord polyp and 249 cases of vocal cord node were excised, 113 cases of foreign bodies in hypopharynx or larynx were removed.91 cases of other laryngeal diseases were treated under video laryngoscope.Conclusion: Video laryngoscope is a new excellent instrument in diagnosis and treatment of laryngeal disease.
9.Electronic laryngoscopic diagnosis and therapy for laryngeal diseases

Objective: To study the diagnostic and therapeutic effects of electronic laryngoscope in treating laryngeal diseases and discuss some problems concerned. Methods: Follow up studies were conducted in 846 patients with laryngeal diseases who were treated electronic laryngoscopically in our department and sex, site of lesion and therapeutic effect were analyzed. Results: The incidence of vocal polyp and nodules in female was significantly higher than that in male, and vise versa in vocal cyst and leukoplakia. Vocal nodules could be located bilaterally, while vocal cyst unilaterally. The operative style applied showed good therapeutic effect. The effective rate for vocal polyp, nodule, and cyst was 99.84%~100%, and that for vocal leukoplakia, 66.67%. Conclusion: Operation under electronic laryngoscope has advantages of rapid performance, small dead angle, high accuracy, few complications, clear image, and little pain, and is beneficial to early diagnosis and treatment of laryngeal diseases. So it is worthy of wide application. [
10.Virtual screening and preliminary validation of anti-tuberculosis components of traditional Chinese medicine

Objective To verify a method of calculation and prediction by screening the antibacterial components of classical Chinese medicinal herbs (TCM) for anti tuberculosis, and preliminarily evaluate the anti tuberculosis effect of the selected components. Methods The components database of Stemona, Bletilla striata and Chuanbei was established. Dock method was used to predict the anti tuberculosis components from the database, and then mycobacterium smegmatis, MIC and inhibition zone methods were used to evaluate the anti-tuberculosis effect of these predicted components. Results Three ingredients were selected. In vitro experiments also showed that the selected ingredients had the effect of anti-mycobacterium smegmatis by MIC and inhibition zone . Conclusions The virtual screening method could decrease consumption and increase the efficiency of finding anti-tuberculosis ingredients of TCM.
