1.Advances in Research on Hepatitis B Virus DNA Integration

Since HBV DNA integration was discovered for the first time in 1980, various methods have been used to detect and study it, such as Southern Blot, in situ hybridization, polymerase chain reaction and so on. HBV DNA integration is thought to be random on the whole although some hot spots of integration were described by some researchers, one of which might be the repetitive sequences of the genomic DNA. Besides, DNA damage, especially double-strand breaks could promote HBV DNA integration into host genome. HBV DNA integration into cells may damage the stability of the genome, cause DNA rearrangement, promote DNA deletion and induce the formation of HCC.
2.Clinical Observation of B Ultrasound-guided Painless Artificial Abortion in 60 Cases

Objective To estimate the clinical application value of B ultrasound-guided visual painless artificial abortion operation. Methods One hundred and twenty women intented to terminate early pregnancy were randomly divided into 2 groups: group A (observation group) with ultrasound-guided visual painless artificial abortion operation and group B (control group) with traditional artificial abortion. Results Compared with the control group,the observation group has the advantages of less bleeding,less operation time, shorter bleeding time and less complications. There were statistical significances between two groups. Conclusion B ultrasound-guided visual painless artificial abortion operation is a safe and effective method, and it is worth promoting.
3.Protective effects of nifedipine on endothelial cells in spontaneously hypertensive rats

BACKGROUND: Research has proved that nifedipine can improve the functions of vascular endothelial cells. However, it still needs further study on its mechanism on dilating vessels.OBJECTIVE: To observe the effects of controlled-release nifedipine to nitrogen monoxide(NO) and induced nitric oxide synthase(iNOS) in spontaneously hypertensive rats(SHR).DESIGN: Comparative observational study based on experimental animals.SETTING: Experimental animal centre in a medical college.MATERIALS: This study was completed in Experimental Animal Centre of Shandong University during April to May 2002. Twenty-one recombinant inbred rats of clean class with body mass of(300 ± 2) grams were provided by Experimental Animal Centre of Shandong University. They were randomly divided into three groups: control group, normal group and low dose group,each with 7 rats.METHODS: Saline(10 mL/kg), 10 mL/kg and 3 mL/kg controlled-release nifedipine solution(0. 3 g/L) were poured into stomach to rats in control group, normal dose group and low dose group once everyday for 15 days respectively. Eyeball was removed to collect blood and heart and lung were removed at the same time to test the content of NO and iNOS in serum after last administration.MAIN OUTCOME MEASURES: Content of NO, comparison of iNOS activity between groups.RESULTS: There was significant difference on NO content between normal dose group and control group, low dose group after 15 days' administration ( P < 0. 01 ) . Compared with control group and low dose group, there was significant difference on the iNOS activity in heart and lung of normal dose group(P <0.01, P <0.05).CONCLUSION: While nifedipine reduces the blood pressure, it can also improve the content of NO and resist the increasing activity of iNOS casued by increased blood pressure.
4.The value of multimodal MRI in preoperative staging and grading of endometrial carcinoma

Objective To analyze the imaging features of endometrial carcinoma derived from multimodal MRI.Methods MRI scan was performed in 45 patients with biopsy-proved endometrial carcinoma,including conventional MRI,diffusion weighted imaging(DWI),dynamic contrast-enhanced magnetic resonance imaging (DCE-MRI),and time-signal curve (TIC).Results The diagnosis rate of DCE-MRI combined with DWI was 93.3%, and that of conventional MRI was 77.8%.Tumor differentiation was negatively correlated with ADC and rADC, and significant difference was found between G2 and G3 as well as G1 and G3 (both P<0.05).There was no significant difference between G1 and G2 (P>0.05).Of the 45 cases, 43 showed TIC type Ⅰ and type Ⅱ, and 2 showed TIC type Ⅲ, while the normal myometrium was mainly type Ⅲ curve.The difference of signal intensity of each phase (D), and enhancement rate (ER) of the lesions were lower than those of the normal myometrium.The difference was statistically significant at the phase of >30 s(P<0.05),the peak time of the former was (50±10) s.Conclusion Multimodal MRI is of great value in preoperative staging and grading of endometrial carcinoma.
5.Advances in nervous system complications and intervention of human and rats anorectal malformation

Anorectal malformations (ARM)comprise a broad spectrum of congenital disorders that account for 25 % of gastrointestinal malformations.Despite numerous technical advances for treatment of ARM,complications such as fecal incontinence and constipation still occur and can greatly deteriorate patients' quality of life.It is recognized that lumbosacral spinal cord anomalies in ARM has been an important factor affecting the fecal function after procedure.Researchers have found that lumbosacral myelodysplasia is the common seen complication of ARM and neural cells decreased in lumbosacral spinal cord by the study on animals and human.Due to numerous factos affecting nerve innervation on annrectus and pelvic floor muscle,this review summarizes the nervous system complications and abnormal intervention of human and rats ARMs.The developments of study on ARM complications and intervention are detailed.Then the new direction of the research about the anorectal malformation nervous system is put forward,and the new strategy of improving the prognosis of anorectal malformation surgery is explored.
6.Specific coalescent peptides binding with CD59 on HeLa cells screened by phage peptide library

Objective:To search for carrier of short active peptide with binding to human CD59 of the surface of HeLa cell.Methods:Ph. D. 12 display peptide library was biopanning with HeLa cell high-expressing CD59. After competitive test, positive phage clones were identified by sandwich ELISA and sequenced.Results:10 phage clones were identified to have higher combination with CD59.Conclusion:The sequence of peptide with binding to CD59 has been obtained.That might lay a basis for design of short peptide of CD59 on tumor sneaking through.
7.Relationship Between Plasma Fibrinogen Level and Ischemic Stroke in Patients with Nonvalvular Atrial Fibrillation

Objective:To investigate whether the increased plasma fibrinogen level is an independent predictor of cardiogenic embolism in patients with nonvalvular atrial fibrillation (NVAF).Methods:Ninety-eight patients with NVAF hospitalized for cardiac causes were evaluated,and 24 of them had a history of ischemic stroke.The relationship between fasting plasma fibrinogen level and ischemic stroke were analyzed.Results:After adjusting traditional cardiovascular risk factors.the patients with NVAF who had the history of ischemic stroke were significantly older than those without(mean [SD] age 73.6 [8.4]vs.69.1[10.3] years,P=0.017).1he prevalence of hypertension was significantly higher(83.3%va.66.2%,P=0.011).The proportion of patients with left ventricular systolic dysfunction was significantly higher(63.1% vs.45.9%,P=0.006),and plasma fibrinogen level increased significantly (mean [SD] 365 [108]vs.312[82] mg/dL,P=0.008).Conclusions:Plasma fibrinogen level is independently associated with a history of ischemic stroke in older patients with NVAF hospitalized for cardiac causes.
8.Effects of preoperative intravenous different doses of pantoprazole on acidity of gastric fluid

Objective To investigate the effects of preoperative iv different doses of pantoprazole on acidity of gastric fluid in patients undergoing elective surgery. Methods Sixty ASA Ⅰ -Ⅲ patients aged 38-80 yr,weighing 44-72 kg undergoing elective surgery were randomly allocated into 3 groups (n = 20 each): group Ⅰ control (group C); group Ⅱ pantoprazole 40 mg(group P1) and group Ⅲ pantoprazole 80 mg(group P2). The patients received intravenous normal saline (NS) 100 ml, pantoprazole 40 and 80 mg in NS 100 ml respectively in group C, P1 and P2 at 30 min before surgery. Gastric content was aspirated using a gastric tube on the patients'arrival in the operating room and at 1,2 h and the end of surgery. The pH of the gastric fluid was measured. The adverse effects including dizziness, diarrhea, PONV and incidence of stress ulcer were recorded. Results The pH was significantly increased in group P1 and P2 as compared with control group but there was no significant difference in pH between group P1 and P2 . There were no significant difference in the incidence of stress ulcer and pH of the gastric fluid≤2.5 among the 3 groups. Conclusion Preoperative intravenous pantoprazole 40 mg can effectively increase pH of gastric juice and is helpful in reducing the incidence of stress ulcer and the risk of acid aspiration.
9.Study on Insoluble Particles in Dazhu Hongjingtian Injection Combined with Different Solutions

Objective:To research the stability of Dazhu Hongjingtian injection combined with four different infusion solutions (0. 9% sodium chloride injection, 5% glucose injection, fructose injection, xylitol and sodium chloride injection) . Methods:The insolu-ble particles and pH value of Dazhu Hongjingtian injection in 0, 0. 5,1,2,4 and 8h after the combination were detected by the methods described in China Pharmacopeia ( 2010 edition ) . Results: Dazhu Hongjingtian injection combined with the four infusion solutions showed no significant changes in appearance and pH value in 8h. The number of insoluble particles in Dazhu Hongjingtian injection combined with 0. 9% sodium chloride injection and 5% glucose injection met the requirement in the pharmacopeia, and that in 0. 9%sodium chloride injection was lower than that in 5% glucose injection. The quantity of insoluble particles in Dazhu Hongjingtian injec-tion combined with fructose injection exceeded the standard level, and showed remarkable change in the fourth hour. The quantity of insoluble particles in xylitol and sodium chloride injection slightly exceeded the standard. Conclusion: Dazhu Hongjingtian injection can be mixed with 0. 9% sodium chloride injection and 5% glucose injection, and fructose injection and xylitol and sodium chloride in-jection are not suitable for the solvent of Dazhu Hongjingtian injection. Although there is no instruction indicating that 0. 9% sodium chloride injection can be used as the suitable solvent for Dazhu Hongjingtian injection, the results of the experiment can be reasonable reference for diabetes patients treated with Dazhu Hongjingtian injection.
10.Comparison of three herbal proparations in airway remodeling in rats with chronic asthmatic

AIM To research the antiasthmatic effects of Zuogui Pills,Yougui Pills and Yupingfeng Powder and their mechanism of action.METHODS Male Brown Norway rats were randomly divided into five groups.OVA was used to induce the model of chronic asthma.Five groups of asthmatic rats were given Zuogui Pills,Yougui Pills,Yupingfeng Powder by gavage once daily for 14 d,respectively.The normal and the model group were given normal saline.Penh value,serum Thl,Th2 inflammatory cytokines,and CROT and ACTH protein levels were determined.Pathological changes of airway remodeling were measured.RT-PCR method was used to measure the expression of CRH mRNA in hypothalamus,and TGF-β1 and Smad3 mRNA in lung tissues.RESULTS Penh values of Yupingfeng Powder group and Yougui Pills group were significantly lower than that of Zuogui Pills group.The Thl inflammatory cytokine of Yupingfeng Powder group was significantly higher than that of Zuogui Pills group,while the Th2 cytokine was significantly lower than that of Zuogui Pills group.In Yougui Pills group,only IL-5 was significantly lower than that of Zuogui Pills group.Serum CROT,ACTH protein and CRH mRNA of Yupingfeng Powder group were significantly higher than those in Zuogui Pills group.No significant difference was found between Yupingfeng Powder group and Zuogui Pills group with regard to TGF-β1 and Smad3 mRNA.Smad3 mRNA of Yougui Pills group was significantly higher than that of the model group.Airway remodeling was significantly reduced by Yupingfeng Powder and Zuogui Pills.The number of goblet cells in Yougui Pills group was significantly lower than that in Zuogui Pills group.CONCLUSION All the three formulaes possess significant antiasthma effects.Regulation effects of Yupingfeng Powder on Thl/Th2 balance,HPA and TGF-β1/Smad3 pathway are strong.Compared with Zuogui Pills,Yougui Pills possesses weaker TGF-β1/Smad3 regulative effects and stronger HPA regulative effects.
