1.SEROLOGICAL INVESTIGATION ON SARS PATIENTS IN GUANGDONG PROVINCE

Objective To detect IgG and IgM antibodies in the sera of SARS patients, in order to find the possible causative association between new coronavirus and SARS; to compare IgG and IgM antibody titers of acute phase with those of convalescent phase, in order to find specific serological reaction of SARS virus and its clinical significance. Methods Clinical specimens of SARS patients in 4 hospitals of Guangdong Province were collected and then detected by IFA and ELISA. The specificity and cross-reaction of each specimen for confirming the new coronavirus were evaluated. Results One hundred and seventeen of 130 clinically diagnosed SARS patients showed viral specific serological reaction, with the positive rate of 90%. The IgG titer increased significantly 10 days after the onset of infection and reached a higher level (above 320) 15 days later, while IgG titer reached the highest level on the 20 th-30 th day. The specific antibodies were negative in all 119 individuals with close contact with patients and 100 normal controls. Conclusion Viral specific IgG antibody was detected in the sera of SARS patients, confirming that the patients were infected by the virus; persistent high titer of IgG antibody in the sera of convalescent patients might probably be a protective mechanism in general population.
2.Clinical significance of detection on peripheral blood T cell subsets, sIL-2R, VEGF and LDH in patients with diffuse large B cell lymphoma

Objective To investigate the changes of peripheral blood T cell subsets and serum soluble interleukin-2 receptor (sIL-2R),vascular endothelial growth factor (VEGF) and LDH in patients with diffuse large B cell lymphoma,and to investigate their relationship of the cellular immune function,clinical stage and disease progression.Methods 35 cases of DLBCL patients were selected as observe group,20 cases of healthy individuals as normal control group.Peripheral blood T cell subsets were detected by flow cytometry.Serum soluble interleukin-2 receptor (sIL-2R) and vascular endothelial growth factor (VEGF) were detected by ELISA assay.Serum levels of LDH were detected by the rate method for measuring.Results The expression levels of CD4,NK cells and CD4/CD8 ratios in all patients with DLBCL were lower than healthy individuals [(68.62±12.82) % vs(73.00±6.12) %,(14.28±7.93) % vs (39.49±12.01) %,(1.03±0.84) vs (1.45±0.28),all P < 0.05].Serum sIL-2R in patients with DLBCL was significantly higher than healthy individuals [(501.50±157.70).μg/L vs (128.30±82.40) μg/L,P< 0.001].Serum levels of sIL-2R with Ⅲ-Ⅳ stage in patients with DLBCL was higher than that in ones with Ⅰ-Ⅱ stage [(618.38±192.22) μg/L vs (432.45±75.48)μg/L,P < 0.05].Serum expression levels of sIL-2R in patients with DLBCL with B symptoms was higher than that in ones with A symptoms [(616.50±183.03) μg/L vs (441.52±103.48) μg/L,P < 0.01].The expression levels of VEGF,LDH in patients with DLBCL were higher than healthy individuals [(304.03±163.05) pg/ml vs (156.25±47.83) pg/ml,(300.74±300.91) U/L vs (177.00±32.14) U/L,both P < 0.05].The high expression level of serum sIL-2R was correlated with low CD4/CD8 ratio (r =0.4635,P < 0.05).The high expression levels of serum sIL-2R was correlated with the high expression levels of serum VEGF,LDH (r =0.447,P < 0.05; r =0.448,P < 0.05).Conclusion High expression levels of serum sIL-2R in DLBCL are closely associated to low cellular immune function,tumor burden in vivo,disease progression and invasion.
3.To Study the change regularity of special antibody in cases of severe acute respiratory syndrome

Objective By studying the change of the special antibodies titer IgG, IgM and nucleocaspid to SARS corona virus in family clustering cases , searching and testing the special antibody to SARS corona virus in the patients kinfolks Method Testing special antibody titer to SARS corona virus of 14 patients from 5 different families and their 10 kinfolks continuously by IFA and antigen capturing ELISA methods, taken samples in the 7 th , 14 th , 30 th , 60 th , 120 th , 180 th , 210 th ,270 th ,360 th day after SARS patients infected by SARS corona virus, dilute them, and then measure titers of three kinds of antibodies ResultsDuring the testing of IgG antibody titer of the 14 SARS patients by IFA method, in the 120 th day, the average titer is up the highest value, it is 1/1 120; in the 180 th day, the average titer begins to descend, it is 1/262 in the 360 th day, the average titer is 1/71 During the testing of IgM titer, the average titer is 1/32, it is obviously lower than it in IgG testing; the IgM titer of the most SARS patients are disappear after 120 days The average value of nucleocaspid antibody titer is lower than it in IgG antibody But there are some differences in 5 cases While IgG antibody titer begins to descend in the 180th day, nucleocaspid antibody titer begins to ascend obviously, it is up to 1/790 210 th day begins to descend In the meantime, we have tested special antibody to SARS corona virus of their 10 kinfolks (include 2 children) who have contacted with SARS patients closely The samples are taken in the 14 th , 30 th , 60 th day, and all the special IgG, IgM and nucleocaspid antibody to SARS corona virus are negative ConclusionBy testing the special IgG antibody to SARS corona virus of the 14 family clustering cases ,we find the special IgG antibody titer of the SARS patient is higher, and it can remain for a long time As for the nucleocaspid antibody titer of some SARS patients ascend obviously after 180days, we think that it provide useful information for vaccine development and it is necessary for us to study more in the clinic There is no special blood serum antibody to SARS corona virus in their 10 kinfolks of the SARS patients
4.Application of phage-displayed peptide libraries

Phage display is a new technology developed in recent years. Random peptides displayed on the filamentous phage surface offer a rich source of molecular diversity in searching for ligands that bind an antibody, receptor and other proteins. The phage display system has been effective in the discovery of various ligands. Potential applications of the peptide library include investigation of the protein protein interactions and discovery of mimetic drug candidates.
5.Correlation between the chemotherapy resistance of diffuse large B-cell lymphoma and cytokines IL-6 and IL-10

Objective To investigate the correlation between expression levels of serum cytokines IL-2,IL-6, IL-8, IL-10, TNF-α and IFN-γ in patients with diffuse large B-cell lymphoma(DLBCL) and chemotherapy resistance. Methods 30 cases of DLBCL patients with chemotherapy resistant, 30 cases of DLBCL patients with chemotherapy sensitive and 20 cases of healthy individuals as normal control group were enrolled. The levels of serum cytokines IL-2, IL-6, IL-8, IL-10, TNF-α and IFN-γbefore, during and after treatment in both DLBCL groups and normal control group were detected by ELISA assay. Results The expression level of serum IL-6 and IL-10 before treatment in DLBCL patients with chemotherapy resistance was significantly higher than that in DLBCL patients with chemotherapy sensitive and normal control group (P < 0.05), however,that after treatment in DLBCL patients with chemotherapy resistance was significantly lower than that before treatment (P = 0.02, P = 0.015). The level of serum IL-6 and IL-10 in patients with DLBCL recurrence into drug resistance was higher than that during of remission (P = 0.004, P <0.001). Before treatment, the expression level of serum IL-6 in patients with Ⅲ-Ⅳ stage in chemotherapy resistant group was significantly higher than that in ones with Ⅰ - Ⅱ stage(P <0.05). Levels of IL-2, IL-8, TNF-α and IFN-γin chemotherapy resistant group, chemotherapy sensitive group and the normal control group were no differences (P >0.05).Conclusion The expression levels of the serum IL-6 and IL-10 were closely correlated with the chemotherapy efficacy of DLBCL, they may be involved in drug resistance of DLBCL.
6.Study on Interactions between Paeonol and Cadherin

Paeonol has inhibitory effect on a variety of tumor cells, whereas cadherin is a kind of glycoprotein that is associated with the occurrence and development of different tumor.In this study, the interactions of paeonol and E-cadherin have been investigated by fluorescence spectrometry and atomic force microscopy (AFM).Fluorescence spectrometry results revealed that the addition of paeonol significantly quenched the fluorescence of E-cadherin.Based on the results of the quenching constant, it was inferred that the interaction of paeonol and E-cadherin was a static quenching process.The thermodynamic parameters ΔH and ΔS were calculated to be -4.3×10.5 J/mol and -1.3×10.3 J/(mol·K), respectively, which proved the involvement of weak interactive forces such as hydrogen bond van der Waals force.AFM results revealed that cadherin molecules were assembled into the long-chain structure.The addition of paeonol could significantly disrupt these assembling structures into short chains, which could be ascribed to the damage of the interdigitation model from the adjacent cadherin molecules.All these results reveal that cadherin is an important target of paeonol to modulate its activity.
8.Biomechanieal characteristics of fixations parallel to the plateau and vertical to the fracture line in the early management of fractures of tibial plateau in rabbits

Objective To evaluate the biomechanical characteristics of fixations parallel to the tibial plateau and vertical to the fracture line in the early management of fractures of tibial plateau in rabbits. Methods Models of oblique split fractures of the medial tibial plateau were created in 30 New Zealand rabbits of 13 months. One tibial plateau was fixed with 3 lag screws (2.0 mm) parallel to the articular surface while the eontralateral plateau was stabilized with 3 lag screws (2.0 mm) vertical to the fracture line. The specimens were harvested at 0, 1, 2, 4, and 8 weeks after operation respectively and tested in a biome-ehanical testing machine. The maximum load of anti-compression at the medial tibial plateau and the maximum work at the maximum load, as well as the load and work as the articular surface displaced downwards for 2mm, were recorded. Results The biomechanical comparison of the 2 methods at the same time points showed that there were no significant differences at 0, 1 and 8 weeks but significant differences at 2 and 4 weeks. The biomechanical comparison of the same method at different time points showed that the biomechanical measures decreased at the early period after operation, then increased and finally kept steady at 4 or 8 weeks. Con-elusions The stability of tibial plateau fractures in rabbits decreases at the early post-operation and gradually increases to the normal level. It takes less time for the fixation parallel to the tibial plateau to increase the biomeehanical stability to the normal level than the fixation vertical to the fracture line. The parallel fixation can provide more biomechanical advantage in stabilization of the fractures than the vertical one.
9.Analysis of three family clusters of severe fever with thrombocytopenia syndrome by exposure risk matrix

Objective:
To analyze three events of severe fever with thrombocytopenia syndrome (SFTS) among family members by exposure risk matrix, so as to provide reference for SFTS prevention and control.
Methods :
The field investigation and clinical data of seven confirmed cases with SFTS reported in Linhai from 2014 to 2016 were collected. Exposure risk matrix was used to list the risk of exposure two weeks before the onset, such as the environment, duration of activity and protection, and evaluate the possible routes of transmission.
Results:
The first event involved three sisters. Two of them had plucked tea together on a hill, and the possibility of infection through tick bites were 60% and 75%, respectively. Another sister was more likely to be infected by ticks in her residence after visiting her sister (no physical contact with the sick sister), with a possibility of 74%. The second event involved two cases (mother and son). They were more likely to be infected by ticks in the current residential area, and the possibility were 100% and 80%, respectively. The third event involved two cases (husband and wife). The wife had 60% possibility of being infected by the ticks on the hill she picked arbutus, and 40% by the ticks in her residence. The husband was most likely to be infected through contacting with body fluids or blood of his wife (44%), then through the ticks on the hill he picked arbutus (33%).
Conclusion
Among seven cases with SFTS reported in Linhai from 2014 to 2016, six cases have more than 50% possibility of being infected by tick bites; one case has higher possibility of being infected by contacting with body fluids or blood of a patient, but tick bites could not be ruled out.
