Objective: To investigate the influence of autologous adipose stem cell matrix gel on wound healing and scar hyperplasia of full-thickness skin defects in rabbit ears, and to analyze the related mechanism. Methods: Experimental research methods were adopted. The complete fat pads on the back of 42 male New Zealand white rabbits aged 2 to 3 months were cut to prepare adipose stem cell matrix gel, and a full-thickness skin defect wound was established on the ventral side of each ear of each rabbit. The left ear wounds were included in adipose stem cell matrix gel group (hereinafter referred to as matrix gel group), and the right ear wounds were included in phosphate buffer solution (PBS) group, which were injected with autologous adipose stem cell matrix gel and PBS, respectively. The wound healing rate was calculated on post injury day (PID) 7, 14, and 21, and the Vancouver scar scale (VSS) scoring of scar tissue formed on the wound (hereinafter referred to as scar tissue) was performed in post wound healing month (PWHM) 1, 2, 3, and 4. Hematoxylin-eosin staining was performed to observe and measure the histopathological changes of wound on PID 7, 14, and 21 and the dermal thickness of scar tissue in PWHM 1, 2, 3, and 4. Masson staining was performed to observe the collagen distribution in wound tissue on PID 7, 14, and 21 and scar tissue in PWHM 1, 2, 3, and 4, and the collagen volume fraction (CVF) was calculated. The microvessel count (MVC) in wound tissue on PID 7, 14, and 21 and the expressions of transforming growth factor β1 (TGF-β1) and α smooth muscle actin (α-SMA) in scar tissue in PWHM 1, 2, 3, and 4 were detected by immunohistochemical method, and the correlation between the expression of α-SMA and that of TGF-β1 in scar tissue in matrix gel group was analyzed. The expressions of vascular endothelial growth factor (VEGF) and epidermal growth factor (EGF) in wound tissue were detected by enzyme-linked immunosorbent assay on PID 7, 14, and 21. The number of samples at each time point in each group was 6. Data were statistically analyzed with analysis of variance for repeated measurement, analysis of variance for factorial design, paired sample t test, least significant difference test, and Pearson correlation analysis. Results: On PID 7, the wound healing rate in matrix gel group was (10.3±1.7)%, which was close to (8.5±2.1)% in PBS group (P>0.05). On PID 14 and 21, the wound healing rates in matrix gel group were (75.5±7.0)% and (98.7±0.8)%, respectively, which were significantly higher than (52.7±6.7)% and (90.5±1.7)% in PBS group (with t values of 5.79 and 10.37, respectively, P<0.05). In PWHM 1, 2, 3, and 4, the VSS score of scar tissue in matrix gel group was significantly lower than that in PBS group (with t values of -5.00, -2.86, -3.31, and -4.45, respectively, P<0.05). Compared with the previous time point within the group, the VSS score of scar tissue at each time point after wound healing in the two groups was significantly increased (P<0.05), except for PWHM 4 in matrix gel group (P>0.05). On PID 7, the granulation tissue regeneration and epithelialization degree of the wounds between the two groups were similar. On PID 14 and 21, the numbers of fibroblasts, capillaries, and epithelial cell layers in wound tissue of matrix gel group were significantly more than those in PBS group. In PWHM 1, 2, 3, and 4, the dermal thickness of scar tissue in matrix gel group was significantly thinner than that in PBS group (with t values of -4.08, -5.52, -6.18, and -6.30, respectively, P<0.05). Compared with the previous time point within the group, the dermal thickness of scar tissue in the two groups thickened significantly at each time point after wound healing (P<0.05). Compared with those in PBS group, the collagen distribution in wound tissue in matrix gel group was more regular and the CVF was significantly increased on PID 14 and 21 (with t values of 3.98 and 3.19, respectively, P<0.05), and the collagen distribution in scar tissue was also more regular in PWHM 1, 2, 3, and 4, but the CVF was significantly decreased (with t values of -7.38, -4.20, -4.10, and -4.65, respectively, P<0.05). Compared with the previous time point within the group, the CVFs in wound tissue at each time point after injury and scar tissue at each time point after wound healing in the two groups were significantly increased (P<0.05), except for PWHM 1 in matrix gel group (P>0.05). On PID 14 and 21, the MVC in wound tissue in matrix gel group was significantly higher than that in PBS group (with t values of 4.33 and 10.10, respectively, P<0.05). Compared with the previous time point within the group, the MVC of wound at each time point after injury in the two groups was increased significantly (P<0.05), except for PID 21 in PBS group (P>0.05). In PWHM 1, 2, 3, and 4, the expressions of TGF-β1 and α-SMA in scar tissue in matrix gel group were significantly lower than those in PBS group (with t values of -2.83, -5.46, -5.61, -8.63, -10.11, -5.79, -8.08, and -11.96, respectively, P<0.05). Compared with the previous time point within the group, the expressions of TGF-β1 and α-SMA in scar tissue in the two groups were increased significantly at each time point after wound healing (P<0.05), except for the α-SMA expression in matrix gel group in PWHM 4 (P>0.05). There was a significantly positive correlation between the expression of α-SMA and that of TGF-β1 in scar tissue in matrix gel group (r=0.92, P<0.05). On PID 14 and 21, the expressions of VEGF (with t values of 6.14 and 6.75, respectively, P<0.05) and EGF (with t values of 8.17 and 5.85, respectively, P<0.05) in wound tissue in matrix gel group were significantly higher than those in PBS group. Compared with the previous time point within the group, the expression of VEGF of wound at each time point after injury in the two groups was increased significantly (P<0.05), and the expression of EGF was decreased significantly (P<0.05). Conclusions: Adipose stem cell matrix gel may significantly promote the wound healing of full-thickness skin defects in rabbit ears by promoting collagen deposition and expressions of VEGF and EGF in wound tissue, and may further inhibit the scar hyperplasia after wound healing by inhibiting collagen deposition and expressions of TGF-β1 and α-SMA in scar tissue.

7.Determination of IL-1beta, IL-4 and IL-10 contents in the seminal plasma of infertile patients and its clinical value.
National Journal of Andrology 2004;10(11):851-854
OBJECTIVETo observe the relationship between the contents of interleukin-1beta (IL-1beta), interleukin-4 (IL-4) and interleukin-10 (IL-10) in the seminal plasma of infertile males and sperm function indexes.
METHODSBy radioimmunoassay (RIA), we determined the contents of IL-1beta, IL-4 and IL-10 in the seminal plasma of 126 infertile and 20 normal males. According to the sperm count, the infertile were divided into three groups: Groups A (sperm count > or = 20 x 10(6)/ml), B (sperm count < 20 x 10(6)/ml) and C (azoospermia). Based on sperm vitality and motility, Group A was subdivided into a normal and abnormal vitality group and a normal and decreased motility group. In line with the serum results of antisperm antibody (AsAb) and semen WBC, the infertile males were divided into AsAb positive and negative, and WBC semen and non-WBC semen groups. According to the assay results of normal males, Groups A and B were each subdivided into normal and decreased groups of sperm penetrating power, intact acrosome rate and terminal swelling rate.
RESULTSThe content of IL-1beta in the seminal plasma of the infertility group was obviously higher, but the content of IL-4, IL-10 significantly lower than that of the normal group (P < 0.01). In the infertility group, there existed significant differences in the contents of IL-1beta, IL-4, IL-10 in seminal plasma between the WBC and non-WBC semen groups, as well as between the AsAb positive and negative groups (P < 0.05 or P < 0.01); and the same was true for the content of IL-4 between the normal and decreased groups of sperm vitality, motility, penetrating power, intact acrosome rate, and terminal swelling rate (P < 0.05 or P < 0.01).
CONCLUSIONThe contents of IL-1beta, IL-4 and IL-10 in seminal plasma are closely related to male reproduction. The increase or decrease of the contents reflects the state of immunity and infection of the reproductive system, and influences sperm functions.

Journal of Clinical Hepatology 2019;35(10):2194-2199
ObjectiveTo investigate the current status of compliance with antiviral therapy with nucleos(t)ide analogues (NAs) among chronic hepatitis B (CHB) patients treated in General Hospital of Ningxia Medical University. MethodsThe CHB patients who received antiviral therapy with NAs at the outpatient and inpatient services of Department of Infectious Diseases, General Hospital of Ningxia Medical University, from May to December, 2017 were enrolled. A questionnaire survey was performed for demographics, family history of CHB, detailed doctor’s advice, daily routines, physical exercise, compliance, awareness of the knowledge about hepatitis B, health belief, and social support, and the influence of various factors on compliance with NAs treatment was analyzed. The Wilcoxon rank-sum test was used for comparison of non-normally distributed continuous data between groups, and the chi-square test or the Fisher’s exact test was used for comparison of categorical data between groups. A multivariate logistic regression analysis was used to investigate the influencing factors for compliance with antiviral therapy with NAs among CHB patients. ResultsA total of 612 questionnaires were distributed, among which 600 were collected, with a questionnaire recovery rate of 98% and a valid rate of 100%. Among the 600 CHB patients, 334 (55.67%) had good compliance with the antiviral therapy with NAs and 266 (44.33%) had poor compliance. Among the 266 CHB patients with poor compliance, 56 (21.05%) did not follow the doctor’s advice and took other drugs, 206 (77.44%) sometimes forgot medication, 187 (7030%) sometimes did not pay attention to medication (time, dose, and frequency), 135 (50.75%) reduced or stopped drugs after symptoms were improved, and 100 (37.59%) stopped the drugs due to symptom aggravation or appearance of other symptoms. The univariate analysis showed that 13 indices were the influencing factors for compliance with NAs treatment among CHB patients (all P<0.05). A multivariate analysis was performed for these 13 indices and the results showed that race (odds ratio [OR]=0.218, 95% confidence interval [CI]: 0.076-0.996, P=0.007), mode of medical payment (OR=0.772, 95%CI: 0.445-0.919, P<0.001), physical exercise (OR=2.55, 95%CI: 1.472-6.545, P=0.020), level of social support (OR=0.836, 95%CI: 0.649-0.947, P<0.001), awareness of the outcome of incompliance (OR=0.577, 95%CI: 0.393-0.886, P<0.001), and self-efficacy (OR=0.094, 95%CI: 0074-0.328, P<0.001) were independent influencing factors for compliance with NAs treatment among CHB patients. ConclusionCompliance with antiviral therapy among CHB patients is closely associated with the mode of medical payment, psychological state, and self-management ability. Related measures, such as improvement of patients’ ability to pay medical expenses, education on disease risk for patients, and reduction of social discrimination, can help to improve treatment compliance.

Chinese Journal of Experimental Ophthalmology 2015;33(1):55-59
Background Cystoid macular edema (CME) is a manifestation secondary to multiple fundus diseases and often leads to the decline of visual acuity.Optical coherence tomography (OCT) is a common diagnostic method for CME,but it can not reflect the metabolism information of retinal pigment epithelium (RPE).Shortwave autofluorescence (SW-AF)may find the abnormality of macula, but its clinical value remains to be confirmed. Objective This study was to investigate the value of SW-AF in the diagnosis of CME.Methods One hundred and eighty-nine eyes of 140 patients who have the relative diseases to CME or received intraocular surgery were included from May 2010 to August 2011 in Lihuili Hospital of Ningbo city.SW-AF,infrared autofluorescence (IRAF),fundus fluorescein angiography (FFA) and OCT were performed in all the eyes under the informed consent of each patient. The sensitivity, specificity and other indicators in diagnostic test of SW-AF for CME were evaluated and compared with OCT.Results SW-AF had certain accuracy in diagnosis of CME with the sensitivity 78.29%,specificity 96.67%,Youden index 0.75,positive predictive value 98.06%,negative predictive value 67.44%.The area under curve (AUC) of SW-AF for the diagnosis of CME was 0.875 with the 95% confidence interval (CI) 0.823-0.926 (P<0.001).Compared with OCT,the differences of SW-AF and IR-AF in the diagnosis for CME was statistically significant(x2 =22.53,91.35,both at P<0.001),and agreement coefficients of diagnosis result between SW-AF、IR-AF or FFA and OCT were 0.67,0.12 and 0.85.Conclusions SW-AF can be used as a new rapid,non-invasive,ancillary technique in diagnosis of CME with certain accuracy, and the diagnostic agreement of SW-AF with OCT is lower than FFA but is higher than IR-AF.
