AIM: To establish a method for the determination of Astragaloside Ⅳ in Oiluyishen Tabletes(Radix Astragali, Herba Pyrolae, Rhizoma Atractylodis Macrocephalae, Poria, Radix Codonopsis, etc.) by HPLC-ELSD. METHODS: Chromatography was performed on a 5?m HIQsilC 18 column(4.6mm?250mm) at 25℃. The mobile phase composition was acetonitrile-water(34∶66). The flow rate was at 1.0mL?min -1. An evaporation light-scattering detector (ELSD) was used as detector with gas flow rate of 2.6 mL?min -1 and drift tube temperature at 100℃. RESULTS: The linear range of Astragaloside Ⅳ was in the range of 2.288~ 22.88?g, r=0.999, the average recovery was 98.8% and RSD was 1.2%. CONCLUSION: This method is simple, feasible and reproducible. It can be used for the quality control of Oiluyishen Tabletes.

Objective:To study the antitumor effect of combination therapy with adenoviral transfer of mouse endostatin gene and cyclophosphamide chemotherapy on mouse Lewis lung cancer. Methods:Mouse endostatin gene was cloned into repli-cation-defective adenovirus by virus recombination technology to construct Ad-mES,adenovirus vector coding mouse endo-statin gene, and then its biological activities were surveyed in vivo.Animal models of mouse Lewis lung carcinoma in C57BL/6mice were established and randomly divided into4groups(n=10):including control, chemotherapy, gene therapy, and chemotherapy+gene therapy group.Mice received cyclophosphamide150mg/kg s.c.3times a day in a21-d cycle for chemotherapy;control group received0.9%normal saline100?l.Ad-mES(6?10 9 pfu/100?l)were administrated by multi-spots intra-tumor injection to every mouse in gene therapy group and combination therapy group. The tumor sizes were mea-sured at different times points. The expression of endostatin after transfer was detected by Western blot and the microvessel density(MVD)of tumor was measured by immunohistochemical examination. Results:Higher titers of recombinant aden-oviruses(Ad-mES)were constructed.Combination therapy with Ad-mES and cyclophosphamide inhibited tumor growth(P

Objective To study the role of OIf-1/EBF associated zinc finger protein (OAZ),a transcription factor encoded by a positional systemic lupus erythematosus (SLE) candidate gene,in the function of mesenchymal stem cells (MSC) of SLE patients by silencing this gene. Methods OAZ mRNA levels of bone marrow MSC obtained from 5 SLE patients and 5 healthy controls were detected by real-time PCR.Bone marrow MSC obtained from 6 SLE patients were incubated with specific siRNAs for 3 days, then cells were harvested for OAZ measurement,Idl-3 and CCL2 mRNA levels were tested by real-time PCR,and levels of CCL2 were detected in culture supernatants using ELISA.Differences between groups were analyzed using t-test or MannWhitney test. Results ① OAZ mRNA levels of bone marrow MSC were significantly elevated in SLE patients (0.013±0.016) compared to healthy controls (0.001±0.000,P=0.009).② After OAZ silencing, the expression levels of OAZ,Id1,Id2 and ld3 mRNA were significantly decreased (△Ct 10.3±0.7,15.2±1.6,8.1±1.4,10.5±0.6 vs 8.7±0.7,14.1±1.2,7.1±1.5,9.8±0.6) (P all <0.05).③ Both the expression levels of CCL2 mRNA (△Ct 2.2±1.1 vs 3.0±1.1 ) and the levels of CCL2 protein in culture supernatants [(341±29) pg/ml vs (304±19) pg/ml] were significantly increased in OAZ silencing group comparing to those in the control group (P all <0.05).Conclusion OAZ gene expression is significantly elevated in bone marrow MSC of SLE patients.OAZ may affect autoantibody production in SLE patients by regulating CCL2 expression.

Objective To investigate the effects of antisense Smad2 oligodeoxynudeotides(ODN) on fibronetin(FN) and collagen Ⅳ(ColⅣ) secretion of rat mesangial cells cultured with high glucose, explore the action of Smad2 in the glomerulosclerosis and to find a new method to retard the progress of glomerular fibrosis. Methods 20-mer antisense, sense and random ODNs were designed and synthesized that were phosphorothioate modified to increase stability. The antisense ODN encompassed the ATG of the rat Smad2 gene. ODN was tranferred transiently into rat mesangial cells through liposome. Rat cells were treated with high glucose. mRNA and protein of Smad2 were detected by RT-PCR and cytochemistry. FN and ColⅣ were examined by ELISA. Results Antisense ODN significantly decreased mRNA and protein expression of Smad2 in rat mesangial cells treated with high glucose(P
