Objective:To construct an expression plasmid of siRNA against gene hIAP-2 and to assess its effect on breast cancer cell line MCF-7.Methods: A hIAP-2 siRNA plasmid was constructed with mU6pro vector containing U6 promotor and its RNAi effect on MCF-7 cells was detected by RT-PCR and Western blot.The effects of the plasmid on MCF-7 proliferation were analyzed by MTT assay,on the cell cycle by flow cytometry,and on the cell morphology by Hoechst staining.The activity of caspase-3 was determined with its substrate Ac-DEVD-pNA and the expression of some related proteins was detected by Western blot.Results: The hIAP-2 siRNA plasmid knocked down hIAP-2 expression in MCF-7 cells specifically and obviously,inhibited tumor cell proliferation and arrested the MCF-7 cells at G_(1) phase.After hIAP-2 gene was silenced,MCF-7 cells became multinuclear and had macronucleus;procaspase-3 was activated and its expression was increased.The protein levels of I?B?(p37) and p21~(waf1) increased,of NF-?B(p65) decreased and Cyt C remained unchanged.Conclusion: hIAP-2 in MCF-7 cells plays a crucial role in cell proliferation and cell cycle progression.Mitotic cell death might be the main way of the cell death;G_(1) arrest after DNA damage might be associated with the p21~(waf1) increase.

Objective To investigate the effects of different times of needle retention on cognitive function in patients with vascular cognitive impairment no dementia (VCIND). Methods One hundred and four VCIND patients were randomly allocated to group A of 35 cases, group B of 34 cases and group C of 35 cases. All the three groups received acupuncture plus medication. Of them, group A was given scalp acupuncture with 30 min retention of needles;group B, with 1 hr retention of needles;group C, with 10 hrs retention of needles. The Montreal Cognitive Assessment (MoCA) score and the ADL score were recorded in the three groups of patients before and after treatment. Results There were statistically significant pre-/post-treatment differences in the MoCA score and the ADL score in every group (P<0.05). There were statistically significant post-treatment differences in the MoCA score and the ADL score between group A or B and group C (P<0.05). Conclusions Acupuncture is an effective way to treat VCIND. It can improve cognitive function in the patients. Scalp acupuncture with 10 hrs retention of needles has a more marked effect.

With the development of global warming,urbanization,ageing of the population,it is paid more attention to the human health impacts on the frequent occurrence of heat wave and urban heat island effect.The study methods and advances in impacts of heat wave on human health were reviewed in the present paper.

Objective To explore the clinical efficacy of cetuximab combined FOLFOX chemotherapy on colon and colorectal cancer.Methods Forty patients with colon and colorectal cancer were randomly divided into observation group and control group,and 20 cases for each group.Patients in control group were treated with oxaliplatin + leucovorin + 5-fluorouracil,and in the observation group were given cetuximab (the first does was 400 mg/m2 at the first week,then the second week and follow weeks was 250 mg/m2 1 time/week for 6 week) plus the treatment as control group.The clinical efficacy,adverse effect and 1-year survival in the two groups were recorded.Results The total effective rate and clinical benefit rate in observation group were 60％ (12/20)and 85％ (17/20) respectively,which significantly higher than the control group (30％ (6/20) and 65 ％ (13/ 20),x2 =18.18,10.67,P ＜ 0.05).Skin rash rate in observation group was 55％ (11/20),which was significantly higher than that of control group (15％ (3/20),x2 =35.16,P ＜0.05).Half-year survival rate and 1-year survival rate of patients in observation group were 75％ (15/20) and 45％ (9/20) respectively,significantly higher than the control group (50％ (10/20) and 25％ (5/20),x2 =13.33,8.791,P ＜ 0.05)~ Conclusion Treatment scheme of Cetuximab combined FOLFOX chemotherapy is proved to be an effect of treating advanced colon and colorectal and be with the low adverse effect.

ObjectiveTo investigate the correlation of TIMI risk score and plasma brain natriuretic peptide (BNP) in patients with Acute myocardial infarction. MethodsTIMI risk score were calculated in 63 patients with acute myocardial infarction;At the same time plasma brain natriuretic peptide were measured by enzyme-linked immune absorption assay(ELISA). Analyze the correlation of TIMI risk score and plasma brain natriuretic peptide. ResultsNine patients of 63 occurred cardiavascular events during hospitalization or follow-up( 14.3％ ). The incidence of MACE increased significantly with the rise of TIMI risk score and BNP level. The TIMI risk score was positively correlated with the plasma BNP level( r =0.73 ,P ＜ 0. 05 ). ConclusionThe higher TIMI risk score and BNP level, the worse the prognosis, it could be use to predict the prognosis.

Systems biology is a new science of the 21st century, which resembles traditional Chinese medicine (TCM) in many aspects such as study method and design. Adoption of systems biology approach would do much help for exploring the scientific connotation of TCM syndrome and the modernization of Chinese herbal medicine. Its technological platforms, such as genomics, proteomics and metabonomics, provide powerful tools for the study of the essence of TCM syndrome and the function of herbal compound recipe. Compared with other molecular biological methods, such as genomics and proteomics, metabonomics method is more direct and more concise, especially for providing an effective method for the study of TCM therapy in preventing and treating kidney diseases.

Objective To evaluate the effects and security of single or combined sufentanil application on sedation and analgesia for mechanically ventilated patients in intensive care unit(ICU).Method A total of 160 ICU patients with mechanical ventilation were randomly allocated into four groups:sufentanil group,sufentanil plus midazolam group,sufentanil plus dexmedetomidine group and fentanyl group.Facial pain scale(FPS),Ramsay score and vital signs were employed to assess the effects of different therapies on organ functions and adverse reactions.Result After the treatment,FPS in all groups were significantly decreased and maintained around 3 points.Ramsay scores were significantly increased(P <0.001)and the score in group given sufentanil combined with midazolam or dexmedetomidine at the time points of30 min,1 h,6 h were even higher.Compared with the preoperative data,indicators of lung function(PaCO2and blood lactic acid),liver function(AST)and renal function(Cr)were decreased in all groups(P <0.05).Respiratory rate in the fentanyl group was significantly higher than the others.Total consumption ofsufentanil in group with combined sufentanil application was significantly lower.No significant differencesin adverse reactions were noticed among the groups(P >0.05).Conclusion For mechanically ventilatedpatients,sufentanil has efficient sedative and analgesic effects with less physiological interference and severe adverse reactions.Combined application is beneficial in decreasing total sufentanil consumption andinhibition to respiratory system,which is worthy of being spread.

OBJECTIVE:To put forward suggestions for the legal regulation on the financial conflict of interest of researchers in drug clinical trials.METHODS:The definition and harm of the financial conflict of interest of the researchers in clinical trials were analyzed,and the legal measures of FDA on regulating and controlling the financial conflict of interest of the researchers were evaluated.RESULTS & CONCLUSIONS:The Chinese legislature should draw useful legal experiences from USA to establish suitable legal measures to regulate and control the financial conflict of interest of researchers in clinical trials,clarify the definition of the financial interest of researchers,require the applicant for clinical trial to disclose his financial interest and the applicants for drug registration to submit the statement about the financial conflict of interest of researchers to the drug administration department.

Aim To study the role of nitric oxide by measuring the expression and activity of nitric oxide synthase in hippocampus of rats tolerance and dependence to the anticonvulsant action of flurazepam.Method Animal models of toleran and dependence to flurazepam were established. Nitric oxide synthase in hippocampus was measured using Western blot and immunohistochemistry, and its activity was evaluated using colorimetry. Result Hippocampal nNOS increased in rats tolerance to flurazepam. In rats dependence to flurazepam, hippocampal nNOS increased but without statistical significance. Total NOS activity increased dramatically both in flurazepam tolerane and dependene rats.Conclusion NO may play a role in rats tolerance and dependence to the anticonvulsant action of flurazepam.
